Abstract
Description
[1]ワクシニアウイルス増殖因子(VGF)及びO1Lの機能を欠損し、かつ、SCR(short consensus repeat)ドメインを欠失したB5Rをコードする遺伝子を有する、ワクシニアウイルス。
[2]SCRドメイン1〜4を欠失したB5Rをコードする遺伝子を有する、[1]に記載のワクシニアウイルス。
[3]B5RにおけるSCRドメイン1〜4の欠失が、B5Rにおける配列番号1により示されるアミノ酸配列に対応する領域の欠失である、[2]に記載のワクシニアウイルス。
[4]SCRドメインを欠失したB5Rをコードする遺伝子が、B5Rのシグナルペプチド、ストーク、膜貫通ドメイン及び細胞質尾部を含むポリペプチドをコードする遺伝子である、[1]〜[3]のいずれかに記載のワクシニアウイルス。
[5]SCRドメインを欠失したB5Rが、配列番号2のアミノ酸配列に対応するB5Rのアミノ酸配列からなる、[1]から[4]のいずれかに記載のワクシニアウイルス。

本発明は、SCRドメインを欠失したB5Rをコードする遺伝子を有する、ワクシニアウイルスを提供する。本発明はまた、VGF及びO1Lの機能を欠損し、かつ、SCRドメインを欠失したB5Rをコードする遺伝子を有する、ワクシニアウイルスを提供する(本明細書中、当該ワクシニアウイルスを「本発明のワクシニアウイルス」ともいう。)。
本発明の医薬組成物には、本発明のワクシニアウイルス及び薬学的に許容される賦形剤を含む医薬組成物が含まれる。
本発明の医薬組成物は、がんの予防又は治療剤として用いることができる。
相同組換え法により組換えワクシニアウイルスを作製するために使用するトランスファーベクタープラスミドDNAを以下の通りに作製した。
国際公開第2015/076422号に基づいてpUC19−VGFベクターを作製した。より具体的には、pUC19−VGFベクター作製のために、LC16mO株のゲノムDNA(Accession No.AY678277.1)を鋳型に使用し、PCRでVGF遺伝子領域を増幅し、そのPCR産物を制限酵素BamHI及びHindIIIで切断し、Invitrogen社のpUC19ベクター(製品コード:54357)の同じ制限酵素部位にクローニングした。
pTagBFP−N vector(FP172、Evrogen社)のDNAを鋳型として、2つのプライマー(配列番号7及び配列番号8)によってBFP遺伝子領域を増幅した。そのPCR産物を制限酵素SfiI及びEcoRIで切断し、pTK−SP−LGベクター(国際公開第2015/076422号。但し、鋳型として使用したLC16mO株のゲノムDNAとして、ゲノムDNA(Accession No.AY678277.1)を使用し、pUC19ベクターとしてInvitrogen社のpUC19ベクター(製品コード:54357)を使用した。また、pVNC110−Luc/IRES/EGFPプラスミドは国際公開第2011/125469号に記載のpVNC110−Luc/IRES/EGFPを使用した。)の同じ制限酵素部位にクローニングし、合成ワクシニアウイルスプロモーター(Journal of Virological Methods、1997、Vol.66、p.135−138)下にBFPを連結したpTK−SP−BFPを構築した。次に、pTK−SP−BFPを制限酵素SphI及びEcoRIで切断し、末端を平滑化した。こうして得られたDNA断片を、pUC19−VGFベクターを制限酵素AccIで切断し末端を平滑化した部位へクローニングすることで、pTN−VGF−SP−BFPを構築した。次に、プロメガ社のpGL4.20ベクター(製品コード:E6751)をHindIIIで切断し、末端を平滑化した。そして、XbaIで切断することによって、ルシフェラーゼLuc2遺伝子をコードするポリヌクレオチド(Accession No.DQ188840の100番目から1752番目)断片を得た。この断片を、pTN−VGF−SP−BFPをAgeIで切断し、末端を平滑化し、さらにNheIで切断した部位へクローニングすることで、合成ワクシニアウイルスプロモータで駆動されるLuc2遺伝子を有するトランスファーベクタープラスミドDNAを構築した。構築したプラスミドDNAをpTN−VGF−SP−Luc2と称する。
上記(2)と同様の方法で、pTK−SP−BFPを制限酵素SphI及びEcoRIで切断し、末端を平滑化して得られたDNA断片を、pUC19−O1Lベクター(国際公開第2015/076422号。但し、LC16mO株のゲノムDNAとして、ゲノムDNA(Accession No.AY678277.1)を使用し、pUC19ベクターとしてInvitrogen社のpUC19ベクター(製品コード:54357)を使用した。O1L遺伝子領域はpUC19ベクターのXbaIサイトに挿入した。)を制限酵素XbaIで切断し末端を平滑化した部位へクローニングすることで、トランスファーベクタープラスミドDNAを作製した。作製したプラスミドDNAをpTN−O1L−SP−BFPと称する。次に、ヒトに対してコドンを最適化した大腸菌LacZ遺伝子を含むポリヌクレオチド(配列番号9)を制限酵素AgeIとNheIで切断した。LacZをコードするポリヌクレオチド断片をpTN−O1L−SP−BFPベクターの同じ制限酵素部位(AgeI部位及びNheI部位)にクローニングしBFPを置き換えることで、合成ワクシニアウイルスプロモータで駆動されるLacZ遺伝子を有するトランスファーベクタープラスミドDNAを構築した。構築したプラスミドDNAをpTN−O1L−SP−LacZと称する。
pB5R(国際公開第2011/125469号。但し、LC16mO株のゲノムDNA(Accession No.AY678277.1)を鋳型に使用した。)のDNAを鋳型として、2つのプライマー(配列番号3及び配列番号4)によってB4R遺伝子領域を増幅した。そのPCR産物を制限酵素NotI及びFspIで切断した。また、pDsRed−Express−N1(Clontech社)のDNAを鋳型として、2つのプライマー(配列番号5及び配列番号6)によってDsRed遺伝子領域を増幅した。そのPCR産物を制限酵素FspI及びMfeIで切断した。この2種類のDNA断片を、制限酵素NotI及びMfeIで切断したpB5Rにクローニングすることで、トランスファーベクタープラスミドDNAを作製した。作製したプラスミドDNAをpTN−DsRed(B5R−)と称する。一方、pB5Rを制限酵素NotI及びNspIで、又は制限酵素NspI及びSacIで切断した。この2種類のDNA断片(前者はB4RとB5Rの1番目から21番目まで(N末部分)に及ぶ領域に相当し、後者はB5Rの236番目から317番目まで(C末部分)とさらにB6Rに及ぶ領域に相当する。前者の領域中の突出末端(20〜21番目の配列に相当)と後者の領域中の突出末端(236番目〜237番目の配列に相当)とは互いに相補的である。)を、制限酵素NotIとSacIで切断したpB5Rにクローニングすることで、トランスファーベクタープラスミドDNAを構築した。作製したプラスミドDNAをpTN−B5RΔ1−4と称する。pTN−B5RΔ1−4には4つのSCRドメインが欠失したB5R蛋白質がコードされる。4つのSCRドメインが欠失したB5R蛋白質は配列番号2に示した配列となる。
国際公開第2015/076422号に記載の方法に基づいてワクシニアウイルスLC16mO株からVGFとO1Lの機能を欠損させた組換えワクシニアウイルス(LC16mO VGF−SP−LucGFP/O1L−p7.5−DsRedと称する。)を作製した。これについて、次世代シークエンサーPacBio RSII(Pacific Bioscience社)を用いて、シークエンシングを行い、得られた配列情報から、Sprai[BMC GENOMICS、2014、Vol.15、No.699、p.1−8]ソフトウェアを利用してウイルスゲノムの再構成を行って塩基配列を決定したところ、配列番号18で示される塩基配列を有していた。また、当該塩基配列の両末端にはループ配列(Virology、1990、Vol.179、p.247−266)が付加されており、両末端ループ配列は配列番号16又は配列番号17で示される塩基配列であった。
EEVは感染初期に細胞上清中に放出され、IMVは感染初期には細胞内に留まるという性質からウイルス感染細胞の培養液中の上清中のウイルスをEEVとして回収した。すなわち、感染後48時間で培養液を回収し、700×g、4℃、10分の遠心で浮遊細胞を落として上清のみを取り出し、EEVとして回収し、力価測定を行った。一方、上清を回収した後に残った浮遊細胞と共に、プレート内の接着細胞をダルベッコ−リン酸緩衝生理食塩水(D−phosphate buffered saline;D−PBS(Wako)で洗浄後セルスクレーパーにより剥離させOpti−MEMを加えて回収し、凍結融解及びソニケーション処理により細胞を破砕し、700×g、4℃、10分遠心後に上清をIMVとして回収した。
EEV又はIMVによる細胞死滅効果への影響を検証した。
まずIMVによる細胞死滅効果を比較するため、96ウェルプレートに4種の卵巣癌細胞株(A2780(ECACC 93112519)、CaOV3(ATCC(登録商標) HTB−75)、RMG−1(JCRB JCRB0172)、SKOV3(ATCC(登録商標) HTB−77))を6.0×103細胞/ウェルで播種し、37℃、24時間培養後60〜80%コンフルエントになったところでB5Rウイルス又はΔSCRウイルスをMOI=0、0.001、0.01、0.1、1で各細胞株に感染させた。B5Rウイルス又はΔSCRウイルスは専らIMV形態を有すると考えられる。感染から120時間後にBZ−X700(キーエンス)を用いた蛍光観察(図2−1)及びCellTiter 96 Aqueous Nonradioactive Cell Proliferation Assay(Promega)を用いた細胞生存率測定(図3のパネルA)を行った。蛍光観察像ではウイルスのVGF遺伝子内に挿入されたEGFP遺伝子の発現により、ウイルス増殖がGFP蛍光として観察される。細胞を播種していないウェルの値を0%生存、細胞を播種してウイルスを添加していないウェルを100%生存として細胞生存率を求めた。
次に、EEVによる細胞死滅効果を測定するため、上記(1)と同様の細胞に各ウイルスをMOI=0、0.001、0.01、0.1、1で感染させ48時間培養した。その後、感染細胞の培養液50μlを回収し、700×g、4℃、10分の遠心で上清のみをEEVとして回収した。回収したEEVを96ウェルプレートに新たに播種した上記4種の卵巣癌細胞株へ感染させた。EEV感染から120時間後、上記(1)と同様に蛍光観察(図2−2)及び細胞生存率測定(図3のパネルB)を行った。
次に、SCRドメインをコードする領域の欠失によるウイルスの免疫回避能の増大をウイルスの中和実験により検証した。EEVは抗B5R抗体と補体のCDC活性により中和される(Journal of Virology、2009、Vol.83、No.3、p.1201−1215)ため、EEV及びIMVにウサギ由来の抗ワクシニアウイルス血清とウサギ補体を加えてウイルス中和を行った。
具体的には、EEV及びIMVの産生用に、ウイルス産生・感染細胞として、EEV及びIMV共にB5Rウイルス/ΔSCRウイルス間での産生量に差が見られなかったSKOV3細胞を使用した。24ウェルプレートにSKOV3細胞を5.0×104細胞/ウェルで播種し、24時間培養後60〜80%コンフルエントになったところで、B5Rウイルス又はΔSCRウイルスをMOI=0.1で感染させた。感染後、上記実施例3に記載の方法で感染後48時間時点のEEV及びIMVを回収した。回収したB5Rウイルス又はΔSCRウイルスそれぞれに由来するEEV 10μl(約1500〜2000PFU)又はIMV 1μl(約5000〜7000PFU)を0、0.2、0.5、1%のウサギ抗ワクシニアウイルス血清(Capricorn、IgG fraction、ELISA抗体価1:1000、ELISAにより抗B5R抗体を検出済。図4−5中、抗VV血清又は単に血清とも称する。)及び0、1、3、10、25%のウサギ補体(Cedarlane Laboratories、以下及び図4−5において、単に補体とも称する。)とをOpti−MEM中に混合して計50μlのウイルス混合液を得、37℃で30分反応させた。次にウイルス混合液を前日に96ウェルプレートへ6.0×103細胞/ウェルで播種しておいたSKOV3細胞へと感染させ、37℃で2時間吸着させた。2時間の吸着後にウイルス混合液を除き、新たな培養液として10%ウシ胎児血清を含むRPMI−1640培地(Wako)を加えて37℃で96時間培養した。
96時間培養後、実施例4に記載の方法に従い、感染細胞中で増殖したワクシニアウイルスの蛍光観察を行った。図4は感染後96時間時点での蛍光観察結果を示す。蛍光観察により緑色に確認される領域は中和を免れて増殖したウイルスを反映する。
図5は、図4で観察した蛍光観察結果をキーエンス社のハイブリッドセルカウントによって数値化し(以下、「ハイブリッドカウント」という。)、その結果を示したものである。ハイブリッドカウントの値が高いほど、中和回避能が高いことを示す。ΔSCRウイルス由来EEVはB5Rウイルス由来EEVに比較して中和回避能が高い傾向を示す(図5パネルA)。一方、IMVはΔSCRウイルスを由来とするものであってもB5Rウイルスを由来とするものであっても大きな差は見られなかった(図5パネルB)。
SCRドメイン1〜4を欠失したB5Rをコードする遺伝子を有する組換えワクシニアウイルスの免疫回避能及び抗腫瘍効果の向上をマウスを用いた前臨床試験により検証した。
具体的には、pmirGLO Vector(Promega)のウミシイタケルシフェラーゼ―ネオマイシン耐性融合遺伝子(Renilla Luciferase−neo:以下、「Rluc」と称することがある。)を恒常的に発現するヒト卵巣癌A2780細胞(5×106細胞)をBALB/c−nu/nuマウス腹腔内へ移植し、10日間増殖させた。次に、EEV産生用にA2780細胞をT−25フラスコへ1.35×106細胞を播種し、24時間培養後にB5Rウイルス又はΔSCRウイルスをMOI=0.05で感染させ、48時間培養後に採取した培養液を700×g、4℃、5分間の遠心分離に供し、上清をEEVとして回収した。EEV投与の前日にnu/nuマウス腹腔内へウサギ抗ワクシニアウイルス血清(Capricorn、IgG fraction。以下及び図7−8中、単に血清とも称し、図6−1、6−2中、抗VV血清と称する。)を100μl投与し、擬似的な免疫環境下とした。回収したEEVを前記nu/nuマウス腹腔内へ500μl(B5Rウイルス:約1.2×105PFU、ΔSCRウイルス:約7×105PFU)投与した。Vivo Glo Luciferin(プロメガ)の投与によりウイルスのFluc発光(ウイルス増殖)を、またViviRen In Vivo Renilla Luciferase Substrate(プロメガ)の投与により移植したA2780細胞のRluc発光(腫瘍増殖)を、in vivoイメージングシステム(Berthold、NightDHADE LB985)を用いて、それぞれ非侵襲的に検出した。
図6−1(ウイルスFluc発光の検出結果)に示されるように、EEV投与後3日目ではB5Rウイルス−EEV+血清投与及びΔSCRウイルス−EEV+血清投与マウスのどちらもウイルス増殖は抑えられているが、EEV投与後7日目ではΔSCRウイルス−EEV+血清投与マウスはほぼ全ての個体で腹腔内での強力なウイルス増殖が認められた。一方で、B5Rウイルス−EEV+血清投与マウスはウイルス増殖が抑えられていた。
EEV投与後の各マウスの生存率を図8に示す。図8に示されるように、Mock(PBS、PBS+血清)群と比べると全てのEEV投与群で生存延長が見られるものの、B5Rウイルス−EEV+血清投与マウスは早期の生存率低下が認められた。一方、ΔSCRウイルス−EEV+血清投与マウスは血清非投与群と中央値において同等の生存延長を示しており(中央値はΔSCRウイルス−EEV+血清投与マウスは77、血清非投与群(ΔSCRウイルス−EEV)は74.5)、B5Rウイルス−EEV+血清投与マウスと比較し有意な差がLog−rank検定によって確認された(P=0.0291)。
SCR欠失による抗腫瘍効果を免疫応答性の同種同系移植モデルにおいて検証した。
精製のために、実施例2(3)で作製したB5R−LLウイルス及びΔSCR−LLウイルスをA549細胞又はRK13細胞に感染させた。5%CO2存在下37℃で2〜5日間培養した後、感染細胞を回収した。細胞を凍結融解し、ソニケーション処理、又はベンゾナーゼ処理した。OptiPrep(アクシスシールド社)を用いて密度勾配遠心分離法により精製した。その後、RK13細胞にて各ウイルスのウイルス力価を測定し、後記(2)の実験に供した。
同種移植モデルとして、マウス大腸癌CT26細胞を5×105細胞でBALBc/AJjclマウスの左腹部及び右腹部へ皮下移植した。ノギスで腫瘍径を測定した腫瘍体積(短径mm×短径mm×長径mm×0.5)の各群の平均値がウイルス投与側(左側)で48〜88mm3、ウイルス非投与側(右側)で12〜115mm3になるまで7日間成長させた。腫瘍成長後に、B5R−LLウイルス又はΔSCR−LLウイルスを各5×107PFUでマウス左腹部に植付けた腫瘍内へ直接投与した。各ウイルスの代わりにPBS(35μL)を腫瘍内投与した群も準備した(各群N=5)。投与は1日おきに計3回、Day 0、2、4の時点で行った。初回投与後20日までノギスで腫瘍径を測定して腫瘍体積の変動を観察した。さらに、初回投与後7日までVivo Glo Luciferin(プロメガ)の投与によりウイルスのFluc発光によりウイルス増殖の推移を観察した。
配列番号2で示されるアミノ酸配列は、4つのSCRドメインが欠失したB5Rタンパク質のアミノ酸配列である。
配列番号3〜8及び10〜15で示されるヌクレオチド配列はプライマーである。

Claims (8)
- ワクシニアウイルス増殖因子(VGF)及びO1Lの機能を欠損し、かつ、SCR(short consensus repeat)ドメインを欠失したB5Rをコードする遺伝子を有する、ワクシニアウイルス。
- SCRドメイン1〜4を欠失したB5Rをコードする遺伝子を有する、請求項1に記載のワクシニアウイルス。
- B5RにおけるSCRドメイン1〜4の欠失が、B5Rにおける配列番号1により示されるアミノ酸配列に対応する領域の欠失である、請求項2に記載のワクシニアウイルス。
- SCRドメインを欠失したB5Rをコードする遺伝子が、B5Rのシグナルペプチド、ストーク、膜貫通ドメイン及び細胞質尾部を含むポリペプチドをコードする遺伝子である、請求項1から3のいずれか一項に記載のワクシニアウイルス。
- SCRドメインを欠失したB5Rが、配列番号2のアミノ酸配列に対応するB5Rのアミノ酸配列からなる、請求項1から4のいずれか一項に記載のワクシニアウイルス。
- ワクシニアウイルスがLC16mO株である、請求項1から5のいずれか一項に記載のワクシニアウイルス。
- 請求項1から6のいずれか一項に記載のワクシニアウイルス及び薬学的に許容される賦形剤を含む、医薬組成物。
- がんの予防又は治療用である、請求項7に記載の医薬組成物。
