Abstract
Description
[材料及び方法]
<細胞源及び細胞培養>
健康なドナーの一次骨格細胞(ドナー1及びドナー3)はクロネテイクス(Clonetics)のヒト骨格筋筋芽細胞(Human Skeletal MuscleMyoblasts(HSMM)であった。
MyoScreen(サイトー、仏国)と呼称されるインビトロの完全自動ヒト筋管モデルを使用して、肥大及び萎縮回復アッセイを行った。このモデルは、ヒト一次筋芽細胞の分化をガイドするミクロ環境の厳密な制御に依存するものである。MyoScreen微小パターンに形成される筋管は、高度に画一化された形態とともに高レベルの成熟性を示す。
オペレッタハイコンテンツイメージングシステムにて画像を10倍倍率で得た。サイトーによるアカペラハイコンテンツイメージングソフトウエア(Perkin Elmer)において開発された専用のアルゴリズムを用いて、画像処理及び分析を行った。ウェルごとに11視野を得た。
筋管分化及び筋管サイズを増加させる肥大化合物を特定するために一次スクリーニングを行った。候補化合物を、ドナー3の細胞においてモノプリケートにて10μMでテストした。
再テストをチェリーピッキングにて行った(一次スクリーニングと同じ化合物群)。各ヒットを6ウェルのレプリケートにおいて一次スクリーニングにおけるものと同じ条件(ドナー3、10μM)でテストした。
用量反応アッセイを2人の健康なドナーにおいて(健康なドナー3及び1)、33μM〜0.015μMの8用量の候補化合物、用量ごとに2ウェルのレプリケートにて行った。
用量ごとにトリプリケートのウェルで対象の化合物を用量反応において数回テストした。結果をコントロール条件(基本レベル)に対してノーマライズし、グラフパッドmプリズムを使用してプロットした。「核カウント」の読み出しにより任意の毒性作用を検出できた。「融合指数」及び「ミオシン面積」の読み出しを、グラフパッドプリズムフィッティングソリューションを使用してEC50値を測定するために使用した。
萎縮誘発剤、すなわち、TNF−α、IL−1β、ミオスタチン、TGF−β、及びデキサメタゾン、並びに1μMトラニルシプロミンの存在下でトリプリケートにて萎縮回復アッセイを行った。
以下の表4に示すように、トラニルシプロミンは、+30%を超える融合指数及び/又は筋管面積の読み出し増加を誘発して、一次スクリーニング及び再テスト時に骨格筋肥大誘発剤として特定された。

[材料及び方法]
<アセチルコリン受容体(AchR)クラスター形成アッセイ>
MyoScreenプロトコルを実施例1の肥大及び萎縮回復アッセイにおいて記載したように行ったが、6日目ではなく9日目に停止した。アッセイの終了時に、トロポニンT及び核に加えて、AchRが特異的抗体を使用して免疫染色された。PerkinElmerのオペレッタハイコンテンツイメージングシステムにて画像を20倍で得た。アカペラハイコンテンツイメージングソフトウエア(Perkin Elmer)において開発された専用のアルゴリズムを用いて、画像処理及び分析を行った。核カウント及び筋管融合指数、AchRの数、AchR平均面積、筋管総面積によってノーマライズしたAchR総面積である、特定の読み出しを各ウェルにおいて計算した。
注目すべきことには、特異的抗AchR抗体で標識すると、分化後6日のMyoScreen筋管は、筋管線維の中途部及び筋管端部の特定領域において筋鞘膜に沿って区切るAchRクラスターを示す。自動AchRクラスターアッセイを開発するため、オペレッタ/アカペラシステム(材料及び方法を参照)を使用して画像におけるクラスターの凝集数及びサイズを分析するために十分な感受性があり堅牢なAchR検出ルーチンを開発した。AchRクラスター形成におけるGSK−LSD1の効果は、2人の健康なドナー(ドナー3及びドナー5)を用いて、神経筋接合部における化合物の正の効果を示すために評価された。処置の7日後、GSK−LSD1は、AchRクラスター総面積を、ドナー5モデルにおいて+260%、及びドナー3において+50%、有意に増加させる(図1を比較のこと)。

- 骨格筋組織喪失及び/又は筋衰弱をもたらす疾患又は障害の処置又は防止に使用される、LSD1阻害剤。

- 前記骨格筋組織喪失及び/又は筋衰弱をもたらす疾患又は障害は、廃用性筋萎縮、食欲不振性飢餓により誘発された萎縮、及び急性筋損傷又はオーバーユース筋損傷を含む筋損傷から選択される、請求項1〜15のいずれかに記載の使用のためのLSD1阻害剤。
- 共に、個別に、又は連続して使用するための組合せ製剤としての、請求項1〜14のいずれかに記載のLSD1阻害剤と骨格筋萎縮を誘発する化合物とを含有する製品。
- 前記骨格筋萎縮を誘発する化合物は治療剤である、請求項22に記載の製品。
- 前記骨格筋萎縮を誘発する化合物は、副腎皮質ステロイド、コルヒチン、クロロキン、ヒドロキシクロロキン、D−ペニシラミン、抗生物質、β遮断薬、アミオダロン、シメチジン、ジドブジン、ビンクリスチン、クロフィブラート、スタチン、フィブラート、シクロスポリン、L−トリプトファン、低カリウム血症誘発薬剤、脂質降下剤、及び、ワクチンなど筋肉内経路で投与される治療剤よりなる群から選択される、請求項23に記載の製品。
- 前記骨格筋萎縮を誘発する化合物は脂質降下剤であり、好ましくはスタチン及びフィブラートから選択される、請求項24に記載の製品。
- 対象における筋量、筋力、及び/又は筋パフォーマンスを増加させるための、請求項1〜14のいずれかに記載のLSD1阻害剤の非治療的使用。
- 動物用飼料組成物の材料又は添加剤としての、請求項1〜14のいずれかに記載のLSD1阻害剤の非治療的使用。
- 対象における骨格筋量の喪失を防止するための、請求項1〜14のいずれかに記載のLSD1阻害剤の非治療的使用。
- 家畜の性能を向上させる方法であって、請求項1〜14のいずれかに記載のLSD1阻害剤を前記家畜に与えることを含む方法。
