【発明の属する技術分野】
本発明は、医薬および農薬の重要中間体である光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンの製造方法に関する。

【従来の技術】
光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンは、医薬および農薬の重要中間体である。該光学活性アミンの製造方法は、J. Am. Chem. Soc., 112, 5741(1990)に1件報告されているのみで、J. Chem. Soc., Perkin Trans. 1, 2039(1985)記載のオキシム誘導体の不斉還元を参考にして合成している。しかしながら、その化学収率および光学純度は、それぞれ、15%、71%ee(S)と低く、該光学活性アミンの工業的に簡便で且つ効率の良い製造方法ではなかった。

【発明が解決しようとする課題】
光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールは、ビス−3、5−(トリフルオロメチル)フェニルメチルケトンの不斉還元反応、または、ビス−3、5−(トリフルオロメチル)ベンズアルデヒドの不斉メチル化反応によって、両鏡像体を高い光学純度で得ることができる。従って、光学純度の高い該アルコールを出発原料として、高い光学純度を保ったままで、α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンへ変換することができれば、工業的に簡便で且つ効率の良い製造方法になるものと考えられる。

しかしながら、一般に、ベンジル位での求核置換反応は、ベンジルカチオンの安定化効果により、SN1型の置換反応も競合するため、部分的にラセミ化が起こることが知られている。α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールのp−トルエンスルホン酸エステルを用いて、加溶媒分解の反応性が調べられているが(J. Am. Chem. Soc., 115, 10091(1993)、J. Org. Chem., 52, 4164(1987))、ラセミ体を用いているため、置換反応の前後での立体化学については言及されていない。また、一般に、脱離基のβ位炭素上にプロトンをもつ基質では、その求核置換反応において、アルケンの副生が問題になる。特に、強い塩基性を合わせ持つ窒素求核試剤では、この副反応が頻繁に起こる。さらに、適当な官能基で保護された窒素求核試剤を用いた場合、その脱保護反応において、ラセミ化が起こらない緩和な条件を選択する必要がある。

このように、本発明に係わる光学純度の高いα−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールを出発原料として、高い光学純度を保ったままで、α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンへ収率良く変換できるかについては全く不明であった。

本発明は、光学純度の高いα−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールを出発原料として、高い光学純度を保ったままで、α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンへ収率良く変換できる製造方法の開発を目的とする。

【課題を解決するための手段】
本発明者等は、上記の課題を解決すべく鋭意検討を行った結果、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールを、塩基の存在下、スルホニル化試剤で、スルホニル化反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールのスルホン酸エステル誘導体を製造し、さらに、窒素求核試剤で、置換反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミン誘導体を製造し、引き続き、脱保護反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンが高い不斉転写率で収率良く製造できることを明らかにした。

【発明の実施の形態】
以下、本発明の光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンの製造方法について詳細に説明する。

本発明の製造方法は、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールを、塩基の存在下、スルホニル化試剤で、スルホニル化反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールのスルホン酸エステル誘導体を製造する過程、該光学活性スルホン酸エステル誘導体を、窒素求核試剤で、置換反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミン誘導体を製造する過程、および、該光学活性アミン誘導体を、脱保護反応することによって、光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアミンを製造する過程の三つよりなる。

本発明の第一過程において、一般式[3]で示される光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールのスルホン酸エステル誘導体を、以下の方法により、工業的に簡便で且つ効率良く製造することができる。

すなわち、一般式[1]で示される光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールを、塩基の存在下、一般式[2]で示されるスルホニル化試剤で、スルホニル化反応することによって製造することができる。

一般式[1]で示される光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールは、どのような方法で製造されたものでもよく、例えば、ビス−3、5−(トリフルオロメチル)フェニルメチルケトンの遷移金属−キラルホスフィン錯体を用いる不斉還元反応、または、ビス−3、5−(トリフルオロメチル)ベンズアルデヒドの典型金属または遷移金属−キラルリガンド錯体を用いる不斉メチル化反応によって、両鏡像体を高い光学純度で得ることができる。

本発明のスルホニル化反応に用いられるスルホニル化試剤としては、メタンスルホニルクロライド、エタンスルホニルクロライド、スルホニルジフロライド、スルホニルジクロライド、トリフルオロメタンスルホニルフロライド、トリフルオロメタンスルホニルクロライド、ペンタフルオロエタンスルホニルフロライド、ペンタフルオロエタンスルホニルクロライド、モノクロロメタンスルホニルクロライド、ジクロロメタンスルホニルクロライド、ベンゼンスルホニルクロライド、p−トルエンスルホニルクロライド、無水メタンスルホン酸、無水トリフルオロメタンスルホン酸、無水ベンゼンスルホン酸、無水p−トルエンスルホン酸等が挙げられる。その中でも、メタンスルホニルクロライド、モノクロロメタンスルホニルクロライド、p−トルエンスルホニルクロライドが好ましく、特に、メタンスルホニルクロライド、p−トルエンスルホニルクロライドがより好ましい。

本発明のスルホニル化反応に用いられる塩基としては、アミン類では、トリメチルアミン、トリエチルアミン、ジイソプロピルエチルアミン、トリn−プロピルアミン、トリn−ブチルアミン、ジメチルラウリルアミン、ジメチルアミノピリジン、N、N−ジメチルアニリン、ジメチルベンジルアミン、1、8−ジアザビシクロ(5、4、0)ウンデセン−7、1、4−ジアザビシクロ(2、2、2)オクタン、ピリジン、2、4、6−トリメチルピリジン、ピリミジン、ピリダジン、3、5−ルチジン、2、6−ルチジン、2、4−ルチジン、2、5−ルチジン、3、4−ルチジン等、または、無機類では、水素化ナトリウム、水素化カリウム、水素化リチウム、水素化カルシウム等の金属水素化物、炭酸ナトリウム、炭酸カリウム、炭酸セシウム、炭酸リチウム等の金属炭酸塩、炭酸水素ナトリウム、炭酸水素カリウム等の金属炭酸水素塩等が挙げられる。その中でも、トリエチルアミン、ピリジン、2、6−ルチジンが好ましく、特に、トリエチルアミン、2、6−ルチジンがより好ましい。

本発明のスルホニル化反応は、無溶媒で行ってもよいが、通常、溶媒が使用される。使用される溶媒としては、n−ペンタン、n−ヘキサン、c−ヘキサン、n−ヘプタン等の脂肪族炭化水素系、ベンゼン、トルエン、キシレン、メシチレン等の芳香族炭化水素系、塩化メチレン、クロロホルム、1、2−ジクロロエタン等のハロゲン化炭化水素系、ジエチルエーテル、テトラヒドロフラン、t−ブチルメチルエーテル、ジオキサン等のエーテル系、酢酸エチル、酢酸n−ブチル等のエステル系、アセトニトリル、プロピオニトリル等のニトリル系等が挙げられる。その中でも、トルエン、塩化メチレン、t−ブチルメチルエーテルが好ましく、特に、トルエン、塩化メチレンがより好ましい。これらの溶媒は、単独または組み合わせて用いることができる。

本発明のスルホニル化反応の温度範囲は、−50〜50℃であり、−40〜40℃が好ましく、特に、−30〜30℃がより好ましい。

本発明のスルホニル化反応においては、反応終了後、通常の後処理操作を行うことにより、粗生成物を得ることができる。得られた粗生成物は、必要に応じて、活性炭、蒸留、再結晶、カラムクロマトグラフィー等の精製操作を行うことにより、目的の一般式[3]で示される光学活性α−メチル−ビス−3、5−(トリフルオロメチル)ベンジルアルコールのスルホン酸エステル誘導体を高い純度で得ることができる。
