Abstract
Description
a)細菌培養物を提供するステップと、
b)対照及び試験培養ウェルのそれぞれの無菌ディスクを、細菌培養物からの接種材料で接種するステップと、
c)組成物を試験培養ウェルに添加するステップと、
d)試験及び対照培養ウェルを、細菌がバイオフィルムを形成する条件下でインキュベートするステップと、
e)試験及び対照培養ウェルのそれぞれからのディスク上の細菌の数又は濃度を決定するステップであって、対照ウェルと比べて試験ウェル中のより少ない数又はより低い濃度が、本組成物が細菌によるバイオフィルム形成の処置又は予防において好適であることを示す、決定するステップである。
方法
TSB中に増殖させた黄色ブドウ球菌(S.aureus)及び緑膿菌(P.aeruginosa)の静止期培養物を、0.5マクファーランド(McFarland)(c.1.5×108CFU/mL)に調整し、1:4の比(1mLの接種材料を3mLの0.1%バクトペプトンで)更に希釈した。生菌数を、各接種調製物からとった。24ウェル培養プレート内に収容した無菌ディスク(直径6mm×1.0mm)を80μLの培養物で接種した。次いで、0.2mlの試験培地を添加し、アッセイプレートをパラフィルムで密閉して、80rpmで振盪しながら摂氏37度で24時間インキュベートし、バイオフィルを形成させた。未使用のウェルにPBSを充填し、湿度を増加させて、蒸発を防止した。
A−「低」と称される2U/g(2U/ml)−5gの蜂蜜+100μlのグルコースオキシダーゼ原液(100U/ml)
B−「高」と称される6U/g(6U/ml)−5gの蜂蜜+300μlのグルコースオキシダーゼ原液(300U/ml)
それぞれの30μlをディスク/バイオフィルムに添加した。
蜂蜜及びグルコースオキシダーゼの両方を含む組成物は、緑膿菌(P.aeruginosa)及び黄色ブドウ球菌(S.aureus)の24時間経過した単一種のバイオフィルムの有意な阻害を実証したが、一方蜂蜜を単独で含む及びグルコースオキシダーゼを単独で含む組成物は、バイオフィルムの阻害を示さなかった。Prontosan(登録商標)ゲルは、黄色ブドウ球菌(S.aureus)に対しては高度に活性であったが、緑膿菌(P.aeruginosa)バイオフィルムに対してはほとんど活性を有さなかった(図1及び2を参照)。
Claims (27)
- 病原性細菌の集団によるバイオフィルムの形成を処置又は予防する方法において使用するための、オキシドレダクターゼ酵素と、前記酵素の基質とを含む組成物であって、前記方法が、前記組成物を患者に投与することを含むことを特徴とする、組成物。
- 請求項2に記載の組成物において、前記オキシドレダクターゼ酵素がグルコースオキシダーゼであり、前記基質がグルコースであることを特徴とする、組成物。
- 請求項1又は請求項2に記載の組成物において、前記病原性細菌の集団が、緑膿菌(Pseudomonas aeruginosa)、黄色ブドウ球菌(Staphylococcus aureus)及び/又は表皮ブドウ球菌(Staphylococcus epidermidis)を含むことを特徴とする、組成物。
- 請求項1〜3のいずれか一項に記載の組成物において、前記病原性細菌の集団が、緑膿菌(Pseudomonas aeruginosa)を含むことを特徴とする、組成物。
- 請求項1〜4のいずれか一項に記載の組成物において、前記組成物が、ラクトペルオキシダーゼを含まないことを特徴とする、組成物。
- 請求項1〜5のいずれか一項に記載の組成物において、前記組成物が、酵素と、基質と、薬学的に許容される担体とからなることを特徴とする、組成物。
- 病原性細菌の集団によるバイオフィルム形成の処置又は予防において使用するための装飾被覆材であって、前記創傷被覆材が、請求項1〜6のいずれか一項で定義された組成物を含むことを特徴とする、創傷被覆材。
- 表面上の病原性細菌バイオフィルムの形成を予防する、又は表面から病原性細菌バイオフィルムを除去する方法であって、オキシドレダクターゼ酵素と、前記酵素の基質とを含む組成物を、前記表面に投与するステップを含むことを特徴とする、方法。
- 請求項8に記載の方法において、エクスビボ又はインビトロで実施されることを特徴とする、方法。
- 請求項8に記載の方法において、インビボで実施されることを特徴とする、方法。
- 病原性細菌の集団による表面上のバイオフィルム形成の処置又は予防のための方法であって、オキシドレダクターゼ酵素と、基質とを含む組成物の治療的量を前記表面へ投与するステップを含むことを特徴とする、方法。
- 病原性細菌の集団によるバイオフィルム形成の処置又は予防のための方法であって、オキシドレダクターゼ酵素と、基質とを含む組成物の治療的量を、必要とする患者に投与するステップを含むことを特徴とする、方法。
- 請求項8〜12のいずれか一項に記載の方法において、前記オキシドレダクターゼ酵素がグルコースオキシダーゼであり、前記基質がグルコースであることを特徴とする、方法。
- 請求項8〜13のいずれか一項に記載の方法において、前記病原性細菌の集団が、緑膿菌(Pseudomonas aeruginosa)、黄色ブドウ球菌(Staphylococcus aureus)及び/又は表皮ブドウ球菌(Staphylococcus epidermidis)を含むことを特徴とする、方法。
- 請求項8〜14のいずれか一項に記載の方法において、前記病原性細菌の集団が、緑膿菌(Pseudomonas aeruginosa)を含むことを特徴とする、方法。
- 請求項8〜15のいずれか一項に記載の方法において、前記組成物が、ラクトペルオキシダーゼを含まないことを特徴とする、方法。

- 病原性細菌の集団によるバイオフィルム形成の処置又は予防のための方法であって、オキシドレダクターゼ酵素と、前記酵素の基質と、薬学的に許容される担体とからなる組成物の治療的量を、必要とする患者に投与するステップを含むことを特徴とする、方法。
- 病原性細菌の集団によるバイオフィルムの形成を処置又は予防するために、創傷を処置する方法であって、オキシドレダクターゼ酵素と、前記酵素の基質と、任意選択的に薬学的に許容される担体とを含む組成物を、前記創傷の部位に投与するステップを含むことを特徴とする、方法。
- 請求項20に記載の方法において、前記組成物を組み込む創傷被覆材を適用するステップを含むことを特徴とする、方法。
- 請求項21に記載の方法において、前記創傷被覆材が、前記組成物を含むフォーム又はガーゼを含むことを特徴とする、方法。
- 請求項20〜22のいずれか一項に記載の方法において、前記酵素及び基質が、別個の成分として、任意選択的に、創傷被覆材の別個の層で投与されることを特徴とする、方法。
- 請求項20〜23のいずれか一項に記載の方法において、バイオフィルムを形成する病原性細菌の集団を含む創傷、又はバイオフィルムを形成する病原性細菌の集団による感染の危険にさらされている創傷を特定する先行ステップを含むことを特徴とする、方法。
- 請求項20〜24のいずれか一項に記載の方法において、前記創傷が慢性創傷であることを特徴とする、方法。
- 請求項20〜25のいずれか一項に記載の方法において、前記創傷を処置する前記方法が、陰圧閉鎖療法を更に含むことを特徴とする、方法。
