Abstract
Description
本開示は、4つのペプチド:
医薬組成物は、最高で4つの本明細書に開示されるペプチド及び薬学的に許容されるビヒクルを含む。「薬学的に許容されるビヒクル」は、ペプチドの生物学的活性に影響しない1種または複数種の物質を含んでよく、患者に投与された場合、有害反応を引き起こさない。医薬組成物は、液体であってもよく、又は凍結乾燥されてもよい。凍結乾燥組成物は、組成物の再構成に使用するのに適する液体、典型的には、注射用水(WFI)と共にキットに提供され得る。医薬組成物は、静脈内、筋肉内、皮内、腹腔内、及び皮下投与を含むが、これらに限定されない任意の適切な経路によって投与することができる。
本明細書に開示される医薬組成物は、いくつかの治療的用途を有する。いくつかの実施形態において、本明細書に開示される医薬組成物は、癌などの過剰増殖性障害の進行を阻害するために患者に投与することができる。そのような阻害は、例えば、新生物又は前新生物細胞の増殖を低減すること、新生物又は前新生物細胞を破壊すること;及び転移を阻害すること又は腫瘍のサイズを減少させることを含むことができる。
TriCo−20(商標)(TRICO−20(商標))及びTriCo−16(商標)(TRICO−16(商標))ファージディスプレイペプチドライブラリー(Creative Biolabs,45−1 Ramsey Road,Shirley,NY 11967)を、可溶性組換えヒトPD−1受容体の結合体を特定するためにスクリーニングした。パニングの第4ラウンド後、特異的結合体についての明白な濃縮が観察され、個々のペプチドは、クローンファージELISAにおいて弱い特異的結合体として確認された。パニングの第5ラウンドにより、さらに濃縮した。表1に、クローンファージELISAにおいて強い特異的結合を示した4つのペプチドを記載する。

簡単に説明すると、Jurkat細胞上の細胞表面PD−1の検出は、ヒトPD−L1−Fc融合タンパク質と細胞をインキュベートし、その後、蛍光標識抗ヒトFc抗体での組換え分子の検出によって達成した。フローサイトメトリーを、PD−1とPD−L1組換えタンパク質との間の結合を検出するために行った。次いで、定量的な結合測定値を、平均蛍光強度(MFI)によって決定した。
細胞ベースのレポーターアッセイを用いて、上記で特定した4つのペプチドの結合が、PD−1とそのリガンドPD−L1との相互作用を阻止するのに十分であったかどうかを評価した。該アッセイの成分には、該アッセイにおいて測定可能な効果をもたらすヒトPD−1及びルシフェラーゼレポーターを安定に発現するJurkat T細胞株、ヒトPD−L1を安定に発現したCHO細胞株、及びPD−1とPD−L1の相互作用を阻止する陽性対照抗PD−1抗体が含まれる。Jurkat T細胞株におけるルシフェラーゼレポーターは、プロモーター領域におけるIL−1、NFAT、又はNF−κB応答エレメントによって誘発される。Jurkat T細胞をCD3で前処理し、該アッセイで使用するためにすぐに凍結保存する。Jurkat T細胞とPD−L1発現細胞株との相互作用は、ルシフェラーゼ構築物が活性化される細胞内メカニズムを阻害し、それによってルシフェラーゼ発現を防止する。Jurkat T細胞上のPD−1又はCHO細胞上のPD−L1のいずれかに結合してそれらの相互作用を十分に防止する分子は、Jurkat T細胞がルシフェラーゼを産生するのを可能にする。CellTiter−Glo(登録商標)(CELLTITER−GLO(登録商標)、Promega)を用いて、ルシフェラーゼ発現を測定した。
ペプチド1〜4を、ヒトPBMCベース破傷風抗原リコールアッセイで試験した。「ペプチドCQ−22」を、陰性対照として使用した。

ペプチドの組み合わせを、異なるPBMCドナー及び破傷風トキソイドの異なるロット番号を用いて、上記の抗原リコールアッセイで試験した。結果を図16、17、18、19、20、21、及び22に示す。これらの結果は、4つのペプチドQP20、HD20、WQ20、及びSQ20の4つのペプチド組み合わせの組み合わせがIL−2産生の増加及びIL−17a産生の減少をもたらすことを実証した。
BIACORE(登録商標)アッセイを、25℃でBIACORE(登録商標)T−200を用いて行った。該アッセイ及び再生緩衝液は、10mMのHEPES(pH7.4)、150mMのNaCl、3mMのEDTA、及び0.05%のP20を含有していた。固定化緩衝液は、10mMの酢酸ナトリウム、pH5.0であった。リガンドを固定化するために使用した流速は、5μl/分であった。動態分析のための流速は、30μl/分であった。
ヒトPD−1受容体の12,000応答単位(RU)及びマウスPD−1受容体の6000 RUを、アミンカップリング法(EDC/NHS)によってCM5チップのフローセル2及びフローセル4上に直接固定化した。非占有部位を、1Mのエタノールアミンでブロックした。スカウティングを、結合の有無を確認するために、25μΜの単一検体濃度で行った。フローセル1をブランクとして保持し、基準減算(reference subtraction)に使用した。リガンドへの検体の結合をリアルタイムで監視した。
スカウティングの結果に基づいて、新しいチップへより高いRUのリガンドを固定し、25μΜの検体濃度とそれに続いて12.5、6.25、3.125、1.562、0.78及び0μΜ濃度に連続希釈して、又は示されるように完全な動態を行った。速いオン速度及びオフ速度により、定常状態の平衡動態によってKDを決定した。
ペプチドの有効性を、B16−F10−LacZ実験的転移モデルで評価した。このモデルにおいて、細胞内酵素のβガラクトシドをコードするLacZ遺伝子を発現するようにトランスフェクトされたB16−F10−LacZ細胞を、同系マウスの尾静脈に注射する。細胞は、循環を通って移動し、肺に定着し、腫瘍を形成する。マウスを移植後2週間で屠殺する。酵素がその基質X−galを切断する場合、生成物は二量体化し、色を変え、エクスビボで検出することができる。その後、肺の表面上の転移性腫瘍の数を、解剖顕微鏡下で腫瘍の手動計数によって定量する。

予防ワクチンアジュバントとしてのペプチド組み合わせの免疫原性を、マラリアのマウスモデルで評価した。プラスモジウム・ヨエリのスポロゾイト周囲タンパク質(AdPyCS)を発現するアデノウイルスに基づくマラリアワクチンで免疫したBalb/cマウスに、200μgのペプチド組み合わせ、抗PD−1mAb、抗PDL1mAb、又は陰性対照ペプチドオボアルブミン(OVA)を、AdPyCSによる免疫付与後1、3、5、及び7日目に与えた(表5)。追加のアジュバントをAdPyCS抗原に添加しなかったことに留意されたい。免疫付与の12日後に脾臓を回収し、脾臓PyCS特異的IFNγ分泌CD8+T細胞数を、ELISpotアッセイによって決定した。ELISpotアッセイのために、脾細胞をPyCSのH−2Kd制限CD8+T細胞エピトープであるSYVPSAEQIペプチド(配列番号5)で刺激したことに留意されたい。

敗血症はT細胞機能及び生存を負に変化させ得るが、これは、PD−1:PDL1相互作用が阻止された場合に逆になり得、生存率が改善される。したがって、CD1マウスを盲腸結紮及び穿刺(CLP)に供して、腹腔内腹膜炎を誘導した敗血症の代表的な、臨床的に関連するモデルでペプチド組み合わせの有効性を評価した。この研究のために、ペプチド組み合わせ又は抗PD−1抗体のいずれか200μgを、手術の2、24、48、72及び96時間後に静脈内投与した。ビヒクル対照群も含めた。各群には6匹のマウスがいた。全てのマウスを、罹患率及び死亡率の兆候について毎日2回チェックした。該ペプチド組み合わせの投与により、該ペプチド組み合わせが2倍高い生存率を示し、ビヒクル対照群を上回って延命効果が増強された(表6)。さらに、該ペプチド組み合わせの群における生存率は、抗PD−1抗体での処置をわずかに上回った。

T細胞の消耗及び免疫寛容におけるPD−1の役割が実証されているB型肝炎ウイルス(HBV)マウスモデルでQP20、HD20、WQ20、及びSQ20ペプチドの組み合わせを評価した(Tzeng et al.,2012;Ye et al.,2015)。PD−1は、持続的HBV感染を有するマウスの肝臓T細胞において上昇しているが、感染をクリアした動物では上昇しない。このモデルにおいて、PD−1/PD−L1の相互作用の抗PD−1mAbによる阻害により、肝臓T細胞による抗原特異的IFNγ産生を増加させ、HBVの持続性を逆転することの両方が示された(Tzeng et al.,2012)。持続的HBVのこのマウスモデルは、QP20、HD20、WQ20、及びSQ20ペプチド組み合わせが、インビボでのT細胞消耗を無くし、ウイルス感染を制御する際に免疫系を助けることができるかどうかを試験する機会を提供した。

からなる群から選択される、請求項1に記載の医薬組成物。 - 前記4つのペプチドのうちの3つのみを含み、該3つのペプチドが、
(a)配列番号1のアミノ酸配列からなるペプチド、配列番号2のアミノ酸配列からなるペプチド、及び配列番号3のアミノ酸配列からなるペプチド;
(b)配列番号1のアミノ酸配列からなるペプチド、配列番号2のアミノ酸配列からなるペプチド、及び配列番号4のアミノ酸配列からなるペプチド;
(c)配列番号2のアミノ酸配列からなるペプチド、配列番号3のアミノ酸配列からなるペプチド、配列番号4のアミノ酸配列からなるペプチド;及び
(d)配列番号1のアミノ酸配列からなるペプチド、配列番号3のアミノ酸配列からなるペプチド、配列番号4のアミノ酸配列からなるペプチド、
からなる群から選択される、請求項1に記載の医薬組成物。 - 前記ペプチドの全4つを含む、請求項1に記載の医薬組成物。
- 過剰増殖性障害の進行を阻害して、感染性疾患を治療するため、又は敗血症を治療するための方法であって、請求項1〜5のいずれかに記載の有効量の医薬組成物を、それを必要とする患者に投与することを含む、方法。
- 前記組成物が、過剰増殖性障害の進行を阻害するために投与される、請求項6に記載の方法。
- 前記過剰増殖性障害が癌である、請求項7に記載の方法。
- 前記癌が黒色腫である、請求項8に記載の方法。
- 前記患者に癌ワクチンを投与することをさらに含む、請求項8に記載の方法。
- 前記患者にキメラ抗原受容体(CAR)T細胞療法を施すことをさらに含む、請求項7に記載の方法。
- 前記組成物が、感染性疾患を治療するために投与される、請求項6に記載の方法。
- 前記感染性疾患がマラリアである、請求項12に記載の方法。
- 前記感染性疾患がB型肝炎である、請求項12に記載の方法。
- 前記組成物が、前記感染性疾患に対するワクチンのワクチンアジュバントとして投与される、請求項12に記載の方法。
- 前記組成物が、敗血症を治療するために投与される、請求項6に記載の方法。
- 過剰増殖性障害の進行を阻害するため、感染性疾患を治療するため、又は敗血症を治療するための医薬の製造における、請求項1〜5のいずれかに記載の医薬組成物の使用。
- 過剰増殖性障害の進行を阻害するため、感染性疾患を治療するため、又は敗血症を治療するために使用するための、請求項1〜5のいずれかに記載の医薬組成物。
