Abstract

Description
本発明は、神経変性疾患、特にシヌクレイノパチーの予防又は治療において使用するための化合物を特定する。本発明はさらに、有効量の前記化合物及び一つ以上の薬学的に許容される助剤を含む医薬組成物を特定する。
シナプスタンパク質α-シヌクレイン(α-syn)の進行的な蓄積は、総じてシヌクレイノパチーと称される、パーキンソン病(Parkinson’s disease:PD)、レビー小体型認知症(Dementia with Lewy bodies:DLB)及び多系統萎縮症(Multiple System Atrophy:MSA)の病因として重要な役割を果たすことが提唱されている。全世界で約1000万人がシヌクレイノパチーの影響を受けていると推定されるが、現在のところ、疾患の進行を抑制する治療法はない。α-synの病的な蓄積をもたらす正確なメカニズムは十分に理解されていないが、α-synの合成、凝集、クリアランスの割合の変化が関与していると推定される。例えば、特定の家族性パーキンソン病においては、α-syn遺伝子の重複によりα-synの合成が増大すること、及び、変異(E46K、A53T、H50Q、G51D)により凝集性向が増大することが説明されている。
さらに、α-synの蓄積が、毒性のあるオリゴマーの形成及び細胞から細胞へのプリオン様の伝播を介して神経変性をもたらすことが提唱されてきた。毒性のあるα-syn種の正確な同定については議論の余地があるが、ほとんどの研究は、大きな凝集体よりもむしろオリゴマーが原因であり得るという点で一致している。さらに、生物物理学的研究により、このプロセスにおいて、α-synの結合とその後の神経細胞膜通過が重要であるとの証拠が得られている。
神経細胞膜におけるα-synと脂質との相互作用は、オリゴマー形成及び細胞毒性のプロセスにおける重要なステップであると提唱されている。従って、分解及びクリアランスの増加、凝集の阻害、又はα-syn合成の低減を標的とする戦略は、合理的な治療戦略を示しうる。
これまでの研究は、抗体、タンパク質分解酵素、及び、α-syn凝集又は繊維化を低減させる小分子を用いることにより、α-syn凝集を標的としてきた。近年、膜におけるα-syn伝播二量体の形成が、毒性のあるα-synオリゴマーの発生の初期段階であり、二量体中の一方のα-synの96~102残基と他方のα-synの80~90残基の間の相互作用が、このプロセスにおいて重要な役割を果たすことが示された。
大きな細胞内繊維を破壊する抗アミロイド剤から、ミスフォールドオリゴマー凝集体の細胞間伝播を標的とする抗アミロイド剤又はプロトフィブリルの「種」に単量体α-synを添加することによってモデル化されたフィブリル増大期を標的とするものに至るまで、α-synを標的とする数多くの治療戦略が提案されてきた。
Lamberto G.Rらは、「Structural and mechanistic basis behind the inhibitory interaction of PcTS on α-synuclein amyloid fibril formation (α-シヌクレインアミロイドフィブリル形成の背後にあるPcTsの阻害作用の基本構造及び基本メカニズム)」(PNAS 106 (50): 21057-21062, 2009)において、凝集阻害剤の同定及び作用メカニズムの研究は、アミロイド形成の病理学的影響を緩和するための探求における基本である、と述べている。フタロシアニンテトラスルホネート(phthalocyanine tetrasulfonate:PcTS)のα-シヌクレインに対する抗アミロイド形成作用の基本構造及び基本メカニズムの特性評価により、特定の芳香族の相互作用が、アミロイド形成のリガンド介在阻害の中心的な役割を果たすことが実証された。これらの知見は、アミロイド形成に関する構造メカニズム及び毒性メカニズム、並びにアミロイド関連病態の治療法としての小分子の可能性を評価するための分子プローブとして、凝集阻害剤を使用することを強調するものである。
WO2013/134371は、ヒト又は動物の身体における毒性のあるタンパク質凝集の初期形成(例えば、Aβオリゴマーの形成)を特異的に遮断する化合物に関している。これらの化合物は、非常に初期の毒性タンパク質凝集を特異的に標的とし、アミロイドβ等の、神経変性疾患におけるタンパク質凝集に関与することが知られているタンパク質に対して高い親和性を有すると言われている。
グリア細胞の反応は、筋萎縮性側索硬化症(amyotrophic lateral sclerosis:ALS)、前頭側頭葉変性症(frontotemporal lobar degeneration:TLD)、ハンチントン病、パーキンソン病及びアルツハイマー病を含む神経変性疾患の一般的特徴である。アストロサイト及びミクログリアは、神経変性プロセスの間に反応性を有するようになり、活性化アストロサイトは、アストロサイト受容体、トランスポーター及びトランスミッターにおける異なる発現;代謝変化;及び、タンパク質、ケモカイン及びサイトカインの合成及び放出の変化、を示し得る。制御されたアストロサイトの活性化は、ニューロンには有益であると考えられているが、過活性のアストロサイトは有害であり得る。神経変性におけるアストロサイト過多は、集中的に研究されてきたが、反応性アストロサイトがどのように神経毒性に寄与するかは未だ正確に決定されていない。従って、アストロサイトの活性化の制御及び静止状態のアストロサイトの反応性アストロサイトへの転換の阻害を可能にする、新規化合物の開発に対する需要が依然として存在する。
Biらは、「Reactive astrocytes secrete Lcn2 to promote neuron death(反応性アストロサイトはLcn2を分泌し、神経死を促進する)」(PNAS, 110 (10): 4069-4074, 2013)において、リポカリン2(lipocalin 2:Lcn2)は、反応性アストロサイトにより分泌される、ニューロンに対して選択的に毒性である誘導性因子として説明されている。Lcn2は、変異ヒトTAR DNA結合タンパク質(TAR DNA binding protein:TDP)43又は肉腫における融合RNA結合タンパク質(fused in sarcoma:FUS)のニューロン発現を有するトランスジェニックラットの反応性アストロサイトにおいて誘導されることが示された。さらには、合成Lcn2が、一次ニューロンに対して用量依存的に細胞毒性であるのに対し、アストロサイト、ミクログリア及びオリゴデンドロサイトに対しては無害であることも説明されている。免疫沈降によりLcn2を部分的に低減させると、条件培地が介在する神経毒性が低減した。これらのデータは、反応性アストロサイトが強力な神経毒性メディエーターであるLcn2を分泌することを示唆している。さらに、近年の文献では、パーキンソン病の患者の黒質(substantia nigra:SN)において、LCN2/NGAL発現が増大することが報告されている(Kim, B. 化合物は、神経変性疾患に関連して、静止状態のアストロサイトを反応性(神経毒性)アストロサイトへ転換する経路の阻害を含む、脳に対する複数の好ましい効果で同定されている。
より正確には、本発明者は、α-synを標的とする新規化合物を同定した。高濃度で、該化合物は膜からα-synを放出させ、従ってその凝集性向に影響を及ぼす。しかしながら、従来技術の化合物とは対照的に、該化合物は、可溶性の単量体形態のα-synのN-末端にも結合し、従って、その内在性タンパク質相互作用パートナーのいくつか(すなわち、重要なカルシウム結合タンパク質であるカルモジュリン)へのα-synの結合に影響を及ぼす。パーキンソン病のトランスジェニック動物モデルにおける該化合物の活性は、既に示されている。該化合物は、トランスジェニック動物の皮質においてα-syn凝集の数を低減させ、ニューロン数(神経栄養効果)を増大させた。
しかしながら、本発明の新規化合物による更なる重要な成果は、全く新しい予想外の更なる活性である。最も顕著なのは、本発明者は、該化合物がトランスジェニックマウスの皮質において選択的にアストロサイトの数を増大させたが、健常対照マウスにおいてはそうでない(該化合物単独ではアストロサイトの産生を誘導しない)ことを見出したことである。この予想外の発見は、パーキンソン病などの神経変性疾患に対抗する全く新しい戦略の基礎となる。
この活性は、アストロサイト形成と挙動に影響する重要な因子であると考えられる、リポカリン2(Lcn2/NGAL)への結合という本発明の新規化合物の機能に関連する。ラットの脳スライスから培養培地中に分泌されたタンパク質の二次元電気泳動及び質量分析により、リポカリン2(Lcn2)は、反応性アストロサイトによって分泌される神経毒性を介在する誘導性因子であると同定された。Lcn2はさらに、変異TDP-43又は肉腫における融合RNA結合タンパク質(FUS)を発現するトランスジェニックラットにおいて、アストロサイト因子として検証された。総じて、本願において提示するデータは、Lcn2は反応性アストロサイトによって分泌される強力な神経毒性因子であることを実証している。
従って、他のパーキンソン病治療法と共通する利点(α-シヌクレイン凝集の低減)に加えて、本願において提唱される小分子は、(i)皮質におけるニューロン数の増大(神経栄養効果)及び(ii)Lcn2タンパク質(反応性アストロサイトによって分泌される強力な神経毒性因子)への競争的結合を介したトランスジェニックマウスの皮質におけるアストロサイト数の選択的増大、を含む、数多くの予想外且つ臨床的に重要な性質を有している。
第一の態様によると、本発明は、神経変性疾患の予防又は治療において使用するための、
a)α-シヌクレイン(α-syn)凝集の形成阻害、及び
b)リポカリン2(Lcn2/NGAL)への結合
が可能な化合物を特定する。

更なる態様によると、該化合物は、場合により、参加ストレス及び/又はミトコンドリア機能障害誘発性の神経毒性、神経炎症の主要なプレーヤーからの保護効果を有する。
さらに他の態様においては、該化合物は、場合により、α-synトランスジェニック(tg)げっ歯類において、濃度依存的に運動障害を改善することができる。α-synトランスジェニック(tg)げっ歯類は、ヒトα-syn遺伝子の配列及び/又は発現が、シヌクレイノパチーを模倣するように変更されている任意のマウス又はラットモデルを含む。現在使用されているこのようなげっ歯類の具体例は、A53T、系統61(Line 61) (TNWT61)、D-系統(D-Line)及びE46Kを含む。
従って、本発明による、神経変性疾患を予防/治療する方法は、α-シヌクレイン(α-syn)凝集の形成阻害及びリポカリン2(Lcn2/NGAL)への結合が可能な化合物の治療有効量を、それを必要とする患者に投与することを包含する。この疾患の治療には、対症療法及び神経保護(進行抑制)治療が含まれ得る。特に、対症療法は、運動、行動、認知、気分、睡眠、感覚症状の回復及び神経炎症の低減を含み得る。
本発明の化合物は、とりわけ、α-シヌクレイン(α-syn)凝集の形成を阻害することが可能である。本明細書において使用される用語「凝集」は、「繊維化」も含む。特に、該化合物は、α-synの生理学的機能を必ずしも阻害することなく、小さな毒性のオリゴマー凝集及び大きな下流のプロトフィブリルの両方の形成を阻害する。治療介入のこの初期の時点で膜に組み込まれた二量体の形成を阻害することにより、不可逆な神経変性プロセスが開始する前の段階で、シナプス機能に対するα-synの有害な作用を逆転させる可能性が大いにある。細胞膜において安定化しているα-syn構造を特異的に標的化することにより、より特異的な分子標的化薬物の設計が可能となる。電子顕微鏡研究により、本発明の化合物が、脂質膜マトリクスにおける球状オリゴマーの形成を低減させ、イムノブロットにより二量体形成が低減したことが実証された。従って、該化合物は、α-syn膜結合並びに伝播するα-syn二量体及び小さいオリゴマーのアッセンブリを低減させることにより達成される、ニューロンにおけるα-syn凝集の形成の予防において有用である。
本発明の化合物は、ヒト及び動物の身体における、毒性のあるタンパク質凝集(例えば、α-synオリゴマー形成)の形成を初期に特異的に阻害する。これらの化合物は、例えば、オリゴマー形成を完全に阻害することにより、又は、既に形成されたオリゴマー(例えば三量体及び四量体)のさらなる成長及び環状構造(例えば五量体、六量体)の形成を阻害することにより、真逆のα-syn凝集を特異的に標的とする。本発明の一つの実施形態において、本発明の化合物は、α-syn二量体又は小さなα-synオリゴマーのN末端ドメインと相互作用する。他の実施形態において、該化合物は、単量体で可溶性形態のα-synのN末端ドメインに結合する。好ましくは、該化合物は、少なくともμMオーダーの親和性でα-synと結合し、より好ましくは少なくともnMオーダーの親和性で結合する(等温滴定熱量測定(ITC)、熱蛍光(ThermoFluor)及びNMR分光法などの生物物理学的方法を用いて測定される場合)。
本発明の化合物は、膜中で折りたたまれた状態のα-synをより選択的に標的化することが実証された。NMR研究によって確認された通り、本発明の化合物の相互作用は、遊離α-synよりも寧ろ膜結合コンフォーマーとである。このことは、該化合物が、通常、細胞質画分において小胞と緩やかに相互作用している生理学的な立体構造のα-synよりも寧ろ、病的な形態のα-synを標的化することを示唆した。さらに、該化合物は、α-syn トランスジェニック(tg)マウスにおいて、行動及びシナプス障害を改善した一方で、非tgマウスにおいては副作用を有しなかった。同様に、神経病理学研究及び超微細構造研究によって、非tgマウスにおいてはシナプス小胞及びシナプス末端が影響を受けていないことが確認された。さらに、該化合物は、α-synトランスジェニック(tg)マウスにおいて、Rabl, R.らの文献(2016年)で説明されているビームウォーキング及びpasta gnawing試験によって評価される、行動-運動成績を改善した。また、該化合物はtgマウス及び非tマウスにおいて副作用を示さなかった。
さらには、本発明の化合物は好ましくは血漿及び溶液中で安定であり、容易に血液脳関門を通過することができる。
本発明の化合物の第二の機能は、リポカリン2(Lcn2/NGAL)に結合することである。好ましくは、該化合物はLcn2/NGALに少なくともμMオーダーの親和性で結合し、よりさらに好ましくは、少なくともnMオーダーの親和性で結合する(等温滴定熱量測定(ITC)、熱蛍光及びNMR分光法などの生物物理学的方法を用いて測定される場合)。Lcn2はまた、がん遺伝子24p3又は好中球ゼラチナーゼ関連リポカリン(NGAL)としても知られ、ヒトにおいてはLCN2遺伝子によってコードされるタンパク質である。Lcn2は、好中球で発現し、呼吸器及び消化管の上皮並びに腎臓及び前立腺において、低レベルで発現している。
本発明者は、等温滴定熱量測定(ITC)及び核磁気共鳴(NMR)分光法を用いて、Lcn2タンパク質への本発明の化合物の可能な結合を調べた。ITC及びNMRにより、リガンド結合が明白に実証され、結合親和性についての定量的情報(KD約700 nM)及び結合部位の位置が明らかになった(図7参照)。興味深いことに、そして最も重要なことに、観察された結合部位は、該化合物の結合は、Lcn2の細胞内同族受容体の1つへの結合と競合的であることを示唆している。
本発明の化合物は、患者(好ましくはヒト患者)に治療有効用量投与される。このような有効用量は、神経変性疾患に関する状態の治癒、予防又は緩和をもたらすのに十分な化合物量に関する。有効用量は、治療を受ける個体の健康状態(health and physical condition)、治療を受ける固体の分類群、組成物の処方、医学的状態の評価及びその他の関連因子によって変化するだろう。
本発明の好ましい実施形態によると、本発明の化合物の単回用量は、約0.01mgから約5.0g、好ましくは約0.05mgから約2g、より好ましくは約0.5mgから約1g、更により好ましくは約1mgから約500mgである。本発明の化合物は、体重1kgあたり、約0.01mgから約5g、好ましくは約0.05mgから2g、さらに好ましくは約0.5mgから1g、更により好ましくは約1mgから約500mg患者に投与することができる。
適した投与経路は、例えば、経口投与、直腸投与、経粘膜投与若しくは経腸投与、筋肉内注射、皮下注射、髄内注射及び髄腔内注射、直接心室内注射、静脈内注射、腹腔内注射若しくは鼻腔内注射を含む非経口送達を含み得る。本発明の医薬組成物において使用される又は本発明の方法を行うための本発明の化合物の投与は、種々の従来の方法、例えば、経口摂取、吸入、局所適用又は皮膚、皮下、腹腔内、非経口若しくは静脈内注射で実施することができる。患者への静脈内投与及び経口投与が好ましい。
本発明で予防又は治療される神経変性疾患は、好ましくはシヌクレイノパチーである。シヌクレイノパチー(α-シヌクレイノパチーともいう)は、ニューロン、神経繊維及びグリア細胞におけるα-シヌクレインの異常蓄積によって特徴づけられる神経変性疾患である。
シヌクレイノパチーは、パーキンソン病、レビー小体型認知症及び多系統萎縮症から成る群から選択されてよい。さらには、本発明の化合物によって治療/予防される神経変性疾患は、筋萎縮性側索硬化症及びハンチントン病である。
本発明に化合物によって達成されうるさらなる効果は、ニューロン増殖の促進、アストロサイトの制御された活性化及び/又は静止状態のアストロサイトから反応性ストロサイトへの転換の阻害である。前記の通り、アストロサイトの制御された活性化は、ニューロンには有益であると考えられているが、過活性のアストロサイトは有害でありうると考えられる。本発明の化合物のこの効果は、恐らくLcn2/NGALへの結合機能に関連している。本発明の化合物のLcn2/NGALへの結合部位は、細胞内同族受容体の1つへのLcn2/NGALの結合と競合的である。本発明の化合物は、Lcn2/NGALの神経毒活性に対する阻害効果が実証されている。
ニューロン細胞培養物中における該化合物の効果をさらに研究するために、ヒト神経細胞芽細胞腫SH-SY5Y細胞株をドーパミン作動性ニューロンのin vitroモデルとして使用し、細胞毒性アッセイを開発した。第一に、RA-PMA最終分化SH-SY5Y細胞は、用量依存的にLcn2/NGAL細胞毒性に対して感受性であることが示された。さらに、H2O2によって引き起こされる活性酸素種(ROS)の存在下で、Lcn2/NGAL細胞毒性はより強くなる。第二に、SH-SY5Y最終分化細胞は、細胞膜上でLcn2/NGAL同族受容体(SLC22A17)を約5倍多く発現していることが示された。この結果は、分化SH-SY5Y細胞がLcn2/NGALに対してより感受性である理由を説明するものである。
さらには、スルホカリックスアレーンが、ヒト分化SH-SY5Y神経芽細胞腫細胞において、神経毒性タンパク質Lcn2/NGALに対する神経保護特性を有することが示された。よりさらには、スルホカリックスアレーンが、ヒト分化SH-SY5Y細胞において、MPP+(ドーパミン作動性ニューロンに選択的に取り込まれ、ミトコンドリア電子伝達鎖の複合体Iを阻害するドーパミントランスポーターの基質)及びH2O2ストレスからも保護したことも示された。酸化ストレス及びミトコンドリア障害は、複数の神経変性疾患の病因に関係している。我々の結果は、スルホカリックスアレーン(500μM及び100μM)が、MPP+(1-メチル-4-フェニルピリジニウム)及びH2O2によって誘導される細胞生存率の低下を軽減することを示している。加えて、典型的な位相差画像は、MPP+及びH2O2によって誘導される形態変化の保護効果を示した。
好ましい実施形態において、本発明の化合物はカリックスアレーン骨格を含む。カリックスアレーンは一般に、フェノール及びアルデヒドのヒドロキシアルキル化物に基づく大員環又は環状オリゴマーとして定義される。カリックスアレーンは、三次元バスケット、カップ又はバケツ型によって特徴づけられる。カリックス[4]アレーンにおける内部容積は約10立方オングストロームである。カリックスアレーンは、幅広の上部縁、細い下部縁及び中央の輪によって特徴づけられる。出発物質としてのフェノールの4つのヒドロキシル基は、下部縁上で環内にある。レゾルシン[4]アレーンにおいて、8つのヒドロキシル基は、上部環において環外に位置する。カリックスアレーンは、メチレン架橋の周りの回転が可能であるため、異なる化学配座で存在する。
第二の態様において、本発明は、上記で定義した化合物の有効量を含有する医薬組成物を特定する。当該医薬組成物は、好ましくは一つ以上の薬学的に許容される助剤を含み、活性な医薬化合物を高い生物学的利用能で投与することができ、化合物の血液脳関門通過を助ける医薬形態である。好適な助剤は、例えば、シクロデキストリンに基づくものであってよい。好適な処方は、例えば、アクリレート、メタクリレート、シアノアクリレート、アクリルアミド、ポリアクテート、ポリグリコレート、ポリアンヒドリド、ポリオルトエステル、ゼラチン、アルブミン、ポリスチレン、ポリビニル、ポリアクロレイン、ポリグルタルアルデヒド及びそれらの誘導体、コポリマー並びにそれらの混合物から成る群から選択されるポリマーで形成された合成ポリマーナノ粒子を組み込んでよい。
従って、好ましい実施形態において、本発明は、医薬活性成分として本発明の化合物の有効量を含有し、又は、唯一の医薬活性成分としてその化合物を含み、一つ以上の薬学的に許容される助剤を含有する医薬組成物に関する。
本発明はまた、本発明の化合物を一つ以上の他の薬学的に活性な化合物と組み合わせて投与することを含む、神経変性疾患の併用療法にも関する。特に、本発明の一つの態様においては、
(i) 本発明の第一の化合物、及び
(ii) 神経変性疾患を予防又は治療するために使用される第二の化合物;
の有効量、並びに、一つ以上の薬学的に許容される助剤を含み、治療を必要とする患者に、別々に、連続で又は同時に投与するための、キットを特定する。場合により、第二の化合物は、レボドパ、ドーパミンアゴニスト、モノアミンオキシダーゼインヒビター、抗コリン作用薬グルタミン酸アンタゴニスト、カテコール-C-メチルトランスフェラーゼ(COMT)インヒビター及びDOPAデカルボキシラーゼインヒビターから成る群から選択されてよい。
(i) 本発明の第一の化合物、及び
(ii) 神経変性疾患を予防又は治療するために使用される第二の化合物;
の有効量、並びに、一つ以上の薬学的に許容される助剤を含み、治療を必要とする患者に、別々に、連続で又は同時に投与するための、キットを特定する。場合により、第二の化合物は、レボドパ、ドーパミンアゴニスト、モノアミンオキシダーゼインヒビター、抗コリン作用薬グルタミン酸アンタゴニスト、カテコール-C-メチルトランスフェラーゼ(COMT)インヒビター及びDOPAデカルボキシラーゼインヒビターから成る群から選択されてよい。
本発明はさらに、ヒト及び/又は動物の身体を治療するための方法に関する。一つの実施形態において、本発明は、それを必要とするヒトに対して、
(i)α-シヌクレイン(α-syn)凝集の形成阻害、及び
(ii)リポカリン2(Lcn2/NGAL)への結合
が可能な化合物の有効量を投与することを含む、神経変性疾患を治療又は予防するための方法に関する。

本発明はさらに、神経変性疾患の予防又は治療において使用するための、カリックスアレーン骨格を含む化合物に関する。好ましくは、神経変性疾患の予防又は治療において使用するための化合物は、スルホカリックスアレーンのナトリウム塩又はその誘導体である。より好ましくは、治療は、約0.01mg~5.0g/kg体重の量で、化合物を患者に投与することを含み、好ましくは、化合物の投与は、静脈内投与又は経口投与である。
更なる実施形態において、本発明は、それを必要とするヒトに対して、カリックスアレーン骨格を含む化合物の有効量を投与することを含む、神経変性疾患の治療又は予防のための方法に関する。好ましくは、本発明は、それを必要とするヒトに対して、スルホカリックスアレーンのナトリウム塩又はその誘導体の有効量を投与することを含む、神経変性疾患の治療又は予防のための方法に関する。さらに好ましくは、治療は、約0.01mg~5.0g/kg体重の量で、化合物を患者に投与することを含み、よりさらに好ましくは、化合物の投与は、静脈内投与又は経口投与である。
以下の非限定的な例で、本発明をさらに説明する。
ミセルに結合したα-シヌクレインとのスルホカリックスアレーン相互作用のNMR研究
実験で使用したリポソームは、1-ヘキサデカノイル-2-(9Z-オクタデセノイル)-sn-グリセロ-3-ホスホ-(1'-rac-グリセロール)(ナトリウム塩)のみを含み、したがってPOPGと命名した。POPGは、Avantis Polar Lipidsより、クロロホルムに溶解した5mg/mlのストックとして、購入した。ストック溶液をガラスバイアルに一定量ずつ分注し、窒素フロー下でクロロホルムをほぼすべて蒸発させ、その後サンプルを真空下で最低45分間乾燥させた。POPGを所望のバッファーに溶解させて1mg/mlとし、室温(RT)で1時間維持し、完全に溶媒化させるために、3回の凍結-融解サイクルに供した。懸濁液をバスソニケーター中で5分間超音波処理し、0.4μmの膜を備えた押出機(Avanti(登録商標) mini-Extruder)で20回循環させた。
高濃度の測定を可能にするため、得られた溶液を2000xgで10分間遠心分離し、上清の75%を廃棄し、ペレットを残り25%に再懸濁して4mg/ml POPGの溶液を得た。この溶液を、更なる測定で使用するまで、一定量ずつ凍結した。凍結及び濃縮手順の両方についてリポソームの完全性を検証するために、動的光散乱(dynamic light scattering:DLS)測定法を実施し、未濃縮の新鮮なリポソームと、濃縮した凍結ストックとを比較した。新鮮なリポソームと凍結ストックされたリポソームとα-synとに対するこれらの測定とNMR測定では、出願人による処理で生じた測定可能な差異を示されなかった。
すべてのNMR測定について、タンパク質を、20mMリン酸、pH7.4、100mM NaClで透析した。280nmにおける吸光度からタンパク質濃度を概算した。サンプルの純度及び安定性は、SDS-PAGEで確認した。NMRスペクトルをVarian Direct Drive 600 MHz及びVarian Inova 800MHzスペクトロメーターにおいて、10% D2Oをロック溶媒として記録した。スペクトルは、NMRPipeを使用して処理した (F. Delaglio, S. Grzesiek, G. W. Vuister, G. Zhu, J. Pfeifer, A. Bax, NMRPipe: A multidimensional spectral processing system based on UNIX pipes. J Biomol NMR 6, 第277-293頁、1995年)。α-シヌクレインは0.12 mMで使用し、POPG-リポソームは0.8 mg/mlで添加した。遊離(apo)α-シヌクレインで得られた測定値は、約0.12mMの同じ濃度を使用した。特定のアミノ酸残基位置について観察された化学シフト変化をマッピングすることによって、スルホカリックスアレーンと単量体(可溶性)α-シヌクレインの相互作用を観察した。図1(下部分)から理解される通り、最も顕著な化学シフト変化は、α-シヌクレインのN末端部分に集まっている。全ての1H-15N相関スペクトルを、120μMのサンプルについてSOFASTパルスシーケンス(P. Schanda, E. Kupce, B. Brutscher, SOFAST-HMQC experiments for recording two-dimensional heteronuclear correlation spectra of proteins within a few seconds. J Biomol NMR 33、第199-211頁、2005年)で記録し、Rance-Kayで、40μMのサンプルについて、増幅されたHSQCSの感度を検出した(J. Cavanagh, A. G. Palmer, P. E. Wright, M. Rance, Sensitivity improvement in proton-detected 2-dimensional heteronuclear relay spectroscopy. J. Magn. Reson. 91, 429-436 (1991); L. E. Kay, P. Keifer, T. Saarinen, Pure absorption gradient enhanced heteronuclear single quantum correlation spectroscopy with improved sensitivity. J Am Chem Soc 114, 第10663-10665頁、1992年)。生理学的状態に近い共鳴帰属は、過去の刊行物から容易に入手可能であった (J. N. Rao, Y. E. Kim, L. S. Park, T. S. J Mol Biol 390, 516-529 (2009)) (BMRB ID 16300)。最新のNMR分光法を用いてLcn2に結合しているスルホカリックスアレーンを調べた(実験条件は、α-シヌクレインNMR実験と類似していた)。
等温滴定熱量測定(Isothermal Titration Calorimetry:ITC)
遊離及び結合したスルホカリックスアレーンのLcn2への結合は、Microcal ITC200 マイクロカロリメーターを使用し、ITCによって決定した。実験を、25°Cで、20mMTris(pH7.4)、50mMNaCl中で行った。参照細胞はMilli Q水を含有した。反応性細胞中でのLcn2の濃度は50μMであった。シリンジ内のスルホカリックスアレーンの濃度は500μMであった。滴定は、撹拌速度800rpmで、300秒間隔で分離された、4μLの19回連続注入で構成された。データ分析は、Origin softwareで行い、単一の結合部位が推定された。

図1は、スルホカリックスアレーンのα-syn構造及び脂質結合に対する効果のNMR解析を示している。α-synの高分子量リポソームへの結合は、顕著なシグナル減衰をもたらす。しかしながら、リポソーム結合状態における、残りの分子内の可塑性は、残基特異的な強度変化をもたらす。α-synの膜結合は、2つの結合様式(N-末端ドメイン又はN末端及び中央のNACドメインのいずれかに作用する)を介して進行する。α-synのC末端(残基125番超)ドメインは、両方の結合様式において配座の柔軟性を保持しており、従って、NMRスペクトルにおいて最も高いシグナル強度を示す。(上) (120mM)のPOPG-リポソーム(0.8mg/ml)(黒)でのシグナル減衰パターン(強度比対残基位置)。スルホカリックスアレーン(赤)の添加により、部分的にリポソームからα-synが放出される(特に、N-末端でシグナル強度の増大を介して観察される)。(下)α-synにおけるスルホカリックスアレーンの結合部位のマッピング。スルホカリックスアレーンの結合は、残基特異的な化学シフト変化をもたらす(NMRスペクトルにおいて観察された)。最も顕著な変化は、N末端ドメイン(1-40)の残基について観察された。

- カリックスアレーン骨格を含む、神経変性疾患の予防又は治療において使用するための、請求項1~11のいずれか一項に記載の化合物。
- スルホカリックスアレーンのナトリウム塩又はその誘導体である、神経変性疾患の予防又は治療において使用するための、請求項12に記載の化合物。
- 治療が、約0.01mg~5.0g/kg体重の量で、化合物を患者に投与することを含み、好ましくは、化合物の投与は、静脈内投与又は経口投与である、神経変性疾患の予防又は治療において使用するための、請求項1~13のいずれか一項に記載の化合物
- (i)カリックスアレーン骨格を含む第一の化合物;
(ii) レボドパ、ドーパミンアゴニスト、モノアミンオキシダーゼインヒビター、抗コリン作用薬グルタミン酸アンタゴニスト、カテコール-C-メチルトランスフェラーゼ(COMT)インヒビター及びDOPAデカルボキシラーゼインヒビターから成る群から選択される第二の薬学的に活性な化合物;
