Abstract
Description
・ 上皮性腫瘍は卵巣の外面を覆う細胞層に由来する。約90パーセントの卵巣腫瘍が上皮性細胞腫瘍である。
・ 胚細胞腫瘍は卵(卵子)を産生する細胞から開始する。
・ 間質性腫瘍は卵巣を一体に保持し、女性ホルモンのエストロゲンおよびプロゲステロンを産生する構造組織細胞から開始する。
a)患者の卵巣がんをスクリーニングし、がんがHR欠損であるか、またはHRの能力があるかどうかを評価するステップ、

PARPは、DNA損傷を検出し、DNA一本鎖切断(SSB)に結合し、DNA修復タンパク質を動員し、修復プロセスを駆動することができるタンパク質のファミリーである。ヒトにおけるタンパク質のPARPファミリーには、PARP1およびPARP2が含まれる。

ASCN再構築の品質を評価するために、品質測定法であるKS品質を導入する。具体的には、各試料について、全てのSNPを2つの2つの群に分け、第1の群はアレル不均衡を有する全てのSNPを含み、第2の群は2つの親アレルの等しいコピー数を有する全てのSNPを含む。各SNPにおけるアレル量dは以下のように変換される:d<0.5である場合、dtr=dであり、その他の場合、dtr=1-dである。KS品質は以下のように定義される

の主な理由が存在する:(1)配列データにおける高いノイズレベルおよび(2)試料中の低い腫瘍含有量。
HRD-LOHスコアは、15Mbより長いが、染色体全体よりも短いLOH領域の数と定義される(Abkevichら、2012年)。HRD-LOHスコアは609個の卵巣腫瘍におけるBRCA1、BRCA2およびRAD51C欠損に関連することが示されている(Abkevichら、2012年)。
特定の最小数のSNP(平均で約1.8Mb)を包含する場合のみカウントする。本発明者らは、609個の卵巣腫瘍の3つのデータセットにおけるBRCA1、BRCA2およびRAD51C欠損とのHRD-TAIスコアの関連について試験し(データは示さず)、HRD-TAI領域のサイズについてのカットオフが11Mbに増加した場合、より顕著になる関連性を見出した。したがって、改変されたHRD-TAImスコアは、(a)サブテロメアの1つにまで及ぶ、(b)動原体を超えない、かつ(c)11Mbより長い、アレル不均衡を有する領域の数と定義される。
