Abstract
Description

i.前記一連のステップ(a)~(c)の反復を少なくとも1回実施するステップであって、ステップ(a)において使用される細胞の集団が、ステップ(c)で単離された細胞および/もしくはその子孫の細胞を含む、またはからなるステップとを含み、随意により、前記少なくとも1回の反復の前に、先行するステップc)で単離された細胞を、前記細胞の数が先行するステップa)における細胞の数と実質的に同等となるまで培養する。

i.クロストリジウム神経毒によって切断可能である指標タンパク質、および
ii.クロストリジウム神経毒に対して結合親和性を有する受容体および/またはガングリオシド(好ましくは、受容体およびガングリオシド)
(b)指標タンパク質の切断を示す細胞を同定するステップと、
(c)ステップb)で同定された細胞を単離するステップと、

- 指標タンパク質がC末端側標識を含み、細胞では全長指標タンパク質が容易に分解されないが、その切断後に、得られたC末端側断片が分解され、C末端側断片の分解がC末端側標識の分解をもたらし、指標タンパク質の切断が、細胞(複数可)をクロストリジウム神経毒と接触させた後のC末端側標識からのシグナルを測定することによって決定され、好ましくは、クロストリジウム神経毒との接触の間またはその後の(例えば、ステップ(a)の間またはその後の)C末端側標識の減少が、指標タンパク質の切断を示し、より好ましくは、クロストリジウム神経毒との接触の間またはその後の(例えば、ステップ(a)の間またはその後の)C末端側標識の減少によって、指標タンパク質の切断が確認される、前記の請求項のいずれか一項に記載の方法。
- 接触後、細胞を溶解し、得られた細胞溶解物を抗体と接触させ、ウエスタンブロットを実施する、前記の請求項のいずれか一項に記載の方法。
- ステップa)の細胞が、クロストリジウム神経毒に対して結合親和性を有する受容体をコードする外因性核酸および/またはクロストリジウム神経毒に対して結合親和性を有するガングリオシドの発現を提供する外因性核酸、好ましくは、前記受容体をコードする外因性核酸および前記ガングリオシドの発現を提供する外因性核酸を含む、前記の請求項のいずれか一項に記載の方法。
- 第1の接触ステップ(例えば、ステップa))の前に、細胞中に、クロストリジウム神経毒受容体もしくはそのバリアントもしくは断片であってクロストリジウム神経毒に結合する能力を有するもの、および/または、ガングリオシド合成経路の酵素もしくはそのバリアントもしくは断片であってこのような酵素の触媒活性を有するもの、をコードする外因性核酸を導入すること含む、前記の請求項のいずれか一項に記載の方法。
- ステップa)の細胞が、ガングリオシド合成経路の酵素(またはそのバリアントもしくは断片であってこのような酵素の触媒活性を有するもの)をコードする外因性核酸を含む、前記の請求項のいずれか一項に記載の方法。
- ステップa)の細胞が、グルコシルセラミドシンターゼ、GalT-I、GalNAcT、GM3シンターゼ、GD3シンターゼ、GT3シンターゼ、ガラクトシルセラミドシンターゼ、GM4シンターゼ、GalT-II、ST-IVまたはST-Vまたはそのバリアントもしくは断片であってこのような酵素の触媒活性を有するもの、好ましくは、GD3シンターゼまたはそのバリアントもしくは断片であってGD3シンターゼの触媒活性を有するもの、より好ましくは、GD3シンターゼをコードする外因性核酸を含む、前記の請求項のいずれか一項に記載の方法。
- 細胞(例えば、ステップ(a)の)が、前記核酸を欠く(好ましくは、前記外因性核酸を欠く)細胞と比較した場合に、より多くの受容体および/またはガングリオシドを発現する(例えば、ガングリオシド合成経路の酵素またはそのバリアントもしくは断片であってこのような酵素の触媒活性を有するものを介して)、前記の請求項のいずれか一項に記載の方法。
- 細胞が、クロストリジウム神経毒に結合する能力を有するタンパク質受容体またはそのバリアントもしくは断片を発現または過剰発現するように遺伝子改変されている、前記の請求項のいずれか一項に記載の方法。
- 受容体が、SV2またはシナプトタグミン(またはそのバリアントもしくは断片であってクロストリジウム神経毒に結合する能力を有するもの)である、前記の請求項のいずれか一項に記載の方法。
- 受容体が、SV2(またはそのバリアントもしくは断片であってクロストリジウム神経毒に結合する能力を有するもの)である、前記の請求項のいずれか一項に記載の方法。
- 受容体が、SV2AまたはSV2C、好ましくは、SV2A(またはそのバリアントもしくは断片であってクロストリジウム神経毒に結合する能力を有するもの)である、前記の請求項のいずれか一項に記載の方法。
- 受容体が、SV2AまたはSV2Cの第4の管腔ドメインである、前記の請求項のいずれか一項に記載の方法。
- 核酸(好ましくは、外因性核酸)が、トランスフェクションによって導入される、前記の請求項のいずれか一項に記載の方法。
- 細胞を、(i)クロストリジウム神経毒受容体もしくはそのバリアントもしくは断片であってクロストリジウム神経毒に結合する能力を有するもの、および/または(ii)ガングリオシド合成経路の酵素もしくはそのバリアントもしくは断片であってこのような酵素の触媒活性を有するものを発現するように遺伝子改変することによって、組換え細胞が生成される、前記の請求項のいずれか一項に記載の方法。
- 選択または単離された細胞が、細胞中に存在する指標タンパク質の20%以上が、切断生成物に変換されているものである、前記の請求項のいずれか一項に記載の方法。
- ステップ(a)および(b)が、それ以前のステップ(b)の反復回において選択された細胞を使用して少なくとも1回繰り返される、請求項2から36のいずれか一項に記載の方法。
- ステップ(a)の各反復で、細胞を、それらが先行する反復回のステップ(a)の間に接触したものよりも少ないクロストリジウム神経毒と接触させる、前記の請求項のいずれか一項に記載の方法。
- ステップ(a)および(b)が繰り返される前に、先行するステップ(b)の反復回において選択された細胞を、このような細胞の数が、最初の組換え細胞の集団中に存在した数とおよそ同一となるまで培養し、および/または前記少なくとも1回の反復の各々の前に、指標タンパク質の切断を示す細胞を単離するという先行するステップにおいて選択された細胞を、このような細胞の数が、最初の細胞の集団中に存在した数とおよそ同一となるまで培養する、請求項37または38に記載の方法。
- 細胞(例えば、ステップaの)が、神経細胞、非神経細胞、神経内分泌細胞、胚性腎臓細胞、乳癌細胞、神経芽細胞腫細胞または神経芽細胞腫-神経膠腫ハイブリッド細胞、好ましくは、非神経細胞である、前記の請求項のいずれか一項に記載の方法。
- 細胞(例えば、ステップaの)が、神経芽細胞腫細胞または神経芽細胞腫-神経膠腫細胞である、前記の請求項のいずれか一項に記載の方法。

- 細胞(例えば、ステップaの)が、NG108細胞である、前記の請求項のいずれか一項に記載の方法。
- ガングリオシドが、GM1a、GD1a、GD1b、GT1bおよびGQ1bから選択され、好ましくは、細胞(例えば、ステップaの)が、GM1a、GD1a、GD1b、GT1bおよび/またはGQ1bを発現または過剰発現するように遺伝子改変されている、前記の請求項のいずれか一項に記載の方法。
- ガングリオシドが、GD1a、GD1bおよびGT1bから選択され、好ましくは、細胞(例えば、ステップ(a)の)が、GD1a、GD1bおよび/またはGT1bを発現または過剰発現するように遺伝子改変されている、前記の請求項のいずれか一項に記載の方法。
- ガングリオシドが、GD1bおよびGT1bから選択され、好ましくは、細胞(例えば、ステップ(a)の)が、GD1bおよび/またはGT1bを発現または過剰発現するように遺伝子改変されている、前記の請求項のいずれか一項に記載の方法。

- クロストリジウム神経毒がBoNT/Aである、前記の請求項のいずれか一項に記載の方法。

- ステップa)(例えば、細胞の集団をクロストリジウム神経毒と接触させるステップまたは指標タンパク質を発現する組換え細胞をクロストリジウム神経毒と接触させるステップ)において、クロストリジウム神経毒が、約0.0001~約100,000pM、約0.0001~約50,000pM、約0.0001~約20,000pM、0.0001~約10,000pM、約0.0001~約1,000pM、約0.0001~約500pM、約0.0001~約300pM、約0.0001~約100pM、約0.0001~約10pMまたは約0.0001~約1pMの濃度で存在する、前記の請求項のいずれか一項に記載の方法。
- 請求項1から51のいずれか一項に記載の方法によって生成された集団に由来する細胞。
- 請求項1から51のいずれか一項に記載の方法によって得ることができる細胞集団。
- 修飾されたまたは組換え神経毒の活性を決定するアッセイであって、
(a)請求項52または53に記載の細胞を修飾または組換え神経毒と、細胞において野生型クロストリジウム神経毒のプロテアーゼドメインが指標タンパク質を切断することを可能にするのに十分である条件下および時間の間、接触させることと、
(b)指標タンパク質の切断に起因する生成物の存在を決定することと
を含む、アッセイ。 - クロストリジウム神経毒製剤の活性を特性決定する、または治療的(および/または美容的)使用のためのクロストリジウム神経毒製剤を同定するin vitro法であって、
a.請求項1から51のいずれか一項に記載の方法によって調製された細胞集団または請求項52もしくは53に記載の細胞集団を提供することと、
b.前記細胞集団をクロストリジウム神経毒製剤と接触させることと、
c.クロストリジウム神経毒製剤との接触後の指標タンパク質の切断のレベルを、クロストリジウム神経毒製剤との接触前の切断のレベルと比較することと、
d.(i)接触後の指標タンパク質の切断のレベルが増大される場合に、クロストリジウム神経毒製剤を治療的(および/または美容的)使用に適していると同定することもしくは(ii)接触後の指標タンパク質の切断のレベルが増大される場合に、活性の存在を同定すること、または
e.(i)接触後の指標タンパク質の切断のレベルが増大されない場合に、クロストリジウム神経毒製剤を治療的(および/または美容的)使用に適していないと同定することもしくは(ii)接触後の指標タンパク質の切断のレベルが増大されない場合に、活性の非存在を同定することと
を含む、in vitro法。
