Abstract
Description

ある実施形態において、前記 PMSA 阻害剤 [18F] DCFPyL は、単一の DCFPyL 前駆体、5-(((S)-6-(tert-ブトキシ)-5-(3-((S)-1, 5-ジ-tert-ブトキシ-1, 5-ジオキソペンタン-2-イル)ウレイド)-6-オキソヘキシル)カルバモイル)-N, N, N-トリメチルピリジン-2-アミニウム・トリフルオロメタンスルホネート(3、図5に示す構造)の放射性フッ素化によって合成することがあり、その後、 t-ブチル基の脱保護及び続く精製を行う。

放射性フッ素化モジュールとは、化合物を放射性フッ素化するためのシステムである。放射線曝露を最小限に抑えるために、そのモジュールは自動化及び遠隔制御して放射性フッ素化を行うことがある。例えば、放射性フッ素化モジュールは、複数の異なるサブ‐モジュールを含むことがあり、各サブ‐モジュールが本明細書に記載した [18F]DCFPyL の合成方法の工程を実行するように構成する。ある実施形態では、放射性フッ素化モジュールは、放射性フッ素化のための乾燥 [18F]フッ化物イオンを調製するための第 1 サブ‐モジュール、DCFPyL 前駆体から [18F]DCFPyL の放射性合成を行う第 2 サブ‐モジュール、及び前記放射性合成反応混合物から [18F]DCFPyL を精製する第 3 サブ‐モジュールを含むことがある。各サブ‐モジュールを、例えばチューブ及び/又はポンプを接続することによって、動作可能に接続することがある。ある実施形態では、放射性フッ素化モジュールは、[18F]フッ化物イオンを生成するためのサブ‐モジュールを更に含むことがあり、その [18F]フッ化物イオンを、放射性フッ素化のための乾燥 [18F]フッ化物イオンを調製するための第 1 サブ‐モジュールが受け取ることがある。
ある実施形態では、DCFPyL 前駆体を、構造(II)を有する化合物 A と構造(III)を有する化合物 B 又はその塩とのアシル化反応によって合成することがある:

本明細書で提供する前駆体を放射性フッ素化するための方法が提供される。ある実施形態では、前記方法は:
(i)カートリッジ中に [18F]フッ化物イオンを捕捉すること;
(ii)カートリッジ中に捕捉させた [18F]フッ化物イオンを放出するために、テトラブチルアンモニウム塩基塩(例えば、テトラブチルアンモニウム炭酸水素塩(TBABC))の溶液でカートリッジから溶出すること;
(iii)前記 [18F]フッ化物イオンを含む溶出液を乾燥させること;
(iv)前記乾燥させた [18F]フッ化物イオンに、有機溶媒又は非プロトン性溶媒(例えば、アセトニトリル)中の DCFPyL 前駆体の溶液を加えること;及び
(v)工程(iv)の混合溶液を加熱すること;
を含む。
品質管理 HPLC 分析のために、当該分野で公知の様々な既知の分析クロマトグラフィー・システムを使用することがある。ある実施形態では、[18F]DCFPyL の化学的及び放射化学的特性を、Agilent 1260 Infinity System を使用して測定することがある。クォータナリ・ポンプ(quaternary pump)、HiP ALS オートサンプラー、及び NaI 放射能検出器を備えた Bioscan Flow-Count インターフェースを追加して、264 nmに設定された Max-Light フロー・セルが付いた DAD UV 検出器、を備えた例示的な Agilent 1260 Infinity System の構成が、Ravert ら(J. Label Compt. Radiopharm 2014, 57:695, J. Label Compt. Radiopharm 2015, 58:180)に記載されており、その全体が参照により本明細書に組み込まれる。ある実施形態では、Agilent OpenLAB クロマトグラフィー・データ・システムを、クロマトグラフィー・データを収集し、分析するために使用する。クロマトグラフィー条件のある例示的な組合せは以下の通りである:Atlantis T3 C18 5 μm 4.6×150 mm(ウォーターズ社、ミルフォード、マサチューセッツ州(Waters Corp., Milford, MA))をアセトニトリル(MeCN):トリエチルアミン(TEA)/リン酸塩緩衝液(pH 3.2)の 10:90 の混合液により流速 2 mL/分で溶出し、UV を 264 nm に設定した。ある実施形態では、以下の化合物を HPLC 分析のための標準として使用することがある:((2S)-2-[[(1S)-1-カルボキシ-5-[(6-フルオロアニルピリジン-3-カルボニル)-アミノ]ペンチル]-カルバモイルアミノ]ペンタン二酸、DCFPyL 標準、((2S)-2-[[(1S)-1-t-ブチルカルボキシレート-5-[(6-フルオロアニルピリジン-3-カルボニル)-アミノ]-ペンチル]-カルバモイル-アミノ]-ジ-t-ブチル・ペンタンジオアート、フッ素化された保護された中間体標準。
本明細書で提供される何れか 1 つの方法から得られる生成物の組成物が本明細書で提供される。そのような組成物を、それらの意図された用途の必要に応じて、単独で、又は他の成分又は化合物と組み合わせて、使用することがある。
また、本明細書の中で提供される何れか 1 つの方法を実施するために使用され得る合成モジュールが本明細書の中で提供される。例示的なモジュールには、実施例に記載されるような RFM、及び本明細書で提供される ELIXYS 修正システムが含まれる。好ましい実施形態において、前記モジュールには、本明細書で提供される何れか 1 つの方法の構成要素が含まれる。他の好ましい実施形態では、前記モジュールは、上記及び実施例に記載のものである。
細胞、器官若しくは組織を接触させること、又は被験体に有効量の本明細書で提供される化合物を投与することを含む、1 つ以上の細胞、器官若しくは組織をイメージングする方法もまた本明細書の中で提供される。ある実施形態では、1 つ以上の器官又は組織には、前立腺組織、腎臓組織、脳組織、血管組織又は腫瘍組織が含まれる。
本明細書で提供される何れか 1 つの組成物を含むキットもまた提供される。ある実施形態では、前記キットは、薬学的に許容される担体及び本発明の組成物を含む、パッケージにした医薬組成物を提供する。他のある実施形態では、前記キットは、本明細書で提供される何れか 1 つの方法を実施するために必要な化合物及び試薬を提供する。提供される何れか 1 つのキットの実施形態において、前記キットは、以下:本明細書で提供される何れか 1 つの方法を実施するための指示、本明細書で提供される何れか 1 つの組成物を調製するための指示、又は本明細書で提供される何れか 1 つの使用の方法のような、使用の方法において本明細書で提供されるような最終的な放射性標識化合物のための指示、の少なくとも 1 つを含むインディシア(indicia)を更に含む。
[ 18 F]DCFPyL の放射性医薬品製造レベルの合成
完全に規制に準拠した品質管理の仕様を備えた 2 つの異なる自動化されたプラットフォーム上での [18F]DCFPyL の放射性合成を以下で説明する。放射性トレーサー合成は、カスタム‐メイドの放射性フッ素化モジュール(RFM)及びソフィー・バイオサイエンス社の ELIXYS 自動放射化学合成装置で行った。前記 RFM 合成は、照射終了時間(end-of-bombardment (EOB))から、平均 66 分以内に完了し、合成終了時(end-of-synthesis (EOS))の平均比活性が 4.4 TBq/μmole(120 Ci/μmole)及び EOS での 30.9 %の平均放射化学収率(average radiochemical yield)であった。前記 ELIXYS 合成は、平均 87 分以内に完了し、EOS において平均比活性 2.2 TBq/μmole(59.3 Ci/μmole)及び 19 %の平均放射化学収率であった。両方の合成モジュールは、全ての標準 USP Chapter <823> 受け入れ試験基準(standard USP Chapter <823> acceptance testing criteria)に適合しながら、大きな mCi 量 の [18F]DCFPyL を生成した。
すべての化学物質と溶媒は ACS 又は HPLC 純度であり、記載されている場合を除き、Sigma-Aldrich Chemical Company(セントルイス、ミズーリ州(St. Louis, MO))又は Fisher Scientific(ウォルトハム、マサチューセッツ州(Waltham, MA))を通じて購入した。((2S)-2-[[(1S)-1-カルボキシ-5-[(6-フルオロアニルピリジン-3-カルボニル)‐アミノ]ペンチル]-カルバモイルアミノ]ペンタン二酸、DCFPyL標準、((2S)-2-[[(1S)-1-t-ブチルカルボキシレート-5-[(6-フルオロアニルピリジン-3-カルボニル)-アミノ]-ペンチル]-カルバモイル-アミノ]-ジ-t-ブチル・ペンタンジオアート、フッ素化された保護された中間体標準、2-[3-[1-t-ブチルカルボキシレート-(5-アミノペンチル)]-ウリエド(uriedo) ]-ジ-t-ブチル・ペンタンジオアート (1) のギ酸塩及び N, N, N-トリメチル-5-((2, 3, 5, 6-テトラフルオロフェノキシ)カルボニル)-ピリジン-2-アミニウム・トリフルオロメタンスルホネート (2) を、記載のように合成した(Chen ら、2011;Banerjee ら、2008;Olberg ら、2010)。このような合成方法の記載は、他に記載がない限り、その全体が参照により本明細書に組み込まれる。
1(0.303 g、0.57 mmole)を 6 mL のジクロロメタンに溶解した。これに0.158 mL の TEA 及び 2(0.272 g、0.57 mmole)を添加した。前記反応混合物を室温で 1 時間撹拌した。次に N2 気流下で溶媒を除去した後、真空乾燥した。前記混合物をアセトニトリルに溶解し、撹拌しながらジエチルエーテルを加えた。混合物を室温で 30 分間保持し、半固体を形成させた。エーテル層を除去し、半固体をアセトニトリルに再溶解した。ジエチルエーテルを撹拌しながら添加して半固体を生成させた。前記混合物を室温で30分間保持し、エーテル層を除去した。前記半固体を真空下で乾燥させ、アセトニトリル/水(1:9 から 5:5(v:v))を用いて C-18 Sep-Pak Vac 35 cc(ウォータズ社)で精製した。画分を集め、凍結乾燥して白色固体(0.322 g、71 %)を得た。1H-NMR (500 MHz, MeOD) δ 9.04 (d, J = 2.3 Hz, 1H), 8.54 (dd, J1= 2.3 Hz, J2= 8.7 Hz, 1H), 8.10 (d, J = 8.7 Hz, 1H), 4.17 (m, 2H), 3.68 (s, 9H), 3.43 (m, 2H), 2.32 (m, 2H), 2.03 (m, 1H), 1.80 (m, 2H), 1.67 (m, 3H), 1.45-1.50 (m, 29H). 13C-NMR (500 MHz, MeOD) δ 174.0, 173.9, 173.6, 166.0, 160.1, 159.7, 149.5, 141.4, 134.3, 121.9 (q, J = 317.9 Hz), 115.8, 83.0, 82.8, 81.9, 56.0, 54.9, 54.3, 41.1, 33.5, 32.6, 29.9, 29.1, 28.5, 28.4, 24.2。元素分析:C34H56F3N5O11S の計算値;C、51.05;H、7.06;N、8.76;実測値:C、50.58;H、6.95;N、8.49。HR-MS C33H56N5O8+ の計算値:650.4123、実測値、650.4138 [M] +。
酸素-18 富化水(98%、Huayi アイソトープ、江蘇省、中国、約 2 mL)をニオブ体、ジェネラル・エレクトリック・メディカル・システム(GEMS、ウプサラ、スウェーデン)(General Electric Medical Systems (GEMS, Uppsala, Sweden) )の PETtrace サイクロトロンの高収率な [18F]フッ化物ターゲット、に充填した。前記ターゲットに 55 μA の陽子ビームを 30 分間照射して、 18O(p,n)18F 核反応により約 61 GBq(1.65 Ci)の水性 [18F]フッ化物イオンを生成した。
すべての化学物質と成分を RFM にロードした後、[18F]フッ化物イオンを、1 mL の高純度水(Fluka)で洗浄することにより事前調整した Chromafix 30-PS-HCO3 SPE カートリッジ(ABX 社、ラーデベルグ、ドイツ)(Chromafix 30-PS-HCO3 SPE cartridge (ABX GmbH, Radeberg, Germany))に送った。[18O] は回収して再利用した。RFM コンピューター・コントロール(National Instruments LabVIEW、オースチン、テキサス州(Austin, TX))の制御下で、前記樹脂カートリッジをテトラブチルアンモニウム炭酸水素塩(TBABC)(600 μL、0.075 M、ABX 社、ドイツ(ABX GmbH, Germany))の溶液で溶出し、マルチポート・キャップで封印した 5 mL の反応バイアルに集めた;前記バイアルは、合成の前に希硝酸で汚れを落とし、HPLC水で洗浄し、80 ℃で一晩乾燥させた。前記カートリッジを MeCN(250 μL)ですすいだ後、標準的な温度加熱ブロックの中で、制御した窒素流(325 mL/分)で 150 秒間、前記溶液を 110 ℃で乾燥させた。前記 [18F]フッ化物イオンを更に乾燥させるために、MeCN(それぞれ 250 μL)を 2 回別々に添加し、それぞれ 150 及び 180 秒間加熱した。
すべての化学物質及び成分を ELIXYS 合成カセットに充填した後、前記 [18F]フッ化物イオンを用量キャリブレーター中の 5 mL の V‐バイアルに送った。自動 ELIXYS 合成シーケンスを開始した。1 mL の高純度水(Fluka)で洗浄することにより事前調整した Chromafix 30-PS-HCO3 SPE カートリッジ(ABX 社、ドイツ)に前記 [18F]フッ化物イオンを窒素で押し込んだ。[18O]水は回収して再利用した。ELIXYS 反応器内で、前記樹脂カートリッジから TBABC(300 μL、0.075 M、ABX 社、ドイツ)の溶液で、ガラス撹拌棒を使って 5 mL 反応器 V-バイアルに溶出した。前記カートリッジを MeCN(600 μL)ですすいだ後、前記溶液を攪拌しながら真空下と 270 秒間の窒素流下、110 ℃で乾燥させた。MeCN(600 μL)を 2 回の別々に添加し、真空下及び窒素流下でそれぞれ 90 秒及び 105 秒間加熱した。
遠隔操作装置及び適切な放射線遮蔽(鉛ガラス)を用いて、[18F]DCFPyL 生成物が入っているバイアルを、明るい光の下で目視検査をした。異物がないことが明らかで無色透明であれば、この製品は、この受け入れ仕様を満たすものとした。
対照標準溶液を分析 HPLC に注入して、システム条件が適していることを確認した(リテンション・タイム(retention time)及び質量を調べる標準曲線との一致を確認する)。放射化学的な同一性を測定するために、[18F]DCFPyL の最終注入マトリックスの一定分量(50 μL)を対照標準溶液の一定分量と混合した。UV検出器で測定した参照物質のリテンション・タイムが、放射線検出器によって測定した [18F]DCFPyL のリテンション・タイムと一致していれば(2 つの検出器システム間のオフセットが適正に較正されていて)、この製品は、この受け入れ仕様を満たすものとした。
放射化学的な同一性試験について記載したのと同じ HPLC システムを用いて、[18F]DCFPyL の適切な容量(50 μL)を、放射能検出システムにおける補正されていないデッド‐タイムの損失(メインピークのため)を避ける注入量で注入した。[18F]DCFPyL の放射化学的純度のパーセントは、[18F]DCFPyL ピークに関連する放射能を、クロマトグラムでアッセイした全活性で割り、100 を掛けることによって決定した。放射化学的純度が 95 % 以上であれば、この製品は、この受け入れ仕様を満たすものとした。QC クロマトグラムのサンプルを図7に示す(DCFPyL-RT = 7.6分、k'= 8.7)。
[18F]DCFPyL の比活性は、較正した一定分量の [18F]DCFPyL のアッセイした放射能(合成終了時の mCi/mL)を、標準質量較正曲線から解釈されるような HPLC-UV で測定した担体 DCFPyL の質量濃度(1 mL 当たりの DCFPyL の μmole)で割ることにより計算した。前記比活性が 1000 mCi/μmole 以上であれば、この製品は、この受け入れ仕様を満たすものとした。
高い比活性では、化学的純度の指標として単純な UV ピーク比を使用することは、質量が典型的には小さくなるにつれて不適切となる。[18F]DCFPyL をうまく合成するために、出発前駆体の 99.5 %以上を合成中に除去しなければならない;即ち、最終生成物マトリックス中に残存する前駆体又はその副生成物は 0.5 %以下であり得る。マトリックスに起因しない他のすべての UV 吸収 HPLC 成分も、同じ許容される残存前駆体濃度未満でなければならない。放射化学的同一性試験について記載したのと同じ HPLC システムを用いて、[18F]DCFPyL の担体質量を測定した。最初の HPLC カラム空隙容量(ボイド・ボリューム(void volume))の後、他のすべての UV ピークを合計し、副生成物とした。これらの副産物の質量濃度が 1.5 μg/mL 以下であれば、この製品は、この受け入れ仕様を満たすものとした。
無水エタノール(6.7 %)1675 μL、アセトニトリル(400 ppm)12.7 μL 及びメタノール(3000 ppm)94.7 μL を加えた 25 mL の HPLC 水の標準溶液の一定分量を分析して、システム適合性を決定した。最終の [18F]DCFPyL 生成物マトリックスの一定分量を注入し、残存溶媒のレベルを、単一点曲線(single-point curve)からの場合と同様に標準と比較して計算した。アセトニトリルのレベルが 400 ppm 以下であり、メタノールのレベルが3000 ppm 以下であり、エタノールのレベルが 10 %以下である場合には、この製品は、この受け入れ仕様を満たすものとした(Intl. Conf. on Harmonisation of Tech. Requirements for Registration of Pharm. for Human Use, 1997)。
一滴の [18F]DCFPyL 最終生成物マトリックスを pH 指示紙(ColorPhast-Indicatorストリップ-pH 2-9;感度 0.3 から 0.5 単位、EMD Chemicals 社、ギブスタウン、ニュージャージ州)に滴下した。ストリップの色を、指標チャートと照合した。pH が 4.5 から 8.5 であれば、この製品は、この受け入れ仕様を満たすものとした。
[18F]DCFPyL 最終ろ過工程(terminal filtration step)に由来する滅菌マイクロフィルターを 5 mL の無水エタノールで洗浄し、濡れたままにし、較正した圧力計(Millipore 社)及び空気圧源に取り付けた。フィルターの遠位端を液体貯蔵器に入れ、ガスの圧力をゆっくりと増加させた。気泡の流れが見られることなく 13 psi 以上の圧力に達した場合に、Millipore Millex FG フィルターの完全性について、この製品は、この受け入れ仕様を満たすものとした。
[18F]DCFPyL 最終生成物マトリックスの一定分量中の放射能含有量(mCi)を、時間(0 分;A)及び再度 15 分後に(B)に、Capintec CRC-15R Radioisotope Dose Calibrator (放射性同位元素用量較正器)(ラムゼイ、ニュージャージ州(Ramsey, NJ))で測定した。半減期を以下の式を用いて算出した。計算された半減期が 105 から 115 分である場合、この製品は、この受け入れ仕様を満たすものとした。
T1/2 =(4.495)/(log A - log B)
適切なガンマ線分光計を用いて、ガンマ・スペクトルを得るのに十分な時間、適切な一定分量を注入してアッセイをした。得られたガンマ・スペクトルについて、18F の発光の特徴ではない識別可能な光電ピークが存在するかを分析した。放射性核種の純度について、観察されたガンマ線の総放出量の99.5%以上が 0.511 及び 1.022 MeV に相当していれば、この製品は、この受け入れ仕様を満たすものとした。
Charles River Laboratories EndoSafe Portable Testing System(Endosafe PTS Reader, Integrated Software Version 7.10, Service Pack 2.0, 及びプリンター;ウィルミントン、マサチューセッツ州(Wilmington, MA))を用いて、[18F]DCFPyL 最終生成物マトリックスのバッチ内のエンドトキシン・レベルを分析した。エンドトキシン・レベルが 11 エンドトキシン単位/mL 以下であれば、この製品は、この受け入れ仕様を満たすものとした。
ラミナ・フロー・フード(laminar flow hood)内で、[18F]DCFPyL 最終生成物マトリックス(各約 100 μL)のサンプルを液体チオグリコール酸培地及び大豆カゼイン消化培地(ベクトン・ディッキンソン社(Becton, Dickinson and Company))に添加した。その培地をそれぞれ 32.5±2.5 ℃及び 22.5 ± 2.5 ℃でインキュベートし、増殖陽性を示す濁度について毎日観察した。14 日間のインキュベーション期間中に増殖が観察されなかった場合、この製品は、この受け入れ仕様を満たすものとした。
周囲環境の条件下で保存したときの放射性トレーサー製品の安定性を実証するために、放射性トレーサー合成の終了後 360 分に、上記の受け入れ試験のサブセットを実施した。
[18F]DCFPyL のマニュアル放射性合成(Chen ら、2011)を、自動化された二重反応器合成プラットフォーム(古い核インターフェース「ダブル FDG 合成モジュール」(an old Nuclear Interface “Double FDG Synthesis Module”))でのルーチンな使用に適合させた。ヒトに使用するための放射性トレーサー合成の最初の評価の一部として、合成終了時での比活性が 192 GBq/μmole(5.2 Ci/μmole)である最終放射性トレーサー製品(2.3 GBq;62 mCi 平均)が適度な量で生産された。表1は、最初の受け入れ仕様と製品を評価した結果を示す。時間が経つ中、この手順は、2 年の期間にわたって実施した 110 件を超える放射化学合成について、合成終了時の平均比活性で、およそ 3 %のやや変動しうる放射化学収率(減衰に対して較正しておらず、推定平均 [18F]フッ化物標的の収量に基づく)、しばしばより低い、であった。
QC 受け入れ仕様:[18F]DCFPyL の元の合成と RFM 及び ELIXYS モジュールでの新しい合成との比較。(収量、比活性及びフィルターの完全性を除くすべての試験を 360 分の有効期限時に繰り返して行い、EOS で実施した試験結果と合致した。)
カスタム化した、高収率、超高比活性の、使用された放射性フッ素化合成モジュールを、PSMA 阻害剤 [18F]DCFPYL 等の [18F]放射性トレーサーを製造するために設計し、構築した。PET 放射性トレーサーの比活性は、質量依存性の受容体局在化又は薬理学的な毒性懸念に対する重要な品質管理放出基準(a critical quality control release criteria)である。超高比活性では、放射性トレーサーの有効期限を延長することができ、放射性トレーサーの単一の製造バッチで、より多くの PET 研究が可能になる。前記放射性フッ素化モジュールによって、PET イメージングに使用される放射性トレーサーの自動及び半自動の放射化学合成が可能になる。現在、10 を超える商業的な放射化学合成モジュールが市販されている。この化学モジュールをユニークにするのは、モジュールを使用して製造される放射性トレーサーの品質である。本発明のカスタム放射性フッ素化モジュールは、今日市販されている商業的な合成モジュールの何れよりも 10 から200 倍優れた比活性を有する。

Claims (86)
- 2-(3{1-カルボキシ-5-[(6-[18F]フルオロ-ピリジン-3-カルボニル)-アミノ]-ペンチル}-ウレイド)-ペンタン二酸([18F]DCFPyL)の合成方法であって、前記方法が以下:
(i)放射性フッ素化 DCPFPyL 前駆体を形成するために、エステル部分保護基を含む DCFPyL 前駆体を放射性フッ素化すること;
(ii)反応混合物中に [18F]DCFPyL を形成するために、工程(i)の放射性フッ素化 DCPFPyL 前駆体のエステル部分保護基をリン酸で脱保護すること;及び、
(iii)[18F]DCFPyL を提供するために、工程(ii)の反応混合物から [18F]DCFPyL を精製すること;
を含む方法。 - 前記保護基が、ベンジル、p-メトキシベンジル、第三ブチル、メトキシメチル、メトキシエトキシメチル、メチルチオメチル、テトラヒドロピラニル、テトラヒドロフラニル、ベンジルオキシメチル、トリメチルシリル、トリエチルシリル、t-ブチルジメチルシリル(TBDMS)及びトリフェニルメチルからなる群から選択される、請求項1に記載の方法。
- 工程(i)及び工程(ii)が 1 つの反応器中で行われる、請求項1又は2に記載の方法。
- 前記合成が、加熱ブロック、2 つのシリンジ・ポンプ、マルチ‐ポート・キャップ、及びバルブ付き試薬添加バイアルを含む放射性フッ素化モジュール(RFM)を使用することによって自動化される、請求項1から3の何れか一項に記載の方法。
- 前記 RFM が温度加熱キャビティを更に含む、請求項4に記載の方法。
- 前記合成が自動放射化学合成装置を使用することにより自動化される、請求項1から3の何れか一項に記載の方法。
- 前記 RFM 又は前記自動放射化学合成装置の構成要素がフッ素を含まない、請求項4から6の何れか一項に記載の方法。
- 前記 DCFPyL 前駆体が 5-(((S)-6-(tert-ブトキシ)-5-(3-((S)-1, 5-ジ-tert-ブトキシ-1, 5-ジオキソペンタン-2-イル)ウレイド)-6-オキソヘキシル)カルバモイル)-N, N, N-トリメチルピリジン-2-アミニウム化合物である、請求項1から7の何れか一項に記載の方法。
- 前記 DCFPyL 前駆体が 5- (((S)-6-(tert-ブトキシ)-5-(3-((S)-1, 5-ジ-tert-ブトキシ-1, 5-ジオキソペンタン-2-イル)ウレイド)-6-オキソヘキシル)カルバモイル)-N, N, N-トリメチルピリジン-2-アミニウム・トリフルオロメタンスルホネートである、請求項8に記載の方法。
- 前記 DCFPyL 前駆体が 5-(((S)-6-(tert-ブトキシ)-5-(3-((S)-1, 5-ジ-tert-ブトキシ-1, 5-ジオキソペンタン-2-イル)ウレイド)-6-オキソヘキシル)カルバモイル)-N, N, N-トリメチルピリジン-2-アミニウム・トリフルオロアセテートである、請求項8に記載の方法。
- 前記 DCFPyL 前駆体が、
に従って合成される、請求項9又は10に記載の方法。 - 前記 DCFPyL 前駆体が、以下:
N, N, N-トリメチル-5-((2, 3, 5, 6-テトラフルオロフェノキシ)カルボニル)-ピリジン-2-アミニウム・トリフルオロメタンスルホネートと2-[3-[1-t-ブチルカルボキシレート-(5-アミノペンチル)]-ウリエド(uriedo)]-ジ-t-ブチル・ペンタンジオアートとのカップリング、
を含む方法によって合成される、請求項9又は10に記載の方法。 - 前記方法が更に、以下:
に従う、又は、N, N, N-トリメチル-5-((2, 3, 5, 6-テトラフルオロフェノキシ)カルボニル)-ピリジン-2-アミニウム・トリフルオロメタンスルホネート2-[3-[1-t-ブチルカルボキシレート-(5-アミノペンチル)]-ウリエド(uriedo)]-ジ-t-ブチル・ペンタンジオアートのカップリングによる、前記 DCFPyL 前駆体を合成することを更に含む、請求項8から12の何れか一項に記載の方法。 - 前記 DCFPyL 前駆体を放射性フッ素化することが、以下
に従って行われる、請求項1から13の何れか一項に記載の方法。 - 前記 DCFPyL 前駆体を放射性フッ素化することが:
(a)カートリッジ中に [18F]フッ化物イオンを捕捉すること;
(b)カートリッジ中に捕捉させた [18F]フッ化物イオンを放出するために、テトラブチルアンモニウム塩基塩の溶液で前記カートリッジから溶出すること;
(c)乾燥させた [18F]フッ化物を形成するために、前記 [18F]フッ化物イオンを含む溶出液を乾燥させること;及び、
(d)前記乾燥させた [18F]フッ化物イオンに、(化合物(3))
を含む、請求項1から13の何れか一項に記載の方法。 - 前記カートリッジが陰イオン交換クロマトグラフィー・カートリッジである、請求項15に記載の方法。
- 前記カートリッジを、カートリッジ中に [18F]フッ化物イオンを捕捉する前に、高純度水で洗浄することによって、事前調整する、請求項15又は16に記載の方法。
- 前記 DCFPyL 前駆体を放射性フッ素化することが、(化合物(3))
- 前記加熱が、約 2 分から約 10 分間である、請求項18に記載の方法。
- 前記加熱が、約 50 ℃で約 6 分間である、請求項18又は19に記載の方法。
- 前記加熱が、DCFPyL 前駆体と前記乾燥させた [18F]フッ化物イオンとの混合溶液に、約 40 W から約 60 W のマイクロ波照射を約 20 秒から約 200 秒間、照射することによるものである、請求項18から20の何れか一項に記載の方法。
- 前記加熱が、DCFPyL 前駆体と前記乾燥させた [18F]フッ化物イオンとの混合溶液に、約 50 W のマイクロ波照射を約 30 秒から約 150 秒間、照射することによるものである、請求項21に記載の方法。
- 工程(c)からの [18F]フッ化物イオンを乾燥させる、請求項15から22の何れか一項に記載の方法。
- 前記 [18F]フッ化物イオンを含む工程(c)の溶出液を約 80 ℃から約 150 ℃の間の温度で乾燥させる、請求項15から23の何れか一項に記載の方法。
- 前記温度が約 110 ℃である、請求項24に記載の方法。
- 前記 [18F]フッ化物イオンを含む工程(c)の溶出液を窒素流下で乾燥させる、請求項15から25の何れか一項に記載の方法。
- 前記乾燥を約 50 秒から約 300 秒間行う、請求項15から26の何れか一項に記載の方法。
- 前記乾燥を約 150 秒間行う、請求項27に記載の方法。
- 更に乾燥させるために、CH3CN を前記乾燥させた[18F]フッ化物イオンに添加する、請求項15から28の何れか一項に記載の方法。
- 前記 RFM 若しくは自動放射化学合成装置、又はその反応部分を、合成前に、希硝酸で汚れを落とし、水で洗浄し、一晩約 80 ℃で乾燥させる、請求項4から29の何れか一項に記載の方法。
- 前記リン酸による脱保護を約 30 ℃から約 55 ℃の間の温度で行う、請求項1から30の何れか一項に記載の方法。
- 前記温度が約 45 ℃である、請求項31に記載の方法。
- 前記温度を約 2 分から約 10 分間維持する、請求項31又は32に記載の方法。
- リン酸による脱保護の後に工程(ii)の反応混合物の pH を、約 2 から約 2.5 の間の pH に調整することを更に含む、請求項1から33の何れか一項に記載の方法。
- 前記反応混合物の pH を、水酸化ナトリウムとリン酸二水素ナトリウム緩衝液を添加することによって調整する、請求項34に記載の方法。
- 前記精製を液体クロマトグラフィーによって行う、請求項1から35の何れか一項記載の方法。
- 前記液体クロマトグラフィーが少なくとも 1 つの C18 カラムを含む、請求項36に記載の方法。
- [18F]DCFPyL を含む溶液を、メタノールとリン酸二水素ナトリウムを含む第 1 の溶出溶液を用いて第 1 の C18 カラムから溶出する、請求項36から37に記載の方法。
- 前記第 1 溶出溶液中のメタノールとリン酸二水素ナトリウムが、pH 2.1 で、15:85 のメタノール:0.01 M リン酸二水素ナトリウムである、請求項38に記載の方法。
- 前記 [18F]DCFPyL を含む溶液を更に第 2 の C18 カラムに供し、エタノールを含む第 2 の溶出溶液で溶出する、請求項38又は39に記載の方法。
- 前記精製する工程が、第 1 又は第2 の C18 カラムから溶出した [18F]DCFPyL をろ過する工程を更に含む、請求項36から40の何れか一項記載の方法。
- 前記ろ過が 0.2 μmの滅菌フィルタを通すことである、請求項41に記載の方法。
- 前記ろ過が無菌バイアル中へのものである、請求項41又は42に記載の方法。
- 前記無菌バイアルを無菌生理食塩水で予め充填している、請求項43に記載の方法。
- 前記精製をアスコルビン酸ナトリウムの存在下で行う、請求項1から44の何れか一項に記載の方法。
- 前記アスコルビン酸ナトリウムを収集リザーバに添加し、及び/又は [18F]DCFPyL 生成物を溶出するために使用する溶液中に添加する、請求項45に記載の方法。
- 前記精製後の [18F]DCFPyL の収量が少なくとも約 20 mCi である、請求項1から46の何れか一項に記載の方法。
- 前記精製後の [18F]DCFPyL の収量が少なくとも約 100 mCi である、請求項47に記載の方法。
- 前記精製後の [18F]DCFPyL の収量が少なくとも 400 mCi である、請求項47に記載の方法。
- 以下:
(i)カートリッジ中に [18F]フッ化物イオンを捕捉すること;
(ii)前記カートリッジ中に捕捉させた [18F]フッ化物イオンを放出するために、テトラブチルアンモニウム炭酸水素塩(TBABC)の溶液で前記カートリッジから溶出すること;
(iii)工程(ii)からの [18F]フッ化物イオンを含む溶出液を乾燥させること;
(iv)工程(iii)からの [18F]フッ化物イオンに、(化合物(3))
(v)工程(iv)の混合溶液を加熱すること;
を含む、DCFPyL 前駆体を放射性フッ素化する方法。 - 前記カートリッジが陰イオン交換クロマトグラフィー・カートリッジである、請求項50に記載の方法。
- 前記カートリッジを、カートリッジ中に [18F]フッ化物イオンを捕捉する前に、高純度水で洗浄することによって、事前調整する、請求項50又は51に記載の方法。
- 前記加熱が、約 30 ℃から約 70 ℃の間である、請求項50から52の何れか一項に記載の方法。
- 前記加熱が、約 2 分から約 10 分間である、請求項53記載の方法。
- 前記加熱が、約 50 ℃で約 6 分間である、請求項54記載の方法。
- 前記加熱が、工程(iv)の混合溶液に、約 40 W から約 60 W のマイクロ波照射を約 20 秒から約 200 秒間、照射することによるものである、請求項50から52の何れか一項に記載の方法。
- 前記加熱が、工程(iv)の混合溶液に、約 50 W のマイクロ波照射を約 30 秒から約 150 秒間、照射することによるものである、請求項56に記載の方法。
- 工程(iii)からの [18F]フッ化物イオンを乾燥させる、請求項50から57の何れか一項に記載の方法。
- 前記 [18F]フッ化物イオンを含む溶出液を約 80 ℃から約 150 ℃の間の温度で乾燥させる、請求項50から58の何れか一項に記載の方法。
- 前記温度が約 110 ℃である、請求項59に記載の方法。
- 工程(ii)からの [18F]フッ化物イオンを含む溶出液を窒素流下で乾燥させる、請求項50から60の何れか一項に記載の方法。
- 前記乾燥を約 50 秒から約 300 秒間行う、請求項50から61の何れか一項に記載の方法。
- 前記乾燥を約 150 秒間行う、請求項62に記載の方法。
- 更に乾燥させるために、CH3CN を前記乾燥させた [18F]フッ化物イオンに添加する、請求項58から63の何れか一項に記載の方法。
- 請求項1から49の何れか一項に記載の方法によって製造された [18F]DCFPyL を含む組成物。
- 前記 [18F]DCFPyL が少なくとも約 50 Ci/μmole、又は少なくとも約 100 Ci/μmole の平均比活性を有する、請求項65に記載の組成物。
- 前記組成物が約 400 ppm 以下の濃度でアセトニトリルを含む、請求項65又は66に記載の組成物。
- 前記組成物が約3000 ppm 以下の濃度でメタノールを含む、請求項65から67の何れか一項に記載の組成物。
- 前記組成物が約 50 ppm 以下の濃度でメタノールを含む、請求項68に記載の組成物。
- 前記組成物が 1 つ以上のクリプタンドを含まない、請求項65から69の何れか一項に記載の組成物。
- 前記クリプタンドがクリプトフィックス(Kryptofix)(登録商標)である、請求項70に記載の組成物。
- 前記組成物がトリエチルアミンを含まない、請求項65から71の何れか一項に記載の組成物。
- 前記組成物が t-ブタノールを含まない、請求項65から72の何れか一項に記載の組成物。
- [18F]DCFPyL の放射化学的純度が約 95 %から約 100 %の範囲である、請求項65から73の何れか一項に記載の組成物。
- 請求項65から74の何れか一項に記載の組成物を含むキット。
- DCFPyL 前駆体及びリン酸を含むキット。
- DCFPyL 前駆体及びテトラブチルアンモニウム炭酸水素塩を含むキット。
- 1 つ以上のクリプタンドを含まない、請求項75から77の何れか一項に記載のキット。
