類似のタンパク質または断片への特異的結合についてのタンパク質足場のスクリーニングは、ヌクレオチド(DNAもしくはRNAディスプレイ)またはペプチドディスプレイライブラリー、例えばin vitroディスプレイを使用して、便利に達成することができる。この方法は、所望の機能または構造を有する個々のメンバーについてペプチドの大きなコレクションをスクリーニングすることを含む。提示されるヌクレオチドまたはペプチド配列は、長さが3〜5000ヌクレオチドもしくはアミノ酸またはそれより長いこともあり、高頻度に5〜100アミノ酸長でありえ、多くの場合、約8〜25アミノ酸長でありうる。ペプチドライブラリーを生成するための直接化学合成法に加えて、いくつかの組換えDNA法が記載されている。1つのタイプは、バクテリオファージまたは細胞の表面でのペプチド配列の提示を含む。各バクテリオファージまたは細胞は、提示される特定のペプチド配列をコードするヌクレオチド配列を含有する。そのような方法は、PCT特許公開第91/17271号、同第91/18980号、同第91/19818号および同第93/08278号に記載されている。

本開示の単離された核酸を、公知の方法による直接化学合成により調製することもできる(例えば、Ausubelら、上掲を参照されたい)。化学合成は、一般に、一本鎖オリゴヌクレオチドを産生し、その一本鎖オリゴヌクレオチドを、相補配列とのハイブリダイゼーションにより、または鋳型として一本鎖を使用するDNAポリメラーゼを用いる重合により、二本鎖DNAに変換することができる。DNAの化学合成を約100塩基またはそれより多い塩基の配列に限定することができるが、より長い配列を、より短い配列のライゲーションにより得ることができることは、当業者には認識される。
本開示は、本開示の核酸を含む組換え発現カセットをさらに提供する。本開示の核酸配列、例えば、本開示のタンパク質足場をコードするcDNAまたはゲノム配列を使用して、少なくとも1つの所望の宿主細胞に導入することができる組換え発現カセットを構築することができる。組換え発現カセットは、所期の宿主細胞におけるポリヌクレオチドの転写を指示する転写開始調節配列に作動可能に連結された本開示のポリヌクレオチドを通常は含むことになる。異種プロモーターと非異種(すなわち、内在性)プロモーターの両方を、本開示の核酸の発現を指示するために用いることができる。

本発明のタンパク質足場化合物、組成物または組み合わせは、任意の好適な助剤(auxiliary)、例えば、これらに限定されないが、希釈剤、結合剤、安定剤、緩衝剤、塩、親油性溶媒、保存薬、アジュバントなどのうちの少なくとも1つをさらに含んでもよい。薬学的に許容される助剤が好ましい。そのような滅菌溶液の非限定的な例およびそれらを調製する方法は、当技術分野において周知であり、例えば、Gennaro編、Remington’s Pharmaceutical Sciences、第18版、Mack Publishing Co.(Easton、Pa.)1990年があるが、これに限定される。当技術分野において周知である、または本明細書に記載される、タンパク質足場、断片または改変体組成物の投与方式、溶解度および/または安定性にとって好適である、薬学的に許容される担体は、慣用的に選択することができる。

本発明は、当技術分野において公知であるような、または本明細書に記載されるような、細胞、組織、器官、動物または患者における疾患をモジュレートまたは処置する方法であって、本発明の少なくとも1つのタンパク質足場を使用して、例えば、細胞、組織、器官、動物もしくは患者に治療有効量のタンパク質足場を投与して、または細胞、組織、器官、動物もしくは患者を治療有効量のタンパク質足場と接触させて、モジュレートまたは処置する方法も提供する。本発明は、細胞、組織、器官、動物または患者における、これに限定されないが悪性疾患を含む、疾患をモジュレートまたは処置する方法も提供する。
本発明による少なくとも1つのタンパク質足場の薬学的有効量を投与するために、多くの公知のおよび開発された方式を、本発明に従って使用することができる。経肺投与が下記の説明の中で使用されるが、他の投与方式を本発明に従って好適な結果で使用することができる。ここで説明されるまたは当技術分野において公知の吸入または他の方式による投与に好適な様々なデバイスおよび方法のいずれかを使用して、担体の中の、溶液、エマルジョン、コロイドもしくは懸濁物としての、または乾燥粉末としての、本発明のタンパク質足場を送達することができる。
非経口投与用の製剤は、一般的な賦形剤として、滅菌水または食塩水、ポリアルキレングリコール、例えばポリエチレングリコール、植物由来の油、水素化ナフタレンなどを含有することができる。注射用の水性または油性懸濁物は、公知の方法に従って、適切な乳化剤または湿潤剤(humidifier)および懸濁化剤を使用することにより、調製することができる。注射用の薬剤は、水溶液、溶媒中の滅菌注射用溶液または懸濁物などの、非毒性で非経口投与可能な希釈剤でありうる。使用可能なビヒクルまたは溶媒として、水、リンゲル液、等張食塩水などが許容される。通常の溶媒または懸濁溶媒として、滅菌不揮発性油を使用することができる。これらのために、天然または合成または半合成脂肪油または脂肪酸、天然または合成または半合成モノまたはジまたはトリグリセリドを含む、任意の種類の不揮発性油および脂肪酸を使用することができる。非経口投与は当技術分野において公知であり、非経口投与は、これらに限定されないが、従来の注射手段、米国特許第5,851,198号に記載されているようなガス加圧式無針注射デバイス、および米国特許第5,839,446号に記載されているようなレーザー穿孔デバイス(laser perforator device)を含み、特許文献は、全体が参照により本明細書に援用される。
本発明は、非経口、皮下、筋肉内、静脈内、関節内(intrarticular)、気管支内、腹部内、嚢内、軟骨内、腔内(intracavitary)、腔内(intracelial)、小脳内、脳室内、結腸内、頸部内、胃内、肝内、心筋内、骨内、骨盤内、心膜内、腹腔内、胸膜内、前立腺内、肺内、直腸内、腎内、網膜内、脊髄内、滑液包内、胸腔内、子宮内、膀胱内、病巣内、ボーラス、経膣、直腸、頬側、舌下、鼻腔内または経皮手段による、少なくとも1つのタンパク質足場の投与に、さらに関する。少なくとも1つのタンパク質足場組成物は、非経口(皮下、筋肉内もしくは静脈内)または任意の他の投与に使用するために、特に、溶液または懸濁物の形態で調製することができ;経膣または直腸投与の際に使用するために、特に、これらに限定されないがクリームおよび坐剤などの、半固体形態で調製することができ;頬側または舌下投与のために、例えば、これらに限定されないが、錠剤またはカプセルの形態で調製することができ;または鼻腔内使用のために、例えば、これらに限定されないが、粉末、点鼻薬(nasal drop)またはエアロゾルまたはある特定薬剤の形態で調製することができ;または経皮使用のために、例えば、これらに限定されないが、皮膚構造を修飾するためのもしくは経皮パッチ中の薬物濃度を増加させるためのジメチルスルホキシドなどの化学的促進剤(全体が参照により本明細書に援用される、Jungingerら、「Drug Permeation Enhancement」、Hsieh, D. S.編、59〜90頁(Marcel Dekker, Inc.、New York 1994年))を伴う、あるいはタンパク質およびペプチドを含有する製剤の皮膚への塗布(WO98/53847)を可能にする、またはエレクトロポレーションなどの、一過性輸送経路を生じさせるための、もしくはイオントフォレーシスなどの、皮膚を通しての荷電薬物の移動を増加させるための電場の印加、またはソノフォレーシスなどの、超音波の適用(米国特許第4,309,989号および同第4,767,402号)を可能にする、酸化剤を伴う、ゲル、軟膏、ローション、懸濁物またはパッチ送達システムの形態で調製することができる(上記刊行物および特許は、全体が参照により本明細書に援用される)。
経肺投与のために、好ましくは、少なくとも1つのタンパク質足場組成物は、肺の下気道または洞への到達に有効な粒径で送達される。本発明によれば、少なくとも1つのタンパク質足場を、吸入による治療剤の投与のための当技術分野において公知の様々な吸入または鼻用デバイスのいずれによっても送達することができる。患者の洞または肺胞内にエアロゾル化した製剤を堆積させることができるこれらのデバイスは、定量吸入器、ネブライザー、乾燥粉末発生器、噴霧器などを含む。タンパク質足場の経肺または経鼻投与を導くのに好適な他のデバイスも、当技術分野において公知である。すべてのそのようなデバイスは、エアロゾルでタンパク質足場を調剤するための投与に好適な製剤を使用することができる。そのようなエアロゾルは、溶液(水性および非水性両方)からなってもよく、または固体粒子からなってもよく。
タンパク質足場組成物を含む噴霧剤は、圧力下で、少なくとも1つのタンパク質足場の懸濁物または溶液を強制的にノズルに通すことにより生じさせることができる。所望のアウトプットおよび粒径を獲得するために、ノズルのサイズおよび構成、印加圧力ならびに液体供給速度を選択することができる。例えば、毛管またはノズルフィードと接続している電場により、エレクトロスプレーを生じさせることができる。有利なことに、噴霧器により送達される少なくとも1つのタンパク質足場組成物の粒子は、約10μm未満の粒径、好ましくは約1μm〜約5μmおよび最も好ましくは約2μm〜約3μmの範囲の粒径を有する。
本発明のタンパク質足場組成物を、ジェットネブライザーまたは超音波ネブライザーなどの、ネブライザーにより投与することができる。通常は、ジェットネブライザーの場合、圧縮空気源を使用して、開口部を通して高速空気ジェットを生じさせる。ガスがノズルを越えて膨張すると、低圧領域が作られ、それにより、液体レザバに接続された毛管を通してタンパク質足場組成物の溶液が引っ張られる。毛管からの液体流は、毛管から出ると剪断されて不安定なフィラメントおよび液滴になり、その結果、エアロゾルが生じる。所与のジェットネブライザーから所望の性能特性を獲得するために、様々な構成、流速、およびバッフルのタイプを用いることができる。超音波ネブライザーの場合、高周波電気エネルギーを使用して、通常は圧電変換器を用いて、振動、機械エネルギーを生じさせる。このエネルギーが、タンパク質足場組成物の製剤に、直接、またはカップリング流体を介して伝達され、その結果、タンパク質足場組成物を含むエアロゾルが生じる。有利には、ネブライザーにより送達されるタンパク質足場組成物の粒子は、約10μm未満の粒径、好ましくは約1μm〜約5μmおよび最も好ましくは約2μm〜約3μmの範囲の粒径を有する。
定量吸入器(MDI)では、噴霧体、少なくとも1つのタンパク質足場、および任意の賦形剤または他の添加剤が、液化圧縮ガスを含む混合物として、キャニスターの中に収容されている。計量弁(metering valve)の作動により、混合物が、約10μm未満、好ましくは約1μm〜約5μm、および最も好ましくは約2μm〜約3μmのサイズ範囲の粒子を好ましくは含有するエアロゾルとして、放出される。ジェットミリング、噴霧乾燥、臨界点凝結などを含む、当業者に公知の様々な方法により産生されたタンパク質足場組成物の製剤を用いることによって、所望のエアロゾル粒径を得ることができる。好ましい定量吸入器は、3MまたはGlaxoにより製造されたおよびヒドロフルオロカーボン噴霧体を用いるものを含む。定量吸入器デバイスで使用するための少なくとも1つのタンパク質足場の製剤は、非水性媒体中の懸濁物、例えば、界面活性剤の補助の下、噴霧体に懸濁された懸濁物として少なくとも1つのタンパク質足場を含有する微細に分割された粉末を、通常は含む。噴霧体は、この目的で用いられる任意の従来の材料、例えば、クロロフルオロカーボン、ヒドロクロロフルオロカーボン、ヒドロフルオロカーボン、または炭化水素(トリクロロフルオロメタン、ジクロロジフルオロメタン、ジクロロテトラフルオロエタノールおよび1,1,1,2−テトラフルオロエタンを含む)、HFA−134a(ヒドロフルオロアルカン−134a)、HFA−227(ヒドロフルオロアルカン−227)などでありうる。好ましくは、噴霧体は、ヒドロフルオロカーボンである。例えば、噴霧体中の懸濁物として少なくとも1つのタンパク質足場を安定化するために、または活性薬剤を化学的分解から保護するなどのために、界面活性剤を選択することができる。好適な界面活性剤は、トリオレイン酸ソルビタン、大豆レシチン、オレイン酸などを含む。一部の事例では、エタノールなどの溶媒を使用する溶液エアロゾルが好ましい。タンパク質の製剤のための当技術分野において公知のさらなる薬剤も、製剤に含めることができる。本明細書に記載されないデバイスによる少なくとも1つのタンパク質足場組成物の経肺投与により本発明の方法を達成することができることは、当業者には認識される。
経口投与用の製剤は、腸壁の透過性を人工的に上昇させるためのアジュバント(例えば、レゾルシノールならびに非イオン性界面活性剤、例えばポリオキシエチレンオレイルエーテルおよびn−ヘキサデシルポリエチレンエーテル)の共投与、ならびに酵素的分解を阻害するための酵素阻害剤(例えば、膵臓トリプシンインヒビター、ジイソプロピルフルオロリン酸(DFF)およびトラジロール)の共投与に依拠する。経口、頬側、粘膜、経鼻、経肺、経膣、膜貫通または直腸投与が意図された、タンパク質とタンパク質足場と少なくとも2つの界面活性剤の組み合わせとを含む親水性薬剤の送達のための製剤は、米国特許第6,309,663号で教示されている。経口投与のための固体型の剤形の活性成分化合物を、スクロース、ラクトース、セルロース、マンニトール、トレハロース、ラフィノース、マルチトール、デキストラン、デンプン、寒天、アルギネート、キチン、キトサン、ペクチン、トラガカントゴム、アラビアゴム、ゼラチン、コラーゲン、カゼイン、アルブミン、合成または半合成ポリマー、およびグリセリドを含む、少なくとも1つの添加剤と混合することができる。これらの剤形は、他のタイプの添加剤、例えば、不活性希釈剤、滑沢剤、例えばステアリン酸マグネシウム、パラベン、保存剤、例えばソルビン酸、アスコルビン酸、アルファ−トコフェロール、抗酸化剤、例えばシステイン、崩壊剤、結合剤、増粘剤、緩衝化剤、甘味剤、矯味矯臭剤、芳香剤なども含有することができる。
生物活性薬剤を経口投与するための製剤であって、マイクロカプセルを生じさせるように1つまたは複数の生体適合性ポリマーまたはコポリマー賦形剤に、好ましくは、1つの生体分解性ポリマーまたはコポリマーにカプセル化されており、活性薬剤が、結果として生じるマイクロカプセルの適正なサイズのため、動物の、他に「パイエル板」または「GALT」としても公知の集合リンパ小節に到達し吸収され、胃腸管を通過した薬剤に起因する有効性の損失のない、製剤。同様の集合リンパ小節を、気管支(bronchei tube)(BALT)および大腸において見いだすことができる。上記組織は、一般に粘膜関連リンパ組織(mucosally associated lymphoreticular tissue)(MALT)と称される。粘膜表面による吸収のために、少なくとも1つのタンパク質足場を投与する組成物および方法は、複数のサブミクロン粒子を含むエマルジョンと、粘膜付着性高分子と、生理活性ペプチドと、水性連続相とを含み、エマルジョン粒子の粘膜付着を達成することにより粘膜表面による吸収を促進する(米国特許第5,514,670号)。本発明のエマルジョンの適用に好適な粘膜表面は、角膜、結膜、頬側、舌下、経鼻、経膣、経肺、胃、腸および直腸投与経路を含むことができる。経膣または直腸投与用の製剤、例えば坐剤は、賦形剤として、例えば、ポリアルキレングリコール、ワセリン、カカオ脂などを含有することができる。鼻腔内投与用の製剤は、固体であってもよく、賦形剤として例えばラクトースを含有することができ、または点鼻薬の水性もしくは油性溶液であってもよい。頬側投与のための賦形剤は、糖、ステアリン酸カルシウム、ステアリン酸マグネシウム、アルファ化デンプン(pregelinatined starch)などを含む(米国特許第5,849,695号)。
経皮投与のために、少なくとも1つのタンパク質足場は、リポソームもしくはポリマーナノ粒子、マイクロ粒子、マイクロカプセルまたはマイクロスフェア(別段の記述がない限り、まとめてマイクロ粒子と称される)などの、送達デバイスにカプセル化される。合成ポリマー、例えば、ポリヒドロキシ酸、例えば、ポリ乳酸、ポリグリコール酸およびこれらのコポリマー、ポリオルトエステル、ポリ無水物、およびポリホスファゼン、ならびに天然ポリマー、例えば、コラーゲン、ポリアミノ酸、アルブミンおよび他のタンパク質、アルギネートおよび他の多糖類、ならびにこれらの組み合わせで作製されたマイクロ粒子を含む、多数の好適なデバイスが公知である(米国特許第5,814,599号)。
長期間にわたって、例えば、単回投与から1週間〜1年の期間、本発明の化合物を被験体に送達することが、望ましいこともある。様々な緩徐放出、デポーまたはインプラント剤形を利用することができる。例えば、剤形は、体液への低い溶解度を有する化合物の薬学的に許容される非毒性塩、例えば、(a)リン酸、硫酸、クエン酸、酒石酸、タンニン酸、パモ酸、アルギン酸、ポリグルタミン酸、ナフタレンモノもしくはジスルホン酸、ポリガラクツロン酸などの多塩基酸との、酸付加塩(acid addition salt);(b)亜鉛、カルシウム、ビスマス、バリウム、マグネシウム、アルミニウム、銅、コバルト、ニッケル、カドミウムなどの多価金属陽イオンとの塩、または例えばN,N’−ジベンジル−エチレンジアミンもしくはエチレンジアミンから形成された、有機陽イオンとの塩;または(c)(a)と(b)の組み合わせ、例えば、亜鉛タンニン酸塩を含有することができる。加えて、本発明の化合物または、好ましくは、今述べたものなどの比較的不溶性の塩を、注射に好適なゲル、例えば、モノステアリン酸アルミニウムの、例えばゴマ油とのゲルに、製剤化することができる。特に好ましい塩は、亜鉛塩、亜鉛タンニン酸塩、パモ酸塩などである。注射用の別のタイプの緩徐放出デポー製剤は、例えば米国特許第3,773,919号に記載されているようなポリ乳酸/ポリグリコール酸ポリマーなどの、緩徐分解性、非毒性、非抗原性ポリマーへのカプセル化のために分散される化合物または塩を含有する。化合物または、好ましくは、上記のものなどの比較的不溶性の塩を、特に動物での使用のために、コレステロールマトリクスシラスティックペレットに製剤化することもできる。さらなる緩徐放出、デポーまたはインプラント製剤、例えば、気体または液体リポソームが、文献で公知である(米国特許第5,770,222号および「Sustained and Controlled Release Drug Delivery Systems」、J. R. Robinson編、Marcel Dekker, Inc.、N.Y.、1978年)。
MUC1nは、上皮細胞により主として発現される、広範にO−グリコシル化されているタンパク質である。分泌および膜結合MUC1nは、毒素、微生物、および外部環境と境界面で発生する他の形態のストレスにより誘導される損傷から上皮細胞の頂端境界を保護する物理的バリアを形成する。膜貫通MUC1n 1(MUC1)は、細胞の内部に、その細胞質ドメインを通ってシグナルを伝達することもできる。MUC1には、ウニ精子タンパク質−エンテロキナーゼ−アグリン(SEA)ドメインの存在を除いて、他の膜結合MUC1nとの配列類似性がない。その点において、MUC1は、単一ポリペプチドとして翻訳され、その後、SEAドメインにおいて自己切断を経る。

本開示は、本開示の1つまたは複数のCARおよび/またはCARTyrinを発現する修飾細胞であって、それを必要とする被験体に投与するために選択および/または増殖された修飾細胞を提供する。本開示の修飾細胞は、室温および体温を含む任意の温度で保管するために製剤化することができる。本開示の修飾細胞は、凍結保存およびその後の解凍のために製剤化することができる。本開示の修飾細胞は、滅菌包装からの被験体への直接投与のために薬学的に許容される担体中で製剤化することができる。本開示の修飾細胞は、最低レベルの細胞機能およびCAR/CARTyrin発現を保証するための細胞生存度および/またはCAR/CARTyrin発現レベルの指示薬とともに薬学的に許容される担体中で製剤化することができる。本開示の修飾細胞は、さらなる増殖を阻害するためのおよび/または細胞死を防止するための1つまたは複数の試薬とともに処方された密度で薬学的に許容される担体中で製剤化することができる。
本開示の誘導性アポトーシス促進性ポリペプチドは、既存の誘導性ポリペプチドより優れている。なぜなら、本開示の誘導性アポトーシス促進性ポリペプチドのほうが、はるかに免疫原性が低いからである。本開示の誘導性アポトーシス促進性ポリペプチドは、組換えポリペプチドであり、したがって、天然に存在しないが、本開示の誘導性アポトーシス促進性ポリペプチドを産生するように組み換えられる配列は、宿主ヒト免疫系が「非自己」と認識し、その結果、本開示の誘導性アポトーシス促進性ポリペプチド、誘導性アポトーシス促進性ポリペプチドを含む細胞、または誘導性アポトーシス促進性ポリペプチドもしくは誘導性アポトーシス促進性ポリペプチドを含む細胞を含む組成物を受容する被験体において、免疫応答を誘導する可能性がある、非ヒト配列を含まない。
本開示のMUC1結合タンパク質足場(Centyrinとも称される)を、MUC1−C/細胞外ドメイン(MUC1−C/ECD)を含む、好ましい標的であるMUC1に特異的に結合するように、生成することができる。
標的の検証:Poseidaにより提供された標的材料(Muc1−C融合タンパク質)をプルダウン実験で試験して、パニング手順におけるそれらの有用性を検証する。
ベンチマークscFv:E.coli宿主、ペリプラズムでの産生、修飾なし、可溶性形態として産生される。
単一濃度結合ELISAによるヒットの同定
クローンアウト(clone out):CISディスプレイ選択の産物をPCRにより増幅し、発現ベクターにクローニングし、E.coliに形質転換する。産生されたクローンを96ウェルプレート(キャンペーン規模に依存して1選択アウトプット当たり少なくとも1プレート)に採取する。
三次スクリーニング:CARTyrin機能スクリーニングの最初の代替物として、膜提示形態のMUC1−Cへの組換えタンパク質結合物質の到達可能性を、FACSを使用して確認する。MUC1−Cがトランスフェクトされた宿主細胞または対照宿主細胞をELISAにおいてCentyrin結合組換えタンパク質の単一希釈物とともにインキュベートする。scFvの細胞への結合を、二次抗3×FLAG−FITCコンジュゲートとのインキュベーション、続いてのフローサイトメーターでの分析により検出する。前の手順でトランスフェクト細胞に選択的に結合すると決定された一方または両方のscFvは、陽性対照としての役割を果すことができる。
パニング:以前のスクリーニングラウンドの結果および必要とされる所望のアフィニティーに依存して、さらなるパニングラウンドを行ってもよい。これらのさらなるパニングラウンドは、アフィニティーをより緩徐な解離速度にさせるために非固定化抗原を組み込む洗浄ステップを含む場合がある。
生物物理学的分析:Centyrin材料の小規模なプレートベースのHisタグアフィニティー精製を可能にするために、抗原選択1回当たり96までの固有ヒットを再整列(re−array)させ、再増殖させてもよい。精製された材料をサイズ排除クロマトグラフィーに付して、どの候補が単量体(非凝集)タンパク質として挙動するかを決定することになる。
固定化組換えタンパク質標的に対するBLIを使用して、選ばれた候補Centyrinについて最終的な結合アフィニティー定数を生成する。データは、候補Centyrinとそれらが選択された対象である組換え標的との間の結合強度の尺度である、解離速度(kd)およびKd値を提供する。
Amaxa 4Dヌクレオフェクターを使用して、4つのpiggyBacトランスポゾンのうちの1つをヒト汎T細胞にヌクレオフェクトした。「模擬」条件を受ける修飾T細胞には、空のpiggyBacトランスポゾンをヌクレオフェクトした。修飾T細胞は、治療剤(CARTyrinをコードする配列)のみを含有するpiggyBacトランスポゾン、または組み込まれているiC9配列と治療剤(CARTyrinをコードする配列)とを含有するpiggyBacトランスポゾンのいずれかを受けた。
MUC1のエピトープに特異的に結合する一本鎖抗体を含む抗原認識領域を有するキメラ抗原受容体(CAR)を生成した。例示的なMUC1−scFv CARの図を図11に示す。
