Abstract
Description
本願は、2016年3月1日に出願された米国仮出願第62/301,806号、2016年6月8日に出願された第62/347,376号、および2017年1月13日に出願された第62/446,026号の優先権の利益を主張し、その内容はその全体が参照によりここに組み込まれる。

図面の簡潔な説明

ErA遺伝子クローニングおよび変異誘発。最初の21アミノ酸残基のシグナルペプチドを欠いているErA(UniProtエントリP06608)のアミノ酸配列に対応するコドン最適化合成遺伝子を、以前に記載されたようにGenscriptにより合成した(Schalk, et al. (2014) J. Biol. 高L−アスパラギナーゼおよび低L−グルタミナーゼ活性を有するErA変異体の設計および特性評価。L−アスパラギナーゼがALLの処置に成功するには、酵素が、可能な限り高いL−アスパラギナーゼ活性(がん細胞を殺すため)であり、可能な限り低いL−グルタミナーゼ活性(有毒な副作用を最小化するため)であることが必要であると仮定された。2つのFDAに認可された細菌のL−アスパラギナーゼのうち、E.クリサンセミ(ErA)由来のものは、大腸菌(EcA)由来のものよりも高いL−アスパラギナーゼ活性を有する。表2を参照されたい。
変異誘発のための残基の選択。4つの残基Ala31、Glu63、Pro123、およびSer254を、L−グルタミナーゼ/L−アスパラギナーゼ比に影響を及ぼし得る候補部位として同定した。触媒作用におけるそれらの役割のために、L−アスパラギナーゼ/L−グルタミナーゼスーパーファミリー(図3のアスタリスクで示され、T12GGT15、H93GTDT97、S62、K168を含む)間のコンセンサスモチーフに存在する残基を、変異の検討から除外した。代わりに、活性部位を取り囲み、必ずしも基質と相互作用しない非保存残基を標的化した。多重配列アラインメント(図3)に基づき、産物AspおよびGluとの複合体中のErAの結晶構造の分析と組み合わせて、Ala31、Glu63、Pro123およびSer254を変異誘発のための4つの潜在的な候補部位として同定した。興味深いことに、これらの4つの残基のうち、ただ1つのGlu63のみが基質と直接相互作用する。
TRAIL(TNF関連アポトーシス誘導リガンド)は、アポトーシスによって細胞死を誘導するタンパク質である。したがって、TRAIL−アスパラギナーゼ融合タンパク質は、これら2つのタンパク質の活性を組み合わせるために作製した。この融合タンパク質を使用して、L−アスパラギナーゼ成分は細胞にアポトーシスを起こさせ、TRAIL成分は細胞死を誘導する。
L−アスパラギナーゼの有効性は、薬のin vivo半減期に関連する;半減期が長ければ長いほど、酵素は血液アスパラギンを加水分解するようにより長く作用する。したがって、L−アスパラギナーゼならびにここに開示される変異体および融合タンパク質のin vivo半減期を増加させるために、タグをL−アスパラギナーゼのN末端に融合させた。タグは、His6−酵母SUMOタグ、His6−ヒトSUMOタグおよびアルブミン結合ペプチドSA21タグを含んでいた。各タグは、L−アスパラギナーゼ酵素の循環時間を増加させた。さらに、タグの組み合わせを試験した。特に、SA21および酵母SUMOタグを組み合わせて、さらに長期にわたる半減期を有する変異体を得た。
PEGの機能は、L−アスパラギナーゼをタンパク質分解および宿主免疫システムから保護することである。L−アスパラギナーゼのマレイミドベースのPEG化を可能にするために、様々なアミノ酸残基をシステイン残基に変異させて、酵素活性にほとんどまたはまったく影響を及ぼさない好適な位置を同定した。注目すべきことに、ErA−WTおよびErA−TM2はシステイン残基を含有しない。したがって、このアミノ酸残基の導入は、折りたたみまたは活性を破壊しないように、酵素の適切な位置になければならなかった。したがって、ErA−TM2の構造を分析して、システインに変異し得る残基の領域を同定した。(1)表面に位置し、(2)オリゴマー化界面から適切な距離にあり(ErA−TM2は四量体であるため、界面付近の残基をPEG化することは酵素の活性に有害であり得る)、(3)外側を指している残基を含む、5つのかかる領域を同定した(表14)。
Claims (45)
- 配列番号1の31、63および254位の1以上でのアミノ酸置換を含む、エルウィニア・クリサンセミ(Erwinia chrysanthemi)のL−アスパラギナーゼ(ErA)変異体。
- 31位でのアミノ酸置換が、イソロイシン、バリン、ロイシン、またはトレオニンを含む、請求項1に記載のErA変異体。
- 63位でのアミノ酸置換が、グルタミン、アスパラギン、またはアスパラギン酸を含む、請求項1に記載のErA変異体。
- 254位でのアミノ酸置換が、アスパラギンまたはグルタミンを含む、請求項1に記載のErA変異体。
- 配列番号2、配列番号3、配列番号4、配列番号5、配列番号6または配列番号7に示されたアミノ酸配列を含む、請求項1に記載のErA変異体。
- 最小で野生型ErA酵素の75%のアスパラギナーゼ反応速度(kcat)を示す、請求項1に記載のErA変異体。
- L−アスパラギンについて250μM未満のKmを示す、請求項1に記載のErA変異体。
- 野生型ErA酵素の60%未満のグルタミナーゼ反応速度(kcat)を示す、請求項1に記載のErA変異体。
- L−グルタミンについて3mMより大きいKmを示す、請求項1に記載のErA変異体。
- ヒスチジンタグ、SUMOタグ、アルブミン結合ドメイン、またはそれらの組み合わせをさらに含む、請求項1に記載のErA変異体。
- PEG化されている、請求項1に記載のErA変異体。

- 請求項1に記載のErA変異体をコードする、核酸分子。
