Abstract
Description
生物科学および関連分野において、選択された要素を微少流体デバイスから搬出することは有用であり得る。本発明のいくつかの態様は、微少流体デバイス中の特定の保持用囲い(holding pen)中の微小物体の群を選択するための、および、微少流体デバイスから選択された群を搬出するための装置ならびに処理を含む。
いくつかの態様において、本発明は、微小物体を微少流体デバイスから搬出する処理を提供する。処理は、微少流体デバイスの包囲体(enclosure)の内側に置かれている保持用囲い中に置かれている微小物体の群を選択することを含み得る。保持用囲いは、包囲体の内側に置かれている複数の保持用囲いのうちの1つであり得る。処理は、選択された微小物体の群を包囲体の内側にあるステージングエリアへ移動させること、および、選択された微小物体の群を、該ステージングエリアから、包囲体中の通路を経て、該包囲体の外側にある場所へ搬出することをさらに含み得る。ある態様において、包囲体中の各保持用囲いは、複数の微小物体を保持するように構成された単離領域を含む。ある態様において、微小物体の群を移動させることは、微小物体の群を、微少流体デバイス内に配置されている他の全ての微小物体から単離することを含む。

Claims (31)
- 複数の保持用囲いを含む包囲体を有する微少流体デバイスから、微小物体を搬出する処理であって、該処理することが、
該微少流体デバイスの該包囲体の内側にある該複数の保持用囲いのうち1つの保持用囲いに置かれている微小物体の群を選択すること;
該微小物体の選択された群を、該包囲体の内側にあるステージングエリアへ移動させること;および、
該選択された群の該微小物体の1つを、該ステージングエリアから、該包囲体における通路を経て、該包囲体の外側にある場所へ搬出すること
を含む、前記処理。 - 移動させることが、微小物体の群を、微少流体デバイスに配置されている他の全ての微小物体から単離することを含む、請求項1に記載の処理。
- 複数の保持用囲いの各々が、複数の微小物体を保持するように構成されている単離領域を含む、請求項1または2に記載の処理。
- 選択された群の各微小物体が、生体細胞である、請求項1〜3のいずれか一項に記載の処理。
- 選択された群が、単一の生体細胞である、請求項4に記載の処理。
- 選択された群が、生体細胞のクローン集団である、請求項4に記載の処理。
- 選択することが、微小物体の群が特定の活性または物理的な特徴を有することを決定することを含む、請求項1〜6のいずれか一項に記載の処理。
- 選択することが、微少流体デバイスの中へ光のパターンを向けることを含み、ここで該光のパターンが、選択された群の微小物体を囲み、および、該選択された群の該微小物体を捕まえるDEP力を起動する;ならびに、
移動させることが、該光のパターンをステージングエリアへ移動させることを含み、ここで該選択された群の該微小物体が、該光のパターンが該ステージングエリアへ移動しても、該光のパターンによって起動された該DEP力によって捕まえられたままである、
請求項1〜7のいずれか一項に記載の処理。 - ステージングエリアが、微少流体デバイスの包囲体によって定義されるチャネルに置かれている、請求項1〜8のいずれか一項に記載の処理。
- ステージングエリアが、保持用囲い内に置かれている、請求項1〜8のいずれか一項に記載の処理。
- 微小物体の選択された群をステージングエリアへ移動させることが、重力によって、該微小物体の群が該ステージングエリアへ引き寄せられることを含む、請求項10に記載の処理。
- 搬出することが、選択された群の微小物体を、搬出デバイスの近位端中の開口を経て引き出すことによって、該選択された群の該微小物体を、包囲体中の通路を経てステージングエリアから引き出すことを含む、請求項1〜11のいずれか一項に記載の処理。
- 引き出すことが、群の微小物体を搬出デバイスの近位端での開口の中へ引き出す圧力差を発生させることを含む、請求項12に記載の処理。
- 搬出することが、搬出デバイスを、包囲体中の通路の上に配置されている搬出インタフェースの中へ挿入することをさらに含み、それによって、該搬出デバイスの近位端での開口が、該通路に近接して設置される、請求項12または13に記載の処理。
- 搬出インタフェースが、通路の上に配置されおよびそれを覆う自閉カバーを含み;ならびに、
挿入することが、該カバー中の割れ目に対し、搬出デバイスの近位端を押し付けることによって、該カバーを開口すること、および、該搬出デバイスの該近位端を、該通路に近接するように移動させることを含む、
請求項14に記載の処理。 - 搬出インタフェースが、通路の上に配置されおよびそれを覆う自己回復カバーを含み;ならびに、
挿入することが、搬出デバイスの近位端で該カバーを貫通すること、および、該搬出デバイスの近位端を、該通路に近接するように位置付けることを含む、
請求項14に記載の処理。 - 微小物体の選択された群が、1μL以下の体積を有する媒体中または他の溶液中に搬出される、請求項1〜16のいずれか一項に記載の処理。
- 搬出することの後に、選択された群のうち搬出されなかった微小物体のいずれについても、ステージングエリアを調べることをさらに含む、請求項1〜17のいずれか一項に記載の処理。
- 搬出することの後に、選択された群のうち搬出されなかった微小物体のいずれについても、搬出デバイスを調べることをさらに含む、請求項12〜18のいずれか一項に記載の処理。
- 搬出することの後に、選択された群のうち搬出されなかった微小物体のいずれについても、搬出インタフェースを調べることをさらに含む、請求項14〜19のいずれか一項に記載の処理。
- 微小物体のうち搬出されなかったいずれのものも、ステージングエリア、搬出デバイスおよび/または搬出インタフェースから除去することをさらに含む、請求項18〜20のいずれか一項に記載の処理。
- 包囲体中の通路が、該包囲体中の第2通路であり、および、ステージングエリアの第2端に近接するように配置されており、
移動させることが、選択された群を、該ステージングエリアの該第2端とその逆側にある第1端との間の該ステージングエリアの中間部分へ移動させることを含み、ならびに、
搬出することが、
第1搬出デバイスから、該包囲体中の第1通路を経て、該ステージングエリアの該第1端の方へ液体を流すこと、およびそれによって、該ステージングエリアの該第1端から該ステージングエリアの該第2端への流れが発生すること、ならびに、
該選択された群の該微小物体を、該ステージングエリアの該第2端から、該包囲体中の該第2通路を経て、該第2通路に近接して配置されている第2搬出デバイスの近位端での開口の中へ引き出すこと
を含む、請求項1に記載の処理。 - 液体媒体を含有するように構成された包囲体、ここで該包囲体が該包囲体を経る通路を含む;
該包囲体の内側に配置されているチャネル、ここで該チャネルが、該チャネルに通じて開かれている保持用囲いを複数有しており、該複数の保持用囲いの各々が、微小物体の群を保持するように、および、微小物体の群を、該複数の保持用囲いの他の保持用囲い中の微小物体から単離するように、構成されている;ならびに、
該通路に近接して該包囲体上に配置されている搬出インタフェース、ここで該搬出インタフェースが、搬出デバイスの第1端中の開口が、該包囲体を経て該通路に近接して位置付けられるように、該搬出デバイスとつなぎ合わせられるように構成されている、
を含む、微少流体装置。 - 包囲体が、少なくとも1つのステージングエリアを含む、請求項23に記載の装置。
- 少なくとも1つのステージングエリアが、包囲体を経る通路に近接して置かれている、請求項24に記載の装置。
- 少なくとも1つのステージングエリアが、チャネル中に置かれている、請求項24に記載の装置。
- 包囲体が、複数のステージングエリアを含み、および、各保持用囲いが、該複数のステージングエリアのうち1つのステージングエリアを含む、請求項24に記載の装置。
- 搬出インタフェースが、包囲体を経る通路を全体的に覆うカバーを含む、請求項23〜27のいずれか一項に記載の装置。
- カバーが、該カバーを一続きの弁に分ける割れ目を含み、
該弁が、互いに接触するように偏られ、それによって、包囲体を経る通路が全体的に覆われ、および、
該弁が、該割れ目に対し押し付けられている搬出デバイスの第1端の力に応答して、該搬出デバイスの該第1端を受け入れるための開口を形成するよう離れていくのに充分な可撓性がある、
請求項28に記載の装置。 - カバーが、自己回復材料を含み、該材料が、搬出デバイスによって貫通され得ひいては該搬出デバイスを受け入れるための穴を形成し得るが、自己回復し、およびそれによって、該搬出デバイスが該材料から除去されるとき該穴を閉鎖する、請求項28に記載の装置。
- 複数の保持用囲いのうちいずれか1つの保持用囲い中の微小物体の群を捕まえるための、および、該捕まえられた微小物体の群を少なくとも1つのステージングエリアへ移動させるための手段をさらに含む、請求項23〜30のいずれか一項に記載の装置。
