Description
【発明の詳細な説明】
産業上の利用分野
本発明は腸内大腸菌、あるいはインフルエンザウィルス、エップシュタインバ
ーウィルス等により引き起こされる感染症を予防するために使用される感染防御
剤に関する。
従来技術とその問題点
従来、病原菌に起因する感染症の治療には抗生物質が用いられており、その効
果は大なるものがある反面、アレルギー症患者に対しては抗生物質の投与がショ
ック死等の原因となることがあるため、その使用にあたっては制限されることが
多い。
また、ウィルスによる感染症に対してはワクチンの投与が一般に行われている
が、ウィルス自体が屡々変更することがあるため、ワクチンが効果を発揮し
ないことも少なくない。特に、インフルエンザウィルスに対しては、流行するウ
ィルスのタイプが毎年異なるため、学童等を対象にした集団ワクチン投与も殆ど
効果がないと考えられている。
したがって、アレルギー症患者に対しても何らの悪影響のない、かつウィルス
の変異によって予防効果が失われないような感染防御剤の開発が要望されている
。
ところで、人乳中に非特異的な赤血球凝集阻止因子が含まれており、牛乳でも
弱いながらも上記因子と同様な活性を有することが報告されている〔サイトウ等
「アグリカルチュアル バイオロジカル ケミストリイ」(Agricultural Biolo
gical Chemistry)36,1437〜1439,(1972)〕ことから、牛乳中にも病原菌やウ
ィルスのレセプターに直接相互作用する物質の存在が期待されていた。
このような物質としては、N-アセチルノイラミン酸を含むオリゴ糖やガングリ
オシドが従来知られており、これら物質は病原菌やウィルスのレセプターと相互
作用することにより、感染を防止すると考えられる。
しかし、これら物質は分子量が小さいため、レセプターの全てと相互作用して
効果を発揮するには可成り多量を投与することが必要となるので実用的でない。
発明が解決しようとする課題
本発明は、叙上の状況に鑑みなされたものであって、腸内大腸菌、インフルエ
ンザウィルス、エップシュタインバーウィルスに起因する感染症の防御効果を有
し、しかもこれらの細菌やウィルスの変異等により同効果が失われることがなく
、かつ安全性の点で何ら問題もない感染防御剤を提供することを課題とする。
以下本発明を詳しく説明する。
発明の構成
本発明に係るこれらの腸内大腸菌あるいはインフルエンザウィルス、エップシ
ュタインバーウィルスに対する感染防御剤の特徴は、常乳(以下、牛乳という)
由来のκ−カゼインもしくはκ−カゼインをプロテアーゼ処理して得られるシア
ル酸結合ペプチドを有効成分として含有することにある。
課題を解決するための手段
シアル酸結合タンパク質は、牛乳中に存在しているシアル酸と結合している糖
タンパク質であって、例えばκ−カゼイン、ラクトフェリン等が知られている〔
コバタ「ザ・グリココンジュゲイツI」アカデミックプレス(1977),p423〕。特
に、κ−カゼインはシアル酸含量が 2g/100g以上であって、これらの腸内大腸菌
あるいはウィルスに対する感染防御効果が高い。
また、シアル酸結合ペプチドは、上記のシアル酸結合タンパク質をプロテアー
ゼで処理して得られる画分であって、特に、κ−カゼインにレンネットを作用さ
せて得られるグリコマクロペプチド(以下GMP と略記する)は、シアル酸含量が
5g/100g以上であるのでこれらの腸内大腸菌あるいはウィルスに対する感染防御
効果が特に高い。
シアル酸結合タンパク質は、公知の方法〔H.A.マッケンジー編「ミクロプロテ
イン」アカデミックプレス社〕によって牛乳から調製し得るが、κ−カゼインを
工業的に調製する方法としては、ナトリウムカゼイネートを低イオン強度下でゲ
ル濾過して分画する方法(特開昭59-91848号)等が知られている。
また、シアル酸結合ペプチドは、牛乳より公知方法で得たシアル酸結合タンパ
ク質にプロテアーゼを作用させ、得られた生成物をゲル濾過、イオン交換クロマ
トグラフィー、アフィニティクロマトグラフィー等の分画手段の一種又もしくは
二種以上を組合わせたものに付してシアル酸結合ペプチドを分取するこ
とにより得られる。
なお、上記プロテアーゼとしては、エンドペプチターゼに属するものであれば
よく、ペプシン、トリプシン、パパイン、キモトリプシン、プロナーゼ、レンネ
ット、パンクレアチン、フィシン等を例示し得る。
シアル酸結合ペプチドのうち、GMP については、κ−カゼイン、牛乳、還元乳
或いはカゼインをpHを 5.5〜6.5 に調整した後、レンネットをタンパク質当り1/
5000〜1/50(重量)の割合で増加し、30〜45℃の温度で30〜3 時間反応させ、得
られた反応液を加熱してレンネットを失活させ、40℃に冷却した後、カルシウム
を5mM 以上添加するとGMP 以外の成分が沈殿するので、上澄を分取して濃縮し、
必要に応じて脱塩後、乾燥することによりGMP を高純度で得ることができる。
本発明では、上述のようにして得られたシアル酸結合タンパク質、特にκ−カ
ゼインを腸内大腸菌、インフルエンザウィルスあるいはエップシュタインバーウ
ィルスに対する感染防御剤の有効成分とする。
これらの有効成分を前記感染防御剤として用いるには、例えばκ−カゼインで
は 2.5mg/kg/日以上投与するとよく、この投与により病原菌やウィルスによる感
染を防止し得る。
本発明における前記感染防御剤は、牛乳由来のκ−カゼインもしくはκ−カゼ
インをプロテアーゼ処理して得られるシアル酸結合ペプチドをそのまま、すなわ
ち粉末あるいは溶液状にして用いられる。また、通常の製剤、例えば錠剤、顆粒
剤、散剤等の形にして用いてもよい。
次に、本発明の感染防御効果について具体的な試験結果に基づいて説明する。
ヒト上皮細胞への大腸菌付着防止効果
試験方法:
ハートインフュージョン培地で培養した大腸菌タイプ055(ATCC 12014)並びに
タイプ0111a,111b(ATCC 29552)を108ヶ/mlとなるようにイーグル培地に懸濁し
た。
一方、イーグル培地中で継代培養しているヒト小腸由来株 407にκ−カゼイン
並びにGMP をそれぞれ0.01%、0.1%及び1%の濃度に添加、含有させた上記大
腸菌懸濁液を加え、30℃の温度に1時間それぞれ反応させた。
反応後、各々試料について顕微鏡下にて細胞(株407)に付着している菌数を、
細胞50ヶ分について計測した。
コントロールにおける菌数をXo、各試料における菌数をXsとし、付着率Aを
下記式により算出した。
A=Xs/Xo×100(%)
結果は表1に示すとおりである。
表1にみられるとおり、κ−カゼインでは 0.1%の濃度で、GMP では0.01%の
濃度で大腸菌の小腸由来ヒト上皮細胞への付着防止効果が認められた。
赤血球凝集阻止(HI)効果
試験方法:
インフルエンザA新潟株及びBシンガポール株を用い、1%の濃度になるよう
にそれぞれ溶解したκ−カゼイン並びにGMP のHI活性を常法により調べた。結果
は表2に示すとおりである。
表2にみられるように、いずれの試料も強いHI活性を示した。
エップシュタイン バーウィルス(EBV)によるリンパ球のトランスフォーメ
ーション阻止効果
健康人より採取した末梢血リンパ球(PBL)を4×105ヶ/mlとなるように、10%
牛胎児血清(FCS)を含むRPI1640 培地に懸濁した。これに、同培地で希釈したEVB
を10%(V/V)となるように添加し、さらに、試料液10μl を添加した。この混合
液を20μl ずつ96穴マイクロタイタープレートに5ウエルずつ分注し、5%炭酸
ガス環境下で37℃にて培養した。2週間後に培地を交換し、3週目に各ウエルで
トランスフォーメーションが生じているか否かについて判定した。結果は表3に
示すとおりである。
表3から分るように、κ−カゼインでは10μg/mlの濃度で、また、GMP では1
μg/mlの濃度でも、EBV によるトランスフォーメーションを阻止する効果を認め
た。
発明の効果
叙上のとおり、本発明おいて有効成分として用いる牛乳由来のシアル酸結合タ
ンパク質であるκ−カゼイン並びにκ−カゼイン由来のグリコマクロペプチドは
いずれも、腸内の細胞への大腸菌の付着防止効果、インフルエンザウィルスに対
するHI活性及びEBV によるリンパ球のトランスフォーメーション阻止効果が認め
られることから、本発明に係る感染防御剤は、腸内での大腸菌感染による下痢の
発生の予防、インフルエンザの流行に対する集団予防或いはEBV によるリンパ球
のガン化等に顕著な効果を発揮するものと考えられる。
また、本発明に係る感染防御剤の有効成分は、牛乳由来の物質である安全性の
観点からも何ら問題がなく、しかも全く無味、無臭であることから、食品に添加
して使用することも可能である。
以下に実施例を示して本発明を具体的に説明する。
実施例
有効成分として牛乳から単離したκ−カゼイン並びにκ−カゼインから分取し
たGMP をそれぞれ用い、試験動物として体重約200gのウイスター系ラット35匹を
5匹ずつ7群に分け、1群をコントロール、他の6群をさらに2分し、κ−カゼ
イン投与区並びにGMP 投与区をそれぞれ3群とし、κ−カゼインとGMP を0.1,0
.5並びに1mg/日の割合で経口ゾンデを用いてそれぞれ強制投与した。
次いで、各群に病原性大腸菌ATCC 12014の一定量を経口投与して下痢の発生率
を調べた。結果は表4に示すとおりである。
表4にみられるとおり、κ−カゼイン並びにGMP 投与群では下痢発生率が顕著
に低く、κ−カゼインでは0.5mg/日の投与で、GMP では0.1mg/日の投与で十分効
