Description
発明の背景
天然のソマトスタチンは、14アミノ酸イソ型(ソマトスタチン−14)および18アミノ酸イソ型(ソマトスタチン−28)双方から構成される。ハイマン(Heiman)ら、ニューロエンドクリノロジー(Neuroendocrinology)、45:429〜436(1987年)。天然のソマトスタチンは半減期が短いので、様々なソマトスタチン類似体が、例えば、先端巨大症の処置を目的として、開発された。レイナー(Raynor)ら、モレキュラー ファーマコル(Molecular Pharmacol.)、43:838(1993年)。5種の異なるソマトスタチンレセプターが同定されその特性が明らかになった。ホヤー(Hoyer)ら、ナウニン−シュミーデベルグス アーク ファーマコル(Naunyn-Schmiedeberg's Arch.Pharmacol.)、350:441(1994年)。ソマトスタチンは、成長ホルモン、グルカゴン、インスリン、アミリン等の、ホルモンの放出、および神経伝達物質の放出の修飾などの、様々な作用を生じる。上記作用のうち、特異的なソマトスタチンレセプターへの結合に関連するものがあった。例えば、成長ホルモンの阻害は、ソマトスタチンタイプ−2レセプター(SSTR−2)よるものであり(レイナー(Raynor)ら、モレキュラー ファーマコル(Molecular Pharmacol.)、43:838(1993年);ロイド(Lloyd)ら、アム ジェー フィジオル(Am.J.Physiol.)、268:G102(1995年))、インスリンの阻害はソマトスタチンタイプ−5レセプター(SSTR−5)よるものであった(コイ(Coy)ら、197:366〜371(1993年))。目的とする生体応答に応答可能な特異的なソマトスタチンレセプターサブタイプに対して選択的である、故に、望ましくない副作用を生じさせる他のレセプターサブタイプとの相互作用を抑制する類似体を有することが好ましい。
発明の要約

治療上有効な量の本発明のペプチドおよび製薬上許容できる担体物質(例えば、炭酸マグネシウム、ラクトース、または治療化合物がそれと共にミセルを形成できるリン脂質)で、ペプチドを必要とする被検者への(例えば、経口、静脈内、経皮的、肺、経膣、皮下、経鼻的、イオン導入、または気管内による)投与を目的とする治療用組成物(例えば、ピル、錠剤、カプセル、または液体)を形成する。ピル、錠剤、またはカプセルは、組成物を被検者の小腸中を未消化な状態で通過させるのに十分な時間、被検者の胃内で胃酸または腸の酵素から組成物を保護できる物質で被覆してもよい。治療用組成物はまた、皮下または筋肉内投与を目的とする生分解性のまたは非生分解性の徐放性配合物の形態であってもよい。例えば、米国特許第3,773,919号及び4,767,628号ならびにPCT出願番号WO 94/00148号を参照。持続的な投与はまた、埋込型または外部型ポンプ(例えば、インフューサイド(登録商標) ポンプ(INFUSAIDTM pump))を用いて上記治療用組成物を投与することにより達成されてもよい。本ペプチドは、患者の就寝前に投与されてもよい。
上記したような病気または疾患を治療するための本発明のペプチドの投与量は、投与形態、被検者の年齢や体重、及び治療される被検者の状態によって変化し、最終的には、主治医や獣医によって決定されるであろう。主治医や獣医によって決定されるペプチドの量を、本明細書では「治療上有効な量」と称する。
インスリン放出の阻害の必要性に関連する病気または疾患、例えば、II型糖尿病を処置するのに、およびソマトスタチンレセプターを検出するのに、例えば、ラジオイメージング(radioimaging)に使用される上記一般式に含まれるペプチドもまた、本発明の概念に含まれる。

発明の詳細な説明
本発明のソマトスタチン類似体の合成および使用は、当該分野における通常の知識を有するものの能力内によく含まれる。特記しない限り、本明細書中に使用されるすべての技術的及び科学的用語は、本発明の属する分野における通常の知識を有するものによって共通して理解されるのと同様の意味を有する。また、本明細書中に記載されるすべての公報、特許出願、特許、及び他の参考文献は参考のため引用される。
当業者は、本明細書の記載に基づいて、本発明を十分利用できると考えられる。したがって、下記の特定の実施例は単に説明するためのものであり、残りの開示を何等制限するものではない
ソマトスタチン類似体の合成
短いペプチドの合成は、ペプチド分野においてよく試験される。例えば、ステュワート(Stewart)ら、ソリッド フェーズ シンテシス(Solid Phase Synthesis)(ピアース ケミカル シーオー(Pierce Chemical Co.)、第二版、1984年)を参照。類似体Iの合成は下記のとおりである。本発明の他のペプチドは、当該分野における通常の知識を有するものによって同様の方法により調製できる。
塩化物イオンの形態のベンズヒドリルアミン−ポリスチレン樹脂(アドバンスド ケムテック インコーポレイテッド(Advanced ChemTech,Inc.)、ルイスヴィレ、ケーワイ(Louisville,KY))(1.1g、0.5ミリモル)を、下記試薬/溶剤をデリバーするようにプログラムされたアドバンスド ケムテック エーシーティー200ペプチド合成器(Advanced ChemTech ACT 200 peptide synthesizer)の反応容器中に仕込んだ:(a)塩化メチレン;(b)塩化メチレンにおける33%トリフルオロ酢酸(それぞれ1及び25分で2回);(c)塩化メチレン;(d)エタノール;(e)塩化メチレン;および(f)クロロホルムにおける10%トリエチルアミン。
中和された樹脂を、塩化メチレンにおけるBoc−S−4−メチルベンジル−Cys及びジイソプロピルカルボジイミド(各1.5ミリモル)と共に、1時間、撹拌し、さらに得られたアミノ酸樹脂を上記洗浄プログラムの段階(a)〜(f)を行った。次に、下記アミノ酸(1.5ミリモル)を同様の操作によって順次カップリングした:Boc−Phe、Boc−O−ベンジル−Thr、Boc−N−ベンジルオキシカルボニル−Lys、Boc−D−Trp、Boc−Phe、Boc−Phe、およびBoc−S−メチルベンジル−Cys。洗浄及び乾燥後、完全樹脂の重量は1.6gであった。

Detailed Description of the Invention
The synthesis and use of the somatostatin analogs of the present invention are well within the ability of those with ordinary knowledge in the art. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this invention belongs. Also, all publications, patent applications, patents, and other references mentioned herein are cited for reference.
Those skilled in the art will be able to fully utilize the present invention based on the description herein. Accordingly, the specific embodiments below are for illustrative purposes only and are not intended to limit the remaining disclosure in any way.
Synthesis of somatostatin analogues.
The synthesis of short peptides is well tested in the peptide field. See, for example, Stewart et al., Solid Phase Synthesis (Pierce Chemical Co., 2nd edition, 1984). The synthesis of analog I is as follows. Other peptides of the invention can be prepared in a similar manner by those with ordinary knowledge in the art.
Benzhydrylamine-polystyrene resin in the form of chloride ion (Advanced ChemTech, Inc., Louisville, KY) (1.1 g, 0.5 mmol) was added to the following reagent / solvent: Was charged into a reaction vessel of an Advanced ChemTech ACT 200 peptide synthesizer programmed to deliver: (a) methylene chloride; (b) 33% trifluoroacetic acid in methylene chloride ( (2 times each at 1 and 25 minutes); (c) methylene chloride; (d) ethanol; (e) methylene chloride; and (f) 10% triethylamine in chloroform.
The neutralized resin was stirred with Boc-S-4-methylbenzyl-Cys and diisopropylcarbodiimide (1.5 mmol each) in methylene chloride for 1 hour, and the resulting amino acid resin was further washed through the washing program. (A) to (f) were performed. Next, the following amino acids (1.5 mmol) were sequentially coupled in the same manner: Boc-Phe, Boc-O-benzyl-Thr, Boc-N-benzyloxycarbonyl-Lys, Boc-D-Trp, Boc. -Phe, Boc-Phe, and Boc-S-methylbenzyl-Cys. After washing and drying, the complete resin weighed 1.6 g.
The resin (1.6 g, 0.5 mmol) was then mixed with anisole (5 ml), dithiothreitol (100 mg), and anhydrous hydrogen fluoride (35 ml) at 0 ° C. and stirred for 45 minutes. Excess hydrogen fluoride was quickly evaporated under a stream of dry nitrogen to precipitate the free peptide and washed with ether. Furthermore, this crude peptide was dissolved in 500 ml of 90% acetic acid and until a constant brown color was observed,2A concentrated solution of / MeOH was added. The column was eluted with a linear concentration gradient from 80% solution A and 20% solution B to 40% solution A and 60% solution B. In this case, solution A is 0.1% trifluoroacetic acid (TFA) in water and solution B is 79.9% acetonitrile, 20% water and 0.1% TFA. Fractions were tested by thin layer chromatography (tlc) and analytical high performance liquid chromatography (hplc) and stored to the highest purity. This solution was repeatedly lyophilized from water to give 95 mg of product as a white dispersible powder.
This product is found to be homogeneous by hplc and tlc. The composition of the cyclic octapeptide was confirmed by amino acid analysis of the acid hydrolyzate and matrix-assisted laser desorption mass spectrometry (MALDI MS) (calculated molecular weight 1077; measured molecular weight 1080).
The synthesis of analog V is as follows. Benzhydrylamine-polystyrene resin in the form of chloride ion (Advanced ChemTech, Inc.) (0.7 g, 0.25 mmol) was advanced programmed to deliver the following reagents / solvents: Charged into a reaction vessel of Advanced ChemTech ACT 200 peptide synthesizer: (a) methylene chloride; (b) 33% trifluoroacetic acid in methylene chloride (twice at 1 and 25 minutes each) (C) methylene chloride; (d) ethanol; (e) methylene chloride; and (f) 10% triethylamine in chloroform.
The neutralized resin was stirred for 1 hour with Boc-S-methylbenzyl-Cys and diisopropylcarbodiimide (1.5 mmol each) in methylene chloride, and the resulting amino acid resin was further purified by the steps of the washing program (a ) To (g) were performed. A column purification similar to that described above yielded 21 mg of cyclic nonapeptide found to be homogeneous by hplc and tlc. The composition of the cyclic nonapeptide was confirmed by amino acid analysis of the acid hydrolyzate and MALDI MS (calculated molecular weight 1192; measured molecular weight 1192).
Synthesis of somatostatin analogs iodinated at tyrosine residues (eg, the chloramine-T method) is well reported and within the ability of those with ordinary knowledge in the art. See, for example, Czenick et al., J. Biol. Chem., 258: 5525 (1993) and European Patent No. 389,180 B1.
Somatostatin receptor binding assay
(1)Human SSTR-2 binding assay:
CHO-K1 (ovary, Chinese hamster) cells were obtained from the American Type Culture Collection (ATCC) (Rockville, MD) (ATCC No. CCL61), and this cell contains molecular organisms. Using standard techniques known in the field of science, Yamada et al., Proc. Natl. Acad. Sci. USA, 89: 251-255 (1992) and ATCC Human SSTR-2 cDNA available from (ATCC No. 79046) was transfected. See Patel et al., Biochem. Biophys. Res. Commun., 198: 605 (1994). Unpurified membrane is set to polytron of setting 6TM(POLYTRONTM) Human SSTR-2 was transfected in 20 ml ice-cold 50 mM Tris-HCl (buffer A) for 15 seconds with a homogenizer (Brinkmann Instruments, Westbury, NY). CHO-K1 cells were prepared by homogenization. The final pellet was resuspended in 10 mM Tris-HCl and kept on ice for receptor binding assays.
Aliquots of this membrane preparation can be obtained at various concentrations (eg, 10-11-10-6M) test peptide, 10 mg / ml bovine serum albumin (fraction V, Sigma Chemical Co., St. Louis, MO), MgCl2(5 mM), 0.05 nM in 50 mM HEPES (pH 7.4) containing trazirol (200 KIU / ml), bacitracin (0.02 mg / ml), and phenylmethylsulfonyl fluoride (0.02 mg / ml) [125I-Tyr] MK-678 (2000 Ci / mmol; c [N-methyl-Ala-Tyr (I125) -D-Trp-Lys-Val-Phe]) for 90 minutes at 25 ° C. The final assay volume was 0.3 ml. Rapid fill with a GF / C filter (previously soaked in 0.3% polyethyleneimine for 30 minutes) using a filtration manifold (Brandel, Geysersburg, MD) Incubation was terminated by rapidfilltration. Each tube and filter was then washed 3 times with 5 ml aliquots of ice-cold buffer A. Specific binding is determined by [(total bound [125I-Tyr] MK-678)-(binding in the presence of 200 nM somatostatin-14)].
The following test peptides were assayed: somatostatin-14, somatostatin-28, analog I, analog II, analog III, analog IV, and analog V. The structures of analogs I to V are shown above. The Ki value for the test peptide was calculated using the following formula:
Ki = IC50/ [1+ (LC / LEC)]
However, IC50Is a radioligand [125I-Tyr] concentration of test peptide required to inhibit 50% of specific binding of MK-678, LC is the concentration of radioligand (0.05 nM), and LEC is the equilibrium of radioligand Dissociation constant (0.155 nM). The Ki values calculated for the test peptides are shown in the column whose heading in Table 1 is “SSTR-2”.
(2)Human SSTR-5 binding assay:
For CHO-K1 cells, using standard techniques known in the field of molecular biology, Yamada et al., Biochem. Biophys. Res. Commun., 195-844 (1993). The human SSTR-5 cDNA described in 1 was transfected. See Patel et al., Biochem. Biophys. Res. Commun., 198: 605 (1994). Unpurified membranes, polytronTM(POLYTRONTM) Prepared by homogenizing CHO-K1 cells transfected with human SSTR-5 in 20 ml ice-cold 50 mM Tris-HCl with a homogenizer (setting 6, 15 seconds). Buffer was added to a final volume of 40 ml and the homogenate was solvated at 39,000 g for 10 minutes at 0-4 ° C.TMSS-34 rotor (SORVALTM Centrifuged with SS-34 rotor). The resulting supernatant was decanted and discarded. The pellet was rehomogenized, diluted and centrifuged further in ice-cold buffer as before. The final pellet was resuspended in 10 mM Tris-HCl and kept on ice for receptor binding assays.
Aliquots of this membrane preparation can be obtained at various concentrations (eg, 10-11-10-6M) test peptide, 10 mg / ml bovine serum albumin (fraction V), MgCl2(5 mM), 0.05 nM in 50 mM HEPES (pH 7.4) containing trazirol (200 KIU / ml), bacitracin (0.02 mg / ml), and phenylmethylsulfonyl fluoride (0.02 mg / ml) [125I-Tyr11] Incubated with somatostatin-14 (2000 Ci / mmol; Amersham Corp., Arlington Heights, IL) for 30 minutes at 30 ° C.
