Abstract
Description
hGHの成長アゴニストから成長アンタゴニストへの変換には、hGHの120番目の位置で、ネイティブなグリシンからアラニンを除く任意のアミノ酸への単一のアミノ酸の変化だけが必要である(Chen et al.,1994)。しかし、この分子は、インビボでの半減期が短いため、成長過剰の状態(先端巨大症など)の治療薬としては使用できない。そこで、研究者らは、hGHアンタゴニストであるhGH G120Kにポリエチレングリコール分子を加え、腎臓からのクリアランスを減少させることで、この問題に対処してきた。FDAが承認した先端巨大症の治療薬であるSOMAVERT(登録商標)は、それぞれ5000ダルトンの分子量を持つ4~6個の直鎖のPEG分子を含む。表面のリジンにランダムに結合するPEGが加わることで(van der Lely and Kopchick,2006)、アンタゴニストのインビボでの半減期が1時間未満から約72時間に延びる(Finn,2009)。しかし、PEG化されたアンタゴニストの膜結合型受容体への親和性は、未PEG化分子に比べて約30倍に低下する(Ross et al.,2001)。受容体親和性の低下にもかかわらず、SOMAVERT(登録商標)は先端巨大症の有効な治療法であるが、通常、1日5~30mgという大量の用量が処方される。
成長ホルモンアンタゴニストの活性および半減期に対する異なるPEG化戦略の予測される効果に関する洞察は、ヒト成長ホルモン(hGH)のPEG化からの結果を分析することによって得ることができる(Finn,2009)。Coxおよび同僚研究者(2007)は、ヒト成長ホルモンの3位のスレオニンをシステインに変異させ(T3C hGH)、そのシステインを20kDaの直鎖PEGに共役させた。増殖アッセイで測定したhGHの活性は約4倍に低下し、半減期は約8倍に増加した。同様の半減期の増加は、Gln141への20kDaのPEGの酵素触媒による付加や、N末端への20kDaのPEGの化学的付加の後にも生じた(Freitas et al.,2013)。化学的に活性のある非ネイティブなアミノ酸に変異させた異なるアミノ酸位置に30kDaの直鎖PEGを付加したところ、最適なケースでは、増殖活性が約10倍失われ、半減期が約10倍増加した(Cho et al.,2011)。141位に直鎖の43kDa PEG(Glnをシステインに変異)を加えると半減期が~30倍になり、hGHのN末端に分岐状の40kDa PEGを加えると半減期が約20倍になることが報告される(Rasmussen et al.,2010)。
hGHと同程度の分子量を持つサイトカインへのPEGの部位特異的な付加は、単一のPEGが半減期の著しい増加を引き起こすことを示す。Rosendahl et al.(2005)は、インターフェロンα-2の5位をシステインに変異させた後に、10kDaのPEG、20kDaのPEG、40kDaのPEGを加えると、インビトロでの生物活性が2~3倍に低下することを報告している。一方、10kDa、20kDaのPEG、40kDaのPEGでは、半減期がそれぞれ14倍、23倍、40倍に増加した。同様の結果は、ヒト顆粒球マクロファージコロニー刺激因子に異なる分子量のPEGを添加した場合にも見られた(Doherty et al.,2005)。Bell et al.(2008)は、インターフェロンαの111位(M111C)に20kDaまたは40kDaのPEGを添加すると、受容体結合活性が3倍に減少することを見出した。また、20kDaと40kDaの置換インターフェロンの半減期は、それぞれ25倍と39倍に増加した。一方、インターフェロンβ-1bに40kDaのPEGを部位特異的に付加しても、半減期は約3倍にしかならなかった(Lee et al.,2013)。
体内での滞留時間を長くするために、抗体断片にPEGを添加することについては、広範な研究が行われる。Lee et al.(1999)は、一本鎖の抗体断片(scFv MAb;26kDa)を、MWが2~20kDaの範囲にある6種類のPEGポリマーと共役した。これらの共役は、PEG化されていない親と比較して、より長い半減期を示した。半減期の延長には、PEGの総量を増やすよりも、PEGポリマーの長さを増やす方が効果的であることがわかった。Li et.al.(2010)は、ジアボディ上のランダムなリジンに共役した2つの離散的な直鎖PEGユニットを加えると、未PEG化または多分散のPEG化製品よりも血中滞留時間が長くなることを示した。Chapman et al.(2002)は、抗体のFab'フラグメント(約50kDa)の半減期は、部位特異的なPEGのサイズと数に直接関係することを示した。
本発明の様々な実施形態において、アンタゴニストhGH G120KのPEG化されたバージョンは、遺伝子工学によってアンタゴニスト配列に組み込まれたシステイン残基にPEGを付着させることによって調製された。システインに対する変異のために選択されたアンタゴニストの位置は、hGHとhGH受容体二量体(hGHR2;Sundstrom et al.,1996)との複合体のX線構造を用いて選択した。hGHR2に結合した時のhGHの構造は、hGHのアンタゴニストであるhGH G120Rが同じ受容体に結合した時の構造とほぼ同じである(Sundstrom et al.,1996)。
本発明では、2つの異なるクラスのポリエチレングリコール(PEG)分子が利用される。PEGの第1のクラスは、重合によって調製され、インビボの半減期を増加させるためにタンパク質を修飾するために使用されてきた(Kling,2013)。このタイプのPEGは、もともと多分散であり、平均分子量の周りに分子量製品の分布があることを意味する。PEGには、20kDaの直鎖PEG(Layson Bio,MPEG-MAL-20,000)、40kDaの分岐状PEG(NOF,Sunbright GL2-400MA)、および直鎖40kDaPEG(NOF,Sunbright ME-400MA)がある。これらのPEGはそれぞれ、変異タンパク質の遊離のスルフヒドリル基に共役するためのマレイミド基を含む。第2のクラスのポリエチレングリコールは、「離散型」PEG(dPEG(登録商標);Quanta BioDesign)である。これらのdPEG(登録商標)は、各dPEG(登録商標)種が特定の構造と分子量を持つ純粋な単一の化合物となるように、段階的に有機化学を用いて調製された純粋な単一のPEG分子である(Povosky et al.,2013)。本発明で使用されるdPEGは、典型的には、遊離チオールにカップリングするためのマレイミド基を含むものであり、以下のものが含まれる:分子量4473ダルトンの3分岐分子で、各分岐の末端にカルボン酸アニオンを有するもの(Quanta BioDesign #10451、MAL-dPEGA);分子量4299ダルトンの中性3分岐分子(Quanta BioDesign #4229、MAL-dPEGB);分子量8324の中性9分岐分子(Quanta Biodesign #10484、MAL-dPEGE);および分子量15,592の中性9分岐分子(Quanta Biodesign #11487、MAL-dPEGF)である。3つに分岐した4473 Daの分子は、抗体フラグメントと結合しており、マウスの血液クリアランスに及ぼす影響が調べられる(Ding et al.,2013)。添加されたdPEG(登録商標)は、50kDaのタンパク質の分子量を約8%しか増加させなかったが、血液クリアランスの「曲線下面積」(AUC)は、PEG化されていないタンパク質のAUCに比べて約2.5倍に増加した。
細胞の破壊
発現した変異体を含む増殖培地250mLを遠心分離して得られた細胞ペレットを10mLのPBSに懸濁し、EDTAを含まないプロテアーゼインヒビターカクテル(Sigma P8849)0.05mLと混合した。この溶液を氷水で冷やし、30秒のバーストで4分間超音波処理した。各々の超音波処理の後、サンプルを氷水混合液中で4℃以下になるまで冷却した。その後、超音波処理した懸濁液を4℃、25,000Xgで30分間遠心分離し、上澄み液を回収して氷上に保存した。
超音波処理した上澄み液を0.3M塩化ナトリウムに調整し、150mMイミダゾールのpH7溶液を加えて5mMイミダゾールとした。このサンプルを、TALON(登録商標)(Clontech)固定化メタルアフィニティー樹脂(IMAC)5mLを充填した栓付きアウトレットを有する重力流カラムに適用した。この樹脂は、上澄み液を加える前に、5mMのイミダゾールと0.3Mの塩化ナトリウムを含む0.05Mのリン酸ナトリウム緩衝液(pH7.0)で平衡化された。その後、カラムの上部も栓をして、室温で30分間、カラムを端から端まで混合した。その後、カラムを排出させ、少なくとも5mLの平衡化バッファーのアリコート5本で洗浄した。洗浄は、溶出液のA(280)nmが減少しなくなるまで続けた。その後、150mMのイミダゾールを含むpH7の平衡化緩衝液でカラムを溶かし、pH7に調整したEDTA二ナトリウムの100mM溶液を加えて、生成物を含む画分を5mM EDTAにした。
IMACで精製した変異体を分子濾過で2mg/mLまで濃縮し、その溶液0.5mLに、15mM還元型グルタチオン+1.5mM酸化型グルタチオンを含む溶液0.05mLを加えた。その後、0.04mgのTEVプロテアーゼ(TurboTev,Accelagen)のアリコートを加え、溶液を室温で2時間インキュベートした後、4℃で一晩インキュベートした。イミダゾールを含む緩衝液は、スピンカラム上で、0.05Mリン酸ナトリウム、pH7.0および0.3M塩化ナトリウムを含む緩衝液に交換した。
脱塩された変異体は、溶液を0.5mMのマレイミド-PEGとし、室温で2時間反応させた後、4℃で一晩インキュベートすることでPEG化された。次に、PEG化された変異体を、スピンカラムバッファーで平衡化された1mLのTALON(登録商標)樹脂を含む重力流IMACカラムに適用し、カラムを同じバッファーの5CVで洗浄した。TALON(登録商標)フロースルーおよび洗浄液には生成物が含まれており、これを遠心濃縮器で0.3mLに濃縮し、0.05Mトリスバッファー(pH8)で平衡化したSuperdex 200 Increase 10/300GLカラム(GE Healthcare)を用いてサイズ排除クロマトグラフィーにより精製し、0.15M塩化ナトリウムおよび10%グリセロールを含んだ。生成物のフラクションを合わせ、A(280)nmでの吸収によるタンパク質濃度の分析とSDS-PAGEによる純度の分析を行った。シングルシステイン変異体には1つのdPEGBが、ダブルシステイン変異体には2つのdPEGBが付加されることが、MALDI質量分析法により確認された。
競合ELISAでは、ビオチン化したhGHを調製する必要があったが、これはBiotin-dPEG12-NHS(Quanta BioDesign)を用いて標準的な方法(Hermanson,2008)で調製した。マイクロタイタープレート(Corning 96ウェルプレート、ハーフエリア、ポリスチレン)に、pH9の0.05M炭酸ナトリウム緩衝液中のhGH受容体(R&D Systems,1210-GR-50;抗体Fc領域を持つキメラとしてクローン化される)の0.125μg/mL溶液を0.05mLでコーティングした。0.125mL PBS,0.05% Tween 20(Wash Buffer)で3回洗浄した後、PBS中の2%BSAで1時間インキュベートしてプレートをブロックした。

3 これらの反応は、二重PEG化された製品には進まなかった。
本発明のPEG化変異体が、hGHによるStat5 Proteinリン酸化の刺激を阻害する能力を、細胞ベースのアッセイで測定した。IM9細胞をRPMI培地で2時間インキュベートした。その後、細胞を100万個/mLの新しいRPMI培地に再懸濁し、0~5000ng/mLの濃度でhGH、PEG化hGH変異体、またはhGH+PEG化変異体のいずれかで処理した。その後、処理した細胞を5%炭酸ガスインキュベーター内で37℃で15分間インキュベートした。その後、細胞をスピンダウンし、1% Triton X-100とオルトバナジン酸ナトリウムを含む緩衝液で溶解し、SDS PAGEゲルにのせた。このゲルを標準的な条件で実行し、タンパク質をPVDF膜に電気泳動で移した。膜をブロッキングした後、ウサギの抗Stat5 Protein抗体とウサギのβ抗-actin抗体(ポジティブセルコントロール)の混合物を用いて4℃で一晩インキュベートした。その後、膜を洗浄し、HRP結合ヤギ抗ウサギ抗体と室温で1時間インキュベートした。最後に、Pierce Supersignal West化学発光基質を用いてバンドを可視化した。PEG化された変異体の定性的な結果は、以下の表3に示される。ウェスタンブロットアッセイによって測定された、hGHによるSTAT5のリン酸化の刺激を阻害する本発明のhGH G120KPEG化二重変異体の相対的な能力は、以下の表4に示される。

ii)システインに変異した前記2つのアミノ酸に共役した前記ポリエチレングリコール分子が、3つのカルボン酸アニオンをそれぞれ含む2つの40kDaの分岐ポリエチレングリコール分子または2つの4.5kDa分岐ポリエチレングリコールのいずれかである、前記ポリエチレングリコール分子と、
を含む、ヒト成長ホルモン受容体アンタゴニスト。 - 請求項10に記載の組成物において、前記ポリエチレングリコール分子が、重合によって調製され、および多分散されているか、または前記ポリエチレングリコール分子が、段階的な有機化学によって調製され、実質的に純粋な単一化合物である、組成物。
- 請求項10に記載の組成物において、前記ポリエチレングリコール分子が、直鎖構造であるか、またはポリエチレングリコール分子が分枝構造である、組成物。
- 請求項10に記載の組成物において、前記ポリエチレングリコール分子が、遊離のスルフヒドリル基に共役するためのマレイミド基を含む、組成物。
- 請求項10に記載の組成物において、前記ヒト成長ホルモン受容体アンタゴニストが、配列ID番号:13、15、17、19、21、および23からなる群から選択されるDNA分子と少なくとも95%の同一性を有するDNA配列によってコード化される、組成物。
- 請求項10に記載の組成物において、前記ヒト成長ホルモン受容体アンタゴニストが、配列ID番号:14、16、20、22、および24からなる群から選択されるアミノ酸配列を有する、組成物。
- ヒト成長ホルモン受容体アンタゴニストに反応する疾患または状態を治療するための方法であって、請求項10に記載の前記組成物の有効量を患者に投与する工程を含む、方法。
- ヒト成長ホルモン受容体アンタゴニストであって、
a)ヒト成長ホルモン受容体アンタゴニストG120Kであって、ヒト成長ホルモン受容体アンタゴニストG120Kの2つのアミノ酸がシステインに変異しており、システインに変異した前記2つのアミノ酸が、N99/T142、N99/H151、およびT142/H151からなる群から選択される、前記ヒト成長ホルモン受容体アンタゴニストG120Kと、
b)ヒト成長ホルモン受容体アンタゴニストG120K変異体の各置換されたシステインに共役したポリエチレングリコール分子と、
を含む、ヒト成長ホルモン受容体アンタゴニスト。 - 請求項17に記載の組成物において、システインに変異した前記2つのアミノ酸に共役した前記ポリエチレングリコール分子が、3つのカルボン酸アニオンをそれぞれ含む2つの40kDaの分岐ポリエチレングリコール分子または2つの4.5kDa分岐ポリエチレングリコールのいずれかである、組成物。
- 請求項17に記載の組成物において、前記ヒト成長ホルモン受容体アンタゴニストが、配列ID番号:19、21、および23からなる群から選択されるDNA分子と少なくとも95%の同一性を有するDNA配列によってコード化される、組成物。
- 請求項17に記載の組成物において、前記ヒト成長ホルモン受容体アンタゴニストが、配列ID番号:20、22、および24からなる群から選択されるアミノ酸配列を有する、組成物。
