本発明は、概して、核酸の捕捉、増幅およびシーケンシングに関する。

前記2つの5’連結標的核酸を、前記捕捉領域に相補的な標的領域および複数のプライマーを含む分子に、前記標的領域と前記捕捉領域の結合は可能にするが前記プライマーと前記プライマー部位の結合は阻害する条件下で曝露させるステップであって、前記標的領域で、前記2つの5’連結標的核酸のうちの少なくとも一方を前記分子に結合させるステップと、
前記条件を、前記プライマー部位と前記プライマーの結合が可能になるように変更するステップであって、前記分子のプライマーを前記2つの5’連結標的核酸の両方の前記プライマー部位と結合させるステップと、
鎖置換型ポリメラーゼを使用して前記プライマーを伸長させるステップであって、前記5’連結標的核酸のそれぞれの分子結合コピーを生じさせるステップと
を含む方法。 - 前記プライマー部位がユニバーサルプライマー部位であり、前記プライマーがユニバーサルプライマーである、請求項1に記載の方法。
