Abstract
Description
本発明は、タンパク質及びポリペプチドの分野に関し、より具体的には、スパイダーシルクタンパク質(スピドロイン(spidroin))並びに他の非スピドロインタンパク質及びポリペプチドの発現及び産生に関する。本発明は、新規なタンパク質であって、所望のタンパク質及びポリペプチドの発現及び産生のための、それ自体で、及び新規な融合タンパク質における部分として、有用であるタンパク質、並びに、これら新規なタンパク質及び融合タンパク質をコードする核酸分子を提供する。本発明はまた、所望のタンパク質又はポリペプチドの発現及び産生方法を提供する。
DNAからのタンパク質及びポリペプチドの産生は、様々な宿主で達成することができるが、一般的な問題は、不溶性タンパク質/ポリペプチド凝集体の形成である。これは、機能性タンパク質/ポリペプチドの産生を深刻に妨げ、又は防ぐ場合もある。この問題は典型的には、低溶解性タンパク質及びポリペプチド、例えば膜関連タンパク質及びポリペプチドで深刻化する。
本発明の目的は、融合パートナー、すなわち、融合タンパク質の部分であって、前記融合タンパク質の別の部分である所望のタンパク質又はポリペプチドの溶解性を向上するために有用である部分を提供することである。
本発明は、タンパク質及びポリペプチドの産生及び発現に関する。この産生による目的に応じて、最終産物は異なり得る。例えば、溶液で又は他の生体分子に関連して、脂質膜に挿入された所望のタンパク質又はポリペプチドを得ることが望ましい場合がある。また、融合タンパク質の一部としての所望のタンパク質又はポリペプチドであって、精製及び検出に適したハンドル(handle)を提供し、及び/又は、所望の特性、例えば安定性及び溶解性を提供し得るタンパク質又はポリペプチドを得ることが非常に望ましい場合もあるということが認識されるものとする。可溶性融合タンパク質に機能的に統合された所望のタンパク質又はポリペプチドを維持することは、所望のタンパク質又はポリペプチドを特徴付け及び研究するのに有用である。
a)好適な宿主において、所望のタンパク質又はポリペプチドを含む本発明に係る融合タンパク質を発現し;並びに
b)融合タンパク質を含有する混合物を得て、そして
b1)融合タンパク質を単離し、ここで、本ステップは以下のステップを含み:
b1a)融合タンパク質の沈殿、好ましくは高塩濃度での塩析による沈殿;並びに
b1b)沈殿した融合タンパク質を水性溶媒中に懸濁すること、ここで、融合タンパク質は水性溶媒に可溶性である;

実施例1-NT及び電荷反転NT突然変異体の発現
NTwt(配列番号11をコードする配列番号12)及びNTD40K/K65D(配列番号2をコードする配列番号8)を有するコンストラクトを、pT7発現ベクターにクローニングし、コンピテントな大腸菌BL21(DE3)細胞に化学的に形質転換した。プラスミド含有細胞を、70 mg/Lカナマイシンを含む10 mLのルリア-ベルターニ(Luria-Bertani)(LB)培地に接種し、37℃及び180 rpmで一晩増殖させた。5 mLの一晩培養物をカナマイシンを含む500 mLのLB培地(1/100)に接種し、細胞をさらに30℃で〜1のOD600まで増殖させた。細胞を、0.5 mMの最終濃度までのイソプロピル β-D-1-チオガラクトピラノシド(IPTG)の添加によって誘導し、発現を20℃で一晩行った。翌日、細胞を遠心分離によって採取し、20 mMのTris-HCl、pH 8、30 mLまでに再懸濁し、−20℃で少なくとも24時間保存した。
NT二量体化プロセスは、単量体の初期会合に重要な役割を果たす残基D40とK65との間の分子間静電相互作用に大いに依存している。本研究では、本発明者らは、ドメインの正味の電荷を維持しながら、野生型NT(NTwt;配列番号11)と比較してこれら残基が入れ替わっている、二重突然変異体(NTD40K/K65D;配列番号2)を設計し、評価した。重要な生物物理学的特性を評価して、溶解性向上融合パートナーとしての突然変異体の適用性を決定した。
NTD40K/K65D(配列番号2)及びNTwt(配列番号11)それぞれについての蛍光発光スペクトルを、分光蛍光光度計(Tecan Safire 2)で、96個の平底ウェルを有するCostar(登録商標)黒色ポリスチレンアッセイプレートを用いて測定した。タンパク質を、0.4 pH単位のステップでpH 5.6〜8.0に調整した20 mM HEPES/20 mM MESで5 μMの濃度に希釈した。試料を280 nm(5 nmのバンド幅)で励起後に、発光スペクトルを1 nmステップで300〜400 nm(10 nmのバンド幅)の間、記録した。トリプトファン蛍光比を339 nm及び351 nmでの強度から計算し、pHの関数としてプロットした。NTwtについて得られたデータを、単量体-二量体平衡のシグモイド挙動による2状態結合モデルにフィットさせた。
NT変異体NTD40K/K65D(配列番号2)及びNTwt(配列番号11)を発現する細胞を、一晩培養し、さらに、15N標識塩化アンモニウム及び70 mg/Lのカナマイシンを含有する1/100〜500 mLの最小培地M9に接種した。細胞を一晩22℃で、1.4のOD600まで増殖させた。タンパク質を、前述のように発現及び精製した。
NTwt(配列番号11)及びNTD40K/K65D(配列番号2)タンパク質を、0.5 Mステップでの0〜7 Mの尿素で補充した20 mM HEPES/20 mM MESで5 μMに希釈した。尿素の各濃度でのタンパク質の安定性を、0.5単位ステップで5.0〜7.5の範囲の一定pH値でのTrp蛍光によりモニターした。それぞれの測定されたpHについて、蛍光比を尿素濃度に対してプロットし、遷移点を決定するために、2状態アンフォールディングモデルにフィットした。NTwt(点線)及びNTD40K/K65D(黒線)についてのデータを、図4におけるpHの関数として、天然状態と変性状態との間の遷移点として示している([den]50%)。
温度スキャンを行い、円二色性(CD)を用いて分析した。実験を、1 mmの経路長を有する300 μL キュベットを用いて410-モデルの円形CD分光計(Aviv biomedical Inc.、Lakewood、NJ、USA)で行った。全ての測定について、NTwt(配列番号11)及びNTD40K/K65D(配列番号2)タンパク質を、pH 5.5又はpH 8.0での5 mMのリン酸緩衝液で10 μMに希釈した。CDスペクトルを260から185 nmまで、25℃で、95℃に加熱した後、再度試料を冷却した後に25℃で記録した。各温度について、データは4回のスキャンの平均として示されている。温度スキャンを、25〜95℃の温度間隔での1℃ステップを記録することによって222 nmで測定し、データを2状態アンフォールディングモデルにフィットした。
それぞれNTD40K/K65D及びNTwtとの融合で、標的ペプチドSP-C33Leu(配列番号56〜57をコードする配列番号58〜59)及びKL4(配列番号60〜61をコードする配列番号62〜63)を有する融合タンパク質コンストラクトを、pT7発現ベクターにクローニングし、コンピテントな大腸菌 BL21(DE3)細胞に化学的に形質転換した。PGB1又はTrxとの融合で同じ標的ペプチド及びタンパク質を有する融合タンパク質コンストラクトを、同じ手順に供した。
高度に可溶性のPGB1ドメインとの比較における、凝集しやすい融合パートナーに対して溶解性を媒介するNT変異体NTwt及びNTD40K/K65Dの能力を試験した。TrxもSP-C33Leuとの融合で評価したが、その乏しい性能のために後に除外した。

実施例3で得られた融合タンパク質を発現する細胞を、80%の振幅で1.5分間、1秒オン及び1秒オフ、3分の合計時間、超音波処理によって溶解した。沈殿によるフルスケール精製中にのみ、超音波処理手順を、5分間氷上に立てた後に繰り返し、試料を50000×gで30分間遠心分離した。塩化ナトリウムを1.2 Mの最終濃度まで上清に添加し、遠心分離を繰り返した。遠心分離からのペレットを、20 mMのTris-HCl、pH 8に溶解させ、そして、融合タンパク質を完全に再溶解させるために、60%の振幅で1.5分間、1秒オン及び1秒オフ、3分、短く超音波処理した。30 mLの溶解溶液に1.7 mLの2 M HCl、次いで1.7 mLの1 M CNBrを添加することによって、CNBr切断を行った。切断反応を一晩室温で行った。翌日、800 mMの塩化ナトリウムを、第2の沈殿ステップにおいて切断反応物に添加し、次いで20000×gで30分間遠心分離した。上清を除去し、ペレットを37℃で乾燥させ、99.9%のエタノールで懸濁した。不溶性物質を、20000×g で30分間の遠心分離により除去した。
SP-C33LeuをNTD40K/K65D融合タンパク質として発現し、クロマトグラフィーステップから独立したプロセスで生成した。図7Dは、融合タグの除去のためにNaCl沈殿/エタノール抽出プロトコル及びCNBr臭化物切断を用いた、NTD40K/K65D-SP-C33LeuからのSP-C33Leuペプチドの精製のSDS-PAGE評価を示す。P1、S1、P2、S2及びTはそれぞれ、不溶性画分、可溶性画分、第1の沈殿後のペレット、第1の沈殿後の上清、及び精製された標的ペプチドを示す。
NTD40K/K65D-SP-C33LeuからのSP-C33Leuペプチドの精製について上述した手順は、NTD40K/K65D-KL4からのKL4ペプチドの精製にも再現可能であった。図8Dは、NaCl沈殿/エタノール抽出プロトコル、及び融合タグの除去のためのCNBr臭化物切断を用いた、NTD40K/K65D-KL4からのKL4ペプチドの精製のSDS-PAGE評価を示す。P1、S1、P2、S2及びTはそれぞれ、不溶性画分、可溶性画分、第1の沈殿後のペレット、第1の沈殿後の上清、及び精製された標的ペプチドを示す。
ESI-MSによる実施例5で得られた精製されたSP-C33Leu(配列番号44)のさらなる特徴付けは、エタノールに溶解した組み換え産生されたペプチドが、正しい共有構造を有することを示した。
一回換気量及び肺気量に対するSP-C及びその誘導体の影響は、呼気終末陽圧(positive end-expiratory pressure)(PEEP)を有する動物モデルを用いて評価することができる(Almlen, Aら、Neonatology 92、194〜200(2007))。
アミロイド凝集メカニズムを広く調査するために、研究者らは、非常に凝集しやすく、かつイン・ビトロでアミロイド様線維を形成することができるポリペプチドである、β17(配列番号27)を設計した。β17は、5つの短いターンによって分離された、7個のアミノ酸をそれぞれ含む6つのβ鎖からなる。残基は、極性、非極性パターンで配置されている。β17は、これまでに、免疫検出のためのmyc-タグとともに発現されており、その結果、ペプチドは可溶性でなく、かつ封入体から精製しなければならなかった。そのような精製プロセスは、変性条件(8 Mの尿素)の使用を必要とし、時間がかかり、かつ不安定なタンパク質を生じる。また、レポーターとしてのThioflavin T(ThT)を用いるβ17繊維形成の研究は、早期凝集に悩まされている。
(A)ゲル濾過
実施例8で得られたβ17ポリペプチド(配列番号27)の流体力学的サイズを、ゲル濾過を用いて特徴づけた。ゲル濾過を、0.3 mL/分で流れる24 mL Superdex-200カラムで行った。試料を、ランニング緩衝液としての150 mMのNaCl有り又は無しで、200 μLのloop及びTBS、5 mM EDTA、pH 8を用いて注入した。カラムを、それぞれ10.25、12.54、13.65及び16.18 mLで溶出した、アポフェリチン(443 kDa)、アルコールデヒドロゲナーゼ(150 kDa)、BSA(66 kDa)及び炭酸脱水酵素(29 kDa)で較正した。
凝集動態を、アミロイド線維に結合する際のThT蛍光についての増強された量子収量に基づいてThT蛍光を用いてモニターした。マイクロプレートウェル(Microplate Corning 3881、96ウェル、低結合、半分の領域、Corning Incorporated Life Sciences、Acton、MA)で、10 μM ThTを含む20 mMのリン酸ナトリウム緩衝液、pH 8.0、0.2 mMのEDTA中で、80 μM NTD40K/K65D-β17を用いて、実験を行った。440 nmの励起フィルター及び480 nmの発光フィルターで、Fluostar Omega又はOptimaプレートリーダー(BMG Labtech、Offenburg、Germany)を用いて、37℃で静止条件下で、ThT蛍光を記録した。
Bri2は、N-末端領域、次いでTMドメイン、リンカー領域、BRICHOSドメイン及びC-末端領域からなるTM糖タンパク質である。機能は比較的不明であるが、当該タンパク質は、アルツハイマー病、Aβ前駆体タンパク質プロセシング、Aβ恒常性、アポトーシス、腫瘍抑制及び男性生殖に関連付けられている。Bri2遺伝子における突然変異は、脳におけるアミロイド線維の蓄積によって引き起こされる家族性イギリス型認知症及び家族性デンマーク型認知症と関連している。状況はアルツハイマー病(AD)と類似しており、アルツハイマー病はアミロイドベータペプチド(Aβ)の蓄積を特徴とし、最終的に脳のプラークを形成する。Bri2 BRICHOSドメインは、抗アミロイドシャペロンとして作用し、アミロイド形成を防ぐと考えられ、従って有望な治療標的として興味深い。
(A)精製Bri2-BRICHOSのSDS-PAGE分析
図16は、実施例10で得られたBri2-BRICHOS タンパク質(配列番号50)の精製のSDS-PAGE分析である。精製ステップからの試料を、還元(パネルA)及び非還元(パネルB)条件下で、NTwt(レーン1)又はNTD40K/K65D(レーン2)との融合でのBri2113-231について分析した。分子量を左に示している。上清をNi-Sepharoseにロードし、フロースルーを回収し(FT)、10 mLのランニング緩衝液での4回の洗浄ステップに続く(W1-W4)。純粋な融合タンパク質を、イミダゾールで溶出し(F)、トロンビンで切断した(CL)。
実施例10で得られたBri2-BRICHOS タンパク質(配列番号50)の立体配座を、本質的に実施例9に記載されたゲル濾過を用いて特徴づけた。それらの単量体立体配座における融合タンパク質の割合を推定するために、ゲル濾過を行った。図17は、Bri2113-231融合タンパク質のオリゴマー状態を決定するためのゲル濾過を示す。
SP-A及びSP-Dは、肺の本質的な可溶性自然免疫タンパク質であり、肺を調査し、病原体に結合して、それらの中和、凝集及びクリアランスをもたらす。それらはまた、様々な免疫細胞の及び炎症性免疫応答の機能の重要な調節因子である。SP-A又はSP-Dのいずれかを欠損したマウスは、RSVを含む病原体感染症に対する増加した感受性、並びに感染性細菌の接種後の過剰炎症応答を示す。ヒト呼吸器疾患のモデルにおける肺内でのそれら分子作用を理解するために、SP-A及びSP-Dの組み換え型を開発するための多大な努力が成されてきた。組み換えSP-A及びSP-Dはまた、様々なヒト肺疾患の治療のための治療可能性を有し得る(Salgado, D.ら、Front Immunol 5、623(2014))。
Aβペプチドは、アルツハイマー病に関するアミロイドプラークの形成に関与している。当該ペプチドは、ベータ及びガンマセクレターゼでのタンパク質分解切断によってアミロイド前駆体タンパク質(APP)から放出され、及び凝集して、オリゴマー又はアミロイドプラークを構成するより大きな線維を形成し得る。Aβミスフォールディング及びフィブリル化の機構、並びに、線維及び中間体オリゴマー状態のイン・ビトロ及びイン・ビボ毒性は、合成ペプチドを用いて広範に研究されてきた。細菌発現系を用いたAβ1-40及びAβ1-42ペプチドの成功した組み換え生産は、変性条件下での封入体からの抽出によって実証された。しかしながらこの方法は、ペプチドのオリゴマー状態に対する制御を可能にせず、分析前に単量体画分を得るためのその後のサイズ排除クロマトグラフィーを必要とする。
膵島アミロイドポリペプチド(アミリン又はIAPP;Uniprot ID P10997)は、ペプチドホルモンであり、膵臓β-細胞からインスリンと共分泌され、かつ血糖値の調節に重要な役割を果たす。IAPPにより形成された膵臓アミロイドが、II型糖尿病の発症に関連していることを研究は示唆している。IAPPは、ProIAPPと呼ばれるプロペプチドとして発現され、当該プロペプチドは刺激によりIAPPにプロセシングされる。ヒトproIAPPはこれまでに、Trx溶解性タグとの融合で組み換え発現され、変性条件下での封入体からの抽出によって精製されてきた。組み換えヒトIAPP(hIAPP)ペプチドは、多数の商業的供給源から入手可能であり、かつ、ペプチドを可溶性に保つために、溶解性タグ、例えばGSTとの融合で、あるいはBSA又はOVAへのコンジュゲーションで、送達される。
カテリシジンは、脊椎動物の好中球の顆粒に見られる抗菌及びエンドトキシン結合タンパク質のファミリーである。このファミリーのメンバーは、カテリン様ドメインとして知られている高度に保存された12 kDaのN-末端を共有する。生物学的に機能的なドメインはC-末端に存在し、セリンプロテアーゼによりプロタンパク質から切断された際に活性化する。唯一のヒト型のカテリシジン、hCAP18は、抗菌ペプチドLL-37に対するプロタンパク質であり、LL-37はプロテアーゼ3による細胞外切断によって放出される。hCAP18は、成熟ペプチドLL-37に匹敵する効率でグラム陰性細菌の増殖を阻害する。しかしながら、hCAP18の組み換え生産は、低収量及び不明な溶解性に関連付けられており、それゆえ、組み換え生産のために新たな戦略が必要とされている。
γ-セクレターゼタンパク質複合体は、四構成要素プロテアーゼであり、アミロイド前駆体タンパク質(APP)のプロセシング及びアルツハイマー病関連ペプチドのアミロイドベータ(Aβ)の生成に関与する。γ-セクレターゼの活性及び特異性の調節は、アルツハイマー病の治療のための潜在的な治療戦略を表す。構成要素の1つである、ニカストリン又はNCTは、大きな細胞外ドメイン(ECD)を有するI型膜貫通糖タンパク質であり、これは、γ-セクレターゼ基質の動員に重要な役割を果たしていると考えられている。全長ニカストリンECDの異種大腸菌産生はこれまでに報告されていない。
GFPは、青から紫外線の範囲の光にさらされると、明るい緑色蛍光を示す。GFPは、生物学的プロセスと干渉することなく細胞全体への拡散を可能にする比較的小さなサイズのために、発現のリポーターとして頻繁に使用される。GFPの多くの異なる突然変異体が操作されており、最も重要には、S65T突然変異が蛍光及び光安定性を劇的に増加させた。増強されたGFP(eGFP)は、S65T突然変異に加えてF64L点突然変異の結果であり、これは、37℃で増加したフォールディング効率を示し、かつ哺乳動物細胞におけるGFPの実用的な使用を可能にした。
精製された可溶性NTD40K/K65D-SP-C33Leu融合タンパク質(配列番号 56)を、実施例3〜4に記載のように得て、サイズ排除クロマトグラフィー(SEC)及び透過型電子顕微鏡(TEM)に供した。TEMのために、精製された可溶性NTD40K/K65Dタンパク質(配列番号2)を対照として使用した。
精製融合タンパク質を、ランニング緩衝液(20 mMのTris、150 mMのNaCl、1 mMのEDTA、pH 8.0)で2 mg/mLに希釈した。Superdex 200カラムをランニング緩衝液で平衡化し、試料の200 μLを0.5 mL/分の速度でカラムに通した。タンパク質の溶出を、280 nmでの吸光度を測定することによって検出した。分子量標準フェリチン(440 kDa)、アルドラーゼ(158 kDa)、コンアルブミン(75 kDa)、オボアルブミン(43 kDa)、炭酸脱水酵素(29 kDa)及びリボヌクレアーゼA(13.7 kDa)(GE Healthcare)を流し、それぞれ8.56 mL、10.65 mL、12.06 mL、12.96 mL、14.26 mL及び15.64 mLで溶出させた。
試料を20 mMのTris、pH 8で希釈した。ネガティブ染色のために、3μlの試料を、グロー放電した炭素被覆銅グリッドに適用し、2 %(w/v)酢酸ウラニルで染色し、風乾した。グリッドを、200 kVで操作したJEOL JEM-2100f透過型電子顕微鏡を用いてチェックした。TVIPS TemCam-F415 4k x 4k CCD-カメラ(Tietz Video and Image Processing Systems GmbH、Gauting、ドイツ)で、60000の通常倍率を用いて、画像を収集した。
欧州特許出願公開第2 644 619号明細書は、溶解性向上部分NTA72R及び当該部分を含む融合タンパク質を開示している。NTA72Rは、6.4のpH未満であっても構成単量体である。融合タンパク質NTA72R-SP-C33Leu(配列番号100)及びNTwt-SP-C33Leu(配列番号57)を大腸菌BL21(DE3)細胞で発現し、実施例3〜4に従って精製した。
NTwt-SP-C33Leu > NTA72R-SP-C33Leu。
修飾されたフィブロネクチン由来RGDループ、FNcc、反復部分及び小瓶状腺スピドロイン(MiSp)からのCT部分を有するスピドロインタンパク質を、それぞれNTD40K/K65D及びZとの融合でクローニングした(配列番号101〜102)。同一の融合タンパク質ではあるが、大瓶状腺スピドロイン(MaSp)からのCT部分を有する融合タンパク質もクローニングした(配列番号103〜104)。修飾されたスピドロインタンパク質の発現レベルを試験するために、融合タンパク質をpT7発現ベクターにクローニングし、コンピテントな大腸菌BL21(DE3)細胞に化学的に形質転換した。
スピドロイン融合タンパク質の発現レベルを試験するために、4RepCTに共有結合したIgG断片sCD40を、それぞれNTD40K/K65D及びZとの融合でpT7発現ベクターにクローニングし(配列番号105〜106)、コンピテントな大腸菌BL21(DE3)細胞に化学的に形質転換した。
それぞれNTD40K/K65D及びZとの融合でソルターゼ認識配列に共有結合したIgG断片sCD40を(配列番号107〜108)、pT7発現ベクターにクローニングし、コンピテントな大腸菌BL21(DE3)細胞に化学的に形質転換した。
Claims (17)
- 配列番号1と少なくとも70%の同一性を有する100〜160アミノ酸残基の部分を含むタンパク質であって、配列番号1の40位に対応するアミノ酸残基が、Lys、Arg及びHisからなる群から選択され;かつ、配列番号1の65位に対応するアミノ酸残基が、Asp及びGluからなる群から選択される、タンパク質。
- 前記配列番号1の40位に対応するアミノ酸残基が、Lys又はArg、好ましくはLysである、請求項1に記載のタンパク質。
- 前記配列番号1の65位に対応するアミノ酸残基が、Aspである、請求項1又は2に記載のタンパク質。
- 以下、
(i)溶解性向上(solubility-enhancing)部分である、請求項1から3のいずれか一項に記載の少なくとも1つの部分;並びに
(ii)所望のタンパク質又はポリペプチドである、少なくとも1つの部分、
を含む融合タンパク質である、請求項1から3のいずれか一項に記載のタンパク質。 - 以下、
(iii)少なくとも1つの所望のタンパク質又はポリペプチド部分と、少なくとも1つの溶解性向上部分との間に配置された、少なくとも1つの切断部位、
をさらに含む、請求項4に記載のタンパク質。 - 請求項1から3のいずれか一項に記載の少なくとも1つの部分の、融合タンパク質の部分としての使用であって、前記融合タンパク質の別の部分である所望のタンパク質又はポリペプチドの溶解性を向上するための使用。
- 所望のタンパク質又はポリペプチドの産生方法であって、以下のステップ:
a)好適な宿主において、所望のタンパク質又はポリペプチドを含む請求項4から5のいずれか一項に記載の融合タンパク質を発現し;並びに
b)融合タンパク質を含有する混合物を得て、そして場合により、
b1)融合タンパク質を単離すること、
を含む、方法。 - 以下:
c)融合タンパク質を切断して、切断産物として残留溶解性向上部分又はその断片から所望のタンパク質又はポリペプチドを切り離し;そして場合により、
c1)所望のタンパク質又はポリペプチドを単離すること、
をさらに含む、請求項7に記載の方法。 - 融合タンパク質を単離する、ステップb1)を含み;かつ、所望のタンパク質又はポリペプチドを単離する、ステップc1)を含む、請求項7から8のいずれか一項に記載の方法。
- 前記方法が、ゲル濾過、クロマトグラフィー又は任意の他の固相吸着に基づく分離を含む分離ステップを含まないことを条件とする、請求項9に記載の方法。
- 融合タンパク質を単離する、ステップb1)が、以下のステップ:
b1a)融合タンパク質の沈殿;並びに
b1b)沈殿した融合タンパク質を水性溶媒中に懸濁すること、ここで、融合タンパク質は水性溶媒に可溶性である、
を含む、請求項7から10のいずれか一項に記載の方法。 - 所望のタンパク質又はポリペプチドを単離する、ステップc1)が、以下:
c1b)有機溶媒中に切断産物を懸濁することによって、所望のタンパク質又はポリペプチドを抽出すること;ここで、所望のタンパク質又はポリペプチドは有機溶媒に可溶性であり;かつ、残留溶解性向上部分又はその断片は有機溶媒に可溶性でない、
