Description

i) 成熟ナイーブB細胞を対象から単離する段階、ならびに
ii) Toll様受容体(TLR)アゴニスト、および/または免疫調節性サイトカインを用いて、エクスビボでB細胞を刺激する段階

Bリンパ球としても知られるB細胞の、任意の供給源が、採取目的のために使用され得る。当業者に公知であるように、そのようなB細胞は、骨髄、脾臓、リンパ節、血液、または、B細胞の供給源である他の同種組織に由来し得る。B細胞の好ましい供給源は、骨髄および血液である。好ましくは、自家B細胞か、または同種B細胞か、または異種B細胞が採取される。
第1日
a. ドナーのアフェレーシス産物が受領され、そして無菌性判定、細胞の計数、生存性判定、およびフローサイトメトリーのためにサンプリングされる。
b. 産物は、低温で一晩保管される。
第2日
a. 産物は、冷却装置から取り出され、十分に撹拌され、そして周囲の温度と平衡化するため30分間放置される。試料は、無菌性判定、細胞の計数、生存性判定、およびフローサイトメトリー(CD20を有するDuraCloneパネル)のために取り出される。
b. 標準的なCliniMacs手順にしたがって、血小板の洗浄除去が実施される。
c ビーズが添加され、そしてインキュベーションが4度で実施される点を除いて標準的なCliniMacs手順にしたがって、振とう機上で30分間インキュベートされる。
d. ビーズとのインキュベーション後、冷却された(4度)媒体を用いて、1回の抗体洗浄が実施され、そして産物は、標準的なCliniMacs手順にしたがって、CliniMacs LSカラムにロードされる:
e. CliniMacsエンリッチメント1.1プログラムを用いて、分離が実施される。
f. CD19が富化された標的画分は、細胞の計数、フローサイトメトリー、安定性判定、および無菌性判定のためにサンプリングされる。
g. CD19が枯渇した(非標的画分)は、細胞の計数およびフローサイトメトリーのためにサンプリングされる。
単離されたら、本明細書に記載されるようなTLRアゴニストかもしくは免疫調節性サイトカインの1種または複数種へと、B細胞を曝露することによって、該B細胞は処理または刺激され得る。そのようなエクスビボでの刺激を用いる、IL-10産生Breg細胞の産生は、本明細書に記載される方法および治療戦略に有用である。
投与される細胞の数は、処置される患部の面積または体積に関連し得、かつ送達方法に関連し得る。
本明細書に記載されるB細胞は、患者への、たとえば本明細書に記載される疾患または異常を有するヒト患者などへの投与のために、ビヒクル中に組み込まれ得る。B細胞を含む薬学的組成物は、当技術分野において公知の手法を用いて調製することができる。たとえば、そのような組成物は、たとえば、生理学的に許容される担体、賦形剤、または安定剤(Remington: The Science and Practice of Pharmacology 22nd edition, Allen, L. Ed. (2013);参照により本明細書に組み入れられる)を用いて、かつ所望の形態において、たとえば水溶液の形態において、調製され得る。
この実施例は、アイソバリック標識多重化プロテオミクスを用いた、創傷治癒に対するB細胞の分子レベルでの効果の、大規模な解析を示す。
以下の材料および方法が、この試験に利用された。
創傷治癒試験は、7~9週齢の雄の野生型C57Bl6/Jマウス(Jackson Laboratories)において実施された。雄のWT C57Bl6/Jは、B細胞単離のためのアイソジェニックなドナーとして使用された。動物は、実験室での標準的な管理条件下で、20~23度の範囲の温度、12時間:12時間の明:暗周期において、餌および酸性化水を自由にとれるようにして維持された。すべての動物の取り扱いは、実験動物の人道的管理に関する公衆衛生局の規範(Public Health Service Policy on Humane Care of Laboratory Animals)にしたがって実施され、かつマサチューセッツ総合病院(Massachusetts General Hospital)の施設内動物実験委員会(Institutional Animal Care and Use Committee)によって承認を受けた。使用する動物の数を減少させるための、および動物の苦痛を最小限にするための、あらゆる取り組みがなされた。
マウスの脾臓は、氷冷EasySep(商標)緩衝液(STEMCELL Technologies)中に採取され、該緩衝液は、リン酸緩衝生理食塩水(PBS)中に2% ウシ胎児血清(FBS)および1 mM エチレンジアミン四酢酸(EDTA)を含んでいた。脾臓は、40 μmのセルストレーナーを通過させて機械的に解離させ、そして脾細胞懸濁液は、市販の細胞単離キット(STEMCELL Technologies)を製造者の使用説明書にしたがって用いた免疫磁気分離により、B細胞またはT細胞の負の選択に関して処理された。
背部皮膚を通る全層切除創が、以前に記述されたように作製された(Wang et al. (2013) Nat Protoc. 8(2):302-9.)。手短に述べると、マウスは、ケタミン(100 mg/kg)およびキシラジン(10 mg/kg)の混合物を用いて麻酔され、そして背部皮膚は、剃毛および除毛された。鎮痛は、皮下注入された0.08 mg/kg ブプレノルフィンを用いて、術前に施された。背部皮膚を広げ、そして5 mmのバイオプシーパンチを折り曲げた皮膚に貫通させて、背中の両側に、2つの対称な創を作製した。それぞれの創は、およそ20 mm2の当初面積を有していた。7 mmの内径を有するシリコンスプリント(Sigma-Aldrich)が、Vetbond組織接着剤(3M)を用いて、創の周囲に取り付けられた。スプリントが取り付けられた創はその後、Tegaderm(商標)透明被覆材(3M)を用いて覆われた。細胞懸濁液含有PBSか、または等量のPBS溶液単独(生理食塩水対照)は、手動ピペットを用いて、創面に直接的に適用された。各マウスはまた、背部皮膚の下に、等用量のB細胞または生理食塩水を用いた2か所の局所皮下注入を受けた。処置を受けた、創および皮下部位のそれぞれは、15~20 x 106個のB細胞含有20 μl PBSの投与を受けた。
組織のサンプリング。全層切除創は、上述のように、マウスの背部皮膚に作製された。20 μlの生理食塩水中に懸濁された2 x 106個の細胞とした、精製B細胞の溶液か、または生理食塩水対照のいずれかで、創は処置された。0日(およそ10分)、1日、4日、および10日という規定の期間後に、マウス(条件1つにつき、n = 3~5匹)を安楽死させ、そして創縁および皮下層を含む創領域が切除され、そして液体窒素中で急速凍結され、その後、溶解まで-80度で保管された。

損傷の0日後(インタクト)、1日後、4日後、10日後、および16日後に採取された創バイオプシーは、4% 緩衝パラホルムアルデヒド中において、24~48時間にわたり4度で固定され、その後1M スクロース溶液中で、さらに24~48時間にわたり4度で凍結保護処理され、そして組織凍結メディウム(tissue freezing medium)(Electron Microscopy Sciences)中に包埋された。創面を通る横断切片は、クリオスタット(Leica Biosystems)を用いて10 μmの厚さで切断され、そしてSuperFrost Plus Goldスライド(Fisher Scientific)上に解凍マウント(thaw-mount)された。抗原の免疫組織化学的検出のため、切片は、トリス緩衝生理食塩水(TBS)、pH 7.4を3回交換することによって洗浄され、その後透徹され、そして、5% ウシ血清アルブミン、5 % FBS、および0.3 % トリトンX-100を含むTBSを用いて、室温で1時間インキュベーションすることにより、ブロッキングされた。切片はその後、ブロッキング溶液中で希釈された以下の一次抗体と、4度で一晩インキュベートされた:APCコンジュゲートラット抗マウスCD45R/B220(クローンRA3-6B2; BioLegend, Inc.)、PEコンジュゲートラット抗マウスCD31(クローンMEC 13.3; BD Biosciences)、Alexa Fluor(登録商標)488コンジュゲートマウス抗チューブリンβ3(クローンTUJ1; BioLegend, Inc.)、Alexa Fluor(登録商標)488コンジュゲートラット抗マウスF4/80(クローンBM8; BioLegend, Inc.)、PEコンジュゲートラット抗マウスCD11b(クローンM1/70; BioLegend, Inc.)、ウサギポリクローナル抗Ki67(Abcam)、およびウサギモノクローナル抗活性型カスパーゼ3(クローンC92-605; BD Pharmigen)。未結合の一次抗体は、TBS中で、それぞれ5分ずつ3回リンスすることによって除去された。コンジュゲート型でない一次抗体が使用された場合には、抗原性部位は、ブロッキング溶液中で1:200に希釈されたAlexa Fluor 488(登録商標)コンジュゲートF(ab')2ヤギ抗ウサギIgG(Thermo Fisher Scientific)と、切片を2時間にわたり室温でインキュベートすることにより、可視化された。切片は、室温で3分間の、2 μg/mlの4', 6-ジアミジノ-2-フェニルインドール二塩酸塩(DAPI; Sigma Aldrich)含有PBSとのインキュベーションによって、対比染色された。切片は、TBS中で7分間、3回洗浄され、そしてフルオロマウント(Novus Biologicals)を用いて包埋された。抗体対照は、組織切片とアイソタイプ抗体とのインキュベーション、および二次抗体が可視化に使用された場合には一次抗体の省略を、含んでいた。対照試料においては、非特異的なシグナルは検出されなかった。染色された組織切片は、20x、40x、および63xの対物レンズを備えるZeiss LSM 710レーザー走査型顕微鏡(Carl Zeiss)を用いてイメージングされた。共焦点画像は、Zenソフトウェア(Carl Zeiss)を用いて、0.1~0.7 μm/ピクセルの解像度、および0.5~2.2 μmの光学的厚さにおいて取得された。
創傷治癒の経時変化のエンドポイントにおける創バイオプシーは、10 mmのバイオプシーパンチを用いて採取され、そして4% パラホルムアルデヒド含有PBS中で24~48時間にわたり4度で固定されて、その後試料は、濃度勾配をつけたエタノールおよびキシレンでの洗浄によって脱水され、そしてパラフィン中に包埋された。創面を通る横断切片は、5 μmの厚さで切断され、そして顕微鏡スライド上にマウントされた。連続切片は、ヘマトキシリンおよびエオシンで染色され、そしてコラーゲン繊維を可視化するためにマッソン・トリクローム染色法で染色された。染色されたスライドは、Aperio CS2スキャナー(Leica Biosystems)を用いて、0.25 μm/ピクセルの解像度でデジタル化された。デジタル化されたスライドは、処置条件について知らされていない実験者による、組織再生のスコア付けのために使用された。
関心対象の細胞マーカーの発現に対する、B細胞適用の時間依存性の効果は、検査された細胞集団それぞれに分けて、SPSS 23(IBM Corporation)における線形混合効果モデリングを用いて評価された。分散安定化のため、各試料および検査された各マーカーについての、ゲーティングされた細胞の割合は、解析の前に、ロジット(オッズの対数)変換された。3元配置(または、あてはまる場合には4元配置)完全実施要因計画を用いて、生存時間(18、45、または93時間)、環境(創、皮下、またはB細胞のみ氷上)、マーカー、および、あてはまる場合には、条件(B細胞での処置または生理食塩水)を固定因子として含有させた。技術的反復物(technical replicate)(実施日)および生物学的反復物(biological replicate)(マウス/試料ID)は、ランダム効果として含有させた。固定因子のレベル間の事後対比は、ダン-シダック法を用いて、多重比較に関して調整された。* p < 0.05; ** p < 0.01; *** p < 0.001; **** p < 0.0001。
B細胞の適用は、創の分子微小環境において複雑な変化を引き起こす
創全体の溶解物のプロテオミクス解析により、全試料および全処置にわたって、最大で9125個のタンパク質が同定された。処理条件間であってかつ全タイムポイントにわたる解析に関し、全試料(n = 動物30匹)中に存在するタンパク質のみが検討された。これは、全部で3809個のタンパク質をもたらした(図12)。
生理食塩水で処置された創(対照、通常の創傷治癒)における、またはB細胞での処置後の創における、機能的ファミリーごとのタンパク質の、経時的な平均的発現は、図2A~2Hに示される。この解析は、タンパク質発現のインデューサーや阻害剤というよりは、ホメオスタティック作用物質としての、B細胞の全体的効果を示す。B細胞の適用は、損傷および治癒の過程で通常減少するかまたは増加するかいずれかであるタンパク質の発現を一定のレベルに維持すること、正常な治癒において観察される炎症ピークを有意に減少させること、抗アポトーシス因子(矢印)の減少および酸化ストレス保護物質の減少を防ぐこと、ならびに増殖を増大させることと関連しており(図2A~図2B)、抗酸化ストレス保護物質の低下および細胞増殖の低下を減少させること、ならびに細胞遊走を低レベルに維持することと関連しており(図2C~図2D)、リモデリングおよび二次皮膚構造に関連するタンパク質を一定のレベルに維持することと関連しており(図2E~図2F)、対照において損傷の初期に観察される、タンパク質分解およびオートファジーのレベルを減少させること、ならびに治癒の後期における血管新生および神経再生に関連するタンパク質のレベルを増加させることと関連していた(図2G~図2H)。
急性創傷治癒における、B細胞適用のインビボ評価についての実験パラダイムは、図3に示される。野生型C57Bl6マウスの背部皮膚に、全4か所の全層創が作製され、そしてアイソジェニック動物から精製された成熟ナイーブB細胞が、創面に直接的に適用された。対照動物は、生理食塩水の適用を受けた。損傷無しの同様の微小環境を作り出すために、内部対照としてB細胞または生理食塩水対照もまた、インタクトな皮膚の下へと皮下注入された。規定の生存期間後に、創、または未損傷の皮膚組織は採取され、解離され、そしてフローサイトメトリー解析のために処理された。右側の散布図は、それぞれの処置カテゴリー由来の細胞懸濁液の、典型的な分布を示す。創試料は、白血球の特徴的な流入(中空の白色矢印)を示し、これは未損傷の組織においてはほとんど存在しない。B細胞は、典型的にはいずれの位置においてもほとんど存在しないが、実験的適用後には、該細胞を容易に多数検出することができる(赤色の塗りつぶされた矢印)。
さまざまな条件間、およびさまざまなタイムポイント間での、B細胞と、創環境の細胞との両方の変化を評価するため、試料はフローサイトメトリーを用いて解析された。B細胞で処置された創および対照の創の細胞懸濁液の、フローサイトメトリーによる本発明者らのゲーティング戦略および解析は、図4に示される。生細胞は、以下の3つの主要なカテゴリーへとゲーティングされた:B細胞(CD19+/B220+リンパ球)と、好中球、単球およびマクロファージ、樹状細胞、ならびにT細胞の混合物を含む非B細胞白血球(CD140a-/B220-白血球)と、線維芽細胞(CD140a+/B220-)。これらの細胞カテゴリーは、活性化のマーカー、およびサイトカイン産生に関して評価された。
創の微小環境への規定の曝露期間後に創面から回収されたB細胞における、活性化マーカーのおよび重要なサイトカインのダイナミクスは、図5に示される。B細胞は、インビボで創ニッチに曝露されたか、または未損傷の皮膚下に注入された(同等の位置における対照)。同じ継続期間にわたり、単離直後から氷上で維持された対照B細胞は、比較のために示される。18時間、2日、4日、および10日を含む期間の後に、サイトカインの細胞内での滞留を誘導するため、創は、4時間にわたってブレフェルジンAで処置された。B細胞はその後、組織を切除および解離することによって回収され、そして、表面マーカーおよび細胞内サイトカインの両方について、フローサイトメトリーによってさらに特徴付けされた。創の微小環境に曝露されたB細胞は、複数の免疫調節性サイトカインを一過性に上方制御し、ピークは適用の2日後である。TGFβおよびIL-6を含むいくつかの免疫調節性サイトカインは、第4日において上昇したままであり、かつIL-10は、第10日まで上昇したままである。N = 動物3~6匹 / 群。
創の微小環境に曝露されたB細胞、皮下対照に曝露されたB細胞、または氷上で維持されたB細胞(曝露無し)における、各マーカーについての平均値の集約としてのヒートマップは、図6に見出される。創に存在する浸潤非B細胞白血球の凝集体における、活性化マーカーのおよび重要なサイトカインのダイナミクスは、図7に示される。全体的に見て、B細胞が創に存在する場合、浸潤白血球では、抗炎症性サイトカインであるIL-10、TGFβ、およびIL-35の産生はより多く、かつ炎症誘発性サイトカインであるTNFαおよびIL-2の産生はより少なかった。この影響は、損傷およびB細胞適用の4日後に最も強く現れ、かつ10日後まで持続した。N = 動物3~6匹 / 群。
図11に示されるように、創傷治癒における、再生に関する外来B細胞の機能には、機能的なTLRシグナル伝達、およびIL-10産生が必須の要素である。全層切除創(治癒の第6日のものがここに示される)は、共通のTLRシグナル伝達アダプターであるミエロイド系分化因子88(myeloid differentiation factor 88)(MyD88)を欠くB細胞か、IL-10を欠くB細胞か、または対照としてWT B細胞を用いて、第0日に処置された。生理食塩水もまた、内部対照として試験動物それぞれに包含させた。WT B細胞は、WT動物において第2~3日まで創閉鎖を一貫して促進した一方で、MyD88-/- B細胞と、IL-10-/- B細胞は、生理食塩水の適用と同様に、創閉鎖に関する恩恵を示さなかった。
この実施例は、マウスTBIモデルにおいて、外部より適用されたB細胞が、損傷後のパフォーマンスを有意に改善したことを示す。
以下の材料および方法が、この試験において利用された。
損傷時のBリンパ球の適用により、CCI後の認知機能の回復が改善する
全部で63匹のマウスが試験を完了し、1匹は死亡した。ローターロッドアッセイにおいて、Bリンパ球を用いた実質内処置は、有意な保護効果を有していた(図15A)。すべての損傷群は、偽損傷動物よりも劣るパフォーマンスを示した一方で、B細胞で処置された損傷マウスは、T細胞または生理食塩水で処置された損傷群よりも優れたパフォーマンスを示し(群に関して、p < 0.01)、かつそれらのパフォーマンスは、B細胞で処置された偽損傷のものと比べて差異は無かった。興味深いことに、連続したトライアルにより、CCI + B細胞群におけるパフォーマンスが、偽損傷対照群の両方と同じく、持続的に改善することが示され、これは、これらの群において手続き学習が生じたことを示唆する。B細胞で処置された偽損傷マウスが、より優れたパフォーマンスを示した(p < 0.01)トライアル3を除き、B細胞で処置されたCCI群と、偽損傷マウスとの間で、有意な差異は存在しなかった。対照的に、損傷時に、生理食塩水かまたは200万個のT細胞のいずれかで実質内処置を受けたCCIマウスは、B細胞で処置されたCCI損傷動物、または偽損傷動物のいずれと比較しても、パフォーマンスの日ごとの改善が最小であることを示し、かつ、落下までの潜時は有意に短いものであった(p < 0.0001)。T細胞の投与を受けたCCI損傷群と、生理食塩水の投与を受けたCCI損傷群との間で、統計学的に有意な差異は存在しなかった。
B細胞での処置と関連して観察された、行動上の改善が、神経病理学的相関を有するかどうかを評価することを目的として、行動評価を受けたすべての動物の脳が、組織学的検査のために、損傷の35日後に採取された(図18)。CCI群における、脳組織へのダメージの解析は、CCIを受けた全マウスにおいて、損傷エリアのキャビテーションを示した一方で、偽損傷群は、脳組織の喪失を示さなかった(図18A)。すべての群にわたる、損傷体積の定量分析(図18B)は、予想されたように、CCIの有意な影響を示した(p < 0.0001)。生理食塩水で処置された損傷群(p < 0.001)、またはT細胞で処置された損傷群(p < 0.0001)と比較して、B細胞で処置されたCCIマウスでは、脳組織の喪失は有意に低下していた。T細胞の投与を受けたCCI損傷群と、生理食塩水の投与を受けたCCI損傷群との間で、損傷体積において有意な差異は存在しなかった。脳全体の連続切片における、損傷面積の分布の解析(図18C)は、処置条件の間における最も著しい差異が、主に海馬を含むレベルである、損傷の後部2/3において観察されたことを示した。実際、標的を絞った体積測定は、生理食塩水で処置されたCCI対照、またはT細胞で処置されたCCI対照のいずれと比較しても、B細胞で処置されたCCIマウスは、損傷を受けた半球において、有意に大きい割合の補填された海馬組織を有していたことを示した(p < 0.0001; 図18D)。
外部より送達されたB細胞が、実質内注入後にインビボで持続するかどうかを決定するため、本発明者らは、CAGプロモーター下でホタルルシフェラーゼを構成的に発現する、精製されたB細胞を使用した。ルシフェラーゼは生物学的半減期が短いため、連続的に産生されていないと検出できない。ルシフェラーゼ発現細胞が死ぬと、シグナルは迅速に消失するので、このアッセイは、実験によって導入されたB細胞の、長期間の生存性を決定するために使用することができる(Zinn et al. (2008) ILAR J 49, 103-115)。外来の生存B細胞のみが光シグナルを生成するので、この方法は、細胞が数100個に減少するまで、細胞の経時的な生存性および持続性を非侵襲性に追跡することを可能にする。結果は、検査されたすべての動物において、注入されたB細胞が、適用後速やかに明確に検出可能であったことを示した(図20A)。頭部における全光量子束の定量分析は、適用された細胞内で活性な酵素活性が、当初ゆるやかに増加し、第3日~第7日にピークに達し、そしてその後、第7日の後で急速に減少し、注入の17日後までに検出不可能なレベルに達したことを示した(図20B)。これらの結果は、外来B細胞が、インサイチューでおよそ2週間という限定された寿命を有するとの見解を支持する。
この試験は、損傷後の構造的および機能的アウトカムを調節するための、前臨床TBIモデルにおけるBリンパ球の直接適用の使用を説明するものであり、これは、本発明者らの知る限り初めてとなる。CCIの際に、精製された(>95%)成熟ナイーブB細胞の単回の実質内送達を行うことで、損傷後の学習障害および記憶障害を有意に減少させることができ、かつ脳組織の喪失量を減少させることができることを、本発明者らは見出した。この結果は、これまで未知であった、脳挫傷モデルにおける内在B細胞の保護効果を示唆する。
この実施例は、ALSの標準的なマウスモデルであるSOD1G93Aマウスにおける、B細胞の静脈内(i.v.)投与の安全性および有効性を例証する。
ALSにおける、B細胞免疫療法の有効性を評価するため、十分に確立された、ALSのB6.SOD1G93Aトランスジェニックマウスモデルを、計画および結果の解釈に関して発表されているガイドラインにしたがって、本発明者らは使用した(Galgano (2017) Cell Transplant 26, 1118-1130; Nordstrom et al. (2014) Ann Neurol 75, 374-381)。ヒトSOD1のG93A変異型のトランスジェニック発現を有するマウスは、ヒトにおけるALSに類似した表現型を示す。該マウスは、片方または両方の肢が進行性に麻痺していき、脊髄からの運動ニューロンの喪失に起因する完全麻痺を有することとなる。トランスジェニックマウスはまた、短命でもある。神経変性症状は、典型的には12~14週齢の頃に出現し始め、そしてマウスは、およそ20~24週齢で死亡する。したがって、このモデルは、急速で悪性である、該疾患の進行を示す。該モデルは非常に安定であってかつ再現性を有しており、この神経変性障害に関する治療上の選択肢の評価を可能にする。
以下の材料および方法が、この試験において使用された。
以下の結果は、すべての動物試験から収集されたデータを要約する。試験は、最後のトランスジェニックSOD1動物を安楽死させた、出生後第150日に終了した。
観察可能な(行動上の、および表現型上の)有害作用(体重減少、アパシー、体調不良、立毛、円背位)は、ナイーブB細胞(細胞およそ2億個/kgの用量で)の投与を受けた対照ノンキャリア動物においては見出されなかった。対照動物は、B細胞での処置とは無関係に、試験期間を通して体重が増加し続けた(図23)。さらに、トランスジェニックSOD1-G93A動物においては、細胞の注入処置に起因し得るそのような有害作用は、症状の発症(処置の第72~90日)前の期間においては観察されなかった。
a. ピーク体重(図24に示される)。ピーク体重は、個々の動物の測定された体重が、その後で連続的な減少を示したタイムポイントとして、定義された。この情報は、生存時間分析を実施するために使用されたものであり、該分析において、ピーク体重の時点は「生存」を表す。対照ノンキャリアマウスは体重減少を示さないため、トランスジェニックSOD1-G93A動物のみがこの分析に使用された。
ALSのトランスジェニックSOD1-G93Aマウスモデルを用いたこの試験において、連続した10週間にわたりマウスへと週1回投与されたものである、精製された同種成熟ナイーブB細胞の静脈内注入は、以下と関連性を有していた:生理食塩水で処置された対照と比較して、(i) 症状の発症における、統計学的に有意な28日間の遅延、(ii) 生存期間の有意な延長、および(iii) 処置を受けた動物の腰髄に存在する、病的な、死んだ、または死につつあるニューロンの、有意な減少。処置に関連する、観察可能な有害な副作用は、細胞200,000個/グラムの用量(またはB細胞200億個/Kgの用量)でB細胞の注入を受けた、野生型動物においてもトランスジェニック動物においても、示されなかった。
治療的有効量のB細胞を含む組成物が、たとえば、本明細書に記載される組成物の任意のものなどが、パーキンソン病を有する対象に投与され得る。パーキンソン病の処置は、治療的B細胞(たとえば、Breg細胞)を、パーキンソン病についての適切な動物モデル(たとえば、Bobela W. et al. Overview of mouse models of Parkinson's disease. Curr Protoc Mouse Biol. (2014)を参照されたい)に投与すること、および当業者に公知の手法にしたがって治療有効性をモニタリングすることによって、本明細書に記載される方法を用いて評価され得る。応答をモニタリングするための方法は、運動機能、疼痛、神経炎症、および黒質ニューロンの死の評価を含む(たとえば、Peng Q. et al. The Rodent Models of Dyskinesia and Their Behavioral Assessment. Front Neurol. (2019)を参照されたい)。
他の神経変性疾患は、B細胞(たとえばBreg細胞)を適切な動物モデルに投与することによって、本明細書に記載される方法を用いて評価され得る。さらなる神経変性疾患は、以下を含む:アルツハイマー病、慢性外傷性脳症(CTE)、前頭側頭型認知症、ハンチントン病、乳児神経軸索ジストロフィー、進行性核上性麻痺、レビー小体型認知症、脊髄小脳失調症、脊髄性筋萎縮症、および運動ニューロン疾患。

本明細書に記載されるB細胞は、炎症性障害または免疫性障害を処置するために投与され得、これはたとえば、嚢胞性線維症、心血管疾患(たとえば、冠動脈疾患または大動脈弁狭窄症)、円錐角膜、球状角膜、変形性関節症、骨粗鬆症、肺動脈性肺高血圧症、網膜色素変性症、または関節リウマチなどである。これらの炎症性障害または免疫性障害の処置は、治療的B細胞(たとえば、Breg細胞)を適切な動物モデルに投与すること、および当業者に公知の方法にしたがって治療有効性をモニタリングすることによって、本明細書に記載される方法を用いて評価され得る。
