Abstract
Description
この出願は、「アルギネートヒドロゲル組成物」のタイトルで2016年3月24日に出願された米国仮特許出願第62/312,998号の利益を主張する。上記出願の全内容が、参照により本明細書に組み込まれる。
イオンで架橋されたアルギネートゲルの1つの欠点は、生理条件下での制限された安定性である。イオンで架橋されたゲルは、一価のカチオン及び他のカチオンとの交換反応によって生理条件下で溶解する傾向がある。ゆえに、イオンで架橋されたアルギネートヒドロゲル及び他の修飾されたアルギネートヒドロゲルに関連する不安定性ならびに他の欠点を有することなく、生物医学的応用における使用に好適なアルギネート組成物を提供することがかなり有利である。

本出願は、本明細書においてより詳細に記載されている、とりわけ、多くの特に驚くべきかつ有益な特徴を有する半浸透性ヒドロゲル組成物の本発明者らの発見に少なくとも部分的に基づいている。ヒドロゲル組成物は、アルギネートマトリックスの周辺においてマルチアームの水溶性ポリマーに共有結合的に架橋された当該アルギネートマトリックスを含む。1つ以上の実施形態において、マルチアームの水溶性ポリマーがアルギネートマトリックスの周辺に浸透していることにより、アルギネートマトリックスの周辺がマルチアームの水溶性ポリマーと連動しており、かつ共有結合的に架橋されている、組成物を提供する。これにより、ヒドロゲルを安定化させることにより、当組成物にいくつかの特有かつ驚くべき特徴を付与する。
アルギネート/アルギン酸
ヒドロゲル組成物は、アルギネートマトリックスを含む。アルギネート(アルギン酸)は、例えば、Laminaria hyperborea、Laminaria digitata、Laminaria japonica、Ascophyllum nodosum、及びMacrocystis pyrifera、ならびにAzotobacter及びPseudomonasを含めた、藻及び細菌の両方の源を含めた任意の好適な源から得られてよく;アルギネートは、褐藻類から最も一般的に調達される。細菌源から得られるアルギネートは、部分的にアセチル化されている。本明細書において提供されているヒドロゲル組成物の調製の際の使用に好適なアルギネートとして、例えば、Tonnesen,H.,et al.,Drug Dev Ind.Pharm.,2002;28:621-630に記載されているものが挙げられる。アルギネートは、(1,4)-結合したβ-D-マンヌロネート(M)及びα-L-グルロネート(G)残基のブロックを含み;本明細書における使用のためのアルギネートは、いずれの好適なM/Gブロック比を有していてもよい。例えば、G-ブロック含有率は、約10%から最大で約80%までの範囲であってよく、または約10%~約60%、約15%~約50%、約20%~約80%、約20%~約60%、もしくは約20%~約55%の範囲であってよい。いくつかの実施形態において、G-ブロック含有率は、約30%~約95%の範囲内であり、または、いくつかの好ましい実施形態において、約40%~約70%の範囲内である。M及びG単位の含有率は、典型的には、製造者によって提供されており、1H NMR分光法によって求めることもできる。アルギネートは、典型的には、約5,000~約500,000g/mol、約7,000~約400,000g/mol、または約10,000~約300,000g/molの範囲の平均分子量(重量平均)を有する。いくつかの実施形態において、好ましい分子量範囲は、約10,000ダルトン~約300,000ダルトンである。アルギネートマトリックスを調製するのに使用されるアルギネートは、共有結合的に架橋された表面層を与えるのに使用されるアルギネートと同じであっても異なっていてもよい。

当ヒドロゲルの形成の際の使用のためのマルチアームの水溶性ポリマーは、中央のコアから放射している2つ以上の線状ポリマー鎖または「アーム」を有する。ゆえに、マルチアームポリマーは、その構成及びコア構造に応じて、単一の分岐点から放射している2つの線状ポリマーアームによる2つのポリマーアーム(典型的には、分岐ポリマーと称される)、3つのポリマーアーム、4つのポリマーアーム、5つのポリマーアーム、6つのポリマーアーム、7つのポリマーアーム、8つのポリマーアーム、9つのアームもしくは10個のアーム、またはこれより多くを有していてよい。いくつかの実施形態において、マルチアームの水溶性ポリマーは、3~10個のポリマーアームを有する。いくつかの実施形態において、マルチアームの水溶性ポリマーは、3~8つのアームを有する。いくつかの実施形態において、マルチアームの水溶性ポリマーは、4つのアームを有する。いくつかの代替の実施形態において、マルチアームの水溶性ポリマーは、6つのアームを有する。いくつかのさらなる実施形態において、マルチアームの水溶性ポリマーは、8つのアームを有する。上記のように、マルチアームのポリマーは、樹枝状ポリマーでも超分岐ポリマーでもない。いくつかの実施形態において、共有結合的連結部自体に含まれるマルチアームのポリマーのアームは、分岐していない(すなわち、線状である)。例えば、共有結合的な架橋連結部に含まれる、アルギネート上の所与の官能基のいくつかの好ましい実施形態において、マルチアームのポリマーの所与のアームに対して1-1の連結部が存在する。
本明細書において提供されているヒドロゲルは、薬理活性物質または生細胞を任意選択的に含んでいてよい。薬理活性剤として、小分子(約1000未満の分子量を有する)、タンパク質、ポリヌクレオチドなどが挙げられる。
ヒドロゲル組成物は、以下のように概して調製されるが、記載されている方法は、当開示に基づいて、本明細書において提供されている組成物、材料、及びヒドロゲルに達するように当業者によって好適に変更されてよいことが認識される。当ヒドロゲル組成物は、典型的には、その周辺において、マルチアームの水溶性ポリマーに共有結合的に架橋されたアルギネートマトリックスを形成するのに有効な条件下で、上記のような誘導体化されたアルギネートマトリックスをマルチアームの水溶性ポリマーと反応させることによって形成される。アルギネートマトリックスは、1から数層の共有結合的に付着された水溶性ポリマーを含むように逐次修飾されてよく、ここで、ポリマー層は、典型的には、官能化されたアルギネートと水溶性のマルチアームのポリマーとの間で交互に存在する。代替的には、アルギネートマトリックスの周辺でのマルチアームの水溶性ポリマーへの共有結合的な架橋結合の形成の後に、さらなる連動層が、任意の好適な水溶性ポリマーとの反応によって共有結合的に付着されてよい。アルギネートマトリックスの共有結合的に付着された周辺を形成するのに利用される反応基対は、任意選択的なさらなる共有結合的に連結された表面層を形成するのに使用される反応基対(複数可)と同じであっても異なっていてもよい。アルギネートマトリックスは、上記のようなヒドロゲル組成物の投与の際に免疫反応を軽減するのに有効な1つ以上の部位を任意選択的に含んでいてもよく、ここで、かかる修飾は、共有結合的に反応性の架橋可能基の導入の前または後のいずれであってもよい。代替的には、いくつかの実施形態において、共有結合的な架橋結合、例えば、ジベンジルシクロオクタントリアゾール環系、もしくはトリアゾール基を含むもの、または修飾トリアゾールは、ヒトまたは動物対象への当ヒドロゲル組成物の投与の際の免疫介在反応を低減/最小化するのに有効であり得る。
当ヒドロゲルは、ストレス条件下(例えば、40℃及び1mMリン酸緩衝生理食塩水)での長期間にわたる顕著な安定性を含めたいくつかの望ましい特徴を有する。1つ以上の実施形態において、当組成物は、その形状及びゲル完全性を維持する(すなわち、良好な機械的安定性を有する)だけでなく、in vivo環境をモデル化する高ストレス条件下に置かれたときに、長期間にわたって半浸透性及び他のレオロジー特性を保持する。より詳細には、本明細書に記載されているアルギネートヒドロゲル組成物は1mM PBS中40℃で保存したとき、最長で少なくとも30日間、例えば、最長で少なくとも45日間、または最長で少なくとも60日間、また、さらには最長で少なくとも90日間にわたって安定であり、また、形状かつその及び完全性を維持する。例えば、当ヒドロゲル組成物の浸透性及び機械的安定性の特徴を記載している実施例6及び7を参照されたい。1つ以上の実施形態において、当ヒドロゲルは、長期間、すなわち、少なくとも30日間、最長で少なくとも45日間、最長で少なくとも60日間、また、実際に最長で3ヶ月間(少なくとも90日間)にわたって、シミュレーションされたin vivo条件下でその半浸透性をさらに維持し-当該ヒドロゲルを驚くべきことに構造的かつ機能的に復元性にする。上記によると、当材料は、治療薬での使用に、特に、他の用途の中でも、生存細胞を送達するのに有利に適している。

3-アジド-アミノプロパンによるアルギン酸の修飾
アルギン酸のカルボキシル基を、異なる量の3-アジド-アミノプロパンによってランダムに修飾して、異なるアジド置換度を有するアルギン酸を得た。アルギン酸アジドポリマーを透析によって精製した。5%、10%、15%及び20%のモル当量のアジドを有する、化学的に修飾されたアルギン酸アジドを以下のように調製した。
アルギン酸(1g、5.05mM)をMES(2-(N-モルホリノ)エタンスルホン酸)(0.1M)及びNaCl(0.3M)緩衝剤にpH6.5で溶解して、0.5%濃度のアルギン酸を有する溶液を得た。3-アジド-1-プロパミン(0.2当量、101.06mg)、EDC.HCl(1当量、967.5mg)及びNHS(0.4当量、232.34mg)を添加し、反応混合物を室温で24時間撹拌した。反応混合物を、次いで、150mmから0mmまで減少する塩勾配を使用して脱イオン水に対して透析した。透析後、生成物を凍結乾燥し、4~8℃で保存した。
アルギン酸(1g、5.05mM)をMES(0.1M)及びNaCl(0.3M)緩衝剤にpH6.5で溶解して、0.5%濃度のアルギン酸を有する溶液を得た。3-アジド-1-プロパミン(0.15当量、75.79mg)、EDC.HCl(0.75当量、725.62mg)及びNHS(0.3当量、174.26mg)を添加し、反応混合物を室温で24時間撹拌した。反応混合物を、次いで、150mmから0mmまで減少する塩勾配を使用して脱イオン水に対して透析した。透析後、生成物を凍結乾燥し、4~8℃で保存した。
アルギン酸(1g、5.05mM)をMES(0.1M)及びNaCl(0.3M)緩衝剤にpH6.5で溶解して、0.5%濃度のアルギン酸を有する溶液を得た。3-アジド-1-プロパミン(0.1当量、50.53mg)、EDC.HCl(0.5当量、483.75mg)及びNHS(0.2当量、116.17mg)を添加し、反応混合物を室温で24時間撹拌した。反応混合物を、次いで、150mmから0mmまで減少する塩勾配を使用して脱イオン水に対して透析した。透析後、生成物を凍結乾燥し、4~8℃で保存した。
アルギン酸(1g、5.05mM)をMES(0.1M)及びNaCl(0.3M)緩衝剤にpH6.5で溶解して、0.5%濃度のアルギン酸を有する溶液を得た。3-アジド-1-プロパミン(0.05当量、25.26mg)、EDC.HCl(0.25当量、241.87mg)及びNHS(0.1当量、58.09mg)を添加し、反応混合物を室温で24時間撹拌した。反応混合物を、次いで、150mmから0mmまで減少する塩勾配を使用して脱イオン水に対して透析した。透析後、生成物を凍結乾燥し、4~8℃で保存した。
4-アームPEG-DBCOの合成
その4つの末端の各々にカルボン酸基を有する4-アームPEG、「4-アームPEG-COOH」、10kD(1グラム)を、40mlのジクロロメタン(DCM)に溶解した。EDC.HCl(5当量、766.8mg)、NHS(2当量、184.14mg)及びトリエチルアミン(5当量、557μl)を4-アームPEG-COOH/DCM溶液に撹拌しながら添加した。ジベンジルシクロオクチンアミン(1.05当量、232.12mg)を次いで添加し、反応混合物を室温で24時間撹拌させた。ジクロロメタンを蒸発によって除去し、DBCO官能化された生成物を、Sephadex g-15カラム及び溶出液としてのエタノールを使用してカラムクロマトグラフィによって精製し、4-アームPEG-DBCOを得た。
アルギン酸-アジドビーズの調製
アルギン酸アジドポリマーの水溶液を(1.5重量%、1.5g/100mL、20%アジド-修飾、実施例1に記載されている)を、25ゲージ針を装着したシリンジを有する自動ポンプを使用してCaCl2の0.1M溶液に滴加した。得られたビーズを収集し、水で洗浄した。
4-アームPEG-DBCOによるアルギン酸-アジドビーズの共有結合的な表面修飾
複数の共有結合的に架橋されたポリマー表面層を有するアルギン酸-アジドビーズの調製
4-アームポリエチレングリコールの単一の共有結合的に架橋された相互浸透表面層を有する、実施例4からの表面修飾ビーズ(「AlgAz-PEGD」)を、所望の層数に応じて、実施例4に記載されているさらなる表面層を逐次添加し、相応に反応官能基のモル比を調整することによってさらに修飾した。共有結合的に架橋された3、5、及び7つの表面層を有するビーズを以下のように調製した。
浸透性研究
浸透性研究を、実施例4及び5からの、共有結合的に表面架橋された修飾されたビーズ(共有結合的に架橋された1、3、5、及び7つの表面層(複数可)を有する)を使用して行った。浸透性研究において使用した蛍光分子量マーカーは、デキストラン-FITC10kD、70kD、150kD、及び250kD、ウシ血清アルブミンFITC(BSA FITC)、ならびに免疫グロブリンG-FITC(IgG FITC)であった。分子量マーカーを、0.1~0.2mg/mlの範囲の濃度で生理食塩水に溶解した。ビーズを、96ウエルプレートにおいて200μLのFITC溶液によって40℃/1mM
PBSで最大90日間インキュベートした。ビーズ内へのプローブの拡散を各時点(18日間、67日間、90日間)において共焦点顕微鏡法によってモニタリングした。
機械的安定性
ビーズの機械的特性を、平行板形状(板直径8mm及びギャップ2mm)を使用して21℃でRSA-G2固体分析装置(TA Instruments)を使用して分析した。0.05nの法線軸方向の力を適用し、ビーズの直径を、分析装置の軸方向の位置の読み取り値によって求めた。ビーズを、5%の動的ひずみ及び1~100rads/sの周波数掃引を適用することによって圧縮モードで分析した。
ホスホコリン及びリシンを含む生体適合性ポリマーの合成
実例的なホスホコリン及びリシンコポリマーを以下に記載のように設計及び合成した。
50mlの二口丸底フラスコに、2-メタクリルオキシホスホコリン(1.18g、4mmol)及びN-メタクリロイルL-リシン(85mg、0.4mmol)、ならびに撹拌棒を入れた。窒素ガスをニードルによって10分間パージした後、脱イオン水(10ml)を付加し;得られた混合物を、透明な溶液が形成されるまで撹拌した。窒素ガスを、反応全体を通して連続してパージした。反応混合物に、水溶性アゾ開始剤、2,2’-アゾビス(2-メチルプロピオンアミジン)二塩酸塩(V-50)を添加した。当該開始剤を、上記の添加後2~3分以内に溶解させた。フラスコを60℃に加熱し、窒素雰囲気下で16時間、制御された温度条件下で維持した。得られた粘性溶液を室温まで冷却させた。粘性溶液をイソプロパノール(200ml)中に注いだ。ポリマーを白色塊として沈殿させた。溶媒をデカントし、乾燥して、真空下で溶媒を除去した。ポリマーを水(30ml)に溶解させ、粘性溶液をイソプロパノール中に滴下により注いだ。ポリマーを収集し、真空下で乾燥し、その後、水にさらに3回溶解させ、その後、イソプロパノールにおいて沈殿させた。ポリマーを真空下で乾燥させて、白色粉末を得た(1.2g)。生成物の分子量(GPC分析)は約300kDであった。
50mlの二口丸底フラスコに、2-メタクリルオキシホスホコリン(1.181g、4mmol)及びN-メタクリロイルL-リシン(170mg、0.8mmol)、ならびに撹拌棒を入れた。フラスコを窒素ガスで10分間パージした後、脱イオン水(10ml)を付加しし;得られた混合物を、透明な溶液が形成されるまで撹拌した。窒素ガスを、反応を通して連続してパージした。水溶性アゾ開始剤、2,2’-アゾビス(2-メチルプロピオンアミジン)二塩酸塩(V-50)を反応混合物に添加した。当該開始剤を、上記の添加後2~3分以内に溶解させた。フラスコを60℃に加熱し、窒素雰囲気下で16時間、この温度にて制御された条件下で維持した。得られる粘性溶液を室温まで冷却させた。粘性溶液をイソプロパノール(200ml)中に注いだ。ポリマーを白色塊として沈殿させた。溶媒をデカントし、乾燥して、真空下で溶媒を除去した。ポリマーを水(30ml)に溶解させ、粘性溶液をイソプロパノール中に滴下により注いだ。ポリマーを収集し、真空下で乾燥し、その後、水にさらに3回溶解させ、その後、イソプロパノールにおいて沈殿させた。ポリマーを収集し、真空下で乾燥した。再び、当該ポリマーを水に溶解させ、イソプロパノールにおいて沈殿させた。3回再沈殿した後、最後にポリマーを真空下で乾燥させて、白色粉末を得た(1.3g)。
実施例8Bに記載されている、ホスホコリン及びリシン(16.6モル%リシン)のメタクリルアミドコポリマー(300mg)を5mlの水に溶解し、pHをNaHCO3粉末(約10mg)の添加によって7.4に調整した。これにDBCO-スルホ-NHSエステル(50mg、0.094mmol))を添加し、その後、室温で3時間撹拌した。pHを、NaHCO3粉末(5mg)を15分以内に添加することによって7.3で維持し、pHを時折モニタリングした。3時間後、反応混合物を(5Kモル重量カットオフ膜を使用して)透析した。溶液を凍結乾燥した。ポリマーを白色のフレーク状材料として得た。
ホスホコリン及びリシンのメタクリルアミドコポリマー、PC-Lys-MA(16.6モル%リシン;300mg)を5mlの水に溶解し、pHをNaHCO3粉末(約10mg)の添加によって7.4に調整した。上記溶液にDBCO-PEG4-NHSエステル(50mg)を添加し、その後、室温で3時間撹拌した。pHを、NaHCO3粉末(5mg)を15分以内に添加することによって7.3で維持し、その後、pHを時折モニタリングした。3時間後、反応混合物を透析した(5Kモル重量カットオフ膜)。溶液を凍結乾燥した。ポリマーを白色粉末として得た。
実施例8Cまたは8Dに記載されているDBCO-PC-Lys-MAポリマーの溶液(2%)を、室温で水中にコポリマーを溶解することによって調製した。4-アームPEGアジドの溶液(10%)を、室温で水中に4-アームPEGアジド(モル中量5K)を溶解することによって調製した。きれいなガラスバイアルにDBCO-PC-Lys-MAポリマー(100マイクロリットル)を入れ、その後、PEGアジド溶液(100マイクロリットル)を添加した。混合物を僅かに旋回させた。すぐに、透明なゲルが得られた。この結果は、DBCO-PC-Lys-MAポリマーが活性であり、かつ、例えば、本明細書に記載されているように、アジド基を含有するアルギン酸ビーズを表面にコーティングするのに好適であり、これにより、生体適合性クリック反応性ポリマーによる最終コーティングが得られることを示唆している。
