上記のバイオセンサ10について、アセトンを検出対象として以下の通り実験を行った。尚、補酵素にNADH、酵素にS−ADHを用いた。実験の諸条件は下記の通りである。
−溶液
pH7.5、0.1Mのリン酸緩衝液(PB)を用いて50μMのNADHを含むリン酸緩衝液(溶液)が調製された。PBの調製は、0.1Mのリン酸水素二カリウムを0.1Mのリン酸二水素カリウムに徐々に加えることにより行われた。溶液流路25の溶液の流速は、200ml/minとした。
−試料ガス
ガスジェネレータ(PD−1B−2、株式会社ガステック)を用いて、アセトン蒸気が調製された。試料流路35の試料の流速は200ml/minとした。試料流路35には最初に高純度空気が流され、測定開始から4分経過後にアセトン蒸気が流された。
−光源部41
UV−LED(DOWA Electronics Materials Co., Ltd製UF4VL-1H33 ピーク波長340nm)を光源部41として用いた。このUV−LEDにUV−LED電源(GS200、横河電機株式会社)を接続した。
−検出器46
検出器46として光電子倍増管(C9692、浜松ホトニクス株式会社)を使用した。
−バンドパスフィルタ45,47
光源部41側に用いられたバンドパスフィルタ45の透過波長は、330〜350nmである。検出器46側に用いられたバンドパスフィルタ47の透過波長は、480〜500nmである。
−フローセル50
溶液流路25の直径が4mmに形成されたPMMA(Polymethyl methacrylate)セルを用いた。
−反射部80
PTFE膜(ADVANTEC社製 T020A047A)を用いた。この膜は、ポア径:0.2μm、空隙率:74%、膜の厚さ:80μm、波長340nmにおける反射率:98%,波長490nmにおける反射率92%である。
(比較例)
実施例の反射部80を設けないこと以外は全て実施例と同じく構成した。
(蛍光強度測定)
以上の条件で行った実施例と比較例の蛍光強度測定を行った。図4は、測定時間に対する蛍光強度を測定したグラフを示す図である。図中の太線は本実施例の測定時間に対する蛍光強度を示す。図中の細線は反射部80を設けていない比較例の測定時間に対する蛍光強度を示す。
