Description
本願は、その内容全体が本願明細書の一部を構成する米国仮出願第61/979,957(出願日:2014年4月15日)に基づく優先権の利益を主張するものである。
鉛(Pb)の放射性同位元素は、治療用および診断用放射性医薬品として有望な用途を有する。例えば、212Pbは有望なα粒子放出源であり、標的化α粒子療法は、この放射の短い光路長(50〜80μm)と高い線エネルギー遷移(100KeV・μm−1)の組み合わせによって、癌治療において多くの利点がある。212Pbで前臨床実験を行うことに関連する1つの課題は、コストが高いこととは別に、212Pb放射性標識治療薬の正確な生体内分布と標的化のアッセイを実行することある。したがって、実行可能な代替策は、そのような実験において代用核種として203Pbを使用することである。203Pbは、好ましい半減期(約52時間)を有し、279keVで80.1%γ線を放出して崩壊し、単光子放出コンピュータ断層撮影法(SPECT)に適合している。これのことにより、この放射性核種は、212Pbをターゲットとする放射性核種療法に適した放射性核種トレーサとして理想的である。従って、203Pbはイメージング、組織分布試験、線量測定データの取得、化学交換の研究に有用である。
鉛(Pb)が放射線(例えば203Pbからの)の非存在下で加熱した水酸化ナトリウムを用いて選択的に溶出できるかどうかを調べるため、各約5mgの銅(Cu)、ニッケル(Ni)およびPbのICP標準を用いてコールド法を実施した。標準液を一緒にプールし、希釈して、タリウム(Tl)なしでターゲット溶液をシミュレートした。
サイクロトロン照射された天然タリウムターゲットを一連の3つのターゲットで使用して、それぞれわずかな改変を加え上記の方法によって、6つの203Pb溶出プロセスを実施した。
目的:以下は、天然のタリウムターゲットおよび存在する他の金属、例えばTl(p、3n)201Pb−201Tlサイクロトロン反応から203Pbを単離するためのプロトコルの例を記載する。
各ステップを実行する際に0〜2PSIの空気圧をカラムに適用することができる。ロードする際には空気圧を0.5PSI未満とする。
1)25mlの硝酸アンモニウム(カラム1)でChelex 100カラムを調製。
2)ガラスウールカラムを準備。
3)pHメーターを較正。
4)プロセスセルにガラスウールカラムとエッチング容器を設置。
5)エッチング容器を慎重に取り外し、新しいエッチング治具を装置に挿入。
6)調製したChelexカラム、ビーカーおよび試薬をプロセスセルに設置。
7)照射された天然タリウムターゲットを得、ターゲット上の放射能を推定する:放射能レンジ=0.5mCi/uAmp時間×uAmp時間〜1.5mCi/uAmp時間×uAmp時間
8)ホットプレート上で約110〜120mlの硝酸を加熱し沸騰させる。沸騰したら、エッチングジグに約90mlを注ぐ。
9)タリウムが完全に溶解するまで、熱1M硝酸中でターゲットをエッチングする。
10)エッチング溶液をガラスウールカラムに通し微粒子を除去。
11)ターゲットとガラスウールカラムを約20mlの熱硝酸でリンス。
12)ガラスウールカラムを廃棄。
13)濃水酸化アンモニウムを用いてエッチング溶液をpH5〜6に調整。必要に応じ1M硝酸で逆滴定。
14)ChelexカラムにpH調整した溶液をロード。
15)Chelexカラムを150〜175mlの1M硝酸アンモニウムで洗浄。
16)150mlのビーカーをホットプレート上に置き、100mlの1M水酸化ナトリウムを加熱し沸騰させる。水酸化ナトリウムが沸騰したらホットプレートを切る。
17)203Pbがカラムから脱離するまで、加熱した水酸化ナトリウム(約90℃の温度)を10−20mlずつ加えカラムから203Pbをストリップ。
18)カラムを廃棄。
19)203Pbストリップ溶液の測定値を取得。セル内イオンチャンバーでカラムを測定することにより完全な脱離を確認。
1)約25mlの硝酸アンモニウムで第2のChelexカラムを調製し、調製したカラムをプロセスセル(カラム2)内に設置。
2)203Pbストリップ溶液を、濃水酸化アンモニウムを用いてpH5〜6に調整。必要に応じ1M硝酸で逆滴定。
3)ChelexカラムにpH調整した溶液をロード。
4)Chelexカラムを150〜175mlの1M硝酸アンモニウムで洗浄。
5)150mlのビーカーをホットプレート上に設置し、100mlの1M水酸化ナトリウムを沸騰するまで加熱。水酸化ナトリウムが沸騰したらホットプレートを切る。
6)203Pbがカラムから脱離するまで、加熱した水酸化ナトリウムを10〜20mlずつ加えカラムから203Pbをストリップ。
7)約25mlの硝酸アンモニウムで第3のChelexカラムを調製し、調製したカラムをプロセスセル内に設置。
8)濃水酸化アンモニウムを用いて第2の203Pbストリップ溶液をpH5〜6に調整。必要に応じ1M硝酸で逆滴定。
9)pH調整した溶液をChelexカラムにロード。
10)Chelexカラムを150〜175mlの1M硝酸アンモニウムで洗浄。
11)硝酸アンモニウムの溶出が完了したら、カラムを精製水60で洗浄。
12)カラムから203Pbが脱離するまで、1M硝酸を10〜20mlずつ加えカラムをストリップ。
1)回収した203Pb溶液を250mL容ガラスビーカー内で蒸発させてホットプレート上で乾燥させる。下記条件を記録。
2)10ccセプタ「S」バイアルの風袋重量を求め、記録。
3)Sバイアルを0.22μmフィルタースタンドに設置し、セプタを取り外す。
4)コンディショニングされたフィルター付き30ccシリンジをスタンドおよびSバイアルに設置。プランジャーを取り外す。
5)きれいなプラスチックピペットを用いて2〜3mLの0.5N HNO3をビーカー内の203Pbに添加。蒸気が発生するまでホットプレート上で加熱し、シリンジに移す。フィルターを通して移送された容積をSバイアルに押し込む。
6)203Pbが約10mlストックされるまで、ステップ(5)を2〜4回繰り返す。
1)単離した203Pbをストックした後、Sバイアルの総重量を計り、空のSバイアルの風袋重量を差し引いた正味重量を求める。
2)「S1」と記された追加のSバイアルの風袋重量を計る。
3)1mlのピペットを用いてSバイアルの内容物を完全に混合する。1mLのピペットを用いて、「S」バイアルから約0.5mLを取り、「S1」バイアルに移す。総重量を計った後、S1風袋重量を差し引いて正味重量を求める。
4)S1バイアルを1/2インチのピッグ(pig)に設置。
5)S1バイアルを圧着し、Capintecイオン化チャンバーステーションに移す。
6)イオン化チャンバーの「放射能レンジ」ダイアルをオートレンジに設定。Capintecのポテンショメータ設定を203Pbの推奨設定である344に設定。バックグラウンドを記録。トングを用いてS1バイアルをCapintecホルダーに移し、チャンバー内の全部を下ろす。総放射能を記録し、バックグラウンドを差し引いて正味のmCiを求める。
7)上で決定した放射能およびSおよびS1バイアルの正味の重量を用いて、Sバイアル濃度(mCi/g)を計算する。

Claims (6)
- イミノ二酢酸を含むキレート樹脂を、Pbを含む溶液と接触させること、および
加熱した水酸化ナトリウム溶液で、キレート樹脂に結合したPbを溶出させることを含んでなり、加熱した水酸化ナトリウム溶液がPbの選択的溶出に適した温度である、方法。 - 加熱した水酸化ナトリウム溶液の温度が約85〜95℃である、請求項1に記載の方法。
- イミノ二酢酸を含むキレート樹脂を、Pbを含む溶液と接触させること、および
水酸化ナトリウムの温度が約90℃である加熱した水酸化ナトリウム溶液で、キレート樹脂に結合したPbを溶出することを含んでなる方法。 - 約85〜95℃、または約85℃以上である、水酸化ナトリウム溶液を用いてキレート樹脂からPbを溶出することを含んでなる方法。
- 203Pb、および0.1μg/mCiよりも少ないNi、および/または0.1μg/mCiよりも少ないCu、および/または0.5μg/mCiよりも少ないZn、および/または0.25μg/mCiよりも少ないFe、および/または0.05μg/mCiよりも少ないTlを含有する組成物。
- 水酸化ナトリウムをさらに含む、請求項5に記載の組成物。
