光波コヒーレンス断層画像化法では、奥行き空間分解能は原理的にコヒーレンス長LC の半分で与えられる。よって、如何にコヒーレンス長LC の短い光源を実現するかが問題となる。これに対して、米国のMITのグループは、レーザ光源にモードロックの技術を用いて、1μm程度のコヒーレンス長を有する光源を実現させているが、実用面では装置が大型、高価、操作が困難などの問題がある(文献〔5〕参照)。そこで、従来、複数の光源を組み合わせてコヒーレンス長を短くする試みがなされてきた(文献〔6〕参照)。

【発明が解決しようとする課題】
しかしながら、従来の複数の光源を組み合わせてコヒーレンス長を短くしようとする試みは、光波コヒーレンス断層画像化法への応用を意図したものでないために、図2に示すようなサイドローブの問題があり、従来この問題に対しては、詳細な検討がなされていなかった(文献〔7〕参照)。

これに対して、本願発明者らはすでに多光波光源による光波断層画像測定用高空間分解能合成光源(特願2001−7501)を提案している。

本発明は、その提案された高空間分解能合成光源をレーザ結晶から得られる蛍光に応用することにより、半導体素子の限られた光学特性を超えて、さらなる高空間分解能を実現することができる光波断層画像測定用高空間分解能合成光源を提供することを目的とする。

【課題を解決するための手段】
本発明は、上記目的を達成するために、
〔1〕励起光を用いて複数の光学結晶又は活性イオンから得られるそれぞれの蛍光を励起光カットフィルターで励起光をカットし、合成された蛍光のみを用いる光波断層画像測定用高空間分解能合成光源であって、前記前記複数の光学結晶が、第1のレーザ結晶と第2のレーザ結晶からなり、前記第1のレーザ結晶と第2のレーザ結晶を直列に配置し、前記第2のレーザ結晶から得られる蛍光を、前記励起光カットフィルターで励起光をカットし、合成された蛍光のみを用いることを特徴とする。

【発明の実施の形態】
以下、本発明の実施の形態について詳細に説明する。

まず、本発明に関連する従来の技術(上記従来技術と重複するが)から順次、詳細に説明することにする。

光波コヒーレンス断層画像化法では、得られる断層データは、生体試料の屈折率分布と光源のコヒーレンス関数との畳み込み積分であるので、コヒーレンス関数において中心のメインピーク幅の半分が光軸方向の空間分解能になる。

通常は、コヒーレンス関数のメインピーク幅(半値全幅)がコヒーレンス長と呼ばれ、この1/2が深さ方向の分解能となる。

よって、光軸方向分解能ΔZ、コヒーレンス長lC 、スペクトル幅Δλなどは次式で示される(図1参照)。

一方、コヒーレンス関数にサイドローブがあり、それが無視できない場合は、バックグランドになり画像の分解能を低下させる。そこで、複数の波長の異なる光源の光波を合成させた場合、中心のメインピークの幅を狭くする事は容易であるが、一般に大きなサイドローブが発生してしまう。

よって、複数の波長の異なる光源の光波を合成させた場合、コヒーレンス長LC とサイドローブ強度IS を同時に減少させるパラメータ条件が重要である。

これに対して、従来、複数の光源の波長、スペクトル幅をパラメータとして検討が行われたが十分ではない。そこで、実現可能な半導体発光素子でのそれぞれの光波の強度を制御することによって、コヒーレンス長LC の減少と同時にサイドローブ強度IS の抑圧を可能とする方法を提案し、すでに特許出願を行った。

最適パラメータは、I1 :I2 :I3 =1.00:2.17:0.65,λ1 =700nm,λ2 =730nm,λ3 =770nm,LC1=14.7μm,LC2=12.4μm,LC3=13.2μmと得られた。また、Δλは式(1)より求められる。

コヒーレンス関数は図3に示すように、サイドローブが抑制されつつコヒーレンス長も単独の光源のコヒーレンス長15μmに比べて7.3μmと約49%に低減されており、本発明の有効性が示されている。具体的には、図4に示すように、それぞれの光源の中心波長、スペクトル幅、強度が最適条件を満たすようにセットして、多光波光源からの出力を光ファイバーカップラーAで結合し、単一のファイバー合成光源とする。

以上に対して、本発明は、多光波光源の各光源にレーザ結晶から発せられる蛍光を用いることである。蛍光をOCTの光源に用いる例は、文献〔1〕に示すようにすでにあるが、複数の蛍光光源を用いて空間分解能を高める方法はまだ報告されていない。これの大きな特徴は、中心波長が生体に有用な近赤外領域にあり、スペクトル幅が半導体素子に比較して、非常に広く発光効率も良いことである。これにより、高空間分解能化が実現される。また、昨今の全固体レーザ技術の進展に伴って小型化が容易であり、コスト的にも実用化に向けて大きな問題はない。

文献〔2〕では、各レーザ結晶での波長可変領域が示されており、生体に対して比較的減衰が少ない近赤外領域をカバーしているのが分かる。ここで、代表的なレーザ結晶、Ti:Sapphire(Ti:サファイア)とCr:Forsterite(Cr:フォーステライト)について、数値検討結果を述べる。

それぞれの蛍光は、レーザ光源として一般的であるNd:YAGの第二高調波である532nmの光波で励起され、Ti:Sapphireでは、中心波長755nm、波長幅(半値全幅)161nmでコヒーレンス長3.11μm、Cr:Forsteriteでは、中心波長1035nm、波長幅(半値全幅)300nmでコヒーレンス長は3.14μmとなり、奥行き方向分解能はその半分となる。

これに対して、図5にTi:SのみとTi:SとCr:Fの強度比を1:1にした場合のコヒーレンス関数の光路差依存性を示した。後者の方が同じピーク値に関わらず全体の幅が狭くなっているのが分かる。

また、図6にTi:SのみとTi:SとCr:Fの強度比を2.2:1にした場合のコヒーレンス関数の光路差依存性を示した。図5と同様に同じピーク値に関わらず後者の方が全体の幅が狭くなり、空間分解能が改善されているのが分かる。このシミュレーション結果と実験結果が対応することは、本願発明者らが論文で実証している(文献〔4〕参照)。

具体構成例を以下に述べる。レーザ結晶は、母体結晶に吸収・蛍光放出のようなレーザ発振の中心的な役割を果たすTi,Cr,Ndなどの活性イオンをある濃度で含むものである。それぞれのレーザ結晶は、その吸収スペクトル、蛍光スペクトルで最適な励起波長、発振波長が決まっている。先の例ではSapphireとForsteriteにそれぞれTiとCrがドープされているものである。

以上、詳細に説明したように、本発明によれば、波長幅や出力で制約の大きい半導体発光素子に比べて、汎用の安価なレーザ結晶を用いた合成光源で高空間分解能化が可能になることから、コストダウン、小型・軽量化、光源の簡素化から安定化・信頼性の向上を図ることができる。
