Abstract
Description
本発明は、CD137に結合する特異的結合メンバーに関する。この特異的結合メンバーは、特異的結合メンバーの定常ドメイン中に位置するCD137抗原結合部位を含み、例えば、癌及び感染症の治療に利用される。
細胞シグナル伝達は、全ての生物の生命に不可欠な部分であり、通常、可溶性リガンド又は表面で発現されるリガンドと相互作用する細胞表面受容体を必要とする。この相互作用は、受容体、リガンド又はその両方に変化をもたらす。例えば、リガンド結合は、受容体を一緒にクラスター化させて二量体又はオリゴマーにする、受容体の構造変化を誘発し得る。次に、このクラスター化効果が、細胞内シグナル伝達経路の活性化をもたらす。CD137などの、腫瘍壊死因子受容体スーパーファミリー(TNFRSF)のメンバーを含む、このように活性化される多くの受容体がある。

実施例1:ヒト、マウス及びカニクイザル抗原の産生、特徴付け及び選択
1.1 組み換え抗原
腫瘍壊死因子受容体スーパーファミリー(TNFRSF)メンバーは、それらの同種リガンドに結合される場合に一緒にクラスター化する多量体を形成する傾向が知られている(Croft,M.2003)。それらの機能性のために集合するこの傾向により、ファージ及び酵母ディスプレイなどのインビトロ選択に使用するため、並びに選択されたタンパク質の特徴付けのために溶液中に集合しない可溶性組み換えタンパク質を産生することが困難になる。
完全長マウスCD137(配列番号184)又はヒトCD137(配列番号180)を発現するDO11.10細胞(National Jewish Health)(それぞれ、「DO11.10.mCD137」及び「DO11.10.hCD137」と示される)を、表2に列挙されるように、選択されたFcabの選択及びさらなる特徴付けのために、その天然形態に最も類似した膜結合構造で抗原を表すために産生した。
ヒトCD137に結合するFcabを発見し、同定された結合剤の多様性を最大にするために、酵母及びファージディスプレイ選択作業の両方を用いた。CD137は、いくつかの非免疫細胞型において低いレベルで発現されるため、活性化T細胞などの、CD137を多量に発現した細胞を選択的に標的とする抗ヒトCD137 Fcabを選択することが決定された。理論に制約されるのを望むものではないが、高度に上方制御されたCD137発現を有する活性化T細胞と異なり、非常に低いか又はごくわずかなレベルのCD137発現を有する細胞は、それらの細胞表面においてモノマー状態のCD137を有する可能性がより高いと仮定され、その場合、タンパク質の大部分が、細胞表面において二量体、三量体、又はより高次の多量体状態で存在することが予想される。
ヒトIgG1のCH3ドメインを示す6つのナイーブファージライブラリーを、ファージディスプレイによる選択のために使用した。6つ全てのライブラリーは、ランダム化ABループ(IMGT番号付けに従って14〜18位に残基を含む)及びランダム化EFループ(IMGT番号付けに従って92〜101位に残基を含む)を含んでいた。ライブラリーの1つは、EFループの101位(挿入される残基は、IMGT番号付けに従って101.4〜101.1位にある)に2つ又は4つのアミノ酸(2つ又は4つのNNKコドンによってコードされる)の挿入を伴うクローンを含んでいた。
ヒトIgG1のCH1からCH3ドメインを示す4つのナイーブ酵母ライブラリーを、酵母ディスプレイによる選択のために使用した。4つ全てのライブラリーが、CH3ドメイン中にランダム化ABループ(IMGT番号付けに従って14から18位に残基を含む)及びランダム化EFループ(IMGT番号付けに従って92から101位に残基を含む)を含んでいた。ライブラリーのうちの2つは、CH3ドメインのABループ中の16位に5アミノ酸残基(IMGT番号付けに従って16.5から16.1位における残基)の挿入をさらに含んでいた。
3.1 「モック」mAb2フォーマットにおける抗ヒトCD137 Fcabの調製
ファージから単離された76個の抗ヒトCD137 Fcabクローン及び酵母選択から単離された9個のクローンを含むIgG1分子からなる「モック」mAb2抗体を、mAb2フォーマットにおけるFcabの特徴付けを可能にするために産生した。AB、CD及びEFループを含むCH3ドメインFcabの部分を、抗ニワトリ卵リゾチーム抗体HelD1.3のCH3ドメインの対応する領域と置き換えることによって、モックmAb2を調製した。HelD1.3抗体の生成が、Tello et al.1993に記載されている。抗体HelD1.3の重鎖及び軽鎖配列はそれぞれ、配列番号186及び173に示される。モックmAb2分子を、HEK293−6E細胞内での一過性発現によって産生した。産生されたタンパク質の量を評価するために、IgGタンパク質含量を、PALL(18−5021)製のプロテインA定量バイオセンサーを用いたOctet QKeプラットフォームを用いて、生体層干渉法によって定量した。タンパク質を、mAb SelectSureカラムを用いて、プロテインA親和性クロマトグラフィーによって精製した。53個のファージで駆動されたCD137 mAb2タンパク質が、検出閾値未満の測定を示し、したがって、さらなる分析に適していないことが分かった。32のmAb2を、mAb Select SuReプロテインAカラム(GE Healthcare、11003494)を用いて精製した:FS22−005、FS22−007、FS22−033、FS22−042、FS22−049、FS22−050、FS22−052、FS22−053、FS22−054、FS22−167、FS22−169、FS22−170、FS22−171、FS22−172、FS22−173、FS22−174、FS22−175、FS22−176、FS22−177、FS22−178、FS22−179、FS22−180、FS22−181、FS22−183、FS22−184、FS22−186、FS22−187、FS22−191、FS22−192、FS22−193、FS22−194、FS22−195。
精製されたモックmAb2分子(クローンFS22−175を除く)のうちの31個を、Octet QKeプラットフォームを用いた生体層干渉法によって、単一点結合実験においてヒト組み換え抗原への結合について試験した。ストレプトアビジンBLIバイオセンサー(PALL 18−5021)を用いて、キネティックバッファー(kinetic buffer)(PALL)中10μg/mlでビオチン化hCD137−mFc−Avi抗原を捕捉した。次に、センサーを、240秒間にわたって、同じキネティックバッファー中で1:1に希釈された精製されたmAb2を含むウェル中に浸漬した後、240秒間にわたって1×キネティックバッファーを含むウェル中に浸漬した。結合ヒットを、緩衝液及び野生型IgG1対照HelD1.3 mAb(G1/HelD1.3)を超えるBLI応答によって定義される単純なBooleanのあり/なし(yes/no)基準で分類した:12のmAb2は、結合せず、19は、CD137被覆センサーに結合した(FS22−007、FS22−033、FS22−042、FS22−049、FS22−050、FS22−052、FS22−053、FS22−054、FS22−169、FS22−172、FS22−173、FS22−174、FS22−179、FS22−180、FS22−181、FS22−183、FS22−187、FS22−194、FS22−195)。
多量体化及びクラスター化が、TNFRシグナル伝達のために必要とされる(Bitra et al,2017)。CD137はクラスター化し、その同種リガンド、CD137Lと相互作用すると、NF−κBシグナル伝達経路を活性化する。アゴニスト分子は、CD137のクラスター化及び活性化を引き起こし、それによって、NF−κBシグナル伝達経路を活性化する際、リガンドを模倣する。いくつかのアゴニスト抗体(例えばウレルマブ)は、結合時にCD137のクラスター化を元々引き起こし得ることが知られている一方、ウトミルマブなどの他のものは、CD137のクラスター化を誘発するために抗体自体のさらなる架橋を必要とする(Fisher et al,2012)。エフェクター細胞におけるFc γ受容体は、インビボでこのような架橋を誘発することが知られているが、これは非効率的であり、治療対象の部位から離れて起こり得る。用量制限毒性は、いくつかの抗CD137抗体による治療に関連付けられているため、固有に作用する(agonise)能力を有さなかった抗CD137結合Fcabを選択するが、CD137のクラスター化を誘発するためにさらなる架橋を必要とするもののみを選択することが決定された。したがって、架橋された抗体によって細胞表面において発現されるCD137のクラスター化時に細胞内のNF−κBシグナル伝達経路の活性化を検出し得るが、抗体が架橋されていなかった場合、活性をほとんど示さなかったアッセイを創出した。次に、このアッセイを用いて、FcabがBLIによって組み換え抗原によって結合することが分かったか否かにかかわらず、モックmAb2フォーマットにおける27個の抗CD137 Fcabクローン、及び抗CD20 mAb2フォーマットにおける6個の抗CD137 Fcabクローンのアゴニスト機能活性を試験した。
活性化T細胞におけるアゴニスト分子を介したCD137のクラスター化は、T細胞活性化及び下流のシグナル伝達を引き起こし、それにより、限定はされないが、IL−2産生をもたらす。FS22−053及びFS22−172が、NFKBレポーターアッセイにおいて活性を有することが確認されたため、CD137を活性化するそれらの能力を、T細胞活性化アッセイにおいて試験した。ヒトCD137を過剰発現するように操作されたDO11.10 T細胞を用いたDO11.10 T細胞活性化アッセイを創出し、T細胞活性化を、IL−2放出を測定することによって評価した。
一連の抗CD137 Fcab(FS22−053、FS22−175、FS22−176、FS22−177、FS22−178、FS22−179)を、より生物学的に関連したmAb2フォーマットにおけるFcabの特徴付けを可能にするために産生した。AB、CD及びEFループを含むCH3ドメインFcabの部分を、抗CD20 2F2クローン(米国特許第8,529,902 B2号からの)のCH3ドメインの対応する領域と置き換えて、CD137及びCD20の両方に結合したmAb2を産生することによって、mAb2を調製した。これらのCD137/CD20 mAb2を、HEK293−6E細胞内での一過性発現によって産生し、mAb Select SuReプロテインAカラムを用いて精製した。
実施例3.4には、抗CD137モックmAb2がプロテインLを用いて架橋されたT細胞活性化アッセイが記載されている。その設定は、かなりの数の分子をスクリーニングするために確実で且つ再現性のある方法を提供したが、架橋される場合に抗体及びmAb2によって形成されるより高次の構造は準最適であり、生理環境の代表ではない。より生物学的に関連した設定は、mAb2分子が、生体系中に存在する標的へのそのFabアームの結合によって架橋される設定である。この細胞ベースのインビトロシステムは、細胞膜に結合するFabを介して抗体及びmAb2の提示を最適化し、抗体のクラスター化を引き起こし、これが、ひいてはT細胞におけるその標的に対するCD137 Fcabアームの親和性を増大する。
上述されるように、3つのクローンが、NF−κBレポーターアッセイ、DO11.10 T細胞活性化アッセイFS22−053及びFS22−172)においてそれらの機能特性に基づいて親和性成熟のために選択され、又はCD137(FS22−033)の異なる領域に結合するものと考えられた。
2つの酵母及び2つのファージディスプレイされたライブラリーを、FS22−033クローンから構築し、一方は、CH3ドメインのABループ中の5つの残基をランダム化することによるものであり、他方は、ELLAプライマーを用いてCH3ドメインのEFループ中の5つの残基をランダム化することによるものである。ELLAプライマーは、各アミノ酸に使用されるコドン及び混合物内のそれらの相対存在量を規定する。システインのみが、混合物から除外され、いずれの他のアミノ酸に対しても偏りがなかった。
4つの酵母ディスプレイされたライブラリーを、FS22−053及びFS22−172 Fcabクローンから構築した。7つの残基(IMGTに従って15〜16.1位における)を、実施例4.1に記載されるものと同じトリヌクレオチド分布で、各クローンのCH3ドメインのABループ中のELLAプライマーを用いてランダム化して、ライブラリーFS22−053 AB及びFS22−172 ABを作成した。5つの残基(IMGTに従って92〜94及び97〜98位における)を、CH3ドメインのEFループ中のELLAプライマーを用いてランダム化して、ライブラリーFS22−053 EF及びFS22−172 EFを得た。
5.1 「モック」及びモデルmAb2フォーマットにおける抗ヒトCD137 Fcabの構築
親FS22−053クローンから得られた16個の親和性成熟したクローン(FS22−053−001からFS22−053−016)、及び親FS22−172クローンから得られた6つのクローン(FS22−172−001からFS22−172−006)を、mAb2フォーマットにおいて調製した。クローンFS22−053−001からFS22−053−007は、さらなる試験及び特徴付けのために下流の精製を可能にするレベルでmAb2フォーマットを発現しなかったため、さらに進めなかった。残っているクローンは、それが得られる親クローンのCH3配列と比較した際に、それらのCH3ドメインにおける少なくとも95%の配列同一性を有していたことが分かった。配列類似性マトリックスパーセンテージを、各アミノ酸位置を、参照配列(親クローンFS22−053又はFS22−172)のものと比較することによって生成した。(図1Dは、親FS22−053 CH3ドメインと比較した、クローンFS22−053−008からFS22−053−016及びFS22−053−017(実施例10.1を参照)のCH3ドメインの配列同一性パーセンテージを示す。図1Eは、親FS22−053 CH3ドメインと比較して、クローンFS22−172−001からFS22−172−006のCH3ドメインの配列同一性パーセンテージを示す。
表3に列挙されるモックmAb2(HelD1.3)フォーマットにおける親和性成熟した抗ヒトCD137 Fcabの機能活性を、実施例3.3に記載される同じNF−κBルシフェラーゼアッセイにおいて試験した。発光を、Gen5 Software、BioTekを用いて、プレートリーダーにおいて、0.5秒間の積分時間で測定した。このアッセイの結果が、表3及び図2に示される。予想どおり、Fcabのいずれも、プロテインL架橋(−XL)なしで活性を示さなかった。全ての親和性成熟したCD137 Fcabは、親CD137 Fcabを上回る大幅な改善を示し、親CD137 Fcabは、このアッセイにおいて陽性である一方、EC50値の計算が可能でなかった(実施例3.3を参照)。FS22−053−008及びFS22−172−003は、プロテインL(+XL)と架橋される場合、最も低いEC50で、各ファミリーからの最良の活性を示した。
表4に列挙されるモデルmAb2(PD−L1 LALA)フォーマットにおける親和性成熟したヒトFcabの機能活性を、実施例3.6に記載されるアッセイと同様の、DO11.10 T細胞活性化アッセイにおいて試験した。
CD137を過剰発現するHEK細胞においてCD137を活性化するFcabの活性が、実施例5.3に示された。CD137を過剰発現するように操作されていない細胞におけるFcabの活性を試験するために、初代ヒトT細胞アッセイが必要とされた。活性化された細胞傷害性CD8+ T細胞は、癌細胞を直接死滅させるのに関与し、それらの細胞表面においてCD137を発現させる(Ye et al,2014)。CD137のクラスター化は、下流のシグナル伝達及びさらなるCD8+ T細胞活性化を誘発するのに不可欠であることが知られている。したがって、CD8+ T細胞活性化アッセイを用いて、CD137のクラスター化及びその後の下流のシグナル伝達を引き起こすFcab(以下に詳述されるようにmAb2フォーマットにおける)の能力を評価した。CD8+ T細胞活性化を、IL−2の放出によって決定した。
他の関連するTNFSFRファミリーメンバーと比較した、ヒトCD137に対する抗ヒトCD137 Fcabの特異性を試験した。Fcabのうちの8つを、モックmAb2(HelD1.3)フォーマットにおいて試験し、他のヒトTNFRSF受容体:CD40、OX40及びGITRへの結合について試験することによって、Biacore T200(GE Healthcare)においてSPRによって測定した。アミンカップリング(アミンカップリングキット、GE Healthcare、BR−1000−50)を用いて、ヒトCD40、GITR及びOX40を、Biacore CM5チップ(GE Healthcare、カタログ番号29149603)中で約1000RUになるまで被覆した。1μMから開始するモックmAb2フォーマット(FS22−053−008/HelD1.3、FS22−053−009/HelD1.3、FS22−053−010/HelD1.3、FS22−053−011/HelD1.3、FS22−053−012/HelD1.3、FS22−053−014/HelD1.3、FS22−172−003/HelD1.3、FS22−172−004/HelD1.3)における抗ヒトCD137 Fcabの希釈物を、HBS−EP+緩衝液(BR100669)中で調製し、30μl/分で3分間にわたって注入し、次に、4分間にわたって緩衝液中で解離させた。30μl/分で12秒間にわたる10mMのグリシンpH2.5の注入によって、チップを再生した。様々なTNFRSFメンバーに特異的な抗体を陽性対照として用いて、Biacoreチップコーティングを確認した。データを二重参照減算し(double reference subtracted)、BIAevaluation 3.2ソフトウェアを用いて分析した。Fcabは、試験されるTNFRSF受容体のいずれにも結合せず、これは、CD137に対するそれらの特異性を実証している。結果として、Fcabがオフターゲットの結合を引き起こすことは予想されない。
ヒト、カニクイザル(cynomolgus)(カニクイザル(cyno))及びマウスCD137に対する抗ヒトCD137 Fcab(FS22−053−008、FS22−053−011、FS22−053−014、FS22−172−004、FS22−172−004)の親和性を、SPRによって測定して、Fcabが動物試験において試験するのに有用であり得るかどうかを決定した。抗ヒトFab捕捉抗体を、製造業者の推奨(GE Healthcare、human Fab capture Kit、#28958325)に従って、6000RUの平均表面密度になるまで、CM5 series Sチップ(GE Healthcare #BR−1005−30)の4つ全てのフローセル上に固定した。30μl/分で60秒間にわたってHBS−EP+緩衝液(GE Healthcare #BR1006−69)中で希釈されたmAb2の3μg/mlの溶液を注入することによって、約150RUになるまで各mAb2を捕捉した。次に、HBS−EP+緩衝液中の様々な濃度のヒト、カニクイザル又はマウスCD137抗原(非ビオチン化ヒト、カニクイザル又はマウスCD137−mFc−Avi又はヒトCD137−Avi−His)を、60μl/分で3分間にわたってチップ上に流し、次に、10分間にわたって解離させた。各抗原濃度の後、チップを、30秒間にわたって30μl/分の流量で10mMのグリシンpH2.1を感染させることによって再生した。参照減算(reference subtraction)のため、最も高い濃度の抗原の前及び最も低い抗原の後、緩衝液HBS−EP+を注入し、ランダムに濃度の1つを2回繰り返した。結合速度を、1:1ラングミュアモデルとフィッティングして、各試料についての平衡結合定数(KD)を生成した。データ分析を、BiaEvaluationソフトウェアバージョン3.2を用いて行った。結果は、表6に示される。
Fcabは、通常、CH3ドメイン中の抗原結合部位とともに2つのホモ二量体Fc鎖を含有する。2つのCH3ドメイン中のこれらの抗原結合部位は、ごく近接しているため、CH3ドメインが、互いに独立してCD137に結合し得るかどうかを決定するために、Fcabの結合の原子価を試験した。ノブ・イントゥ・ホール(knob-into-hole)突然変異(ノブ:T22W、ホール:T22S L24A Y66V)(Atwell S et al,1997)を用いて、FS22−172−003の1つの鎖を、野生型Fcの1つの鎖と組み合わせることによって、単一抗原結合CH3ドメインを含有するヘテロ二量体Fcabを構築した。その結果として、各ヘテロ二量体は、一方の鎖中にFS22−172−003(配列番号139)からのCH3及び他方の鎖中に野生型CH3(配列番号4)を含有していた。Fcabを、mAb2フォーマットにおいて調製した。
実施例2に記載されるように、高いCD137発現レベルで細胞に優先的に結合するようにCD137に結合するFcabを選択した。FS22−172−003の2価結合、したがって強い結合を、実施例5.6においてSPRによって確認した:FS22−172−003は、モノマーCD137と比較して、二量体CD137への高い結合力及びより強い結合を示した。
インビボマウスモデルにおいて抗CD137 Fcabの活性を試験するために、マウスCD137に特異的に結合するFcabを生成し、特徴付けした。
ヒトCD137に結合するFcabの選択について前に使用されたヒトIgG1のCH3ドメインを示す6つのナイーブファージライブラリーを、マウスCD137に結合するFcabの選択に使用し、組み換えマウス二量体CD137又は完全長マウスCD137を発現する細胞を抗原として使用した。
ヒトCD137に結合するFcabの選択について前に使用されたヒトIgG1のCH1からCH3ドメインを示す4つのナイーブ酵母ライブラリーを、マウスCD137に結合するFcabの選択に使用した。
7.1 BLIによる抗マウスCD137 Fcabの特異性の決定
マウスCD137に対する抗マウスCD137 Fcabの特異性を、HelD1.3「モック」mAb2フォーマットにおいて試験し、他のマウスTNFRSF受容体(CD40、OX40、GITR)へのFcabの結合を試験することによって、Octet QKeシステムにおいてBLIによって測定した。10ng/μlのマウスCD40、GITR、OX40受容体(全てR&D Systemsから入手し、Thermoscientific #21328からのEZ−Linkスルホ−NHS−SS−ビオチンキットを用いてビオチン化した)を被覆するためのストレプトアビジンバイオセンサー(PALL ForteBio 18−5021)。モックmAb2フォーマットにおける抗マウスCD137 Fcabを、キネティックバッファー(PALL 18−1092)中で、少なくとも1μMの最終濃度になるまで1:1で希釈した。抗原で被覆されたセンサーを、180秒間にわたってmAb2溶液中に、続いて180秒間にわたって1倍キネティックバッファー中に浸漬した。TNFRSF受容体のそれぞれについての抗体を、陽性対照として使用した。FcabクローンFS22m−055、FS22m−063、FS22m−066、FS22m−075、FS22m−135、FS22m−055、FS22m−063、FS22m−066は、試験されるTNFRSF受容体のいずれにも結合せず、したがって、マウスCD137に対するそれらの特異性を実証している。
マウスCD137配列(配列番号184)を発現するHEK.FRT.luc細胞を、実施例3.3において上述されるのと同じ方法に従って産生した。前に選択された抗マウスCD137 Fcabを含有するmAb2を、実施例3.3に記載される方法に従って、この細胞株、HEK.FRT.luc.mCD137を用いてスクリーニングした。56のmAb2を試験し、そのうちの29がNF−kB活性について陽性であった。LALA突然変異(G1AA/Lob12.3)を有するヒトIgG1 Fcを含有するLob12.3を、陽性対照抗マウスCD137 mAbとして使用し、発光の増加を示し、アッセイの有効性を確認した。LALA突然変異を有するヒトIgG1 Fcを含有するHelD1.3も、このアッセイにおいてヒトIgGモックFabからの干渉を除外するために、陰性対照ヒトIgGアイソタイプとして使用した。EC50を、可能であれば計算し、古典的なシグモイド活性キネティクス(sigmoidal activity kinetics)を示したmAb2を優先して、活性のプラトーに達しなかったmAb2を無視した。mAb2を、EC50及びプロテインLの架橋時の活性の倍率変化の順にランク付けした。FS22m−063を、架橋時の最良のEC50(1.44nM)及び架橋時の活性の最も高い倍率変化(27倍)を有するそれに基づいて選択した。図5は、HelD1.3モックmAb2フォーマットにおける抗マウスCD137 Fcab FS22m−063が、HEK mCD137 NF−kBレポーターアッセイにおいてプロテインLと架橋される場合、CD137のクラスター化及びNF−kBシグナル伝達を引き起こすことを示す。
FS22m−063 Fcabが、インビトロでCD137のクラスター化及び活性化を引き起こすことが可能であることが示されたため、インビボでCD137を活性化するそれらの能力を試験することが望ましかった。
抗マウスCD137 Fcab、FS22m−063、及びPD−L1に特異的なFab領域を含むmAb2を、実施例7.1において産生されたモデルmAb2と同様の方法を用いて調製し、MC38同種マウス腫瘍モデルにおいてインビボ抗腫瘍活性について試験した。
同種マウスモデルは、治療標的を阻害する抗腫瘍効果を試験するための適切なマウスシステムとして認められ、ヒト治療薬の開発を有効にするために広く使用されている。MC38腫瘍が、高度に免疫原性であり、且つ抗CD137抗体単独療法に応答し(Kocak et al,2006)、PD−L1を発現する(Juneja et al,2017)ことが知られているため、MC38同系腫瘍モデルをこの実験において使用した。

FS22m−063 Fcabが、インビトロでCD137のクラスター化及び活性化を引き起こすことが可能であることが示されたため、別のFab標的、この場合、インビボで免疫細胞のみに見られる標的であるPD−1を介してCD137を活性化するそれらの能力を試験することが望ましかった。
抗マウスCD137 Fcab、FS22m−063、及びPD−1に特異的なFab領域を含むmAb2を、実施例7.1において産生されたモデルmAb2と同様の方法を用いて調製し、MC38同種マウス腫瘍モデルにおいてインビボ抗腫瘍活性について試験した。
以下の差異を伴い、実施例8.2に記載されるようにMC38同系腫瘍モデルをこの実験において使用した:
9〜11週齢のC57BL/6雌マウス(Charles River)にマイクロチップを埋め込み、固有の識別子を与えた。各コホートは、12匹のマウスを有していた。各動物に、100μlの無血清培地中で、背部右脇腹に皮下注射される1×106個のMC38結腸癌細胞を与えた。
FS22m−063 Fcab及びPD−L1 Fab(実施例8)を含むmAb2の有効性が示されたため、腫瘍特異的抗原(TAA)、この場合、メソテリン(MSLN)へのそのFabアームの結合を介してmAb2を架橋することによってCD137を活性化するmAb2フォーマットにおけるCD137 Fcabの能力を試験することが望ましかった。このような腫瘍標的化手法は、mAb2の架橋として、T細胞活性化を腫瘍微小環境に限局するのに有益であることが予想され、したがって、CD137のアゴニズムは、MSLNが発現される場合にのみ起こる。
ヒトCD137に結合するFcabのいくつかが、マウスCD137にも結合したことが意外にも分かったため(ヒト、マウス及びカニクイザルCD137への結合の特異性については実施例5.5を参照)、これらのクローンを改善することができたかどうかを調べることが決定された。
マウス−ヒト交差反応性クローンFS22−053−014は、それが、SPRによってマウス二量体CD137抗原に結合することが可能であることが示されたため、選択された(実施例5.5を参照)。しかしながら、さらなる配列分析の際、それは、DGモチーフをもたらす野生型G99位置とのQ98D突然変異の結果として、そのCH3ドメインのEFループ中の翻訳後アスパラギン酸塩異性化をもたらし得る潜在的な配列ライアビリティを有していた。FS22−053系統中の他の親和性成熟したクローン(表12を参照)は、EFループの同じ位置にQ98E修飾を代わりに含んでいた。Q98Dは、製造業者の推奨に従って、QuickChange II突然変異誘発キット(Agilent、カタログ番号200523)を用いた部位特異的突然変異誘発によってQ98Eへと突然変異され、それにより、クローンFS22−053−017が得られた。以下の表12は、クローンFS22−053−008、FS22−172−003及びFS22−172−004のCH3ドメインのEFループ中の高い頻度で存在するLEモチーフ、並びにクローンFS22−053−014のCH3ドメインのEFループ中のDGモチーフを示す。
12.1 ヒトCD137 DO11.10 T細胞活性化アッセイにおけるモックmAb2フォーマットのFS22−053−017 Fcabの活性
FcabクローンFS22−053−017をサブクローニングし、HelD1.3「モック」mAb2として発現させ、次に、実施例3.4に記載されるようにヒトCD137 DO11.10 T細胞活性化アッセイにおいて、同様にHelD1.3 mAb2フォーマットにおけるFS22−053−014クローンと比較した。G1−AA/20H4.9を抗CD137陽性対照として使用し、G1−AA/D1.3をIgG対照として使用した。mAb2を、プロテインL架橋なしで試験するか、又はプロテインLと1:4の比率で架橋させた。
また、FS22−053−017クローン(HelD1.3モックmAb2フォーマットにおける)を、実施例3.4に記載されるようにマウスCD137 DO11.10 T細胞活性化アッセイにおいて、マウスCD137結合FcabクローンFS22m−063(同様にHelD1.3モックmAb2フォーマットにおける)、並びに親FS22−053−014クローン(HelD1.3モックmAb2フォーマットにおける)に対して比較した。mAb2分子を、4:1のモル比(mAb2:プロテインL)でプロテインLと架橋させた。
FcabクローンFS22−053−017の平衡解離定数(KD)を、Biacore T200システムにおいてSPRによって、クローンFS22−053−014の平衡解離定数(KD)と比較した。hCD137−mFc−Avi抗原については、以下の方法を使用した:抗ヒトFab分子を、9,000〜11,000応答単位(RU)の表面密度になるまでCM5チップ上に固定した。抗体を、HBS−EP緩衝液中で4μg/mlになるまで希釈し、30μl/秒の流量で抗Fab分子によって捕捉した。8つの異なる濃度のhCD137−mFc抗原を使用した:200nM;66.67nM;22.22nM(2回含まれていた)、7.41nM;2.47nM;0.82nM、0.27nM:0.091(HBS−EP+緩衝液中で希釈された)を、捕捉されたmAb2上に流した。
実施例5に記載されるように、PPY配列モチーフが、2つの、独立して選択されるFcabのABループにおいて同定され、このモチーフは、分析される全ての親和性成熟したクローンにおいて保存された。したがって、CD137及び全タンパク質構造への結合におけるこのモチーフの関与を理解することが望ましかった。アラニンスキャニングは、タンパク質−タンパク質相互作用における特定の残基の重要性を決定するのに使用される一般的な生物学的技術である。簡潔に述べると、3つのアミノ酸のそれぞれは、順に及び次に一緒に、アラニン残基で置換された。アラニンが、最も化学的に不活性な、嵩高くない残基であると考えられ、したがって結合を助ける可能性が低いと考えられる。
突然変異体クローンを、親クローン(FS22−172−003及びFS22−053−008)における部位特異的突然変異誘発によって産生した。ABループ内のアミノ酸置換の位置は、表16に要約され、全ての他の残基は、それらのそれぞれの親に保存される。実施例3に記載されるように、次に、mAb2変異体を、HEK293−6E細胞内で一過性発現させ、mAb Select SuReプロテインAカラムを用いて精製した。突然変異体の発現収率は、親と同様であり、したがって、これは、アラニン置換がタンパク質産生に影響を与えなかったことを示唆する。
2つの親クローン及び8つの突然変異体クローンの結合速度を、ForteBio製のOctet QKeシステムを用いて比較した。二量体ビオチン化hCD137−mFc−Aviを、10ug/mlでストレプトアビジンセンサー上に捕捉し、次に、200nMの濃度での各mAb2との相互作用を分析した。二量体抗原へのmAb2の結合は、親クローンと比較して、全てのFS22−172−003突然変異体クローンについて低下しており、クローンFS22−172−003_AAA、及びFS22−172−003_APYは、全ての結合を喪失しており、クローンFS22−172−003_PPA、及びFS22−172−003_PAYは、親と比較して有意に減少した(5.5倍及び4.4倍低い応答単位)いくらかの結合を保持していた。FS22−053−008の結合も影響され、変異体PAY及びAAAは、全ての結合を喪失しており、変異体APY及びPPAは、親FS22−053−008クローンと比較した際に、抗原への減少した結合及び会合プロフィールの減速を示す。このデータは、PPYモチーフが、CD137抗原への両方のmAb2の結合にとって重要であることを示唆する。
結合におけるPPYモチーフの関与を考慮すると、CH3ドメインにおけるタンパク質構造予測を評価することは、インシリコでFcabをモデリングするのに有益であった。構造ホモロジーモデリング及びその後の立体配座探索を、MOEソフトウェアスイートの2019.0101バージョン(Chemical Computing Group ULC)を用いて行った。タンパク質データバンク[PDB]構造5JIIにおけるFc領域を、FS22−172−003及びFS22−053−008の両方のFcab領域のための構造テンプレートとして使用した。挿入が、選択された構造テンプレートに関してAB及びEFループ領域中に存在するため、残基側鎖最適化を用いたデノボループ探索を適用して、AB及びEFループ構造を生成した。得られたホモロジーモデルを、エネルギー最小化し、骨格結合長さ、角度、二面角及びキラリティーを含む幾何学的基準に従って採点した。基準を合格したスコアを有する構造ホモロジーモデルを、MOE内で実施されるような、LowModeMDシミュレーション方法を用いた立体配座サンプリングのための基礎として使用した。
