Abstract
Description
レチノイン酸およびその誘導体は、大きく、レチノール、レチナール、トレチノイン(オールトランスレチノイン酸)、イソトレチノイン(13-シスレチノイン酸)、アリトレチノイン(9-シスレチノイン酸)を含む第一世代、エトレチナート、アシトレチンを含む第二世代、アダパレン、ベキサロテン、タザロテンを含む第三世代に分かれる。(レチノール、レチナールデヒド、全トランスレチノイン酸、13-シスレチノイン酸、9-シスレチノイン酸、エトレチナート、アシトレチン、アダパレン、ベキサロテン、タザロテン)
本明細書で用いられるように、用語「ALDH1A」とはレチナールデヒドデヒドロゲナーゼファミリー1メンバーA(Aldehyde dehydrogenase family 1 member A)をいう。
ALDH1A阻害剤(またはALDH1Aファミリー阻害剤)とは生物化学反応においてALDH1Aおよびそのファミリーのタンパク質の化学反応を遮断するか、その速度を低下させることができる物質である。また、当該用語はALDH1Aの発現または活性を低下させる阻害剤、たとえばアンチセンスRNA、miRNA、または抗体を含む。
RA分解酵素CYP26A1阻害剤とは生物化学反応においてCYP26A1タンパク質によるレチノイン酸の分解を遮断するか、その速度を低下させることができる物質である。
また、本発明において、自閉症を治療または予防する候補薬物(または潜在的治療剤)を選別する方法を提供する。
(1)本発明は、新規な自閉症治療の標的を提供するため、本発明によれば、自閉症を治療する薬物を選別・開発することができる。
1.被験者およびサンプル
被験者の募集および評価はいずれも中南大学生命科学院、医学遺伝学国家重点実験室審査委員会の規則に従い、そしてヘルシンキ宣言の原則に厳格に遵守した。被験者の募集および診断の詳細はいずれも従来の報告に従った(Xiaら, 2013; Wangら, 2016)。すべての血液サンプルの採取はいずれも書面の同意書を受け取った。
3名の漢族の患者のコピー数多型(男性、年齢:3-6歳)には、Human660W-Quadマイクロアレイチップが使用された(Illumina)。チップ分析はGenomeStudio v2011.1(Illumina)によって行われ、従来の報告に従った(Navaら, 2013)。ヒト参照ゲノムhg18を参照としてコピー数多型の作図を行った。
標準化方法(Andersonら, 1984)に従い、EBVウイルスで血球を形質転換させて不死化リンパ球を得、そして10%牛胎児血清(Biochrom)を含有するRPMI-1640(Gibco)培地において培養した。HEK-293FT (Life Technologies)、HEK-293 (ATCC)、SH-SY5Y (ATCC)、H1299(ATCC)およびA549 (ATCC)の細胞はいずれも10%牛胎児血清およびペニシリン/ストレプトマイシン(Life Technologies)を含有するDMEM(Corning)培地で培養した。報告された方法に従ってマウス線維芽細胞を準備・構築した(Xuら, 2005)。初代ニューロン細胞は妊娠18日のラット(Sprague Dawley)の胎児の大脳の前頭前皮質から分離し、そして10%牛胎児血清を含有するDMEM/F12培地で培養した。翌日に、細胞の培地をB-27補充物(Gibco)およびGlutaMax(Gibco)を含有する無血清神経基礎培地(Gibco)に換えて培養した。細胞は5% CO2を含有する飽和湿度のインキュベーターで培養した。すべての細胞系はいずれも定期的にマイコプラズマの検出を行った。

本課題におけるプラスミドの構築は制限酵素による消化および連結反応(NEB)を含み、いずれも従来のクローニング方法によって行われた。酵母ツーハイブリッドスクリーニングに使用されたプラスミドはGateway LRクロナーゼ(ThermoFisher)でメーカーのプロトコールに従って反応させて得られた。関連するプラスミドの点突然変異は部位特異的突然変異法によって導入された。さらに、複数のエレメントを含有するプラスミドはギブソン・アセンブリ法によって構築された。本工程において、UBE3A融合発現タグはいずれもN末端に位置した。
ベイトタンパク質としてUBE3Aを使用し、pDEST32-UBE3A(ThermoFisher)およびpDEST22骨格に基づいたヒト由来cDNAライブラリー(ThermoFisher)を酵母菌株Mav203(ThermoFisher)に共形質移入した。陽性クローンは同時にウラシル、ヒスチジン、ロイシンおよびトリプトファンの4つの成分が欠乏している培地(Clontech)において生存することができ、X-Gal(Sigma)が存在する条件において青色を呈色することもできる。
GST、GST-UBE3Aおよび短縮されたGST-UBE3Aタンパク質をBL21感受性細胞(NEB)を誘導発現し、グルタチオンアガロース(GE Healthcare)によって精製した。ALDH1A1-His6、ALDH1A2-His6およびALDH1A3-His6タンパク質はNi-NTAアガロース(Qiagen)によって精製した。精製後のALDH1A2-His6およびGST-UBE3Aタンパク質はプルダウン緩衝液(50 mM Tris-Cl、pH 8.0、200 mM NaCl、1 mM EDTA、1% NP-40、1 mM DTT、10 mM MgCl2)において4℃で2時間インキュベートした。ビーズをプルダウン緩衝液で4回洗浄し、イムノブロット法によって分析した。ほかのタンパク質のプルダウン分析は同様の手順に沿った。
ユビキチン化系のすべての構成要素を従来のクローニング方法によって二重発現系ベクター骨格pACYCDuet-1(Novagen)に導入した。HA-UB、UBCH7およびUba1を1つ目のマルチクローニングサイトに挿入し、T7プロモーター/乳糖オペロンおよびリボソーム結合部位の下流に連結させ、各構成要素の間にいずれもShine-Dalgarno(SD)配列によって分画することによってマルチシストロン性のエレメントを形成した。UBE3Aを2つ目のマルチクローニングサイトに挿入し、pACYC-HA-UB-UBCH7-Uba1-UBE3Aプラスミドを生成した。BL21感受性細胞を電気形質転換法によってpACYCおよびpET22b-ALDH1A2-His6プラスミド(Novagen)で形質転換し、そしてクロラムフェニコールおよびアンピシリン(Sigma)でスクーニングした。大腸菌はOD600吸光値が0.8に達した時点で、0.25mMのイソプロピル-β-D-1-チオガラクトピラノシド(IPTG、Sigma)で18℃の条件において16時間誘導培養した。細胞を収集してRIPA緩衝液(150 mM NaCl、50 mM Tris-Cl、pH 7.4、1% NP-40、0.1% SDS)に再懸濁させた後、細胞をVibra-Cellリアクター(SONICS)で超音波処理し、遠心によって沈殿を除去した。上清をNi-NTAアガロースによって精製した。Usp2cc処理群では、Ni-NTAビーズに結合した精製タンパク質をUsp2cc酵素と4℃で一晩インキュベートした。ALDH1A2タンパク質のユビキチン化レベルをイムノブロット法によって分析した。
内因性または外因性タンパク質を発現する細胞をプロテアーゼ阻害剤混合物(Roche)を入れたIP緩衝液(50 mM Tris-Cl、pH 7.5、150 mM NaCl、1 mM EDTA、1% NP-40、10%グリセリン)において分解させ、そしてVibra-Cellリアクターで超音波処理した。遠心後、細胞破片を除去し、上清に特異性抗体およびプロテインGアガロースビーズ(Merck Millipore)を入れて4℃で一晩インキュベートした。使用された一次抗体は、正常のウサギIgG抗体(sc-2027、Santa Cruz)、Flag抗体(F1804、Sigma)、ALDH1A2抗体(sc-367527、Santa Cruz)、HA抗体(H6908、Sigma)である。免疫沈殿を収集した後、2×SDS-PAGE仕込み緩衝液を入れ、95℃で10分間変性させた。出発タンパク質、免疫沈殿収集タンパク質およびほかの細胞分解サンプルをSDS-PAGEゲルで分離させ、PVDF膜(Bio-Rad)に転写させた。膜をUBE3A抗体(sc-166689、Santa Cruz、1:500希釈)、Flagタグ抗体 (F1804、Sigma、1:8000希釈)、HAタグ抗体(H6908、Sigma、1:4000希釈)、ALDH1A2抗体(sc-367527、Santa Cruz、1:500抗体)、ALDH1A1抗体(15910-1-AP、Proteintech、1:500抗体)、ALDH1A3抗体(25167-1-AP、Proteintech、1:500希釈)、Hisタグ抗体(H1029、Sigma、1:4000希釈)、GSTタグ抗体(66001-1-lg、Proteintech、1:5000希釈)、Mycタグ抗体(sc-40、Santa Cruz、1:1000希釈)、アクチン抗体(A2228、Sigma、1:8000希釈)、GAPDH抗体(sc-32233、Santa Cruz、1:4000希釈)といった特異的抗体でイムノブロットを行った。
SH-SY5Y細胞(ATCC)に特定のプラスミドを形質移入した後、さらに24時間培養し、その後4%パラホルムアルデヒド(Sigma)で固定した。細胞を破裂させた後、一次抗体(Flagタグ抗体、F1804、Sigma)を入れて4℃で一晩インキュベートした後、Alexa Fluor 488が結合した蛍光二次抗体(A11029,ThermoFisher)で室温で1時間インキュベートした。細胞核を4',6-ジアミジノ-2-フェニルインドール(DAPI、ThermoFisher)で二重染色した。ラットの初代ニューロン細胞に体外で10日目にリン酸カルシウム形質移入法によって特定のプラスミドを形質移入し、DIV12の時にDMSO(Sigma)または1μMテトロドトキシン(TTX、上海Aladdin社)および100 μM D-(-)-2-アミノ-5-ホスホノ吉草酸(D-APV、Tocris)で24時間処理した。細胞を固定した後、前記のプロトコールに従ってそれぞれ一次抗体としてGluR1抗体(sc-55509、Santa Cruz)、PSD95抗体(ab18258、Abcam)およびAlexa Fluor 488(A11029、ThermoFisher)、Alexa Fluor 647(A21245、ThermoFisher)が結合した二次抗体で染色を行った。Olympus FV1200共焦点顕微鏡によって写真を撮影し、撮影解像度は1024×1024ピクセルであった。同実験において共焦点顕微鏡の設定パラメーターを一定に維持した。蛍光強度分析の定量はImage-Pro plus(Media Cybernetics)ソフトを使用した。
ウイルスが定位注射されたマウスから取り出された大脳は4%パラホルムアルデヒドを含有するPBSで4℃で一晩固定し、さらに30%ショ糖(Sigma)を含有するPBSに浸漬した。Leica CM3050 Sクリオスタット(Leica Biosystems)によって厚さ40 μMの大脳冠状切片を切り出し、切片をPBST(0.3% Triton X-100)で室温で15分間処理した。その後、脳部切片を3%正常ヤギ血清(BOSTER)によってブロッキングし、Flagタグ抗体(14793, Cell Signaling, 1:800希釈)で4℃で一晩インキュベートした。切片をCy3結合二次抗体(111-165-045, Jackson ImmunoResearch)で室温で1時間インキュベートした後、DAPIで細胞核を二重染色し、封入剤Mowiol(Sigma)で封じた。蛍光写真はOlympus FV1200共焦点顕微鏡によって撮影した。
UBE3Aで形質移入されたHEK-293FT細胞または形質移入されていない細胞において、Flagでカップリングされたアガロースカラム(Sigma)によって外来的に発現されたALDH1A2-Flagタンパク質を集め、さらにALDH1A酵素活性検出緩衝液(0.1 M ピロリン酸ナトリウム、pH 8.0、1.0 mM EDTA、2.0 mM DTT)に配合されたFlag短鎖ペプチドで溶離させた。ALDH1A2酵素の脱ユビキチン化の有無についての研究において、それぞれUSP2cc酵素またはウシ血清アルブミン(BSA)でタンパク質を溶離させて4℃で一晩インキュベートした。酵素活性反応系は酵素活性検出緩衝液に溶解させた2.5 mM NAD+、20 mM DTTおよび100 μMプロピオンアルデヒド(Sigma)を含み、脱水素酵素活性はBioTek Synergy Neo分光光度計において波長340nmで室温の条件で3分間おきに1回検出された。すべてのサンプルに対して吸光値がプラトー期に達したら反応を停止させた。NADHを標準品とし、ALDH1A2の脱水素酵素活性の計算方法は、(反応時間内でNADHの総生成量(nmol)×サンプルの希釈倍数)/(反応時間×反応体積)である。基質であるオールトランスレチナールデヒド(Sigma)を測定する場合、アセトアルデヒド脱水素酵素活性分析キット(Cayman)が使用された。それは蛍光定量に基づいた方法によって検出・分析された。つまり、100 μMのオールトランスレチナールデヒドが存在する時、酵素活性は蛍光基の吸収波長530-540 nmおよび放出波長585-595 nmにおけるパラメーターを検出することによってレチナールデヒドの吸収値に対する干渉を最低限にした。
ALDEFLUORアッセイはALDEFLUORキット(STEMCELL Technologies)によってメーカーが提供したプロトコールに従って行われた。つまり、1×106個の不死化リンパ球を蛍光基質であるBODIPY-アミノアミノアセトアルデヒドジエチルアセタール(BAAA-DA)(1.5 μM)と37℃で30分間インキュベートした。各細胞を二つに分けた。半分は蛍光分析に使用され、もう半分の細胞はまずキットで提供されたALDH阻害剤であるジエチルアミノベンズアルデヒド(DEAB)で前処理し、フローサイトメトリー分析において陰性対照とされた。
pGL4.22-RARE-TK-luciferaseプラスミドの構築方法は、3X RARE(RA-応答エレメント)をpGL4.22ベクター(Promega)にクローニングすることを含む。RA受容器細胞系の構築方法は、H1299細胞にpGL4.22-RARE-TK-luciferaseプラスミドを形質移入し、そしてピューロマイシン(1 μg/mL、Sigma)で安定形質移入細胞株を選別することを含む。RA受容器細胞をRAドナーとして不死化リンパ球と細胞数1:1でVP-SFM(virus production serum-free medium、Gibco)培地において共培養した。1 μMのオールトランスレチナールデヒド(Sigma)を入れて8時間処理した後、培養細胞のルシフェラーゼ活性を検出した。
初代ニューロン細胞および体外で12-14培養した形質移入のニューロン細胞を1 μM TTX(Aladdin)および100 μM D-APV(Tocris)で24時間処理し、室温の条件においてパッチクランプのデータを記録した。ここで、パッチクランプ内液(単位mM)は、20 KCl、5 MgCl2、20 HEPES、110 グルコン酸カリウム(K-gluconate)、0.6 EGTA、2 Na2-ATP、0.2 Na3-GTPを含み、pH 7.3で、290 mOsmで、内液の電気抵抗が約3-6 MΩである。培養された細胞を外液に置き、ここで、外液の成分(単位mM)は、129 NaCl、5 KCl、1 MgCl2、25 HEPES、2 CaCl2、30 グルコースを含み、pH 7.3で、310 mOsmである。細胞外液には、さらに、1 μM TTXおよび100 μM ピクロトキシン(Tocris)が含まれ、電圧-70mVの条件においてmEPSCを記録した。結果はMini Analysisソフト(Synaptosoft)によって分析した。
初代ニューロン細胞を所定の化合物で(1 μM TTXおよび100 μM D-APVで24時間、あるいは0.5 μM ATRAで8時間)処理した後、RNAsimple total RNAキット(Tiangen)によって全RNAを抽出した。ReverTra Ace qPCR RT Master Mix (Toyobo)によって逆転写してcDNAサンプルを得た。SYBR Green Master Mix(Toyobo)によってCFX96 real-time PCR装置(Bio-Rad)によってqRT-PCR検出を行った。所定の遺伝子の転写相対量はΔΔCt方法によってGapdhを内部参照として校正した。本操作においてQPCRプライマー配列は以下の通りである。

ビオチン標識細胞膜表面タンパク質分析は従来報告された方法に従った(Aotoら, 2008)。つまり、DMSOまたは1 μM TTXおよび100 μM D-APVで24時間処理した後、初代PFCニューロン細胞をPBSで洗浄し、さらにビオチン溶液(1 mg/ml EZ-Link Sulfo-NHS-SS-Biotin, Pierce)と4℃で2時間インキュベートした。0.1 Mグリシンを入れて反応そ停止させた後、PBSで3回洗浄し、ビオチンで標識された細胞を細胞分解液(25 mM MgCl2、1%NP-40、1% Triton X-100、10% グリセリンおよびプロテアーゼ阻害剤を含有するPBS)において分解させた。遠心で細胞破片を除去した後、上清をUltraLink Streptavidin樹脂(Pierce)と4℃で一晩回転させながらインキュベートした。ビオチンで標識されたタンパク質を遠心で収集し、そして細胞分解緩衝液で3回洗浄した。タンパク質を2×SDS-PAGE仕込み緩衝液で75℃で30分間変性させ、そしてGluR1抗体(13185、Cell Signaling、1:500希釈)およびGluR2抗体(13607、Cell Signaling、1:500希釈)でイムノブロット分析を行った。
マウスは標準の明期12時間/暗期12時間のサイクルで、各ケージに3-5匹ずつ飼育し、自由に飲食・飲水ができるようにした。すべての行動学実験は明期12時間の条件において行われた。すべての動物学の研究は厳格に中国科学院生化細胞所動物維護・使用委員会(IACUC)の規約に従った。すべての実験におけるマウスはC57BL/6背景のオスマウスであった(SLAC、中国)。
所定の遺伝子はいずれもSynapsin I(SynI)プロモーターによって駆動され、そしてN末端にFlagタグが融合した。アデノ随伴ウイルス(AAV)の外被タンパク質の血清型はAAV2/9で、上海禾元生物技術公司(Obio)によってパッケージングされた。AAV-SynI-Flag-UBE3AおよびAAV-SynI-Flag-UBE3A-T508Eウイルスの力価は約1.5×1013コピー数/mLで、AAV-SynI-Flag-EGFPウイルスの力価は約9.5×1012コピー数/mLであった。
4週齢のマウスに連続6週間で1日おきに低投与量(0.1 mg/g)、または高投与量(0.3 mg/g)のDSF(Sigma、オリーブオイルに溶解)を胃内投与した。対照群では、溶媒のオリーブオイル(Aladdin)のみを胃内投与した。毎週マウスの体重を測定した。胃内投与から6週間後、マウスの行動学のテストを始めた。対照群およびDSF群のマウスの大脳を解剖し、液体窒素で快速凍結後、組織を均質化し、後記のHPLC-MS/MS方法によってATRAを定量した。
従来報告された手順によってサンプルの調製、レチノイドの抽出および後続のHPLC-MS/MS分析を行った(Kaneら, 2010)。赤色光の条件において、快速凍結されたマウスの大脳を氷の上で2 mLの0.9%生理食塩水で均質化した。13-シスレチノイン酸-d5 (20 ng/mL、TRC)を内部参照として組織均質液に入れた。その後、均質液に1.5 mLのエタノールに溶解させた0.025 Mの水酸化カリウムを入れ、さらに7 mLのn-ヘキサンを入れた後、均一に混合して水相を抽出し、さらに120 μLの4 M塩酸で中和した。その後、RAおよび極性レチノイドを含有する油相をさらに入れた7 mLのn-ヘキサンで抽出した。抽出物を窒素ガスの雰囲気において蒸発させ、乾燥した抽出物を50 μLのアセトニトリルで再懸濁させた。サンプルを2.1×100 mm Supelcosil ABZ+PLUS column(3 μm、Sigma)カラムによってHPLC分析を行ったが、移動相はそれぞれAは0.1%ギ酸を含有する水で、Bは0.1%ギ酸を含有するアセトニトリルであった。検出装置はAB Sciex 4000 QTRAP LC-MS/MSシステムで、APCIカチオンモードでATRA成分を定量し、各サンプルにおける含有量を標準ATRA曲線によって校正した。
セルフグルーミング実験に使用されるマウスは厚さが約0.5-1cmのフィラーが敷かれたケージにおいて事前に10分間馴化させた。その後、10分間内で、マウスのセルフグルーミングに使用された時間は二重盲検の実験者によってストップウオッチで記録された。
従来報告された手順で三部屋式社会性行動試験を行った(Sztainbergら, 2015)。つまり、社会性行動試験を行う透明のアクリルボックスを三つの同じ大きさの部屋に分け、仕切りの板に移動可能なドアを取り付けた。そして、左右の部屋にそれぞれ逆さにした鉄網カップを置いた。実験の2日前に、二つのカップにそれぞれ親が違う、年齢・性別が被験マウスとマッチするC57BL/6マウスをストレンジャーマウスとして置き、毎日1時間馴化させた。被験マウスをランダムに分け、そして試験開始前に試験部屋で事前に1時間馴化させた。各被験マウスを中央の部屋に置いて10分間自由に探索させた後、部屋のドアを閉めた。第一段階で、ストレンジャーマウスを1匹ランダムに左か右のカップに置き(位置の偏向性がないように)、もう一方のカップに非生命の物体を置いた。各被験マウスを開放の三つの部屋で10分間探索させ、手動で被験マウスがそれぞれストレンジャーマウスおよび物体との交流時間を記録した。第二段階では、被験マウスをストレンジャーマウスの部屋に続いて5分間置いた後、もう1匹のストレンジャーマウスを先に物体が置かれたカップに置いた。被験マウスを続いて10分間探索させ、手動で被験マウスがそれぞれ馴染んだ動物および新しい動物との交流時間を記録した。観測者が被験マウスの群分けを知らないようにした。
マウスをランダムに分けた後、オープンフィールド装置(Med Associates)に置いて30分間探索させた。Ethovision自動記録ソフト(Noldus)によってマウスの中央区域(区域の各辺の長さ1/3〜2/3)にかけた時間を記録し、そしてその中央区域で移動した距離の総距離における比率を算出した。
マウスを高架式十字迷路(Med Associates)のクローズドアームに置き、記録を開始し、計5分間記録した。上方に設置されたカメラで撮影して記録し、ANY-mazeソフト(Stoelting)によってマウスの5分間内でオープンアームとクローズドアームに入った回数を記録して分析した。
マウスをランダムに分けた後、Rotamex rotarod装置(Columnbus Instruments)でマウスの運動協調性を記録した。同じ日に、被験マウスを3回テストし、試験は毎回5分間持続し、ロータロッドは4回/秒から40回/秒まで加速した。各試験の間隔は30分間以上であった。被験マウスのロータロッドから落下するまでの時間を赤外検出システムによって自動的に記録し、最後の試験の結果で最終の比較を行った。
すべてのデータ分析はGraphPad Prismソフトで行われた。データの有意性検定はそれぞれスチューデント両側t検定、一元配置ANOVA(ボンフェローニ事後検定またはダネット事後検定)が使用された。すべてのデータは平均値と分散で表示された。各データ統計の詳細はいずれも注釈で示された。すべてのデータは3回以上の独立の反復試験から採取された。サンプル量は事前に予想しなかった。すべてのマウスはランダムに各群に分けた。
前期は臨床で自閉症表現型と診断された患者3名を集めたが、彼らの末梢血に対してゲノムDNAの抽出およびBeadChipチップによる検出を行ったところ、染色体15q11.2-14の多型が発見された(図1a-b)。当該多型領域には、一連の遺伝子が含まれるが、一般的に実証された15q重複症候群において重要な役割を担うUBE3A遺伝子以外、これらの自閉症との関連性が不明である(図1b-c)。後続の機能研究のために、EBVウイルス感染の方法によってこれらの患者の末梢血リンパ球で不死化細胞系を構築した。中では、健常者のボランティアの対照細胞と比べ、この三つの自閉症細胞のUBE3Aタンパク質の発現レベルが顕著に増加した(図1d)。
今まで機序の点からASDの発症とUBE3Aの過剰活性化の関連性を説明できるようなUBE3Aの既知の基質がなかったため(Glessnerら, 2009)、まず、ヒトUBE3Aタンパク質をエサに、酵母ツーハイブリッドの方法によって未知の基質タンパク質を選別した。中では、多くの陽性クローンはいずれもRA合成の律速酵素ALDH1A2の遺伝子のcDNAを持つことが見出された(図2a)。レチノイン酸RAはビタミンA(レチノール、retinol)の活性代謝物として、ヒトを含む高等動物の発育と成長に必要である。中では、RAの合成は、レチノールからレチナールデヒドへ触媒するレチノールデヒドロゲナーゼ(RDH10)、およびレチナールデヒドからレチノイン酸へ触媒するレチナールデヒドデヒドロゲナーゼ(ALDH1A1、2、3ファミリー)を含む二つのステップで完成される。免疫共沈実験によって、内因または外因的に発現されたUBE3AがALDH1A2タンパク質とHEK-293FT細胞において複合体を形成することが証明された(図2b-c)。免疫蛍光実験において、それぞれFlagとRFPタグが付いたUBE3AとALDH1A2タンパク質は神経膠腫細胞SH-SY5Yの細胞質内に共局在化した(図2d)。GSTプルダウン実験によって、さらに、組み換えタンパク質UBE3AとALDH1A2が体外で直接結合し(図2e)、そしてUBE3AのN末端(1-280番目のアミノ酸)を介して結合することが証明された(図2f)。ALDH1A2タンパク質はほかのファミリーのメンバーであるALDH1A1および3と高い配列の類似度を有するため、UBE3Aがそれぞれ体外と体内で直接ALDH1A1および3と結合することが証明された(図2g-j)。GSTプルダウン実験において、UBE3AとALDH1A2の結合領域に対する3つの異なるペプチド断片(ペプチド-1、ペプチド-2、ペプチド-3)を入れ、異なる程度で競争的にGST-UBE3AとALDH1A2タンパク質の間の相互結合を遮断することができた(図2k)。ここで、ペプチド-1、ペプチド-2、ペプチド-3の配列は以下の通りである。

次に、UBE3AがALDH1A2に対してユビキチン化修飾を行うことができるか、さらに検証した。まず、大腸菌においてユビキチン化系を構築し(詳細は方法の部分を参照する)(Keren-Kaplanら, 2012)、ALDH1A2およびユビキチン化反応に必要なメンバーを細菌に共形質転換し、IPTGでタンパク質の発現を誘導した。図3aに示すように、UBE3Aタンパク質が存在する条件において、ALDH1A2タンパク質は高度にユビキチン化され、そして当該ユビキチンバンドは脱ユビキチン化酵素USP2の触媒コア部分であるUSP2ccタンパク質によって消されたが、これはALDH1A2がUBE3Aによってユビキチン化修飾を行われることを示唆した。HEK-293FT細胞において、UBE3Aタンパク質の過剰発現も同様にALDH1A2のユビキチン化を促進した(図3b)。
従来の報告では、一部のUBE3Aの基質タンパク質、たとえばHHR23A(Kumarら, 1999)、およびRING1B(Zaaroor-Regevら, 2010)は、ヒトパピローマウイルス遺伝子E6が欠失した条件において、ユビキチン化修飾およびプロテアソームに依存する分解が可能であることがわかった。しかし、p53タンパク質にE6が欠失した場合、UBE3Aは単独でそれに対してユビキチン化修飾およびさらなるタンパク質の分解を行うことができない(Ansariら, 2012)。しかしながら、HEK-293FT細胞において、UBE3Aタンパク質の発現レベルの増加とともに、内因性ALDH1A2の発現レベルに変化が現れなかった(図4a)。同時に、異なるUbe3aタンパク質レベルを有するマウスMEF細胞において、Aldh1a2タンパク質の発現レベルに同様に変化がなかった(図4b)。不死化リンパ球において、自閉症患者由来の細胞および健常者由来の細胞を含め、ALDH1A2タンパク質レベルも同じように維持した(図3i)。これらの結果から、UBE3AによるALDH1A2のユビキチン化修飾はほかの基質のように、基質の分解を促進することがないことが示された。
さらに、UBE3Aによるユビキチン化修飾後のALDH1A2の脱水酵素活性、特にレチナールデヒドからレチノイン酸へ触媒する活性が影響を受けるか、検証する必要がある。その結果、対照細胞サンプルと比べ、UBE3Aタンパク質を過剰発現するHEK-293FT細胞から集められた高度にユビキチン化されたALDH1A2タンパク質のレチナールデヒドデヒドロゲナーゼ活性が半分低下したが、当該タンパク質をUSP2cc脱ユビキチン化酵素で体外で処理すると、活性が顕著に回復したことがわかった(図5a-c)。これによって、UBE3A段階のALDH1A2ユビキチン化修飾が確かに顕著にレチナールデヒドデヒドロゲナーゼ活性を下方調節したことが示された。同様に、ALDH1A2のもう一つの基質であるプロピオンアルデヒドを検出したところ、UBE3Aは同様にALDH1A2のプロピオンアルデヒドヒドロゲナーゼ活性を低下させ、USP2ccタンパク質はこの現象を逆転させることができることがわかった(図5d-e)。そして、ALDH1A2のK269/K370/K415の三つの突然変異体が完全に脱水素酵素活性を失ったが(図5f)、これはこの三つのリシン残基の完全性がALDH1A2の脱水素酵素活性、ひいてはALDH1Aタンパク質ファミリーの脱水素酵素活性には非常に重要であることを示唆した(図5g)。
成年の神経系において、RAが内部安定状態のシナプス可塑性で重要な役割を果たすことがわかってきた(Chenら, 2014)。ニューロン細胞において、シナプス伝達が遮断されると、シナプスのカルシウムイオンレベルがすぐに低下し、RAがALDH1A脱水素酵素の合成過程によって活性化される(Chenら, 2014; Aotoら, 2008)。RAはシナプスに局在化するRARαタンパク質と結合することによって、RARαによるタンパク質の翻訳過程の遮断を解除し、調節されるタンパク質はAMPA受容体を含み、よってシナプス伝達が上方調節される(Aotoら, 2008)。ラットPFC皮質からの初代ニューロン細胞において、D-APVおよびTTXでシナプス活性を抑制すると、細胞におけるRAレベルが上昇することによって、RAREによって駆動されるヒト化レニラ・レニフォルミス緑色蛍光タンパク質(hrGFP)の顕著な発現が誘導される(図6a)。しかし、ニューロン細胞において、UBE3A-IRES-turboRFPおよびRARE-hrGFPレポータープラスミドを共形質移入すると、D-APVおよびTTXによる処理はhrGFPタンパク質の発現を誘導しなくなった(図6b)。しかし、UBE3Aリガーゼ活性が欠失した突然変異体T508EをRARE-hrGFPレポータープラスミドと共形質移入すると、D-APVおよびTTXによる処理は対照群と類似するレベルまでhrGFPタンパク質の発現を誘導するようになった(図6b)。これらの結果によって、UBE3Aの過剰活性化がシナプス伝達の遮断によるRA生成を阻止することが証明された。
大脳におけるPFC領域は、決定、移動の認知、社会性行為、学習や社会的コミニュケーションを含む、大脳の様々な執行機能および高等認知過程を調節する。最近、PFC領域の解剖学的構造およびほかの脳領域の連結構造の異常は、一般的に、自閉症患者の大脳に存在することが見出され、これはPFCの機能異常が自閉症の病因に密接に関連することも示唆した(Stonerら, 2014; Chowら, 2012)。過剰活性化されたUBE3Aが自閉症行為の発生を誘導することを検証するために、それぞれEGFP、UBE3AおよびT508Eがパッケージングされたアデノ随伴ウイルス(AAV)を定位注射の方法によってマウスの大脳内側のPFC領域に入れた(図7a-b)。大脳を凍結切片した後の免疫蛍光の結果から、UBE3AおよびT508EのPFC領域におけるタンパク質の発現レベルが同等であったことが示された(図7c)。
さらに、UBE3Aの過剰発現によるマウスの自閉症の表現型の誘導はRAの生成過程の損傷によるものであることを検証するために、PFCにUBE3Aを担持するAAVウイルスを注射したマウスを分けて溶媒(オリーブオイル)またはATRA(3 mg/kg、毎週5日連続で投与した)を計4週間経口投与した後、マウスに行動学実験を行った(図8a)。マウスのセルフグルーミング実験において、溶媒経口投与群と比べ、ATRAを経口投与したマウスはのセルフグルーミング行為にかけた時間が顕著に減少した(図8b)。同時に、溶媒群と比べ、ATRAを投与したマウスは社会的相互作用にも完全な逆転が現れ、社会性動物と交流する時間が非社会性の物体よりも倍近く多かった(図8c)。図8dに示すように、これらのマウスの社交的新規性の認知の欠失もATRA投与後緩和が現れ、ストレンジャーマウスとの交流時間が馴染んだマウスよりも倍近く多かった。そのため、4週間のATRA投与は大幅にUBE3A過剰発現マウスの自閉症の中核症状を改善することができた。
単独のALDH1A活性の阻害がマウスの自閉症の表現型の発生を誘導することができるか、検証するために、4週齢の野生型C57BL/6マウスにAldh1a阻害剤であるジスルフィラム(DSF)を経口投与した。投与時間は計6週間で、投与量はそれぞれ0.1または0.3 mg/gで、1日おきに投与した。DSFを6週間投与した後、マウスの大脳を取り出し、HPLC-MS/MS方法によってそのATRAレベルを検出した(Kaneら, 2010)。図9aに示すように、DSFを6週間投与したマウスの大脳組織におけるATRAのレベルは溶媒群と比べ、それぞれ30%(投与量0.1 mg/g)と60%(投与量0.3 mg/g)低下した。これはDSFの投与は投与量に依存してマウスの大脳におけるATRAの合成を阻害することができることを示した。同時に、マウスの体重を毎週測定したところ、各群間で体重に有意差がなかった(図9b)。これはここのDSF投与量がマウスに顕著な毒性作用を引き起こさないことを示した。
(1)UBE3AがALDH1A2と結合してユビキチン化することによって、ALDH1A2の脱水素酵素の活性を抑制する。同様に、ALDH1AのほかのファミリーメンバーであるALDH1A1およびALDH1A3もUBE3Aの基質であることが証明された。

Claims (17)
- ALDH1A、またはその作動剤、またはその触媒産物、および/またはその触媒基質の使用であって、自閉症の治療および/または予防に使用される薬物組成物または製剤の製造のために使用されることを特徴とする使用。
- 前記ALDH1Aは、ALDH1A1、ALDH1A2、ALDH1A3、またはこれらの組み合わせからなる群から選ばれることを特徴とする請求項1に記載の使用。
- 前記ALDH1A作動剤は、N-(1,3-ベンゾジオキソール-5-イルメチル)-2,6-ジクロロベンズアミド(N-(1,3-Benzodioxol-5-ylmethyl)-2,6-dichlorobenzamide)、N-(1,3-ベンゾジオキソール-5-イルメチル)-2,6-ジクロロベンズアミド(N-(1,3-Benzodioxol-5-ylmethyl)-2,6-dichlorobenzamide)、6-メチル-2-(フェニルアゾ)-3-ピリジノール(6-methyl-2-(phenylazo)-3-pyridinol)、2-(ベンゾ[d][1.3]ジオキソール-5-イル)-N-(5,6-ジヒドロ-4H-シクロペンタ[c]イソオキサゾール-3-イル)アセトアミド(2-(benzo[d][1,3]dioxol-5-yl)-N-(5,6-dihydro-4H-cyclopenta[c]isoxazol-3-yl)acetamide)およびこれらの各誘導体からなる群から選ばれることを特徴とする請求項1に記載の使用。
- 前記の作動剤は発現促進剤、タンパク質分解阻害剤を含むことを特徴とする請求項1に記載の使用。
- レチノイン酸またはレチノイン酸類似体、またはその溶媒和物、またはその薬学的に許容される塩の使用であって、自閉症の治療および/または予防に使用される薬物組成物または製剤の製造のために使用されることを特徴とする使用。
- 前記レチノイン酸類似体は、レチノール、レチナールデヒド、全トランスレチノイン酸、13-シスレチノイン酸、9-シスレチノイン酸、エトレチナート、アシトレチン、アダパレン、ベキサロテン、タザロテン、またはこれらの組み合わせからなる群から選ばれることを特徴とする請求項5に記載の使用。
- レチノイン酸合成原料化合物、またはレチノイン酸分解酵素阻害剤の使用であって、自閉症の治療および/または予防に使用される薬物組成物または製剤の製造のために使用されることを特徴とする使用。
- 前記レチノイン酸分解酵素阻害剤は、リアロゾール(liarozole)、ケトコナゾール(ketoconazole)、タラロゾール(talarozole)、またはこれらの組み合わせからなる群から選ばれることを特徴とする請求項7に記載の使用。
- ALDH1A阻害剤の使用であって、自閉症動物モデルの構築に使用される製剤の製造のために使用されることを特徴とする使用。
- 前記ALDH1A阻害剤は、ジスルフィラム(disulfiram)、4-(N,N-ジエチルアミノ)ベンズアルデヒド(DEAB)、WIN-18446、A37(CM037)、NCT-501塩酸化物、CVT-10216、またはこれらの組み合わせからなる群から選ばれることを特徴とする請求項9に記載の使用。
- 自閉症を補助診断および/または予後する方法であって、
(1)血液、体液からなる群から選ばれる、被検サンプルを提供する工程と、
(2)マーカーの濃度、含有量、および/または活性を検出する工程と、
(3)基準値または検量線と比べることによって、補助診断および/または予後を行う工程とを含み、
ここで、前記マーカーはレチノイン酸、ALDH1A、またはこれらの組み合わせからなる群から選ばれることを特徴とする方法。 - 被検物質の副作用リスクを評価する方法であって、前記の副作用リスクは自閉症を誘発または発症させるリスクで、かつ前記方法は、
(a) 被検物質を提供する工程と、
(b) 前記被検物質が施用される試験群では、前記被検物質の検出される指標に対する影響を測定し、かつ前記被検物質が施用されていない対照群では、同一の検出される指標のデータを測定し、ここで、前記の検出される指標はレチノイン酸のレベルまたは濃度、ALDH1Aの発現量または活性、あるいはこれらの組み合わせからなる群から選ばれる工程と、
(c) 試験群の前記検出される指標の測定結果Tを対照群における前記検出される指標の測定結果Cと比較する工程とを含み、
測定結果Tが測定結果Cよりも顕著に低い場合、被検物質が妊婦および/または嬰幼児に副作用を発生させるリスクを有することを示唆することを特徴とする方法。 - 前記被検物質は、薬物、日用化学品、またはこれらの組み合わせからなる群から選ばれことを特徴とする請求項12に記載の方法。
- 薬物の組み合わせまたは前記薬物の組み合わせを含むキットであって、前記薬物の組み合わせは、
(a) 薬学的に許容される担体と、クロトリマゾール、モンテルカスト、モンテルカストナトリウムからなる群から選ばれる第一の活性成分とを含有する第一の薬物組成物、および
(b) 薬学的に許容される担体と、ALDH1A、ALDH1A作動剤、ALDH1A触媒産物、ALDH1A触媒基質、またはこれらの組み合わせからなる群から選ばれる第二の活性成分とを含有する第二の薬物組成物
を含むことを特徴とする薬物の組み合わせまたはキット。 - (a) クロトリマゾール、モンテルカスト、モンテルカストナトリウムからなる群から選ばれる第一の活性成分と、
(b) ALDH1A、ALDH1A作動剤、ALDH1A触媒産物、ALDH1A触媒基質、またはこれらの組み合わせからなる群から選ばれる第二の活性成分と
を含むことを特徴とする薬物組成物。 - 自閉症を治療または予防する候補薬物を選別する方法であって、
(a) 被験の候補物質を提供する工程と、
(b) 前記被験の候補物質が施用される試験群では、前記被験の候補物質の検出される指標に対する影響を測定し、かつ前記被験の候補物質が施用されていない対照群では、同一の検出される指標のデータを測定し、ここで、前記の検出される指標はレチノイン酸のレベルまたは濃度、ALDH1Aの発現量または活性、あるいはこれらの組み合わせからなる群から選ばれる工程と、
(c) 試験群の前記検出される指標の測定結果Tを対照群における前記検出される指標の測定結果Cと比較する工程とを含み、
測定結果Tが測定結果Cよりも顕著に高い場合、前記被験の候補物質が自閉症を治療または予防する候補薬物として有用であることを示唆することを特徴とする方法。 - UBE3AとALDH1Aファミリータンパク質の結合を遮断することができるポリペプチドまたは抗体または化合物の使用であって、自閉症の治療および/または予防に使用される薬物組成物または製剤の製造のために使用されることを特徴とする使用。
