本発明は、眼内レンズ材料に対して行う光安定性試験方法及び試験装置に関する。

【従来技術】
プラスチックの材料や金属の材料が日光中の紫外線に長時間露光されると変質、変色を引き起こし、劣化が進行し強度が低下することが知られている。このため、従来より各材料に対して、各種の耐候性試験やその試験装置が提案されている。このような中、眼内レンズにおいても紫外線等に対する耐候性に関する試験が求められている。

【発明が解決しようとする課題】
しかしながら、プラスチック材料や金属材料の耐候試験と異なり、眼内レンズは生体内にて使用するため、所定温度(例えば37℃)を維持しなければならない。しかし紫外線を照射するためのランプはかなりの熱が発生するため、所定温度を維持しながら長期間試験を行うことは難しい。

本発明は、上記従来技術の問題点に鑑み、所定温度を維持し、正確な試験を行うことのできる光安定性試験方法を提供することを技術課題とする。

【課題を解決するための手段】
上記課題を解決するために、本発明は以下のような構成を備えることを特徴とする。

(1) 光源からの紫外光によって露光される眼内レンズの光に対する安定性を試験する光安定試験方法において、眼内レンズと生理食塩水とを透明容器に入れた後に該透明容器を密閉する過程と、冷却手段と加熱手段とを備える恒温水槽内に前記透明容器を取付け手段を介して設置する過程と、前記光源が発する紫外光の照射強度に基づいて定められる期間だけ前記透明容器内の眼内レンズに紫外光を照射する過程と、前記眼内レンズに紫外線を照射することによって生じる前記光源の発熱により上昇する恒温水槽内の水を,前記冷却手段を常時作動させることにより冷却し続けるとともに,同時に冷却し過ぎた前記恒温水槽内の水を前記加熱手段により加熱することにより恒温水槽内の水温を人の体温と同程度(37℃±2℃)に維持する過程と、を備えることを特徴とする。

【発明の実施の形態】
以下に本発明の実施の形態を図面に示しながら説明する。図1は実施の形態で使用される試験装置の該略構成図である。

1は眼内レンズの光安定性試験を行うための試験装置本体である。2は水槽であり、中には水が入っている。10は眼内レンズ11を入れておくためのバイアルである。使用する眼内レンズ11は特に限るものではなく、アクリル系、ビニル系、シリコン等、すべての眼内レンズを用いることができる。また、本実施の形態で使用されるバイアル10は容量5mL、300nm以上の波長で透明、化学的に活性がなく安定なものを使用している。

3は水槽2に入れられたバイアル取付台である。バイアル取付台3は内部に図示なき回転駆動装置を備え、上部の天板3aを図示するA方向(水平方向)に回転させることが可能である。光安定性試験期間中は天板3aを常時回転させることにより、すべてのバイアル10にできるだけ同じ条件にて紫外光が照射されるようにしている。本実施の形態では天板3aを回転させるものとしているが、光源5を光軸を中心に回転させても同じ効果を持つ。

4は天板3a上に設置され、バイアル10を固定保持するための保持具である。(図中、保持具4は3個のみ示しているが、実際は天板3a上に3個×5列並んでおり、計15個のバイアル10を固定保持することができる)。

また、バイアル10は光安定性試験中に雑菌等の混入による細菌汚染を防ぐため、上部を密閉するための蓋10aを使用している。このため、上方から照射される紫外光がバイアル10内に入れられた眼内レンズ11へ十分あたるように、保持具4はバイアル10を若干傾倒した状態にて固定保持するようになっている。

本実施の形態では、保持具4によりバイアル10を傾倒した状態で保持するものとしているが、これに限るものではなく、バイアル10の蓋10aを紫外光の透過し易い透明材料にて形成することにより、バイアル10を垂直に立てた状態にて固定保持し、光安定性試験を行うこともできる。また、バイアル10に眼内レンズを入れた後、開口部を熔閉することにより蓋10aを使わずに使用することも可能である。

5は紫外光を照射する光源であり、本実施の形態ではメタルハライドランプを使用している。光源5は図示なき固定部材により恒温水槽2の中央上方部に設置され、バイアル取付台3を照明するようになっている。また、光源5の前方には光源5から照射される紫外光のうち、所定の波長のみを選択的に透過させるためのフィルタ6が取り付けられている。また、使用する光源は、メタルハライドランプの他にキセノンアークランプ、高圧水銀ランプを使用することができる。

本発明における光安定性試験は、実際に眼内レンズを眼内に入れた状態と同様の状況にて行うことが求められる。したがって角膜や房水等にて吸収されない近紫外線部分のみを眼内レンズに照射するために、フィルタ6は300nm未満の波長を吸収し、300nm以上の波長を透過する特性を持つものが使用される。

7は恒温水槽2に設置される冷却器である。冷却器7は光源5からの紫外光照射により、上昇した恒温水槽内の水温を下げるための役割を持つ。本実施の形態で使用される冷却器7は恒温水槽2内に入れられた流水管7a内に所定温度に維持された冷却水を流動させることにより、槽内の水温を下げるようにしている。8は加熱器であり、図示なき温度センサを備え、冷却器7によって下げられた恒温水槽2内の水温を上げ、所定温度に維持するようになっている。また、加熱器8は攪拌部8aを備え、恒温水槽2内の水を攪拌することが可能である。

眼内レンズは生体内で使用されるため、光安定性試験も人間の体温と同じ温度状態にて行うことが求められる。したがって、光安定性試験期間中は温度管理を厳密に行わなければならない。本実施の形態では光源5によって温められた槽内の水温を、所定温度(ここでは37℃±2℃)を下回る温度になるように冷却器7によって冷やし、所定温度を下回った恒温水槽2内の水を加熱器8によって温め所定温度を維持するようにしている。

表1は冷却器7、加熱器8を使用せず、光源5を所定照射強度(30mW/cm2)にて点灯させたときの恒温水槽2内の水温の経時変化を示した表である。

この表に示すように、点灯してから2.5時間後には恒温水槽2内の水温は45.9℃まで上昇している。また、表2は上記2.5時間時点(水温45.9℃)に冷却器7を作動させたときの水温の経時変化を示している(冷却水の温度は25℃に設定)。

表2に示すように2時間後には28.3℃の状態でほぼ恒温状態となった。また、表3は上記2時間時点(水温28.3℃)に加熱器8作動させたとき水温の経時変化を示している(加熱器8は37℃を維持するように設定)。

表3に示すように加熱器8の作動後、1時間後には略37℃に達し、その後略37℃を維持して恒温状態となった。このように冷却器7を常時作動させた状態で加熱器8を適宜作動させることにより、光源5による加熱下におかれていても37℃付近を維持できるようになっている。

また、本実施の形態では、水温を一定にするために冷却器7、加熱器8の2つを使用しているが、これに限るものではなく、冷却と加熱機能が一体となった装置を使用することもできる。

次に光安定性試験方法について説明する。

試験の目的は300nm〜400nmの波長範囲に眼内レンズ材料が露光されたときの光安定性を評価することである。試験はISO/DIS 11979-5の規定に沿って行う。式1は紫外線の露光時間(照射時間)を算出するための式である。

ここで、I1は生体内のUV−A光度であり、0.5mW/cm2とする。Xは一日当たりの太陽光線露光時間であり、3時間とする。T1は眼内レンズの生体内露出時間であり、20年とする。nは強度係数であり、1(太陽光線の強い地域で考慮すべき最大強度)とする。I2は実際に使用する光源の照射強度(mW/cm2)である。T2は紫外線の露光時間である。式1においてI2を適宜設定することにより、T2を決定することができる。ただし、眼内レンズ材料を極端に早く光分解させないために、I2は30mW/cm2を超えない範囲とする。また、光安定性試験期間中の設定温度は37℃±2℃以内とする。

次に、以上の条件を用いて行う光安定性試験の流れを説明する。
初めに所定照射強度I2にて光源5を点灯させるとともに、冷却器7、加熱器8を作動させ、恒温水槽2内の水温を略37℃に維持させる。水温が37℃を維持しているのを確認後、約2mLの生理食塩水を入れたバイアル10に眼内レンズを浸し、蓋10aを用いてバイアル10を密封した後、15個のバイアル10を保持具4に固定保持する。

保持具4にバイアル10を固定保持した後、取付台3の図示なき回転駆動装置を作動させ、天板3aを回転させる。露光期間T2だけ紫外線を照射させ、眼内レンズを露光させた。露光期間T2後、バイアル10内の生理食塩水を分析し、移動成分の有無を確認する。

本実施の形態では、恒温水槽内に水を使用しているがこれに限るものではなく、設定温度によってはアルコール等の親水性溶媒や油等の疎水性溶媒を使用することもできる。

また、本実施の形態では、300nm未満の波長を吸収し、300nm以上の波長を透過するフィルタを使用しているが、さらに可視光以上の波長を透過させないフィルタを併用することで、恒温水槽内の水温の上昇を抑えることができる。これにより、所定温度を維持するための電力を抑えることができる。

次に、以下に実施例を挙げる。
<実施例1>
光源5を点灯させ、眼内レンズ11上の紫外線照射強度が30mW/cm2になるように光源と眼内レンズ間の距離を調節する。次に、冷却器7(水温25℃)、加熱器8を作動させ、恒温水槽2内の水温を略37℃に維持させた。水温が37℃を維持しているのを確認後、約2mLの生理食塩水を入れたバイアル10に眼内レンズ11を浸し、蓋10aを用いてバイアル10を密封した後、15個のバイアル10を保持具4に固定保持した。

保持具4にバイアル10を固定保持した後、天板3aを回転させ、露光期間15.2日だけ眼内レンズ11を露光させた。途中、恒温層2内から蒸発した水を補うため37℃の水を適量加えた。露光期間15.2日後、バイアル10内の生理食塩水を分析し、移動成分の有無を確認した。露光期間中の恒温水槽2内の水温は略37℃を維持し、温度管理が良好に行われた。
<実施例2>
照射強度を20mW/cm2、露光期間を22.8日とした以外は実施例1と同様の操作を行った。実施例2においても長期間に関わらず露光期間中の恒温水槽2内の水温は略37℃を維持し、温度管理が良好に行われた。
<実施例3>
照射強度を10mW/cm2、露光期間を45.6日とした以外は実施例1と同様の操作を行った。実施例3においても長期間に関わらず露光期間中の恒温水槽2内の水温は略37℃を維持し、温度管理が良好に行われた。

【発明の効果】
以上のように、本発明によれば冷却器を常時作動させた状態で加熱器を適宜作動させることにより、光源による加熱下におかれていても所定温度を維持することができ、眼内レンズの光安定性試験を正確に行うことができる。
