Description
本出願は、2015年10月26日に出願されたシンガポール特許出願第10201508831S号の優先権を主張する。
本明細書で使用される以下の言葉及び用語は、示された意味を有するものとする。
本発明の例示的で非限定的な又は好ましい実施形態をここに開示する。
本発明の非限定的な例及び比較例を、具体的な例を参照することによって、より詳細に説明するが、それは、本発明の範囲を限定するものと決して解釈されるべきではない。
塩化カルシウム(CaCl2)溶液(720mL、1M)を、Lf水溶液(1.8mL、1〜30mg/mL)へ添加する。次いで、炭酸ナトリウム(Na2CO3)溶液(480mL、1M)を激しく撹拌しながら1回で注入する。1分後に撹拌を停止する。得られた粒子の分散液を遠心により分離し、脱イオン水を用いて2回洗浄する。
例1−1の吸収されたLfを含むCaCO3マイクロ粒子をポリ(L−リジン)(PLL)溶液(2mL、2mg/L)に浸し、15分間連続的に振盪する。次いで、マイクロ粒子を遠心によって回収し、残分PLLを脱イオン水による2回の洗浄により取り除く。
図1の(b)は、シェル形成の成功を示す、例1−1のCaCO3と比較して滑らかな表面を有する吸収されたLfを含むマイクロ粒子の表面に堆積された得られたPLL−(BSA−TA)4シェルのSEM画像を示す。
例1−2のマイクロ粒子を遠心により回収し、脱イオン水(1mL)に再分散する。次いで、分散液に、HCl溶液(1M)を約pH3まで滴下する。得られたカプセル化されたLfを含むシェルを遠心により回収し、脱イオン水で2回洗浄する。図1の(c)は、CaCO3コアが完全に分解した、カプセル化されたLfを含むPLL−(BSA−TA)4シェルのSEM画像を示す。
模擬胃液(SGF)は、塩化ナトリウム(NaCl)溶液(4mL、150mM)(HClを用いてpH3に調整した)をペプシン溶液(1mL、7.1mg/mLのペプシン、150mMのNaCl)と混合することにより調製する。
保護されていないDQ Red BSAタンパク質を含むブランクサンプルを例1−1及び1−3に記載の方法に従って調製する。PLL−(BSA−TA)4及びPLL−(BSA−TA)8シェル内にカプセル化されたDQ Red BSAタンパク質のサンプルを例1−1及び1−3に記載の方法に従って調製する。
P.E.=(1−Ix/I0)*100%
Ixは、PLL−(BSA−TA)4又はPLL−(BSA−TA)8シェル内にカプセル化されたタンパク質の蛍光強度であり、I0は、処理時間の同じ時間における保護されていないタンパク質の蛍光強度である。
ポリ(L−アルギニン)(PARG)をPLLの代わりに固定材として使用する。PARG−(BSA−TA)4シェルにカプセル化されたLfを例1−1から1−3に記載の方法にしたがって調製する。加えて、固定層の無いLfカプセル化(BSA−TA)4シェルを、PLL溶液にマイクロ粒子を浸す工程を省くこと以外は例1−1から1−3に記載の方法によって調製する。図7は、CaCO3溶解後のこれらのサンプルのSEM画像を示す。PARG−(BSA−TA)4シェルは、図1に示すPLL−(BSA−TA)4シェルと同じサイズ、形状及び一体性である。むしろ、サンプル内に高分子電解質の固定層の無いシェルは見られない。
図8は、図1の(c)(例1−3に記載された)のサンプルであるが、pH〜1のHClに溶解したもののSEM画像を示す。このサンプルにおいて崩壊したシェル構造が観察され、安定したシェルは見られない。従って、(BSA−TA)シェルは健康的な人の胃液のpHレベルでは安定であるが、より高い胃の酸性度の原因となる特定の疾患を有する人の胃液では分解し得る。
Claims (37)
- a.任意に活性成分化合物を含むコア、
b.少なくとも2つの交互の層を含むシェル構造であって、前記交互の層は、タンパク質層又はポリフェノール層から選択される該シェル構造及び
c.任意に、前記コア及び前記シェル構造を結合する高分子電解質層であって、前記高分子電解質層は、前記タンパク質層又は前記ポリフェノール層に隣接して配置される該高分子電解質層
を含み、
前記シェル構造は、胃の条件下での分解に耐性があるが、1種以上の腸の酵素によって分解することが可能であるように選択される
コアシェル複合材。 - 前記シェル構造の最外層は、ポリフェノール層である、請求項1に記載のコアシェル複合材。
- 前記高分子電解質は、ポリ(L−リジン)、ポリ(L−アルギニン)、キトサン、デキストラン、デンプン、セルロース又はその誘導体から選択される請求項1又は2に記載のコアシェル複合材。
- 前記タンパク質は、アルファs1カゼイン、ペプシン、ウシ血清アルブミン(BSA)、オボアルブミン、リゾチーム、ウシ血液由来のヘモグロビン、キモトリプシン、β−ラクトグロブリン、トリプシン、ユビキチン、レクチン、ラクトペルオキシダーゼ、ウサギの筋肉からのミオシン、アクチン、カッパ−カゼイン、ベータ−カゼイン、アルファラクトアルブミン、ウシ頸部靱帯からのエラスチン又はフェリチンから選択される請求項1〜3の何れか1項に記載のコアシェル複合材。
- 前記タンパク質層は、胃の消化酵素によるタンパク質分解に耐性があり、30%未満の前記胃の酵素で切断可能なアミノ酸残基対総アミノ酸残基の割合を有することを特徴とする上記請求項の何れか1項に記載のコアシェル複合材。
- 前記タンパク質層は、35%を超える総切断可能残基対総アミノ酸残基の割合を有することを更に特徴とする請求項1〜5の何れか1項に記載のコアシェル複合材。
- 前記最外のポリフェノール層は、少なくとも1種の胃の消化酵素と反応し、それによって前記胃の消化酵素の層をそれに結合させることを特徴とする請求項2〜6の何れか1項に記載のコアシェル複合材。
- 前記胃の消化酵素がペプシンである請求項5に記載のコアシェル複合材。
- 前記タンパク質は、アルファs1カゼイン、ペプシン、ウシ血清アルブミン(BSA)、オボアルブミン、リゾチーム、ウシ血液由来のヘモグロビン、キモトリプシン、β−ラクトグロブリン、トリプシン、ユビキチン、レクチン、ラクトペルオキシダーゼ、ウサギの筋肉からのミオシン、アクチン、カッパ−カゼイン、ベータ−カゼイン、アルファラクトアルブミン、ウシ頸部靱帯からのエラスチン又はフェリチンから選択される上記請求項の何れか1項に記載のコアシェル複合材。
- 前記タンパク質は、BSA又はペプシンから選択される請求項9に記載のコアシェル複合材。
- 各ポリフェノール層は、独立に、没食子酸、カテキン類、エピカテキン類、プロアントシアニジン、アントシアニジン、ガロイル化カテキン類、フラボノイド、イソフラボノイド、ネオフラボノイド、フラボン、又はタンニンから選択されるか、又はその誘導体である上記請求項の何れか1項に記載のコアシェル複合材。
- 前記最外のポリフェノール層は、タンニン、タンニン酸又はその誘導体である請求項2〜11の何れか1項に記載のコアシェル複合材。
- 前記活性成分は、薬学的に活性な化合物又はその塩、食品添加物、多糖類、ホルモン、生物活性剤、タンパク質、酵素、ペプチド、ポリペプチド、プレバイオティクス、植物化学物質、プロバイオティクス、ビタミン、並びにそれらの混合物及び錯体からなる群から選択される上記請求項の何れか1項に記載のコアシェル複合材。
- 前記シェル構造の総厚さは、5nmから100nmまでである上記請求項の何れか1項に記載のコアシェル複合材。
- 前記複合シェルは、実質的に球体の形状であり、約0.5μmから約10μmまでの直径を有する上記請求項の何れか1項に記載のコアシェル複合材。
- 前記胃の条件は、約pH3及び少なくとも1種の胃の酵素の存在によって特徴づけられる上記請求項の何れか1項に記載のコアシェル複合材。
- 前記胃の酵素がペプシンである請求項16に記載のコアシェル複合材。
- 前記腸の酵素は、膵臓又は小腸から分泌される1種以上の酵素である上記請求項の何れか1項に記載のコアシェル複合材。
- 前記腸の酵素は、トリプシン、キモトリプシン、エラスターゼ、エンテロキナーゼ、カルボキシペプチダーゼ、アミノペプチダーゼ、ジペプチダーゼ及びその混合物の群から選択される請求項18に記載のコアシェル複合体。
- 前記腸の酵素は、トリプシン、キモトリプシン又はその混合物である請求項19に記載のコアシェル複合材。
- a)少なくとも1つのタンパク質層及び少なくとも1つのポリフェノール層を交互に堆積し、それによってカプセル化された活性成分を包む多層のシェル構造を形成する工程
を含み、
前記タンパク質層は、前記シェル構造が胃の条件下での分解に耐性があるが、1種以上の腸の酵素によって分解可能であるように選択される
コアシェル複合材を調製する方法。 - 前記活性成分は、任意に前記堆積工程の前に高分子電解質によってカプセル化され、更に、任意に、前記活性成分は、固体の支持体に吸収される請求項21に記載の方法。
- 前記タンパク質層は、胃の消化酵素によるタンパク質分解に耐性があり、30%未満の、前記胃の酵素によって切断可能残基対総アミノ酸残基の割合を有することを特徴とする請求項21又は22に記載の方法。
- 前記タンパク質層は、35%を超える総切断可能残基対総アミノ酸残基の割合を有することを特徴とする請求項21〜23の何れか1項に記載の方法。
- 前記固体の支持体は、多孔質構造を含む請求項22に記載の方法。
- 前記方法は、前記堆積工程の前、間又は後に前記固体の支持体を除去する工程を更に含む請求項22〜25の何れか1項に記載の方法。
- 前記除去工程は、前記固体の支持体を化学的に分解することを含む請求項26に記載の方法。
- 前記堆積工程(a)は、交互に、活性成分をタンパク質溶液に混合して前記タンパク質層を形成し、次いで、ポリフェノール溶液中に混合して前記タンパク質層上に前記ポリフェノール層を形成し、それによって前記多層のシェル構造を形成することを含む交互層堆積である請求項21〜27の何れか1項に記載の方法。
- 前記堆積工程は、前記高分子電解質層に隣接して前記タンパク質層を堆積することを含む請求項22〜28の何れか1項に記載の方法。
- 前記堆積工程は、前記高分子電解質層に隣接して前記ポリフェノール層を堆積することを含む請求項22〜28の何れか1項に記載の方法。
- 前記堆積工程は、最外層として前記ポリフェノール層を堆積することを更に含む、請求項21〜30の何れか1項に記載の方法。
- 前記高分子電解質は、ポリ(L−リジン)、ポリ(L−アルギニン)、キトサン、デキストラン、デンプン、セルロース又はその誘導体から選択される請求項21〜31の何れか1項に記載の方法。
- 前記タンパク質は、アルファs1カゼイン、ペプシン、ウシ血清アルブミン(BSA)、オボアルブミン、リゾチーム、ウシ血液由来のヘモグロビン、キモトリプシン、β−ラクトグロブリン、トリプシン、ユビキチン、レクチン、ラクトペルオキシダーゼ、ウサギの筋肉からのミオシン、アクチン、カッパ−カゼイン、ベータ−カゼイン、アルファラクトアルブミン、ウシ頸部靱帯からのエラスチン又はフェリチンから選択される請求項21〜32の何れか1項に記載の方法。
- 前記タンパク質は、BSA又はペプシンから選択される請求項33に記載の方法。
- 各堆積された前記ポリフェノール層は、独立に、没食子酸、カテキン類、エピカテキン類、プロアントシアニジン、アントシアニジン、ガロイル化カテキン類、フラボノイド、イソフラボノイド、ネオフラボノイド、フラボン、又はタンニンから選択されるか、又はその誘導体である請求項21〜34の何れか1項に記載の方法。
- 治療に用いるための請求項1〜22の何れか1項に記載のコアシェル複合材。
- 医薬の製造における請求項1〜22の何れか1項に記載のコアシェル複合材の使用であって、前記医薬はヒト又は動物の小腸へ活性成分を送達するように構成されている使用。
