マウス3T3線維芽細胞およびヒトB.J.皮膚線維芽細胞のトランスフェクション:

ヒトB.J.細胞およびマウス3T3線維芽細胞を、TransIT mRNAトランスフェクション試薬と複合した1μg FGF2またはFGF7 IVT mRNAを使用してトランスフェクションした。Tri-Reagent(Thermo Fisher、USA、製造元の指示)を使用したトランスフェクション後のさまざまな時点で、マウスおよびヒト線維芽細胞またはブタ上皮シートから全細胞RNAを単離し、mRNAを従来のRT-PCR(Protoscript First Strand cDNA合成キット、New England Biolabs、メーカーの指示に従って)によりcDNAに逆転写した。次に、cDNA試料を従来のPCRおよびqPCRにかけた。プライマーは、Invitrogenから入手した。

FGF2およびFGF7 mRNA変異体が長期間にわたってマウス線維芽細胞で安定であるかどうかを評価するため、配列番号1-5および18-22をコードする異なるmRNAを使用して3T3細胞をトランスフェクションする。RNAを異なる時点(トランスフェクション後24-120時間)で単離し、トランスフェクションした細胞内のmRNAの存在を定量的RT PCRによりトランスフェクション後120時間まで測定した。
FGF2およびFGF7 mRNA変異体が長期間にわたってヒト皮膚線維芽細胞で安定であるかどうかを評価するため、配列番号1-5および18-22をコードする異なるmRNAを使用して3T3細胞をトランスフェクションする。RNAを異なる時点(トランスフェクション後24-120時間)で単離し、トランスフェクションした細胞内のmRNAの存在を定量的RT PCRによりトランスフェクション後120時間まで測定した。
組換えFGF2またはFGF7タンパク質がIVT mRNAと同様の仕方でFGF発現およびその後の活性を調節できるかどうかを評価するため、マウス3T3またはブタ初代線維芽細胞を1ng/mlの組換えFGF2またはFGF7タンパク質の存在下でインキュベートするか、または異なるFGF2およびFGF7 mRNA変異体をトランスフェクションした。配列番号1および2および18、19および21をコードするmRNAを使用してマウス3T3細胞およびブタ初代線維芽細胞をトランスフェクションした。その後、FGF2およびFGF7タンパク質のレベルを、タンパク質のトランスフェクション/添加の24時間後に決定した。

− シチジン残基が5-ヨードシチジン残基で置換され、および/または
− ウリジン残基がプソイドウリジン残基で置換され、および/または
− ウリジン残基が1-メチルプソイドウリジン残基で置換され、および/または
− ウリジン残基が2-チオウリジン残基で置換され、および/または
− ウリジン残基が5-メチルウリジン残基で置換され、および/または
− アデノシン残基がN6-メチルアデノシン残基で置換される

Claims (15)
- 5'CAP領域、5'非翻訳領域(5'-UTR)、ヒトFGFをコードするコード領域、3'非翻訳領域(3'-UTR)およびポリアデノシンテール(ポリAテール)を有する、局所的な皮膚低栄養状態の治療に使用するための、線維芽細胞成長因子(FGF)メッセンジャーRNA(mRNA)であって、
該FGF mRNAのコード領域はヒト線維芽細胞成長因子2(FGF2)をコードするか、または
該FGF mRNAのコード領域はヒト線維芽細胞成長因子7(FGF7)をコードする
FGF mRNA。 - 局所的な皮膚低栄養状態が、皮膚弛緩症、慢性萎縮性先端皮膚炎、Pasini-Pierini型進行性特発性皮膚萎縮症、穿孔性皮膚炎に起因する瘢痕、萎縮性白化、脂肪性壊死、放射線皮膚炎、線条状伸展、萎縮性皮膚状態、糖質コルチコイド(GC)誘発性皮膚萎縮、萎縮性瘢痕および皮膚潰瘍からなる群から好ましくは選択される、請求項1に記載の使用のためのFGF mRNA。
- ポリAテールが、少なくとも60個のアデノシン一リン酸、好ましくは少なくとも100個のアデノシン一リン酸、とりわけ少なくとも120個のアデノシン一リン酸を含む、請求項1または2に記載の使用のためのFGF mRNA。
- 5'-UTRまたは3'-UTRまたは5'-UTRおよび3'-UTRが天然FGF2またはFGF7 mRNAとは異なるmRNAであり、好ましくは、5'-UTRまたは3'-UTRまたは5'-UTRおよび3'-UTRが少なくとも1つの安定化配列、好ましくは一般式:(C/U)CCANxCCC(U/A)PyxUC(C/U)CC(配列番号38)(ここで「x」は、NxおよびPyxで独立して、0〜10、好ましくは0〜5の整数、特に0、1、2、4および/または5である)の安定化配列を含む、請求項1〜3のいずれか1項に記載の使用のためのFGF mRNA。
- 5'-UTRまたは3'-UTRまたは5'-UTRおよび3'-UTRが天然FGF2またはFGF7 mRNAとは異なるmRNAであり、5'-UTRおよび/または3'-UTRが、好ましくはアルファグロビン、ベータグロビン、アルブミン、リポキシゲナーゼ、ALOX15、アルファ(1)コラーゲン、チロシンヒドロキシラーゼ、リボソームタンパク質32L、真核生物伸長因子1a(EEF1A1)、オルソポックスウイルスに存在する5'-UTR要素、およびこれらの混合物から選択され、特にアルファグロビン、ベータグロビン、アルファ(1)コラーゲン、およびこれらの混合物から選択される、FGFとは異なるヒトmRNAの5'-UTRおよび/または3'-UTRである、請求項1〜4のいずれか1項に記載の使用のためのFGF mRNA。
- GC/AU比が、FGF2 mRNAの場合は、少なくとも51.7%、好ましくは少なくとも52%、より好ましくは55%、さらにより好ましくは少なくとも58%、特に少なくとも60%であり、FGF7 mRNAの場合は、少なくとも39.5%、好ましくは少なくとも43%、より好ましくは45%、さらにより好ましくは少なくとも50%、特に少なくとも55%である、請求項1〜5のいずれか1項に記載の使用のためのFGF mRNA。
- FGF2 mRNAのコドン適応指数(CAI)が少なくとも0.77、好ましくは少なくとも0.8であり、および/またはFGF7 mRNAのCAIが少なくとも0.75、好ましくは少なくとも0.77である、請求項1〜6のいずれか1項に記載の使用のためのFGF mRNA。
- FGF2をコードするmRNAが、好ましくは配列番号30、特に配列番号31であり、FGF7をコードするmRNAが、好ましくは配列番号12、特に配列番号13である、請求項1〜7のいずれか1項に記載の使用のためのFGF mRNA。
- 皮下、皮内、経皮、表皮、または局所、特に表皮に投与される、請求項1〜8のいずれか1項に記載の使用のためのFGF mRNA。
- 請求項1〜9のいずれか1項に記載のFGF2 mRNAまたはFGF7 mRNAを含む、局所的な皮膚低栄養状態、好ましくは萎縮性瘢痕および糖質コルチコイド(GC)誘発性皮膚萎縮の治療での使用のための医薬製剤。
- 薬学的に許容される担体、好ましくはポリマーベースの担体、特に、直鎖および分岐PEIおよびビロマーを含むカチオン性ポリマー;脂質ナノ粒子およびリポソーム、ナノリポソーム、セラミド含有ナノリポソーム、プロテオリポソーム、カチオン性両親媒性脂質、例えば、:SAINT-Lipids、天然および合成由来のエキソソーム、天然、合成および半合成の層状体、ナノ粒子、リンケイ酸カルシウムナノ粒子、リン酸カルシウムナノ粒子、二酸化ケイ素ナノ粒子、ナノ結晶粒子、半導体ナノ粒子、乾燥粉末、ポリ(D-アルギニン)、ナノデンドリマー、デンプンベースの送達システム、ミセル、エマルジョン、ゾルゲル、ニオソーム、プラスミド、ウイルス、リン酸カルシウムヌクレオチド、アプタマー、ペプチド、ペプチド結合体、ベクタータグ、好ましくは小分子標的結合体、またはウイルスキャプシドタンパク質、乳酸−グリコール酸共重合体(poly-lactic-co-glycolic acid (PLGA) polymers);好ましくはカチオン性ポリマーおよびリポソーム、特にカチオン性ポリマーを含む、請求項10に記載の使用のための医薬製剤。
- 請求項1〜9のいずれか1項に記載の使用のためのFGF mRNAを患者に投与するためのキットであって、
− 請求項1〜9のいずれか1項に記載の使用のためのFGF2および/またはFGF7 mRNA、および
− 皮膚送達装置
を含む、キット。 - 皮膚送達装置が、
− 針ベースの注射システムからなる群から好ましくは選択される皮内送達装置であるか、または
− 経皮パッチ、中空および中実マイクロニードルシステム、微細構造経皮システム、電気泳動システム、およびイオン導入システムからなる群から好ましくは選択される経皮送達装置であるか、または
− 無針注射システム、レーザーベースのシステム、特にエルビウム(Erbium)YAGレーザーシステム、および遺伝子銃システムからなる群から好ましくは選択される表皮送達装置である、請求項12に記載のキット。 - 局所的な皮膚低栄養状態、好ましくは萎縮性瘢痕および糖質コルチコイド(GC)誘発性皮膚萎縮を治療する方法であって、請求項1〜9のいずれか1項に記載のFGF2 mRNAおよび/またはFGF7 mRNAをそれを必要とする患者に有効量で投与することを特徴とする方法。
- FGFメッセンジャーRNA(mRNA)をヒト皮膚に接触させる、特に老化した皮膚のための化粧品スキンケア方法であって、該mRNAは、5'CAP領域、5'非翻訳領域(5'-UTR)、ヒトFGFをコードするコード領域、3'非翻訳領域(3'-UTR)およびポリアデノシンテール(ポリAテール)を有し、該FGF mRNAのコード領域はヒト線維芽細胞成長因子2(FGF2)をコードするか、または該FGF mRNAのコード領域はヒト線維芽細胞成長因子7(FGF7)をコードする、方法。
