【発明の属する技術分野】
本発明は円二色性蛍光励起スペクトル測定装置、特にその蛍光の集光効率と試料の利用効率の改善に関するものである。

【従来の技術】
多くの化学物質において、その絶対構造、立体構造に関する知見は、極めて基本的かつ重要不可欠な情報となっている。特に薬物、毒物、生体物質などの生理活性物質においては、その生理活性が直接にそのキラリティ、すなわち絶対構造、立体構造に依存することから、重要な研究課題とされている。
しかしながら、キラリティのみ異なる光学異性体は、化学的性質は実質的に同一であり、各種物理的特性も極めて近似しているため、これら薬物、毒物、生体物質などの生理活性物質のキラリティを解析する手段としては、X線結晶構造解析、円二色性スペクトルくらいに限られてくる。特に円二色性スペクトルは、取り扱いが比較的平易であることから、この研究には重要不可欠な手段として広く用いられている。

ところが、例えば生理活性物質などのように試料によっては、この円二色性スペクトル測定のために十分な量を確保することが容易ではない場合も多く、この円二色性スペクトル測定検出感度の向上、あるいはより感度の高い代替の測定手段の実現が強く望まれていた。その代替となり得る手段として、蛍光検出円二色性測定装置が知られている。

【発明が解決しようとする課題】
ところが、従来の蛍光検出円二色性スペクトル測定装置は、もっぱら蛍光検出による選択性の向上にのみ着目されていて、その潜在的な感度向上の可能性については全く考慮されておらず、大きな改善の余地が残されていた。すなわち、蛍光を検出する場合の検出感度は、試料から放射される蛍光の集光効率に依存するため、可能な限りその集光効率を高める設計がなされることが望ましいが、従来の蛍光検出円二色性スペクトル測定装置ではそれがなされておらず、大きな改善の余地が残されていた。

本発明は前記従来技術の課題に鑑みなされたものであり、その目的は検出感度の向上を図ることができると共に、試料の利用効率の改善を図ることができる円二色性蛍光励起スペクトル測定装置を提供することにある。

【課題を解決するための手段】
前記目的を達成するために、本発明にかかる円二色性蛍光励起スペクトル測定装置は、光源と、該光源から出た光束のうち、所定波長の励起光束のみを通過させる励起光波長選択手段と、該励起光波長選択手段からの光束を所定の変調周波数で交互に左右の円偏光とする円偏光変調器と、試料が入れられ、該円偏光変調器を通過した左右の円偏光が交互に照射されるサンプルセルと、該サンプルセル内の試料から放射された蛍光のうち、所定波長の蛍光のみを通過させる蛍光波長選択手段と、該蛍光波長選択手段からの蛍光を蛍光強度信号とする蛍光測定手段と、該蛍光強度を検出した電気信号のうち、該変調周波数に同期して交流成分を取り出して円二色性蛍光励起スペクトルとする信号処理手段と、を備え、サンプルセルと蛍光測定手段の間に集光系を介さず、かつ、該サンプルセルと蛍光測定手段を近接させたことを特徴とする。

なお、前記装置において、前記蛍光波長選択手段はカットオフフィルタであり、該カットオフフィルタにより励起光の散乱光の波長域の光を選択的に吸収して除去することが好適である。
また、前記装置において、前記サンプルセルを透過した光の通過コース上に設けられ、該透過光を除去する光トラップを備えることが好適である。

【発明の実施形態】
以下、図面に基づき本発明の好適な実施形態を説明する。
図1には本発明の一実施形態にかかる円二色性蛍光励起スペクトル測定装置が示されている。

同図に示す円二色性蛍光励起スペクトル測定装置10は、光源12と、励起光波長選択手段である分光器14と、ピエゾエラスティックモジュレータ(PEM)よりなる円偏光変調器16と、サンプルセル18と、蛍光波長選択手段であるカットオフフィルタ19と、蛍光測定手段であり、光増倍管(PMT)よりなる検出器20と、プリアンプ22と、直流アンプ24と、交流アンプ26と、ロック・インアンプ28と、直流アンプ30と、A/D変換器32と、I/O装置34と、分光器ドライバ36と、PEMドライバ38と、PMT印加電圧制御回路40と、制御手段であるCPU42と、記憶手段44を備える。

そして、光源12の発光方向前方には分光器14が設置され、この分光器14により波長走査され順次所定波長の単色光となり、さらに所定の振動方向の直線偏光となった光束は、分光器14の前方に設置された円偏光変調器16を通過する。この円偏光変調器16では、左右円偏光が所定の変調周波数信号s1(本実施形態においては図2(a)に示すように50kHz)で交互につくられて、サンプルセル18内の試料に照射される。

ここで、サンプルセル18内の試料に励起光束L1である左右円偏光の不等吸収が起こると、円偏光変調器16の変調周波数信号s1に対応して蛍光L2にも強弱の波が生じ検出器20に到達するが、試料から放射された蛍光L2は、カットオフフィルタ19により励起光束L1の散乱光が取り除かれ、目的の蛍光波長域の光のみが検出器20に入射する。
以上のようにして本実施形態にかかる円二色性蛍光励起スペクトル測定装置10を構成することにより、試料から放射された蛍光を測定することができる。

なお、本実施形態では円偏光変調器16を用いているため、検出器20の出力信号には、直流成分信号s3にわずかの交流成分信号s4が含まれており(図2(b)参照)、この交流成分信号s4には、左右の円偏光で励起したときの蛍光強度の差(励起スペクトルの差)が反映している(同図(c)参照)。
このため、本実施形態においては、例えばこの励起スペクトルの差を求めることにより、円二色性蛍光励起スペクトル(図3参照)を得ることができる。

また、分光器14、円偏光変調器16の動作は、それぞれ分光器ドライバ36、PEMドライバ38により管理されている。
このような円二色性蛍光励起スペクトル測定装置10においては、もともと微弱な蛍光のさらに差を解析しなければならないため、通常の蛍光測定装置と比較しても一層の蛍光検出効率の向上が要求される。
以下、本実施形態の特徴的事項について詳細に説明するが、まず本実施形態における蛍光の集光効率の改善方法について説明する。

サンプルセルと検出器の配置
従来においては、サンプルセル18の設置場所に空間を確保してその扱いを容易にするため、図4(a)に示すように、検出器20をサンプルセル18より離し、このサンプルセル18と検出器20との間の光路中に、集光系である集光レンズ46,48を設置していた。

しかしながら、従来のように検出器20をサンプルセル18より離すと、集光レンズ46,48の開口角は限られていて十分大でなく、その分、集光効率も限られていた。また、集光系は最低2枚のレンズ46,48を設けていたため、この集光レンズ46,48の表面での反射によっても、試料から放射された蛍光L2の約20%程度をロスしていた。

そこで、本実施形態においては、同図(b)に示すように、前記集光系である集光レンズ46,48を介さず、カットオフフィルタ19を介して検出器20をサンプルセル18に近接させている。
この結果、試料から放射された蛍光L2の大部分を検出器20に入射させることができる。これにより、蛍光L2の集光効率を最大とすることができるので、検出感度の向上を図ることができる。

カットオフフィルタ
従来より、一般的な蛍光分光光度計などの円二色性蛍光励起スペクトル測定装置では、検出する蛍光L2の波長を選択するために回折格子を用いた分光器を組み込むのが普通である。
しかしながら、このような分光器を組み込むことにより、検出する蛍光L2の波長を可変することができる、蛍光の波長純度をコントロールすることができる、などの利点があるものの、光学素子の効率のために光のロスが避けられず、その分、機構が複雑となりコストがかかるなどのマイナス面もあった。

そこで、本実施形態においては、励起光L1の散乱光さえ取り除くことができれば、それより長い波長の光の全部を用いても何等問題なく、むしろ光の量が増えて感度を向上させるためには好都合であることから、検出器20側の分光器として前記図4(b)に示すようにカットオフフィルタ19を用いることが好適である。
このカットオフフィルタ19は、入射した光のうち、励起光L1の散乱光の波長成分のみを吸収して、それより長波長の蛍光のみを検出器20に入射させるので、検出器20に入射される蛍光L2の量を増やすことができ、検出感度の向上を図ることができる。

また、このカットオフフィルタ19の厚さは数mm程度と薄いため、その後段に設置される検出器20をサンプルセル18にできる限り近接させて、その開口比を大きく確保することにも役立つ。これにより、集光効率の向上を図ることができるので、検出感度の向上を図ることもできる。

光トラップ
試料を透過した光束は迷光となり、測定の妨害となるおそれがあった。また、本実施形態のように励起光として円偏光を用いた場合には、試料を透過した円偏光に反射が生じると偏光状態が変わり、最悪、逆方向の円偏光となって再度サンプルセル18に戻るおそれがあった。そして、このような逆方向の円偏光がサンプルセル18に入射すると、試料の励起に関わり、左右の円偏光励起による蛍光強度の差を小さくし、折角の信号を小さくしてしまう。

そこで、本実施形態においては、これらの影響を取り除くため、前記図4(b)に示すように、サンプルセル18からの透過光L3を除去することが可能な光トラップ50を設けている。これにより、前述のようなノイズを大幅に低減することができると共に、最大信号の確保を行うことができる。

凹面鏡
前記図4(b)に示すように、試料からの蛍光L2は、励起光L1の照射方向に関わらず、全包囲に放射されるが、検出に用いられるのは、そのうちの1方向である検出器20方向でしかなく、その他は無駄となる。
そこで、本実施形態においては、サンプルセル18を間に挟み検出器20の反対側に凹面鏡52を設置して、その焦点をサンプルセル18のほぼ中央部に位置させる。これにより、検出器20の反対側に放射された蛍光L2も、この凹面鏡52で入反射されてサンプルセル18に戻り、そのまま検出器20に入射することができる。

この結果、検出器20に入射する蛍光L2の量を増やすことができるので、検出感度の向上を図ることができる。

内面反射型の筒形反射鏡
本実施形態においては、前記図4(b)に示す凹面鏡52に代わり、図5に示すように、サンプルセル18として円筒形セルを用い、該円筒形セルの周囲を一部除いた内面反射型の筒形反射鏡で囲み込み、光の利用効率を上げることも好適である。

すなわち、サンプルセル18として円筒形セルを用いたときの光利用効率をさらに上げるためには、図5に示すように、円筒形セルの周りに内面反射型の筒形反射鏡を設置することが好適であり、その1つは、同図(a)に示すように内面反射型の円筒形鏡54の一部(例えば検出器20の口径とほぼ同様)を切り欠いたものを用いて、試料から実質的に全方位へ放射された蛍光を、この円筒形鏡54にて入反射させて、前記図1に示す検出器20に入射させることができる。

もう1つは、同図(b)に示すような、内面反射型の楕円筒形鏡48を用いることができる。そして、この楕円筒形鏡56の切り抜き部分(例えば検出器20の口径とほぼ同様)から遠い方の焦点が、サンプルセル18である円筒形セルの中心に一致するように設ける。これにより、サンプルセル18である円筒形セルから放射された蛍光を、もう1つの焦点に集光した後、効率的に前記図1に示す検出器20に導くことができる。

この結果、試料から放射された蛍光のうち、検出器20方向だけでなく、実質的に全方向に放射された蛍光を検出器20に入射させることができる。これにより、検出器20に入射する蛍光の量を増やすことができるので、検出感度の向上を図ることができる。
なお、前記円筒形鏡54、楕円筒形鏡56などの内面反射型の筒形反射鏡としては、金属製の筒体の内面を研磨したものを用いることができる。

また、サンプルセルとして無蛍光ガラス製の円筒形セルを用い、その外周面に、前記筒形反射鏡としてアルミなど反射率の高い金属を蒸着、メッキすることでも前記円筒形鏡54、楕円筒形鏡56と同様の効果を得ることもできる。しかも、このようにサンプルセル18の外周面に鏡面を設けることにより、サンプルセルをキズや劣化などから有効に保護することもできる。

導光パイプ
本実施形態においては、試料から放射された蛍光L2を、できる限り効率的に検出器20に集光させるためには、前記図4に示すように、サンプルセル18、カットオフフィルタ19および検出器20を直接近接させることも好適であるが、このほか、図6に示すように、サンプルセル18とカットオフフィルタ19との間の光路中に、その内面が鏡面研磨された導光パイプ58を設けることも好適である。

この結果、サンプルセル18から図中右方向に放射された蛍光L2の一部は、該導光パイプ58の内面にて反射せずに直接カットオフフィルタ19を介して検出器20に入射するものもあるが、また導光パイプ58の内面に入射した蛍光L2も、その内面にて1回ないし複数回入反射して、カットオフフィルタ19を介して検出器20に入射することができる。これにより、前記図4に示すサンプルセル18、カットオフフィルタ19および検出器20を直接近接させる構成と同様に、試料から放射された蛍光L2の集光効率の向上を図ることができるので、検出感度の向上を図ることができる。

サンプルセルの形状の違いによる蛍光の集光効率の改善
試料と空気の屈折率の違いにより、サンプルセル18として角形セルを用いた場合、サンプルセル18から外部に蛍光が出るとき、図7(a)に示すように、蛍光L2が開くように屈折する。このため、検出器20に入射される蛍光L2は、その検出器20の口径とサンプルセル18からの距離とで幾何学的に決まる開口立体角と比較して少なくなる。
そこで、本実施形態のように、同図(b)に示すようにサンプルセル18として円筒形セルを用いることにより、その回転軸方向の事情は変わらないが、径方向では屈折がないため、その分だけ、高い光の利用効率を確保することができるので、検出感度の向上を図ることができる。

以上のようにしてサンプルセル18、カットオフフィルタ19、検出器20、光トラップ50、凹面鏡52、内面反射型の筒形反射鏡54,56、導光パイプ58などを適宜構成して蛍光の集光効率の向上を図ることにより、蛍光のように微弱な光を検出する場合でも検出感度の向上を図ることができる。
つぎに、本実施形態における試料の利用効率の改善について説明する。

サンプルセルの形状の違いによる試料の利用効率の改善
図8(a)に示すように、サンプルセル18の励起光束L1の通らない部分(図中黒い部分)は、測定に関与しない部分であり、限られた試料の量から、より濃い試料溶液を調整するためには無駄な部分となる。
そこで、本実施形態においては、サンプルセル18として円筒形セルを用い、同図(b)に示すように、励起光束L1とほぼ同じ形状の円形に形成された端部であるセル窓板より励起光束L1を入射させることにより、同図(a)に示すようにサンプルセル18として角形セルを用い、側方のセル窓板より励起光束L1を入射させた場合に比較し、その無駄な部分(図中黒い部分)を低減することができるので、試料の利用効率の改善を図ることができる。
また、それ以外にもサンプルセル18として円筒形セルを用いると、つぎのような利点が得られる。

すなわち、円筒形セルの方が角形のものに比較し、前記励起光束L1の入射するセル窓板の歪みを少なく作成することができる。この歪みは、本実施形態において励起円偏光の偏光状態を乱し、ベースラインの曲がりの原因となるものであるため、サンプルセル18としてこの歪みの少ない円筒形セルを用いることにより、得られるスペクトルの質的な向上を図ることもできる。

サンプルセルの高低調節手段
生体成分のような試料では、その絶対量を確保することが困難であることが多く、測定には限られた微少量しか用いられないことが一般的である。一方、測定で得られる信号強度は濃度に依存する。
したがって、感度を確保するためには、利用できる試料をできるだけ少量の溶媒に溶解してできる限り濃い試料溶液60とし、測定に供するのが好ましい。より具体的には、測定において励起光に照射されない試料部分を最小にするようにし、それに必要なギリギリの量の試料溶液60を調整するのが好ましい。

そこで、本実施形態においては、図9に示すように、その底部62aが高さ調節自在で、該底部62aにサンプルセル18が設けられたセルホルダ62と、セルホルダ底部62aを図中上下させてサンプルセル18の高さを調節可能な高低調節手段64を備えることが好適である。
そして、高低調節手段64を駆動させて、励起光束L1をカバーすることができるギリギリの高さまでサンプルセル18を持ち上げることにより、サンプルセル18として円筒形セルを用いた場合は勿論、たとえこれに角形のものを用いた場合でも、試料溶液60を該励起光束L1をカバーすることができる最低限の量で済ませることができる。

以上のようにしてサンプルセル18、セルホルダ62、高低調節手段64などを構成することにより、励起光束L1をカバーすることができる必要最低限の試料量で蛍光測定を行うことができるので、試料の利用効率の改善を図ることができる。

なお、本発明の円二色性蛍光励起スペクトル測定装置としては、上記実施形態のものに限られるものでなく、種々の態様の変更が可能であり、例えば前記図1に示すI/O装置34に、可搬型コンピュータのインターフェースをケーブル接続し、同図に示すCPU42の制御機能、記憶手段44の記憶機能などの各機能を、可搬型コンピュータ内蔵のCPU、リムーバブルディスク、ハードディスクなどの記憶手段で代用することも可能である。これにより、測定終了後にはその可搬型コンピュータのみを外部へ持ち出して上記データ処理を行うことができるので、作業性の向上などを図ることもできる。
つぎに、本実施形態において可能な種々の信号処理方法について説明する。

円二色性蛍光励起スペクトル
図10に一般的な(吸収)円二色性スペクトルの1例を示す。これは2R,3R−ビスナフトイルブタンジオールの1.78ppmのアセトニトリル溶液について測定されたものである。この試料については極めて明瞭な円二色性スペクトルが得られているが、この試料を100倍希釈した17.8ppbの溶液については、図11に示すように、最早ノイズの大きさと同じ程度の信号レベルとなって、このスペクトルの測定の主たる目的である構造解析に用いることはできない。

本実施形態の1つである円二色性蛍光励起スペクトル測定装置は、このような希薄な試料について、なお解析に利用できる高感度な円二色性蛍光励起スペクトルに利用できる。図12に示すのは、先の17.8ppbの試料について得られた円二色性蛍光励起スペクトルで、S/N比で比べて(吸収)円二色性スペクトルの20倍の感度が得られている。因みに本実施形態によらない図4に示したような従来の円二色性蛍光励起スペクトル測定で測定した円二色性蛍光励起スペクトルに比べて、S/N比で6倍の感度が得られている。

ところで、この図12に示した円二色性蛍光励起スペクトルでは、その縦軸が規格化されておらず、試料間の直接比較ができない。これを補うために、本実施形態では以下に述べる方法によって円二色性蛍光励起スペクトルの大きさの規格化を行うことが可能である。
すなわち、前述のように直流アンプ24で分離された直流成分信号s3も、そのままA/D変換器32、I/O装置34を介してCPU42に入力されるが、CPU42に入力された直流成分信号s3は、そのまま全蛍光励起スペクトル(同図(b)参照)として記憶手段44に入力され記憶される。

全蛍光励起スペクトルを取得後、CPU42は記憶手段44に適宜アクセスして全蛍光励起スペクトルを読み出す。また、CPU42は読み出された全蛍光励起スペクトルを所定の波長区間(本実施形態においては220nm〜260nm)で積分して0でない積分値Iを得る。
積分値Iを取得後、CPU42は、記憶手段44に適宜アクセスして円二色性蛍光励起スペクトルを読み出す。また、CPU42は読み出された円二色性蛍光励起スペクトルを前記積分値Iで割り算することにより、この円二色性蛍光励起スペクトルの大きさを規格化する。

全蛍光励起スペクトルを所定の波長区間で積分して得られた積分値Iは、0でない1つの値であるから、ノイズとは無縁である。この積分値Iで円二色性蛍光励起スペクトルを割り算することにより、ノイズを増やさずに、その大きさを規格化することができる。これにより、種々の試料間で円二色性蛍光励起スペクトルを適正に比較することもできる。

円二色性蛍光励起スペクトルの利点
円二色性蛍光励起スペクトルの縦軸を規格化する方法として、従来、左右の円偏光励起による蛍光強度の差を、蛍光強度の和で割った量を蛍光検出円二色性スペクトルとして測定していた。本実施形態でも、この蛍光検出円二色性スペクトルを、先の円二色性蛍光励起スペクトルを全蛍光励起スペクトルで割ることにより、得ることができる。この間の関係は下記数1で表される。

ただし、ΔAは吸収円二色性スペクトル、Sは蛍光検出円二色性スペクトル、ΔFは円二色性蛍光励起スペクトル、Aは左右の円偏光に対する試料物質の吸収係数の平均値である。また、通常の円二色性分散計において(吸収)円二色性スペクトルΔAは、試料を透過した左右円偏光の強度差ΔIと、その平均値Iより下記数2で表される。

いずれのスペクトルでも(吸収)円二色性スペクトルと比較して1桁ないし2桁高い感度を得ることができる。これは、例えば通常の紫外可視分光光度計と分光蛍光光度計で期待される感度の違い、あるいは、HPLCにおいて紫外可視検出器と蛍光検出器で期待される感度の違い(至適な試料についてはいずれも蛍光のほうが1桁ないし3桁高い感度が得られる)に匹敵するものであり、微少量しか用意することができないような試料の測定に、莫大な恩恵をもたらすものである。

さらに、これら2者を比較すると、円二色性蛍光励起スペクトルはΔF、蛍光検出円二色性スペクトルはSを測定することになる。この蛍光検出円二色性スペクトルは、前記数1のように後で係数を演算してΔAに直すことができ、その解釈方法も確立しているという利点がある。
一方、Sは全蛍光強度(直流成分信号s3)で割り算している分だけノイズが大きくなり、さらに蛍光のない波長領域では分母が0(もちろん分子も0)となるので、0/0を演算する結果、ノイズがより拡大してスペクトルの質が著しく低下するという宿命をもっている。

これに対し、円二色性蛍光励起スペクトルは、ΔFは割り算しない分だけノイズが極めて少なく、蛍光のない波長領域でもノイズが拡大することなく、スペクトルの質が確保され、試料の量に対する感度の点では、顕著な利点を有する。
また、蛍光性夾雑物や、キラリティに関与しない蛍光性官能基の影響を受けないので、その分、選択性の点でも有利である。
また、蛍光で検出することで選択性も向上し、化合物の特定の部位に特異的な解析が可能となる。さらに、生のままでは蛍光をもっていない物質についても、蛍光の官能基を化学的に導入することにより、高感度に測定解析する道が開けたこととなり、その効果は極めて大きい。

【発明の効果】
以上説明したように本発明にかかる円二色性蛍光励起スペクトル測定装置によれば、サンプルセルと蛍光測定手段の間に集光系を介さず、かつ、該サンプルセルと蛍光測定手段を近接させたので、試料から放射された蛍光の大部分を蛍光測定手段に入射させることができる。これにより蛍光の集光効率の向上を図ることができるので、検出感度の向上を図ることもできる。
なお、前記装置において蛍光波長選択手段として励起光の散乱光の波長域の光を選択的に吸収して除去することが可能なカットオフフィルタを用いることにより、回折格子を用いた分光器の場合に比較し光のロスが少なくなるので、検出感度の向上を図ることができる。
また、前記装置においてサンプルセルを透過した光の通過コース上に設けられ、該透過光を除去する光トラップを備えることにより、励起光の散乱光が蛍光測定手段に入射してしまうのを防ぐことができるので、該励起光の散乱光による測定妨害を大幅に低減することができる。
さらに、透過光が反射して逆の円偏光となって再び試料の励起に関わり、正当な円二色性蛍光励起スペクトルの取得を妨害することを防ぐことができる。
また、前記装置においてサンプルセルを間に挟み蛍光測定手段の反対側に、その鏡面をサンプルセルに向けて設けられ、該サンプルセル内の試料から放射された蛍光のうち、蛍光測定手段の反対方向に放射された蛍光を入反射して蛍光測定手段へ導く凹面鏡を備えることにより、あるいは、該凹面鏡に代わり、実質的に前記サンプルセル外周面のほぼ全周に亘り、その鏡面をサンプルセルに向けて設けられ、該サンプルセル内の試料から放射された蛍光を入反射して、サンプルセルと蛍光測定手段の間の光路中へ導く内面反射型の反射鏡を備えることにより、蛍光測定手段方向とは異なる方向へ放射された蛍光でも集光して蛍光測定手段へ入射させることができるので、検出感度の向上を図ることができる。
また、前記装置において、サンプルセル内の試料の高さと励起光波長選択手段からの励起光束のビーム径とがほぼ一致するように、高低調節手段によりサンプルセルの高さを調節することにより、サンプルセル内に入れる試料量を励起光束のビーム径とほぼ同一の高さの必要最低限の量で蛍光測定を行うことができるので、試料の利用効率の改善を図ることができる。
さらに、前記装置においてサンプルセルは円筒形よりなるものを用い、例えば励起光束のビームの形状ととほぼ同様の円形である円筒形セルの端部より励起光束を入射させることにより、角形よりなるものを用いた場合に比較し、励起光束の照射されない試料部分を大幅に低減することができるので、必要最低限の試料量で蛍光測定を行い、試料を利用効率の向上を図ることができると共に、角形のものに比較し励起光束が入射されるセル窓板の歪みが少ないので、得られるスペクトルの質的な向上を図ることもできる。
