腎不全(末期腎臓疾患、ESRD)は、治療または療法がない衰弱性疾患である。ESRDに苦しむ患者は、透析に進み、腎臓機能を回復するために腎臓移植を必要とする。大部分の腎臓疾患およびESRDは、糸球体で始まり、蛋白尿を伴う。ESRDは、患者に大きな負担を負わせ、世界的な医療管理系は効果的な療法およびより良い治療選択肢のための緊急の必要性を要する。
発明は一般的に、インビトロで有足突起を培養する方法に関する。該方法は、有足突起細胞の第1の培養物(例えば、始原有足突起または条件付きで不死化された有足突起細胞株などの有足突起細胞株)を、許容条件または増殖条件下で維持して有足突起の集団を産生する工程、該有足突起の集団(またはその画分)を、非許容条件または分化条件下で、部分的に分化した有足突起の集団を得るのに充分な時間、維持する工程、および次いで該部分的に分化した有足突起(またはその画分)を、非許容条件または分化条件下で、最終的に分化した有足突起の集団を得るのに充分な時間、第2の培養物において維持する工程を含む。
〔1〕腎臓疾患を治療するための候補化合物を同定する方法であって、該方法が、
(a) 複数のユニットを含むアレイを提供する工程、ここで、それぞれのユニットは、

〔10〕有足突起培養物において、健常な最終的に分化した有足突起を同定する方法であって、
(a) 蛍光部分にコンジュゲートしたF-アクチン特異的結合剤を使用した、全細胞に対するF-アクチン張力線維の平均蛍光強度割合、ここで、約0.75以下の平均蛍光強度割合は、健常な最終的に分化した有足突起を示す;
(b) 1細胞当たりの区分けされたF-アクチン張力線維の平均数、ここで、1細胞当たり少なくとも約100の区分けされたF-アクチン張力線維は、健常な最終的に分化した有足突起を示す;
(c) 1ウェル当たりの区分けされたF-アクチン張力線維の平均数、ここで、前記ウェルは、約0.32cm2の表面積を有し、1ウェル当たり少なくとも約100,000の区分けされたF-アクチン張力線維は、健常な最終的に分化した有足突起を示す;

1. 概要
発明は一般的に、インビトロにおいて有足突起を培養する方法に関する。該方法は、有足突起細胞の第1の培養物(例えば、始原有足突起、増殖有足突起、有足突起前駆細胞または有足突起細胞株、例えば条件付きで不死化された有足突起細胞株)を、許容条件または増殖条件下で維持して有足突起の集団を産生する工程、有足突起の集団(またはその画分)を、非許容条件または分化条件下で、部分的に分化した有足突起の集団を得るのに充分な時間、維持する工程、次いで該部分的に分化した有足突起(またはその画分)を、非許容条件または分化条件下で、最終的に分化した有足突起の集団を得るのに充分な時間、第2の培養において維持する工程を含む。
明細書において使用される場合、単数形「a」、「an」および「the」は、内容が明確にそうではないと指示しなければ、複数の参照物を含む。
一局面において、発明は、インビトロにおける有足突起培養物およびかかる培養物を使用して産生される有足突起に関する。該有足突起は、哺乳動物(例えば、ネズミ科(マウス、ラット)、ヒト)または魚類(例えば、ゼブラフィッシュ)もしくは昆虫(例えば、ショウジョウバエ)などの非哺乳動物などの任意の所望の動物種由来であり得る。
A. 候補化合物のスクリーニング
明細書に記載される有足突起培養を利用して、腎臓疾患を治療するための候補化合物をスクリーニングし得る。
別の局面において、発明は、明細書に記載される培養方法により産生された有足突起を含む最終的に分化した有足突起などの健常な有足突起を同定する方法を提供する。好ましい態様において、該方法は、明細書に記載される培養などの培養中の健常な有足突起(例えば、最終的に分化した有足突起)を同定するために使用される。該方法は、個々にまたは任意の所望の組合せのいずれかで、セクションB1〜B6に記載されるパラメーターの1つ以上を決定する工程を含む。
本発明はまた、患者における、腎臓疾患または腎臓疾患の素因の診断に関する。
培養された有足突起は、げっ歯類およびヒトの糸球体疾患の重大な局面を信頼性高く再現する、実証されたモデル系を提供する。しかしながら、有足突起をインビトロで培養することは、培地またはハイスループットアッセイ環境における有足突起の使用を妨げるいくつかの問題により長い間妨げられていた。有足突起は、インビトロでの培養では不均一な形態を示し、頑強であり、再現性が高く、異なるウェル、プレートおよび異なる日数の間で変動性が低いデータを得ること困難にする。インビトロでこれらの細胞を成育させて適切に分化させるためには長い培養時間が必要であるので、有足突起細胞数はまた、ウェルごと、およびプレートごとに大きく変動する。さらに、分化した有足突起は、現在使用されている細胞培養系において、制限された増殖する能力を示した。これらおよび他の問題は、培地またはハイスループットアッセイ環境における使用にこれらの細胞を適用することを妨げてきたので、新規の薬物および薬物候補、シグナル伝達経路の発見のために、および疾患を有する患者の体液を用いるこれらの細胞を治療することによる疾患の診断において、これらの細胞を用いてハイスループットスクリーニング(HTS)および他のかかるアッセイを実施し得る。
F-アクチン張力線維の強度および数により、有足突起アクチン細胞骨格における変化を測定した。画像からF-アクチン張力線維を区分けし、細胞の残りの強度と対比して、強度の割合を得た。1細胞当たりの区分けされたF-アクチン張力線維の数による細胞に対するF-アクチン張力線維の強度割合により、PANにより誘導された有足突起の変化を定量化し得た。前記のパラメーターを使用して、有足突起接着点の変化もモニタリングし、PANに対する用量応答様式の変化も定量化し得た。また、有足突起アクチン細胞骨格および接着点の定量化のための前述のパラメーターも、ミゾリビンによる用量応答様式のPAN処理からの保護を測定することが見出された。
有足突起細胞培養
ヒトおよびマウス有足突起を、以前に記載されたように培養した(Saleem et al., JASN 13, 630-638 (2002); Mundel, Reiser et al., Experimental Cell Research, 236, p248 (1997))。例えば、マウス系統H-2Kb-tsA58由来のマウス有足突起を、ラット尾部コラーゲンコート皿上、10%熱不活性化FBS、ペニシリンおよびストレプトマイシン(100U/ml)、ならびに10U/mlのリコンビナントマウスγインターフェロンを補充したRPMI培地中、許容条件下、33℃で培養した。次いで細胞をトリプシン処理し、γインターフェロンを含まない培地中、37℃での分化のために再度平板培養した。細胞を0〜7日間分化させて、96ウェルおよび384ウェルプレートについてそれぞれ1500/ウェルまたは500/ウェルの密度で、ラット尾部コラーゲンコートマルチウェルプレートに再度播種した。次いで、細胞を、固定および分析の前に、種々の試薬を用いたその後の実験のために、マルチウェルプレート中でさらに5〜14日間分化させた。始原有足突起を単離および培養するための方法は当該技術分野で周知である。例えば、Kabgani et al., Primary Cultures of Glomerular Parietal Epithelial Cells or Podocytes with Proven Origin, Plos One, PLoS ONE (2012) 7(4): e34907. doi:10.1371/journal.pone.0034907.参照。
分化した有足突起を96ウェルまたは384ウェルマルチウェル(multiwall)光学プレート(PerkinElmer, Boston, MA)で培養し、様々な量のピューロマイシン、PAN、LPS、アドリアマイシンおよび他の薬剤で処理した。いくつかの場合において、細胞は、損傷剤(例えばPAN)による損傷を防ぐために、化学物質および他の薬剤(例えばミゾリビン)で共処理した。他の場合において、傷害発生剤の添加の0〜48時間後に救済剤を添加した。続いて、4%パラホルムアルデヒドおよび2%スクロース(細胞を破壊または培養培地を除去しないように細胞培養培地に直接添加した)を含む溶液で細胞を固定し、1時間後、PBS中0.3% Triton X-100を用いて10分間、室温で透過性処理を行った。Alexa fluor 594ファロイジン(Invitrogen)でF-アクチンを可視化し、核/細胞質染色のためにHCS CellMask Blue (Invitrogen)を使用し、接着点を検出するために抗パキシリン抗体クローンY113を使用した。alexa fluor 488コンジュゲート二次抗体を使用して、抗パキシリン抗体をさらに検出した。適切なフィルターと共にOpera XL (Perkin Elmer)を使用して共焦点顕微鏡検査を行った。種々の細胞パラメーターの定量化のために、その後の画像分析に画像を使用した。アッセイの頑強性のためにウェルを複製してアッセイを行った。
Columbus 2.3.2ハイコンテンツスクリーニング画像データ保存および分析システム(PerkinElmer)を使用して画像を分析した。「Find Nuclei」分析モジュール、方法C、ならびに0.85〜0.90の共通の閾値および190μm2〜250μm2より大きい面積を使用して、CellMask Blue染色された核を検出した。「Find Cytoplasm」分析モジュール、方法D、ならびに0.20の個々の閾値を使用して、細胞質を検出した。100の最少平均細胞強度および細胞面積について1500μm2を有する細胞を、分析のために選択した。「Find Spots」分析モジュール、方法B、ならびにそれぞれ0.40および0.505の検出感度およびスプリット係数を使用してF-アクチン線維を区分けした。いくつかの場合において、それぞれ0.45および0.46のスプリット係数を適用した。「Select Populations」分析モジュールを使用して、パラメーター「バックグラウンドに対する相対スポット」および「総スポット面積」パラメーターにより、健常細胞集団と損傷細胞集団を分類した。次いで、1ウェルあたりの健常な細胞のパーセンテージを決定するために、健常細胞集団 対 損傷細胞集団を使用した。1ウェル当たり約100〜1500個の細胞を分析した。

Claims (12)
- 腎臓疾患を治療するための候補化合物を同定する方法であって、該方法が、
(a) 複数のユニットを含むアレイを提供する工程、ここで、それぞれのユニットは、
(i) 有足突起細胞を、許容条件下で、有足突起細胞が少なくとも1回倍加するのに充分な時間、第1の細胞培養物において維持し、それにより有足突起の集団を産生する工程;
(ii) (i)で得られた該有足突起の集団を、非許容条件下で、部分的に分化した有足突起の集団を得るのに充分な時間維持する工程;および
(iii) (ii)で得られた該部分的に分化した有足突起を再度播種し、(ii)で得られた該部分的に分化した有足突起を、非許容条件下で、最終的に分化した有足突起の集団を得るのに充分な時間、第2の細胞培養物において維持する工程
を含む、インビトロで有足突起を培養する方法により得られる1つ以上の最終的に分化した有足突起を含む;
(b) 候補化合物と、1つまたは複数の前記最終的に分化した有足突起を、該アレイ中の少なくとも1ユニットにおいて接触させる工程;および
(c) 1つまたは複数の最終的に分化した有足突起の細胞特性を決定する工程
を含み、ここで、適切な対照に対する細胞特性の変化が、前記化合物が腎臓疾患を治療するための候補であることを示す、方法。 - 細胞特性が、膜透過性、形態、生存力、または有足突起マーカーの発現である、請求項1記載の方法。
- 前記アレイが、マルチウェルプレートを含み、それぞれのウェルが、1つ以上の最終的に分化した有足突起を含む、請求項1または2記載の方法。
- 前記有足突起マーカーが、F-アクチンである、請求項2記載の方法。
- F-アクチンの発現レベルが、蛍光部分にコンジュゲートしたF-アクチン特異的結合剤を使用した蛍光染色により決定される、請求項4記載の方法。
- F-アクチンの発現レベルが、ELISAまたはウエスタンブロットにより決定される、請求項4記載の方法。
- 前記形態が、アクチン細胞骨格の平均長さである、請求項2記載の方法。
- 細胞形態が、光学顕微鏡検査または電子顕微鏡検査により分析される、請求項7記載の方法。
- 前記候補化合物が、最終的に分化した有足突起の生存力を向上させる、請求項1〜8いずれか記載の方法。
- 有足突起培養物において、健常な最終的に分化した有足突起を同定する方法であって、
(a) 蛍光部分にコンジュゲートしたF-アクチン特異的結合剤を使用した、全細胞に対するF-アクチン張力線維の平均蛍光強度割合、ここで、約0.75以下の平均蛍光強度割合は、健常な最終的に分化した有足突起を示す;
(b) 1細胞当たりの区分けされたF-アクチン張力線維の平均数、ここで、1細胞当たり少なくとも約100の区分けされたF-アクチン張力線維は、健常な最終的に分化した有足突起を示す;
(c) 1ウェル当たりの区分けされたF-アクチン張力線維の平均数、ここで、前記ウェルは、約0.32cm2の表面積を有し、1ウェル当たり少なくとも約100,000の区分けされたF-アクチン張力線維は、健常な最終的に分化した有足突起を示す;
