本発明によるコロナウイルスワクチン及び/又はインフルエンザワクチンを皮内送達するための装置及び方法は、角質層を通して下層の表皮層、又は表皮層及び真皮層へと穿孔するように適合された複数のマイクロニードル(又はそのアレイ)を有するマイクロニードル部材(又はシステム)を有する皮内送達システムを含む。
いくつかの変数は、特定の活性剤に利用されるアレイの種類において役割を果たす。例えば、異なる形状(例えば、矢じり、フック、円錐形、又はワシントン記念塔に類似した形状)は、より高い活性剤充填能力を可能にする場合があるが、微小突起の長さは、貫通のためのより大きな駆動力を提供するために増加させてもよい。角質層は約10~40ミクロンの厚さを有し、微小突起は、角質層を貫通して活性剤の送達を行うのに適切なサイズ、厚さ及び形状を有しなければならない。微小突起は角質層を貫通し、基材は皮膚の表面と接触する。
出願人が所有する米国特許出願公開第20190070103号明細書の図4(A)~図4(B)、図5(A)~図5(E)に示されているように、本開示の皮内活性剤送達システムは、本体と、本体内で移動可能なピストンとを有するインパクトアプリケータをさらに含んでもよく、ピストンの表面は皮膚に対してパッチに衝突し、微小突起が角質層を穿孔する。アプリケータは、10ミリ秒以下で少なくとも0.05ジュール/cm2、又は10ミリ秒以下で約0.26ジュール/cm2、又は10ミリ秒以下で約0.52ジュール/cm2の衝撃エネルギー密度でマイクロニードルアレイを角質層に適用するように適合される。
固体コーティングを形成するために上記の微小突起部材に適用されるコーティング製剤は、生体適合性担体内に溶解され得るか又は担体内に懸濁され得る少なくとも1つの生物学的活性剤を有する液体、好ましくは水性製剤からなる。次いで、製剤を微小突起上にコーティングし、乾燥させ、滅菌し、包装する。生物学的活性剤は、インフルエンザワクチン又はコロナウイルスワクチン、例えば、SARS-Cov-2サブユニットワクチンであってもよい。

COVID-19に対する開発中のワクチンを選択(ソース:前出のKalomara)
・ワクチン抗原は、抗原に対する二糖の質量比が約0.5、1又は2:1の二糖、例えばスクロース又はトレハロースで安定化されてもよい。使用されてもよい他の二糖は、タンパク質を安定化するのに十分な量のラクトース及びマルトースである。
・コーティング厚さは、約50マイクロメートル~約100マイクロメートルの範囲である。
・抗原又は抗原の組合せを含有する水性製剤の粘度は、約50~約300cPの範囲であり得る。
・他の賦形剤としては、酒石酸、クエン酸及びヒスチジンが挙げられる。
・pH範囲は、4.4~7.4である。
乾燥コーティングされた製剤の改善された物理的安定性は、治療剤自体の貯蔵寿命又は有効期間の増加の利益を提供するだけでなく、本発明の組成物並びに製剤化及び送達方法に従って安定化されると、治療剤は、より広範囲の可能な製剤において、より多様な治療剤送達手段と共に有用になるという点で有効性を高める。
本発明の活性剤とデバイスとの組合せは、COVID-19、他のコロナウイルス及びインフルエンザに対するワクチン接種を含む様々な疾患及び状態を治療するために使用することができる。患者は、本明細書の他の箇所に記載されているアプリケータデバイスを使用することによって、約5mcg~約500mcgのワクチン/抗原を含むワクチンでコーティングされたマイクロアレイパッチを自己投与してもよい。パッチは、上腕、手首の近く、大腿、胸又は背中など、一般に平坦で過剰な毛髪がない皮膚の選択された領域に適用される。パッチ装着時間は、約1分~約30分、又は約5分~約20分、又は約10分で変化してもよい。その後、患者は、パッチを取り外してごみに捨てる。
以下に記載されるように、三価インフルエンザワクチン経皮パッチは、三価インフルエンザワクチン筋肉内(IM)注射製剤を超える3つの重要な利点、(1)保存剤を含まない、(2)室温保存、及び(3)用量節約を伴って首尾よく開発された。より重要なことに、このパッチシステムは、前臨床試験及び臨床試験において安定かつ有効であることが証明された。
材料
一価スプリットビリオンインフルエンザウイルス株抽出物を卵インキュベーションから誘導した。各一価株溶液を、方法の項で以下に記載されるように使用前にさらに処理した。スクロース(ロット番号27412A、高純度低エンドトキシングレード)及びトレハロース(ロット番号26554A、高純度低エンドトキシングレード)を、Ferro-Pfanstiehl(オハイオ州クリーブランド)から購入し、受領した状態のまま使用した。界面活性剤をいくつかの供給業者から購入し、受領した状態のまま使用した-Tween80、ロット番号58217、(ICN Biomedicals Inc.、オハイオ州オーロラ);Zwittergent3-14、ロット番号B36399(Calbiochem、カリフォルニア州サンディエゴ);Triton X100、ロット番号QC2755S4D1(89521)、(Union Carbide Corporation、テキサス州ヒューストン);Pluronic F68、ロット番号16H1147、(Sigma、ミズーリ州セントルイス)。
レオメトリー
濃縮されたコーティング製剤の粘度は、円錐平板粘度計(Brookfield Eng.Lab.、CAP2000)を使用して決定した。各測定は、70μLの液体試料を必要とした。粘度は、各液体試料についていくつかの剪断速度及びいくつかの温度で決定した。
コーティング製剤とチタン基材との間の接触角は、接触角計(Tantec Inc.、イリノイ州シャンバーグ)を使用して、金属チタンシート上に5μLの液滴を配置することによる半角測定法に基づいて決定した。
SEMを使用して、微小突起上のコーティングの形態及び配置を決定した。コーティングされたチタンアレイを、炭素両面テープでアルミニウムスタッドに接着し、SEM(日立、S-2460N)の真空チャンバ内に配置した。
出発物質、コーティング溶液及びコーティングされたアレイ中のインフルエンザHA含有量の定量化のために、一元放射免疫拡散アッセイを適合させた。この受動拡散法では、洗剤で処理した後、試料溶液及び参照ワクチンがウェルから放射状に拡散し、ゲルマトリックス中に均一に分散している特異的抗体と反応する。抗原-抗体相互作用は、抗原(HA)ウェルの周りの沈降の明確な輪によって表される。輪の直径は、平衡に達するまで増加し続ける。平衡条件下では、沈降輪の直径はHAの濃度に比例する。完全拡散後、各試料溶液及び参照ワクチンについて沈降円を測定する。試料中の赤血球凝集素含有量は、Influenza Reference Centerによって提供された国際基準に対して決定され、μg/mLで較正される。
ヘアレスモルモット(HGP)の血清から抗インフルエンザ特異的抗体の存在を検出するための間接ELISA法を開発した。以前に、オバルブミンでコーティングされたアレイで免疫したHGPにおける抗オバルブミン抗体価を決定するための間接ELISAを開発した。インフルエンザワクチンでコーティングされたアレイについて、インフルエンザワクチンで免疫したHGPからの血清のエンドポイント力価を特異的に決定するための同様のアッセイを開発した。エンドポイント力価は、非免疫化対照HGP血清(n=10)の平均ODを3標準偏差上回るODを有する免疫化HGP血清試料の、非線形回帰によって決定される逆希釈として定義される。
原材料、コーティング溶液及びコーティングされたアレイのタンパク質含有量を、Pierce(イリノイ州ロックフォード)から購入したキットを使用してBCAアッセイによって測定した。一連の段階希釈標準液をワクチン原材料から直接調製した。未知の試料を、アッセイの標準的な作業範囲内にある濃度まで水で希釈した。標準及び試料を96ウェルプレートにロードし、プレートリーダ(Molecular Devices、SpectraMax250)に入れ、30秒間振盪し、37℃で30分間インキュベートした。吸光度を562nmで測定し、標準の平均値を以下の形式の4パラメータ式に当てはめた。
いくつかの試料の総タンパク質含有量を、タンパク質標準としてウシ血清アルブミン(BSA)を使用する改変ローリーアッセイによって測定した。ローリー法は、タンパク質と銅イオンとの反応の結果として形成される青色錯体の形成、及びその後のタンパク質-銅錯体によるフォリン-チオカルトー試薬の還元に基づく。青色の強度は、試料中に存在するタンパク質の量に比例し、750nmで分光光度的に測定される。
インフルエンザワクチンHAタンパク質試料を、InvitrogenプレキャストNuPAGEゲル上でSDS-PAGEによって分離した。XCell IIブロットモジュール「Novex Western Transfer Apparatus」(Invitrogen)を使用するための説明書に従って、分離したタンパク質をPVDF膜にブロットした。ブロットしたPVDF膜を、希釈した抗HA一次抗体又は抗HA抗血清でプローブした。非特異的結合部位を、5%ミルク+0.1%Tween20を含むPBSによってブロッキングした。ウエスタンブロットを、HRPコンジュゲート化二次抗体及びAmersham PharmaciaからのECL検出試薬を使用することによって可視化した。
界面活性剤Triton-X100の濃度を、比色アッセイ及びHPLC法の2つの方法によって液体試料中で測定した。比色アッセイは、青色沈殿物を形成するコバルトチオシアン酸アンモニウムとの錯体の形成を含んでいた。次いで、沈殿物を二塩化エチレン中に抽出し、吸光度を分光光度的に測定した。HPLC法は、C4カラム及び直線アセトニトリル勾配を用いる逆相法であった。
2つの種類のTFFシステム、すなわち、Pellicon XL、再生セルロース膜(Millipore、50cm2、30kD MWCO)を備えた実験室規模のTFFシステム(Millipore、Labscale)及び0.1ft2の30kD MWCOポリエーテルスルホンPES膜を備えたより大規模なTFFシステム(Pall、Centremate(商標))を、スプリットビリオンインフルエンザ抽出物のダイアフィルトレーション及び濃縮に使用した。タンジェンシャルフロー濾過を第1の工程として使用して、一価株のHA含有量を濃縮する方法として塩及び他の低分子量種を除去した。ダイアフィルトレーションによる低分子量材料の除去のために注射用滅菌水を使用した。一価バルク中に存在するTriton X-100などの界面活性剤を効果的に除去するために、さらなるTFF洗浄工程を使用した。この洗浄工程は、1/4~10ダイアボリュームの注射用滅菌水を使用して濃縮する前のダイアフィルトレーションからなっていた。ダイアフィルトレーション及び洗浄の後、各ワクチン溶液の体積を元の体積の1/20~1/50に減少させ、HA濃度を5~10mg HA/mLに増加させた。これは、ワクチンをTFF濃縮によって効果的に濃縮することができる濃度限界であった。溶液中の不溶性粒子からの膜のファウリングによって引き起こされた可能性が最も高い背圧の増加のために、TFFによる一価株のさらなる濃縮は不可能であった(考察の項を参照)。TFFシステムからのHA濃縮物の回収率は高く、典型的には、濃縮前後のBCAタンパク質アッセイ及びSRID効力によって測定して95%超であった。TFF濃縮の後、一価株を収集し、製剤化し、次いで、HA濃度をさらに増加させる手段として凍結乾燥した。
前臨床試験のために、TFF濃縮後、一価株を20mLガラスバイアルに充填し、液体窒素で急速凍結し、マニホールド式凍結乾燥機(Virtis、25EL Freezemobile)に置いた。チャンバ圧力が定常状態(約50mTorr)に達するまで、溶液を2~5日間凍結乾燥させた。第I相材料の臨床製造のために、5mLの製剤化されたTFF濃縮物を20mLガラスバイアルに充填し、Stoppering Tray Dryer(Labconco、FreezeZone)で凍結乾燥した。再構成された凍結乾燥粉末のBCAタンパク質アッセイ及びSRIDによって決定されたように、凍結乾燥後の回収率も高かった(90%超)。
一価バルクワクチンのHA純度を、総タンパク質及び溶液中に存在する全固体に対して決定した。一価バルクの総タンパク質を、ウシ血清アルブミンを参照標準として使用したローリーアッセイを用いて測定した。次いで、試料の既知のHA含有量を測定した総タンパク質で割ることによって、総タンパク質に対するHA純度%を計算した。全固体に対するHA純度%は、一価バルクの一部を蒸発乾固させて、溶液中に存在する固体の総重量を決定し、この値を試料の既知のHA含有量に分割することによって決定した。
チタン微小突起アレイを、光/化学エッチングによって製造し、制御された製造プロセスを使用して形成した。例えば、欧州特許第0914178(B1)号明細書を参照されたい。
コーティングの製剤パラメータ
この新規な経皮微小突起パッチシステムは、微小突起アレイ上にコーティングされた固体製剤を特徴とした。したがって、コーティングプロセスを可能にする液体製剤の開発は、安定した性能向上固体製剤の前駆体であった。
各バルク溶液は、受領した状態のままでは濁っており、おそらく水不溶性脂質、脂質-タンパク質複合体及び凝集タンパク質による不溶性粒子の存在を示唆している。バルク溶液中の赤血球凝集素抗原(HA)濃度は約0.1~0.2mg/mLと低く、HA純度は様々であり、典型的には全固体(低分子量溶質、タンパク質及び不溶性粒子)の20±5%及び総タンパク質含有量(HA及び非HAタンパク質)の40±10%の範囲であった。微小突起アレイ上のHA抗原をコーティングするために、バルク溶液を再製剤化してHA濃度及び純度を高める必要がある。非HAタンパク質及び粒子は免疫学的応答に寄与する可能性があるので、緩衝液、塩及び界面活性剤、例えばTriton-X100(ワクチン製造中にウイルス粒子を分割するために使用される)を含む低分子量材料を除去することによってのみHA純度を改善することができる。低分子量種の除去は、ダイアフィルトレーションによって達成した。
30kDの膜を含むTFFシステムでは、一価バルクを最初に濃縮して、その体積を元の体積の1/20~1/50に減少させ、10ダイアボリュームの10mMリン酸緩衝液で洗浄した。しかしながら、このプロセスは、HA純度%の15%未満のわずかな増加、及びTriton-X100(625ダルトンのMW)の濃度の有意な増加をもたらした。より高分子量のTriton-X100ミセルの形成は、このプロセスがTriton-X100を効果的に除去できなかった理由である。
凍結乾燥の前に、スクロース又はトレハロースを凍結乾燥保護剤としてTFF濃縮物に添加した(1:1の凍結乾燥保護剤:HA重量比)。これらの二つの二糖安定剤の効果を、製剤を10サイクルの凍結/解凍(液体窒素によって凍結し、室温で直ちに解凍)に供することによって評価した。ELISAによって決定されるように、10サイクルの凍結/解凍の前後のHA効力は変化せず(データは示さず)、トレハロース又はスクロースによる抗原安定性の保存が示唆された。タンパク質に長期安定性を提供するために固体バイオ医薬製剤では通常、より高い重量比の凍結乾燥保護剤が必要とされるが、コーティングされた形態を微小突起の先端でサイズがコンパクトに保つために製剤の全固体含有量を制限することがより重要であり、これは微小突起先端の貫通効率にとって重要である。その後、凍結乾燥のために、スクロースをTFF凍結濃縮物に1:1のスクロース:HA重量比で添加した。得られた凍結乾燥ワクチンの固体組成物は、30+5%のHA、30+5%のスクロース、10+5%のTriton-X100、並びに30+15%の残りの画分を構成する非HA関連タンパク質及び固体粒子を含有していた。
液体コーティング溶液を調製するために、凍結乾燥した一価製剤のそれぞれを、元の凍結乾燥前の体積の1/4~1/5の注射用滅菌水で再構成して、HA濃度を40~50mg HA/mLにさらに増加させた。これにより、再構成ワクチンの微細懸濁液が得られた。次いで、再構成された一価溶液のアリコートをそれらのSRID効力値に基づいて1:1:1のHA比で組み合わせて三価コーティング溶液を生成し、各株は約14~15mg HA/mLの濃度で存在した。ここでも、三価液体コーティング製剤は、3つの重要なコーティング製剤パラメータ、すなわちワクチン濃度、粘度及び表面活性を満たすように調製された。
コーティング溶液のワクチン濃度は、各パッチを製造するのに必要な製造時間を最小限にするために努力して目標用量を達成するのに必要なコーティング通過の回数、すなわち回転ドラム上の膜への浸漬の回数を最小限にするために、可能な限り高くなるように配合された。しかしながら、コーティング溶液の粘度及び安定性は、コーティングに使用されるワクチン濃度を制限した。この実施例では、60mgHA/mL以上のHA濃度を有するコーティング溶液は、粘度が高すぎてドラム上に連続薄膜を形成できないことが分かり、コータ内の連続剪断下で経時的に凝固した。このため、HAの濃度は40~50mg総HA/mLの間で維持した。以下の表は、予備製剤化/製剤化プロセスの異なる段階における全固体に対するHA濃度及び純度をまとめたものである。

抗原、非HAタンパク質/粒子及びTriton-X100の全体的な濃度によって制御されるコーティング溶液の粘度は、コーティングプロセス中の微小突起先端上の薄膜の流れに影響を及ぼす。微小突起先端の各浸漬は、いくらかのコーティング溶液をとることができる。溶液粘度が低すぎると、微小突起先端の溶液が乾燥する前にコーティング溶液膜に滴下して戻る場合がある。溶液の粘度が高すぎると、液体の流れが遅すぎて、必要に応じて微小突起を均一にコーティングすることができない。許容可能なコーティング形態を達成するために、液体製剤の粘度範囲は0.20~1.50ポアズであることが実験的に決定された。3つのHA/スクロース(重量比1:1)濃度、50、40及び35mg/mLのインフルエンザワクチン製剤の様々な剪断速度での粘度を図2に示す。予想通り、コーティング溶液の粘度は、製剤中のHAの濃度に直接関係することが分かった。50mg/mLのHA濃度では、コーティング溶液は、剪断速度の全範囲の間にコーティングのための所望の粘度を示し、その十分に高い濃度のために最小数のコーティング通過も必要とした。
コーティング溶液はまた、微小突起先端を効果的に濡らすために適切な表面活性を示すべきである。濡れ性は、液体の表面張力及び基材の表面エネルギーに応じて、コーティング溶液が微小突起の表面上に付着、接着及び拡散する能力を測定し、接触角測定によって決定することができる。濡れ性が悪いと、流体の取込みが妨げられるか、又は不均一な局所的なコーティングが生じる。表面活性剤を含有する液体製剤は、溶液と基材との間の接触角を減少させることによって、表面張力に影響を及ぼし、表面濡れ性を改善することができる。チタン基材上の純水の接触角(80°)と比較して、コーティング製剤(HA及びスクロースの等しい重量比)は、HA濃度にかかわらず、26°~36°の範囲の接触角で良好な濡れ性を示した。HA抗原及び/又はTriton-X100は、製剤中の表面活性剤である可能性がある。さらに、Tween80、Pluronic F68及びZwittergent3-14などのいくつかの界面活性剤をコーティング製剤に添加した場合(1%まで)、チタン表面上の接触角は同じままであった(データは示さず)。この知見は、コーティング製剤が本質的に表面活性であり、コーティングプロセスに有利であろうことを再び示唆している。
しかし、コーティングに適した物理的特性にもかかわらず、50mg/mLのHA/スクロースコーティング製剤は乳白色の懸濁液である。目に見える粒子がない、この微細懸濁液は、主にナノ粒子を含有する場合がある。この懸濁液は、1ヶ月間冷蔵した後、相分離(粒子の沈降)が観察されなかったため、物理的に安定であると考えられた。さらに、溶液を7,000rpmで2分間遠心分離した後、明らかな粒子沈降はみられなかった。典型的には乳化剤(又は界面活性剤)によって安定化される安定なエマルジョン、水中油又は油中水のように、ナノ粒子の懸濁液は、Triton-X100によって安定化される可能性がある。
コーティング装置は、コーティング溶液リザーバと、コーティング溶液と接触しているステンレス鋼ドラムとを備えていた。ドラムを回転させて、コーティング製剤の連続薄膜(厚さ約100μm)を生成し、その中にチタンアレイ上の微小突起先端を浸漬した。浸漬深さを正確に制御して、微小突起の先端のみをコーティング製剤でコーティングした。微小突起の先端にコーティングされた比較的少量の製剤、製剤の高い固体含有量、及びアレイの非常に高い表面積のために、微小突起表面上の液体コーティングは、周囲条件下でコーティング後5秒未満で風乾されると予想される。コーティングされたワクチンの量は、アレイが薄膜に浸漬された回数によって制御され、BCA及び/又はSRIDによってモニタリングされた。
三価経皮送達システムの第I相臨床製造には、125~500mcg HA/mLの範囲の濃度の三種類の一価株A/ニューカレドニア(H1NI)、A/パナマ(H3N2)及びB/山東を使用した。バルクウイルス抽出物の各一価株およそ2リットルをダイアフィルトレーションし、次いで、TFF装置で10mg HA/mLに濃縮した。次いで、濃縮された一価溶液を、1:1重量比のHA:スクロースで個別に製剤化し、凍結乾燥して粉末形態にした。次いで、3つの凍結乾燥粉末を再構成し、合わせて、42mg HA/mL(各一価株に対して14mg HA/mL)の濃度の1:1:1三価コーティング溶液を生成した。このコーティング溶液は、30mcg HA三価(すなわち、一価株当たり約10μg)の目標用量をアレイ当たり最小数の浸漬でコーティングするために許容可能な粘度及び濡れ性を示した。コーティング後、許容可能なシステムを窒素パージされたヒートシール箔パウチに包装し、2~8℃で保存した。代表的なシステムを臨床バッチから選択し、SRIDアッセイによってロットリリースについて試験した。試験したすべてのシステムは、>8mcg HA/パッチのロットリリース規格を満たした。バッチ全体を通して無作為に選択した20のシステムの平均は、相対標準偏差が6%以内で、11.0mcg A/ニューカレドニア、13.3mcg A/パナマ及び12.2mcg B/山東であった。
予備製剤化プロセス全体(ダイアフィルトレーション/濃縮、凍結乾燥及び再構成)を通して抗原の安定性を維持することが最も重要である。すべての加工応力以外に、Triton-X100濃度が0.1~0.3mg/mLから3~5mg/mLに10倍を超えて増加したため、コーティング製剤中に存在する高濃度Triton-X100のHAの抗原性への影響が懸念される(TFFプロセスの項を参照)。
前臨床免疫原性データは、ヒトと同様の皮膚構造を有するヘアレスモルモットから得た。この有効性動物モデルにおける正の免疫応答は、パッチ製剤を用いた第1相ヒト試験に入るという本発明者らの決定を促した。経皮経路による投与(2つのパッチデザインについて1株当たり7~8mcg HA)は、ヘマグルチン阻害)(HAI)セロコンバージョン率に関して筋肉内(IM)経路による投与(1株当たり15μg HA)よりも優れていた(表2)。これは、50%未満の抗原でさえ、パッチによって誘導された免疫応答(一次免疫後28日目)がIM注射によるものと同等又はそれよりも優れていたことを示唆している。
表2:前臨床免疫原性結果の要約
マイクロニードルパッチによって投与された三価インフルエンザ抗原の有効性と、標準的な筋肉内経路を介した三価ワクチンの送達とを比較するために、第1相臨床試験(単一施設、非盲検、無作為化)を実施した。この試験では、健常な男性及び女性(18~40歳、約30名の対象/群)を、各抗原株(A/ニューカレドニア(H3N2)、A/パナマ(H3N2)、B/山東)、12μg/各)でコーティングしたマイクロニードルパッチ又は市販のIM送達ワクチン(各株15μg)のいずれかで処置した。投与後、パッチを5又は15分間装着した。

三価インフルエンザワクチン経皮パッチは、前臨床及び第I相ヒト臨床試験において首尾よく開発され、有効であることが証明された。TFFシステムによるダイアフィルトレーション/初期濃度、凍結乾燥、及びコーティング用の高HA濃度(約40~50mg HA/mL)溶液を調製するための再構成からなる独自の予備製剤化プロセスを確立した。この予備製剤化プロセスは非常に効率的であり、抗原の損失はほとんどなかった(プロセス収率85%超)。予備製剤化プロセスに従って調製された後続のコーティング溶液は、チタン微小突起の先端へのコーティングのための許容可能な物理的及び化学的安定性を有するように最適化された。パッチ製剤は、現在利用可能な製剤を超える3つの重要な利点、すなわち保存剤を含まない、室温保存及び用量節約を示した。上記の経皮パッチの成功に基づいて、当業者は、そのような技術が、(類似又は異なる賦形剤を使用して)コーティング溶液中に製剤化され、治療有効量で微小突起アレイに適用され得る任意のワクチンに拡張され得ることを理解するであろう。
抗原が合成ペプチドであることを除いて、コロナウイルスワクチンパッチは、一般に実施例1に従って調製される。この非限定的な例では、そのような合成ペプチドは5つのペプチド抗原である。5つのペプチドを1:1:1:1:1の比で混合する。次いで、混合物をペプチド当たり約50~約100mcgの用量で微小突起パッチ上にコーティングする。混合物は、約pH3~約9.5で製剤化される。5つのペプチドは以下の通りである。

Claims (29)
- ヒト患者の角質層を貫通又は穿孔するように適合された複数の微小突起を含む皮内送達システムであって、前記微小突起は、先端から基部まで測定された各微小突起の長さの約10%~80%を覆う固体製剤コーティングをその上に有し、前記コーティングは、治療有効量のワクチンを含み、前記ワクチンの少なくとも95%が、前記システムを前記ヒト患者の前記角質層に適用した後約20分以内に前記システムから放出される、皮内送達システム。

ii.その上に固体製剤コーティングが配置されており、前記コーティングは治療有効量のワクチンを含む、前記微小突起、
iii.約10μm~約500μmの幅及び約30~約70度の先端角を有する、前記微小突起
を含む皮内送達システムを提供する工程、及び
b.前記微小突起を前記患者の皮膚の選択された領域に適用する工程を含み、
