4mmの直径のディスクが、4mmの生検パンチで、ECMシート材料(腎被膜、「RC」の4層ラミネート)から切り出された。RCディスク移植組織は、臍帯血由来のECFC(10μlの中の約300,000個の細胞)および臍帯血由来のMSC(10μlの中の約100,000個の細胞)を組み合わすことにより準備された20μlの懸濁液に24時間、37℃でインキュベートされた。その後、外科医が既知の免疫不全の(SCID)マウスの頭蓋冠欠損モデルの欠損へそれを注入することを可能にするために、細胞がシードされたRCディスクは、外科のフィールドへ渡された。モデルについては、各マウスに左右同形の4mmの直径の欠損が空けられた。左右同形の欠損のうちの1つはコントロール(処理されない)として使用された。また、他方は処理物質を受け取った。2本のチタン線が、イメージ可能な参照として対向する移植組織ディスクの外側端に結び付けられた。縫合は、欠損治療で空けられた頭部および尾部の穴、および移植組織ディスクの対になった穴部を通って行われた。移植組織ディスクは縫合結び目で頭頂骨に固定された。対照(空間)治療部位は、縫合で同様に結ばれたが、移植組織を受け取らなかった。その切り口は閉じられ、マウスにエリザベスカラー(手術の後に1週間着用された)が取り付けられた。研究されたハツカネズミは、移植後2、4、8、12および16週にmicroCT(小規模コンピューター断層撮影)で生体内イメージを得た。microCT走査は、治療された部位と治療されない欠損のための骨被覆の相違の測定のために使用された。
細胞がシードされたECM移植組織ディスクで治療された欠損については、移植後16週での骨被覆%は平均52.6%(n=6)となった。これらのグループの対応する治療していない欠損については、移植後16週での骨被覆%は平均23.7%となった。同様な実験が行われた実験が行なわれた。一群のハツカネズミ(n=7)はECM移植組織ディスク(追加の細胞はない)のみを受け取り、他の側には治療はしなかった。移植後16週で、治療していない側の骨被覆は約22%であり、治療した側の骨被覆は約35%であった。
Claims (1)
- 内皮先祖細胞(EPC)および間充織幹細胞(MSC)並びに固体のキャリア材料を含む、患者の損傷したかまたは病気の骨を治療するための組成物であって、
EPCとMSCをそれぞれ少なくとも2:1の細胞数比で含み、
該固体のキャリア材料は、コラーゲンの細胞外マトリックス(ECM)組織材料を含み、該ECM組織材料が該ECM組織材料のための源組織からの生来の生物活性物質を保持し、該生来の生物活性物質が、成長因子、グリコサミノグリカン類、プロテオグリカン類、および糖タンパク質を含み、該生来の生物活性物質が、該ECM組織材料のコラーゲン繊維の間、上および/または内に点在した固体として該ECM組織材料に保持されて、該ECM組織材料の乾燥重量での少なくとも1%を構成しており、
当該組成物中のEPCとMSCを合わせた合計数の少なくとも50%が該固体のキャリア材料に付着されている、組成物。
