【発明の属する技術分野】
本発明は、表面プラズモンの発生を利用して物質の特性を分析する表面プラズモン測定装置等の全反射光によるエバネッセント波の発生を利用した測定装置に関するものである。

【従来の技術】
金属中においては、自由電子が集団的に振動して、プラズマ波と呼ばれる粗密波が生じる。そして、金属表面に生じるこの粗密波を量子化したものは、表面プラズモンと呼ばれている。

従来より、この表面プラズモンが光波によって励起される現象を利用して、被測定物質の特性を分析する表面プラズモン測定装置が種々提案されている。そして、それらの中で特に良く知られているものとして、 Kretschmann配置と称される系を用いるものが挙げられる(例えば特開平6−167443号参照)。

上記の系を用いる表面プラズモン測定装置は基本的に、例えばプリズム状に形成された誘電体ブロックと、この誘電体ブロックの一面に形成されて液体試料などの被測定物質に接触させられる金属膜と、光ビームを発生させる光源と、上記光ビームを誘電体ブロックに対して、該誘電体ブロックと金属膜との界面で全反射条件が得られるように種々の角度で入射させる光学系と、上記界面で全反射した光ビームの強度を測定して表面プラズモン共鳴の状態、つまり全反射減衰の状態を検出する光検出手段とを備えてなるものである。

なお上述のように種々の入射角を得るためには、比較的細い光ビームを入射角を変化させて上記界面に入射させてもよいし、あるいは光ビームに種々の角度で入射する成分が含まれるように、比較的太い光ビームを上記界面に収束光状態であるいは発散光状態で入射させてもよい。前者の場合は、入射した光ビームの入射角の変化に従って、反射角が変化する光ビームを、上記反射角の変化に同期して移動する小さな光検出器によって検出したり、反射角の変化方向に沿って延びるエリアセンサによって検出することができる。一方後者の場合は、種々の反射角で反射した各光ビームを全て受光できる方向に延びるエリアセンサによって検出することができる。

上記構成の表面プラズモン測定装置において、光ビームを金属膜に対して全反射角以上の特定入射角で入射させると、該金属膜に接している被測定物質中に電界分布をもつエバネッセント波が生じ、このエバネッセント波によって金属膜と被測定物質との界面に表面プラズモンが励起される。エバネッセント光の波数ベクトルが表面プラズモンの波数と等しくて波数整合が成立しているとき、両者は共鳴状態となり、光のエネルギーが表面プラズモンに移行するので、誘電体ブロックと金属膜との界面で全反射した光の強度が鋭く低下する。この光強度の低下は、一般に上記光検出手段により暗線として検出される。なお上記の共鳴は、入射ビームがp偏光のときにだけ生じる。したがって、光ビームがp偏光で入射するように予め設定しておく必要がある。

この全反射減衰(ATR)が生じる入射角、すなわち全反射減衰角θspより表面プラズモンの波数が分かると、被測定物質の誘電率が求められる。すなわち表面プラズモンの波数をKsp、表面プラズモンの角周波数をω、cを真空中の光速、εm とεs をそれぞれ金属、被測定物質の誘電率とすると、以下の関係がある。

すなわち、上記反射光強度が低下する入射角である全反射減衰角θspを知ることにより、被測定物質の誘電率εs 、つまりは屈折率に関連する特性を求めることができる。

なおこの種の表面プラズモン測定装置においては、全反射減衰角θSPを精度良く、しかも大きなダイナミックレンジで測定することを目的として、特開平11−326194号に示されるように、アレイ状の光検出手段を用いることが考えられている。この光検出手段は、複数の受光素子が所定方向に配設されてなり、前記界面において種々の反射角で全反射した光ビームの成分をそれぞれ異なる受光素子が受光する向きにして配設されたものである。

そしてその場合は、上記アレイ状の光検出手段の各受光素子が出力する光検出信号を、該受光素子の配設方向に関して微分する微分手段が設けられ、この微分手段が出力する微分値に基づいて被測定物質の屈折率に関連する特性を求めることが多い。

また、全反射減衰(ATR)を利用する類似の測定装置として、例えば「分光研究」第47巻 第1号(1998)の第21〜23頁および第26〜27頁に記載がある漏洩モード測定装置も知られている。この漏洩モード測定装置は基本的に、例えばプリズム状に形成された誘電体ブロックと、この誘電体ブロックの一面に形成されたクラッド層と、このクラッド層の上に形成されて、液体試料に接触させられる光導波層と、光ビームを発生させる光源と、上記光ビームを上記誘電体ブロックに対して、該誘電体ブロックとクラッド層との界面で全反射条件が得られるように種々の角度で入射させる光学系と、上記界面で全反射した光ビームの強度を測定して導波モードの励起状態、つまり全反射減衰状態を検出する光検出手段とを備えてなるものである。

上記構成の漏洩モード測定装置において、光ビームを誘電体ブロックを通してクラッド層に対して全反射角以上の入射角で入射させると、このクラッド層を透過した後に光導波層においては、ある特定の波数を有する特定入射角の光のみが導波モードで伝搬するようになる。こうして導波モードが励起されると、入射光のほとんどが光導波層に取り込まれるので、上記界面で全反射する光の強度が鋭く低下する全反射減衰が生じる。そして導波光の波数は光導波層の上の被測定物質の屈折率に依存するので、全反射減衰が生じる上記特定入射角を知ることによって、被測定物質の屈折率や、それに関連する被測定物質の特性を分析することができる。

なお、表面プラズモン共鳴測定装置もしくは漏洩モード測定装置等の全反射を利用した測定装置としては、光を界面に全反射条件が得られる入射角で入射させ、その光によるエバネッセント波の発生により、界面で全反射した光の状態の変化を測定することにより被測定物質の特性分析等を行うに際して、前述の全反射減衰を生じる特定入射角の測定をする装置のほか、複数の波長の光ビームを界面に入射させ、角波長毎の全反射減衰の程度を検出する装置、あるいは、光ビームを界面に入射させるとともに、この光ビームの一部を、界面入射前に分割し、この分割した光ビームを界面で反射した光ビームと干渉させて、該干渉の状態を測定する装置等種々のタイプがある。

また、上述した表面プラズモン共鳴測定装置等の全反射光を利用した測定装置は、創薬研究分野等において、所望のセンシング物質に結合する特定物質を見いだすランダムスクリーニングへ使用される場合があり、効率よくスクリーニングを行うために、誘電体ブロックと試料保持部とが一体化した複数の測定チップと該複数の測定チップを一次元もしくは二次元状に支持する支持台とからなるセンサユニットや、誘電体ブロック自体が、該誘電体ブロックの上面に開口を有する検体ウェルを1次元状もしくは2次元状に複数備えたセンサウェルユニットを構成するもの、および、該センサユニットやセンサウェルユニットの複数の検体ウェルに対して並列的に複数の光ビームを入射させ、各測定チップ(もしくはウェル)の界面における反射光を個別に検出する測定装置が提案されている。

【発明が解決しようとする課題】
ところで、上記測定装置においては、測定チップが配置されている周囲の温度や湿度等の環境値が変化すると試料液の屈折率等が変化するため、環境値の変動により、本来同一の検出結果が得られるべき試料であっても、実際に検出される検出結果が異なるものとなる場合が生じ、検出結果の信頼性が低下するという問題がある。

この問題を解決するために、本出願人は、環境値を検知する環境値検知手段を備え、環境値に左右されない検出結果を得ることができる測定装置を既に提案している。しかしながら、複数の測定チップ(もしくはウェル)について同時に測定する測定装置の場合、測定チップの位置により環境値に差があるために、各測定チップ毎に環境値検知手段を備える必要が生じ、測定装置のコストアップおよび装置の複雑化を引き起こしかねないという問題がある。

本発明は上記の事情に鑑みて、コストアップ、装置の複雑化を抑制した、周囲の環境値に左右されずに、全反射光の状態を精度良く検出することができる信頼性の向上した測定装置を提供することを目的とする。

【課題を解決するための手段】
本発明の測定装置は、光ビームを発生させる光源と、
前記光ビームに対して透明な誘電体ブロック、この誘電体ブロックの一面に形成された薄膜層、およびこの薄膜層の表面上に試料を保持する試料保持機構を備えてなる3以上の複数の測定チップと、該複数の測定チップを支持する支持体からなるセンサユニットと、
前記光ビームを前記誘電体ブロックに対して、該誘電体ブロックと前記薄膜層との界面で全反射条件が得られる入射角で入射させる入射光学系と、
前記界面で全反射した光ビームの強度を検出する光検出手段とを備えてなる測定装置において、
前記センサユニット近傍における、2つ以上かつ前記複数の測定チップ数より少数箇所の環境値をそれぞれ検知する環境値検知手段と、
該環境値検知手段により検知された前記少数箇所の環境値から前記複数の測定チップの各々における環境値を求める環境値演算手段と、
該各測定チップの環境値に基づいて該各測定チップについての前記光検出手段による検出結果を補正する補正手段とを備えたことを特徴とするものである。

前記センサユニットは、測定チップと支持体が別個に形成され、測定チップが支持体の測定チップ保持孔に保持されてなるものであってもよいし、測定チップと支持体が一体的に形成され、測定チップの試料保持部がウェル状に形成されたセンサウェルユニットであってもよい。

前記環境値は、複数個の環境値の組み合わせでもよいし、1つの環境値であってもよい。環境値としては、例えば温度あるいは湿度などが挙げられる。また、環境値は、温度や湿度そのものであっても、所定値からの差、変動量等であってもよい。

上記各全反射減衰を利用した測定装置において、「光検出手段による検出結果を補正する」とは、光ビームの強度に基づいて求められた値、例えば全反射減衰角の角度等を補正することも含むものである。また、多数の受光素子がアレイ状に設けられた光検出手段と、該受光素子の並設方向に関して微分する微分手段を用いる場合等には、該微分手段が出力する微分値を補正することも含むものである。

本発明による全反射光を利用した測定装置において、光検出手段により前記界面で全反射した光ビームの強度を測定して試料の分析を行うには種々の方法があり、例えば、光ビームを前記界面で全反射条件が得られる種々の入射角で入射させ、各入射角に対応した位置毎に前記界面で全反射した光ビームの強度を測定して、全反射減衰により発生した暗線の位置(角度)を検出することにより試料分析を行ってもよいし、D.V.Noort,K.johansen,C.-F.Mandenius, Porous Gold in Surface Plasmon Resonance Measurement, EUROSENSORS XIII, 1999, pp.585-588 に記載されているように、複数の波長の光ビームを前記界面で全反射条件が得られる入射角で入射させ、各波長毎に前記界面で全反射した光ビームの強度を測定して、各波長毎の全反射減衰の程度を検出することにより試料分析を行ってもよい。

【発明の効果】
本発明の測定装置は、センサユニットの測定チップ数よりも少数の環境値から各測定チップにおける環境値を求める環境値演算手段と、各測定チップについての検出結果を環境値に応じて補正する補正手段とを備えているので、各測定チップ毎に環境値検知手段を備える必要がなく、各測定チップにそれぞれ環境値検知手段を備える場合と比較して、コストアップ、装置の複雑化を抑制しつつ、各測定チップについて、環境値に左右されない検出結果を得ることができ、測定装置の信頼性を向上させることができる。

【発明の実施の形態】
以下、図面を参照して本発明の実施の形態を詳細に説明する。図1と図2はそれぞれ、本発明の第1の実施の形態による表面プラズモン共鳴測定装置の平面形状、側面形状を示すものである。

本実施の形態の測定装置においては、誘電体ブロックの平坦な上面に開口する検体ウェル(液溜め)101が、該誘電体ブロックに1次元状に複数備えられ、該各検体ウェル101の内底面に薄膜層として金属膜12が形成されてなるセンサウェルユニット100が備えられている。センサウェルユニット100は、例えば透明樹脂等で構成されており、金属膜12上には特定物質と結合するセンシング物質14が固定されてなる。また上記ウェル101底面の各々に下方に突出した誘電体ブロックの光入出射部102が形成され、この光入出射部102と上記金属膜12との界面102aに光ビームLが入射されるようになっている。

さらに、本表面プラズモン共鳴測定装置は光ビームLを出射するレーザ光源2と、該光源2から出射された光ビームLをセンサウェルユニット100の矢印P方向に並んだ複数(本例では8個)のウェル101の底面に、並列的に入射させる光ビーム入射手段1と、各ウェル101の界面102aで反射した光ビームLをそれぞれ受光する矢印P方向のウェル数と同数の8個の光検出手段17A〜Hと、該光検出手段17A〜Hからの出力を受ける信号処理部20と、該信号処理部20に接続された表示部25と、環境値検知手段である温度検知手段51〜54とを備えている。

光ビーム入射手段1は、レーザ光源2から発散光状態で発せられた光ビームLを平行光とするコリメータレンズ3、光ビームLを分岐するハーフミラー4a〜gおよびミラー4h、分岐された光ビームLを図2で示される面内のみで径を拡大するシリンドリカルビームエキスパンダ5a〜h、光ビームLを反射するミラー6a〜h、該ミラー6a〜hで反射された光ビームLを、図2に示される面内のみで集光するシリンドリカルレンズ7a〜hとを備えてなる。

次に、測定原理を説明する。

図1に示す通り、1つのレーザ光源2から発散光状態で発せられた光ビームLはコリメーターレンズ3で平行光とされた後、ハーフミラー4a〜gとミラー4hとによって8本に分岐され、分岐された8本の光ビームLが各々ウェル101の底面、つまり光入出射部102と金属膜12との界面102aに入射する。

このとき、分岐された各光ビームLは、それぞれシリンドリカルビームエキスパンダ5a〜hにより図2に示される面内のみで径が拡大され、それぞれミラー6a〜hで反射して進行方向を変えた後、それぞれシリンドリカルレンズ7a〜hにより図2に示される面内のみで集光される。それにより各光ビームLは、それぞれ光入出射部102と金属膜12との界面102aに対して、種々の入射角成分を持った状態で入射する。なおレーザ光源2は、直線偏光である光ビームLがp偏光状態で上記界面102aに入射するようになる向きに配設されている。その他、光ビームLを界面102aに対してp偏光で入射させるには波長板で光ビームLの偏光の向きを制御するようにしてもよい。

光ビームLは、上述のように集光されるので、界面102aに対して種々の入射角θで入射する成分を含むことになる。なお、この入射角θは、全反射角以上の角度とされる。そのため、界面102aで全反射した反射光ビームLには、種々の反射角で全反射された成分が含まれることになる。なお、上記光ビーム入射手段1は、光ビームLを界面上に点状に集光させずにデフォーカス状態で入射させるように構成してもよい。そのようにすれば、界面上のより広い領域において光ビームLが全反射されるので、全反射減衰の状態の検出誤差が平均化されて全反射解消角の測定精度を高めることができる。

界面102aで全反射した光ビームLは光検出手段17A〜Hにより検出される。本例における各光検出手段17A〜Hは、それぞれ複数のフォトダイオード17a、17b、17c……が1列に並設されてなるフォトダイオードアレイであり、図1の図示面内において、平行光化された光ビームLの進行方向に対してフォトダイオード並設方向がほぼ直角となる向きに配設されている。したがって、上記界面102aにおいて種々の反射角で全反射した光ビームLの各成分を、それぞれ異なるフォトダイオード17a、17b、17c……が受光することになる。なお、下記の説明において、各検出手段17A〜Hは単に光検出手段17と記載する。

図3は、(a)光ビームLの界面102aへの入射角θと上記光強度Iとの関係、および(b)入射角θと光強度Iの微分値dIとの関係を示すものである。図3(a)に示すように、界面102aにある特定の入射角θSPで入射した光は、金属膜12と試料との界面に表面プラズモンを励起させるので、この光については反射光強度Iが鋭く低下する。つまりθSPが全反射減衰角であり、この角度θSPにおいて反射光強度Iは最小値を取る。反射光強度Iの最小値の位置変動の経時的な変化、すなわち全反射減衰角の経時的な変化を測定することにより、試料液中の特定物質とセンシング物質14との結合の有無を得ることができる。また、図3(b)に示すように、反射光強度Iの微分値dI=0となる位置は、全反射減衰角の位置変動に伴い変動する。したがって、測定開始時に微分値dI=0近傍であった測定点についての経時変化を観測することにより、間接的に全反射減衰角の変化を得ることができ、特定物質とセンシング物質14との結合の有無を得ることができる。

本実施形態の信号処理部20は、高精度の測定を行うため、上述のように反射光強度Iの微分値dIを求め、該微分値dIの経時変化を測定することにより、全反射減衰角の経時変化を間接的に観察し、特定物質とセンシング物質14との結合の有無を得るものである。ただし、ここでは、光検出手段17から得られた光強度Iの微分値dIを測定値Sとし、補正手段22において、この測定値Sに対して所定の補正を施した補正値S’を求め、該補正値S’を真の微分値dI’として結合の有無を得て、検出結果を表示部25に出力する。

次に、上記構成の表面プラズモンセンサーにおける実際の測定方法について説明する。

まず、センサウェルユニット100の各ウェル101にそれぞれ試料液を分注する。このとき、8つのウェル101のうち検体ウェルW1〜W7には被検体を含む試料液を分注し、残りの1つのウェルWcには検体ウェルの注入される試料液の溶媒のみを注入する。このウェルWcは、各検体ウェルWn(n=1、2、…7)における溶媒による屈折率の変化(バルク効果)を差し引くためのコントロールとして用いられる。

上述の測定原理で説明した通り、8つのウェルWnおよびWcの各界面102aに対して光ビームLを並行して入射させ、該界面102aからの全反射光をそれぞれ光検出手段17で検出する。

光検出手段17が受光した光強度Iが信号処理部20に入力されるとともに、温度検出手段51〜54が検知した検知温度も信号処理部20に入力される。なお、光強度の測定および温度検知は、所定時間が経過する毎になされる。

信号処理部20は、まず、温度演算手段21において、温度検知手段51〜54により検知された検知温度から各ウェルWnおよびwcにおける擬似検知温度を求め、次に、補正手段22において、各ウェルWnおよびWcの温度に基づいて、光検出手段17から出力された光強度Iの微分値dIを測定値Sとして該測定値Sに対し所定の補正を施し、補正値S’(=真の微分値dI’)を得、該補正値S’に基づいて被検体とセンシング物質14との結合の有無を判定し、判定結果を表示部25に出力する。

以下、温度演算手段21および補正手段22の作用を説明する。

図4は、温度演算手段22による温度演算方法を説明するための図である。図4中のグラフの縦軸は温度T、横軸はセンサウェルユニット100のP方向位置を示す。温度演算手段22においては、温度検知手段51〜54により検知された検知温度、ここでは図4中○印で示した4つの測定点に基づいて、同図中実線で示すような2次近似曲線を求め、該2次近似曲線から同図中×印で示した各ウェルWnおよびWcにおける擬似検知温度Tn(n=1、2…7)およびTcを得る。この手法によれば、ウェル数よりも少ない温度検知手段で、各ウェルの温度を近似的に求めることができ、各ウェル毎に温度検知手段を備える必要がないため、コストアップおよび装置構成の複雑化を防ぐことができる。

図5は、温度Tに対するドリフトの傾きαの関係の一例を示すものである。表面プラズモン共鳴測定装置では、光検出器手段により検出される反射光が一定温度においても徐々に位置変化を示すドリフトという現象がみられる。図5に示すように、このドリフトの傾き(変化の度合い)は各温度毎で異なり、温度が高いほど傾きが大きくなる傾向にある。本実施形態のように、複数のウェルを備えたセンサウェルユニットにおいては、検体ウェルWnとコントロールウェルWcとの間に温度差が生じる場合があり、上述のように、温度が異なるとドリフトの傾きαが異なるため、バルク効果の除去の際にはこの温度差を考慮する必要がある。

補正手段22は、予め求められた温度とドリフト傾きとの関係を記憶しており、入力された測定値Sおよび各ウェル毎の温度TnおよびTcに基づいて、補正式(1)に基づいた補正を行い、補正値Soutを出力する。
