【発明の属する技術分野】
本発明は、化学物質を検出するフロースルー型装置及びこれを用いる電気化学発光免疫分析装置に関する。

【従来の技術】
試料中の興味ある検査対象物質を検出する方法として、検査対象物質と特異的に結合する磁性微粒子を用いる方法がある。この方法では、例えば、免疫測定等、試料中の検査対象物質の含有量が極微量であり、検査対象物質を選択的に検定する必要がある場合に、検査対象物質を磁性微粒子に結合させ磁力を用いて選り集めている。一方、自動化に適した化学測定システムとしてフローセルを用いるフロースルー方式があり、この方式では、フローセルをクリーニング・コンディショニングする初期段階と、測定段階との間の迅速な交代が可能である。このクリーニング・コンディショニングする初期段階では、流体力学的なクリーニングが利用される。流体力学的なクリーニングでは、フローセルに洗浄溶液を流通させるフローセル内部をクリーニングするので、フローセル内部には常時液体が存在し空気に曝されていないため、測定に際し外部因子の混入がない。また、装置を分解することなしに複数の異なる試料を測定でき、測定のスループット向上が期待できる。従来技術の一例として、特公平7−6912号に記載のエレクトロケミルミネセンス測定装置がある。

【発明が解決しようとする課題】
フローセルを用いるフロースルー方式の化学分析装置では、フローセルのクリーニング(洗浄)の効率の向上が、測定精度の向上の点で非常に重要であり、クリーニングが充分でない場合、次の測定に於いて過去の測定の履歴が影響を及ぼし、測定精度を低下させるという問題がある。特に、異なる試料の測定を連続して行なう自動分析装置では、異なる試料の間の検体混入(キャリーオーバー)が起き、正確な測定ができないという問題がある。

しかし、検査対象物質を磁性微粒子を用いて捕捉した後に検出(測定)をフロースルー方式により行なう場合、測定後の磁性微粒子のクリーニングが非常に困難である。磁性微粒子は、フローセルの僅かな隙間、境界部、及び傷等に入り込み、そのままクリーニングにより除去されずにフローセル内に残留する場合があり、フローセルのクリーニングに必要な時間が長くなり一検査あたりの所要時間が増大し、更にクリーニングに必要な緩衝液の総量が著しく増加するという問題がある。

本発明の目的は、上記従来技術の問題を解決し、フローセルの洗浄を効率良く行ない洗浄時間の短縮を図り、異なる試料の間の検体混入(キャリーオーバー)が少なしく、分析精度の向上、分析所要時間の短縮、計測のスループット向上、低コスト化を実現するフロースルー型検出器及びこれを用いる電気化学発光免疫分析装置を提供することにある。

【課題を解決するための手段】
本発明のフロースルー型検出器は、流路を具備するフローセルと、流路をはさみ対向する電気音響変換器と、各電気音響変換器から発する音響波の定在波の節の位置を変化させる制御を行なう制御手段と、流路内に気泡を生成する手段(水溶液の電気分解により気泡を生成する電極が、流路の底面に形成される)とを有し、定在波の節の位置を変化させて気泡の位置を変化させることに特徴がある。

上記制御手段は、各電気音響変換器を独立して駆動する交流電圧に関し、次の何れかの制御を行なう。(1)交流電圧の周波数を、周期的に変化させること、(2)交流電圧の周波数を、正弦的に周期的に変化させること、(3)交流電圧の周波数を、予め定めた2つの周波数の間で線形的に周期的に変化させること、(4)交流電圧の位相差を制御すること、(5)交流電圧の位相差を一定に制御すること、(6)交流電圧の間の位相差を周期的に変化させること、(7)交流電圧の位相差を線形的に周期的に変化させること、(8)交流電圧の周波数差を制御すること、(9)交流電圧の周波数差を、予め定めた一定の周波数の差を有し周期的に変化させること。即ち、周波数変調、位相変調、うなりによる変調等により電気音響変換器を駆動制御する駆動制御部を具備し、定在波の節の位置を変化させる。

また、各電気音響変換器を駆動する交流電圧は、矩形波であることが好ましく、各電気音響変換器と上記流路の間の部材を、高分子ゴム、特に6フッ化プロピレンとフッ化ビニリデンの共重合体で構成するのが好適である。

更に、本発明のフロースルー型検出器は、作用電極、参照電極が配置されたセル下基板と、対向して電気音響変換器が配置された中間基板と、対向電極が配置されたセル上基板とが、積層されて構成される流路を具備するフロースルー型検出器と、各電気音響変換器を独立に変調して電圧駆動制御する駆動制御部とを有し、各電気音響変換器から発する音響波の定在波の節の位置を変化させて気泡を捕捉し、作用電極の面に物理的な力を作用させて洗浄を行なうことに特徴があり、各電気音響変換器を駆動する交流電圧が、矩形波であることが好適である。

【発明の実施の形態】
(第1の実施例)
図1は、本発明の第1の実施例のフロースルー型検出器の構成を示す図である。本実施例のフロースルー型検出器のフローセルは、作用電極11、及び参照電極12が配置されたセル下基板111と、電気音響変換器15が配置された中間基板部材112−1と電気音響変換器16が配置された中間基板部材112−2からなる中間基板112と、対向電極13−1、13−2が配置されたセル上基板113とが、図1に示すz方向に積層されて構成される。中間基板部材112−1、112−2は、フッ素ゴム平板を金型により打ち抜いて形成し、フローセル流路を形成すると共にガスケットとして使用しても良い。溶液は、図1に示すy方向の矢印114よりフローセル内に流入し、セル下基板111、中間基板112、及びセル上基板113により形成されるフローセル流路を通り、矢印115の方向に流出する。フローセル流路を通る溶液に接する、作用電極11、参照電極12、対向電極13−1、13−2の電極の電位はポテンシオスタット14により制御され、電気音響変換器15、16は、駆動制御部(回路)17により各々独立に制御され駆動される。

参照電極12は、フローセル内の液相の電位と同電位となる電極である。ポテンシオスタット14は、参照電極12と作用電極11の電位差が予め設定された電位差となる様に、対向電極13−1、13−2に電圧を印加して液相の電位を変化させる機能を持つ。電気音響変換器15、16から発生する音響波の進行方向が、作用電極面と平行であり、音響波の進行方向とフローセル内の溶液の流れ方向とが交差する様に、電気音響変換器15、16を、図1に示す様にx方向で対向させて配置する。駆動制御部17は、関数発生器、増幅器を含み、予め設定したプログラムにより、電気音響変換器16、15を駆動する交流電圧波形(V1(t)、V2(t))を制御して、フローセル流路に照射する音響波の状態を変化させる。一般に交流電圧波形、V1(t)、V2(t)は、V01、V02を振幅、f1、f2を周波数、α1、α2を位相、tを時間変数として、(数1)〜(数2)で与える。

(第2の実施例)
次に、本発明のフロースルー型検出器その洗浄方法、特に作用電極11の表面に存在する物質の除去方法について詳細に説明する。先ず、フローセル流路に電解質溶液、例えば、水酸化カリウム水溶液、水酸化ナトリウム水溶液等のアルカリ溶液を導入する。次に、液相の電位に対する作用電極の電位を、水の電気分解電位以上に設定し、作用電極11の表面で水溶液の電気分解を行ない、気泡を発生させる。電気音響変換器16、15を駆動し、位相、周波数、及び振幅が等しい音響波を発生させ、作用電極11の表面に接する液相に音響波の定在波を形成させる。即ち、電気音響変換器16、15を駆動する交流電圧波形を、(数1)〜(数2)に於いて、V01=V02=V0、f1=f2=f0、α1=α2=αとして、(数3)とする。

図2は、フローセル流路に形成される定在波の状態を説明する図である。図2(A)は、フローセル流路の作用電極11の表面を含む断面図であり、図2(B)は図2(A)に示す点線21に於ける作用電極11の表面に垂直な断面図である。図2に於いて、22−1、22−2は、電気音響変換器15、16により液相に形成される定在波を横波表示で表わす。図2では、音響波の定在波の節の位置を、点線23−1、23−2、23−3、23−4で示し、定在波の隣り合う節の間の距離24は、音響波の波長の1/2に相当する。電気分解で生じる殆どの気泡の直径は10μmを超えるので、定在波の周波数が500kHz以上となる様にすると、電気分解で生じる気泡は、定在波の音圧の低い領域に集まる。

本実施例では、定在波の節の部分であって作用電極11の表面近傍の領域25(図2に示す白丸)に於いて、定在波の音圧が極小となる。この結果、作用電極11の表面で発生する気泡は、白丸25の領域に集まり捕捉され、更にフローセル流路中に形成される定在波の音圧により、作用電極11の表面方向(−z方向)への圧力を受ける。この圧力により作用電極11の表面へ押し付けられる気泡は、作用電極11の表面に存在する物質に対して、物理的な力を及ぼす(作用する)。フローセル内の溶液を流れ方向26(y方向)に流す時、気泡は、定在波の節23−1〜23−4に沿って作用電極11の表面を方向27(y方向)に流れる。この結果、気泡は、作用電極11の表面に存在する物質を方向27(y方向)に押し流す様に働き(作用を与え)、作用電極11の表面の物質を効率よく除去できる。

(第3の実施例)
図3は、「周波数変調法」による音響波の定在波の節の位置(気泡が捕捉される位置)の変化を説明する図である。図4は、音響波の定在波の節の位置の変化の詳細を説明する図であり、図2(A)に示す点線21に於ける作用電極11の表面に垂直な断面図である。電気音響変換器16、15を駆動してフローセル流路内を照射する音響波の周波数を、周期的に変化させる「周波数変調法」について、以下説明する。本実施例では、定在波を形成している音響波の周波数を、音響波の周波数と比較し十分大きな周期Tで変化させて、音響波の定在波を保持したまま定在波の節の位置を変化させる。

以上説明した様に本実施例では、定在波の節(左第1節、右第2節)とこれらに捕捉された気泡は、x方向の範囲443、444を1秒間に(1/T)回だけ往復移動し、作用電極11の表面の範囲443、444に存在する物質を押し流す。周波数変調法では、定在波中間面の位置が重要である。音響波の周波数が増加する場合は、音響波の定在波の節の位置(気泡が捕捉される位置)は、常に定在波中間面に近づく方向に移動する。また、音響波の周波数が減少する場合は、音響波の定在波の節の位置(気泡が捕捉される位置)は、常に定在波中間面より離れる方向に移動する。

図6は、「周波数変調法の改良法」により定在波の節の空間的分布を任意に変化させることを説明する図であり、図2(A)に示す点線21に於ける作用電極11の表面に垂直な断面図である。第3の実施例では、(数5)に示す周波数変調した交流電圧波形により、同一の位相で電気音響変換器16、15を駆動したので、2つの音響波の位相が一致しており、定在波中間面は常に振幅が最大な部分である。例えば、第3の実施例に示す周波数変調法の改良として、(数12)〜(数13)に示す周波数変調した交流電圧波形V1(t)、V2(t)の位相α1、α2に位相差を(α2−α1)を持たせ、音響波の定在波の節の空間的分布を任意に変更できる。
