- 重鎖可変領域および軽鎖可変領域を含む抗原結合性タンパク質もしくは完全ヒト抗CD38抗体、またはそれらの抗原結合性断片であって、前記重鎖可変領域および前記軽鎖可変領域が、それぞれ、配列番号3および4(例えば、本明細書において、3H10m1という);それぞれ、配列番号5および4(例えば、本明細書において、3G8m1という);それぞれ、配列番号6および4(例えば、本明細書において、3E3m1という);それぞれ、配列番号7および2(例えば、本明細書において、3G3という);それぞれ、配列番号9および2(例えば、本明細書において、3E11という);それぞれ、配列番号10および2(例えば、本明細書において、3H10という);それぞれ、配列番号11および12(例えば、本明細書において、3H10Nという);それぞれ、配列番号13および12(例えば、本明細書において、3H10NSという);それぞれ、配列番号1および4(例えば、本明細書において、3E10という);または、それぞれ、配列番号3および12(例えば、本明細書において、3H10m1gという)のアミノ酸配列を含み、必要に応じて、前記重鎖可変領域の54~56位のNGRモチーフが、SGRモチーフに置き換わっている、抗原結合性タンパク質もしくは完全ヒト抗CD38抗体、またはそれらの抗原結合性断片。
- Fab断片を含む、請求項1から4のいずれか一項に記載の抗原結合性断片。
- 一本鎖抗体を含み、前記重鎖可変ドメイン、前記軽鎖可変ドメインが、ペプチドリンカーで互いに接続されている、請求項1から4のいずれか一項に記載の抗原結合性断片。
- IgG1、IgG2、IgG3またはIgG4クラスの抗体である、請求項1から4のいずれか一項に記載の抗原結合性タンパク質または完全ヒト抗CD38抗体。
- ヒトおよびカニクイザル由来のCD38タンパク質に結合する、先行する請求項のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片。
- CD38タンパク質を発現する細胞に結合する、先行する請求項のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片。
- CD38タンパク質を発現するヒト骨髄腫細胞に結合する、先行する請求項のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片。
- 先行する請求項のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片、および薬学的に許容される賦形剤を含む医薬組成物。
- 請求項3に記載の抗体の重鎖可変領域を含む第1のポリペプチドをコードする第1の核酸であって、前記抗体の重鎖可変領域の前記アミノ酸配列が、配列番号3、5、6、7、9、10、11または13のアミノ酸配列と少なくとも95%の配列同一性を有する、第1の核酸、および請求項2に記載の抗体の軽鎖可変領域を含む第2のポリペプチドをコードする第2の核酸であって、前記抗体の軽鎖可変領域の前記アミノ酸配列が、配列番号4または12のアミノ酸配列と少なくとも95%の配列同一性を有する、第2の核酸。
- 請求項1から10のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片をコードする1つまたは複数の核酸。
- 請求項12に記載の第1および第2の核酸に作動可能に連結している1つまたは複数のプロモーターを含む1つまたは複数の発現ベクター。
- 請求項13に記載の1つまたは複数の核酸に作動可能に連結している1つまたは複数のプロモーターを含む1つまたは複数の発現ベクター。
- 請求項14または15に記載の1つまたは複数の発現ベクターを持つ宿主細胞。
- 前記抗体の重鎖可変領域を含む第1のポリペプチド、および前記抗体の軽鎖可変領域を含む第2のポリペプチド、または前記抗原結合性タンパク質、抗体もしくは抗原結合性断片を調製するための方法であって、請求項16に記載の宿主細胞の集団を、前記第1のポリペプチドおよび前記第2のポリペプチド、または前記抗体もしくは抗原結合性断片を発現するために好適な条件下で培養するステップを含む方法。
- 前記宿主細胞の前記集団から、発現した前記第1のポリペプチドおよび発現した前記第2のポリペプチド、または発現した前記抗体もしくは抗原結合性断片を回収するステップをさらに含む、請求項17に記載の方法。
- CD38を発現する細胞を死滅させるための方法であって、(i)エフェクター細胞の集団を、(ii)CD38を発現する標的細胞の集団と、(iii)請求項1から10のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片の存在下、前記CD38を発現する細胞を死滅させるために好適な条件下で接触させるステップを含み、必要に応じて、in vitroの方法である方法。
- CD38過剰発現またはCD38陽性がんと関連する疾患を有する対象を処置するための方法であって、前記対象に、請求項1から10のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片を含む有効量の治療用組成物を投与するステップを含む方法。
- CD38陽性がんを有する対象を処置するための方法であって、前記CD38陽性がんが、B細胞白血病、B細胞リンパ腫またはB細胞骨髄腫を含み、前記対象に、請求項1から10のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片を含む有効量の治療用組成物を投与するステップを含む方法。
- CD38の発現と関連する疾患を有する対象を処置するための方法であって、前記CD38の発現と関連する前記疾患が、多発性骨髄腫(MM)、バーキットリンパ腫(BL)を含む非ホジキンリンパ腫(NHL)、B慢性リンパ性白血病(B-CLL)、全身性エリテマトーデス(SLE)、BおよびT急性リンパ性白血病(ALL)、急性骨髄性白血病(AML)、慢性リンパ性白血病(CLL)、びまん性大細胞型B細胞リンパ腫、慢性骨髄性白血病(CML)、ヘアリー細胞白血病(HCL)、濾胞性リンパ腫、ワルデンストレームマクログロブリン血症、マントル細胞リンパ腫、ホジキンリンパ腫(HL)、形質細胞性骨髄腫、前駆B細胞リンパ芽球性白血病/リンパ腫、形質細胞腫、巨細胞骨髄腫、形質細胞性骨髄腫、重鎖骨髄腫、軽鎖またはベンス・ジョーンズ骨髄腫、リンパ腫様肉芽腫症、移植後リンパ増殖性障害、免疫調節障害、関節リウマチ、重症筋無力症、特発性血小板減少性紫斑病、抗リン脂質症候群、シャーガス病、グレーブス病、ウェゲナー肉芽腫症、結節性多発動脈炎、シェーグレン症候群、尋常性天疱瘡、強皮症、多発性硬化症、抗リン脂質症候群、ANCA関連血管炎、グッドパスチャー病、川崎病、自己免疫性溶血性貧血、および急速進行性糸球体腎炎、重鎖病、原発性または免疫細胞関連アミロイド症、ならびに意義不明の単クローン性ガンマグロブリン血症であり、前記方法が、前記対象に、請求項1から10のいずれか一項に記載の抗原結合性タンパク質、抗体または抗原結合性断片を含む有効量の治療用組成物を投与するステップを含む、方法。
