Abstract
Description
本発明のもう一つの目的は、上記高親和力のPDL-1分子の製造方法と使用を提供することである。
もう一つの好適な例において、前記PDL-1分子のアミノ酸配列は配列番号1で示されるアミノ配列に基づき、かつ配列番号1で示されるアミノ配列に対して1個または複数個のアミノ酸残基の突然変異またはアミノ酸残基の挿入を行うことによって前記PDL-1分子を得る。
もう一つの好適な例において、前記PDL-1分子のアミノ酸配列は配列番号1で示されるアミノ配列と少なくとも90%(好ましくは少なくとも92%、より好ましくは少なくとも94%)の配列相同性を有する。
もう一つの好適な例において、前記PDL-1分子とPD-1分子の親和力は野生型PDL-1分子とPD-1分子の親和力の少なくとも100倍、好ましくは少なくとも200倍、より好ましくは少なくとも500倍である。
もう一つの好適な例において、前記PDL-1分子における突然変異のアミノ酸残基の部位は1、36〜40、47〜48および/または95〜101番目のアミノ酸残基のうちの1個または複数個で、ここで、アミノ酸残基の番号に配列番号1で示される番号が使用される。

本発明の第三の側面では、多価のPDL-1複合体であって、少なくとも2つのPDL-1分子を含み、かつそのうちの少なくとも1つのPDL-1分子は本発明の第一の側面に記載のPDL-1分子であるか、あるいは少なくとも1つの本発明の第二の側面に記載の融合タンパク質を含む複合体を提供する。
本発明の第五の側面では、本発明の第四の側面に記載の核酸分子を含むベクターを提供する。
本発明の第六の側面では、本発明の第五の側面に記載のベクターを含有するか、あるいは染色体に外来の本発明の第四の側面に記載の核酸分子が組み込まれた宿主細胞を提供する。
もう一つの好適な例において、前記薬物組成物はさらにImmTACおよび/またはHATacを含む。
本発明の第八の側面では、疾患を治療する方法であって、治療が必要な対象に本発明の第一の側面に記載のPDL-1分子、本発明の第二の側面に記載の融合タンパク質、または本発明の第三の側面に記載のPDL-1複合体、または本発明の第七の側面に記載の薬物組成物を施用することを含む方法を提供する。
もう一つの好適な例において、前記疾患は腫瘍である。
本発明の第十では、本発明の第一に記載のPDL-1を製造する方法であって、
(i) 本発明の第六に記載の宿主細胞を培養することによって、本発明の第一の側面に記載のPDL-1分子を発現させる工程と、
(ii) 前記のPDL-1分子を単離または精製する工程と、
を含む方法を提供する。
本発明者は幅広く深く研究したところ、意外にPD-1分子に対して高親和力を有する可溶性PDL-1分子が有効にリンパ球の殺傷能力を向上させることを見出した。そのため、本発明はPD-1に対する親和力が野生型PDL-1分子のPD-1に対する親和力の少なくとも2倍である可溶性の高親和力PDL-1分子を提供する。
具体的に、本発明に係るPDL-1分子は配列番号1で示されるアミノ酸配列に突然変異を含有する。より具体的に、前記PDL-1分子のアミノ酸配列は配列番号1で示されるアミノ配列と少なくとも90%の配列相同性を有する。
別途に定義しない限り、本明細書で用いられるすべての技術と科学の用語はいずれも本発明が属する分野の当業者が通常理解する意味と同様である。
本発明の実施またはテストにおいて本発明における記載と類似または等価の任意の方法と材料を使用することができるが、本明細書で好適な方法と材料が挙げられた。
野生型PDL-1分子:本発明に係る野生型PDL-1分子とは、野生型PDL-1分子の細胞外領域で、そのアミノ酸配列とヌクレオチド配列はそれぞれ図1aと図1bに示す。
PBMC:末梢血単核球(PBMC)は円形の細胞核を有する血球で、たとえばリンパ球や単核球が挙げられる。これらの血球は免疫系の感染に抵抗して侵入者に適応する要素である。リンパ球類はT細胞(CD4とCD8陽性は約75%)、B細胞およびNK細胞(合計約25%)からなる。
高親和性T細胞受容体:そのリガンドとの親和力が相応する野生型T細胞受容体とそのリガンドの親和力の少なくとも2倍であるT細胞受容体をいう。
PD-1は活性化したT細胞およびB細胞で発現される免疫抑制性受容体である。PDL-1はPD-1のリガンドで、B7ファミリーに属し、IgVおよびIgC様領域、膜貫通領域および細胞質領域の尾部を有し、PDL-1はリンパ球における受容体PD-1と相互作用し、免疫応答の負調節において重要な役目を果たす。本発明者は幅広く深く研究したところ、意外にPD-1分子に対して高親和力を有する可溶性PDL-1分子が有効にリンパ球の殺傷能力を向上させることを見出した。そのため、本発明はPD-1に対する親和力が野生型PDL-1分子のPD-1に対する親和力の少なくとも2倍である可溶性の高親和力PDL-1分子を提供する。
可溶性の高親和力PDL-1分子を得るために、本発明で使用される野生型PDL-1分子は膜貫通領域を含まない。そのため、本発明の一つの好適な例において、前記PDL-1分子は可溶性のものである。
もちろん、本発明の野生型PDL-1を発現する遺伝子または少し修飾された本発明の野生型PDL-1を発現する遺伝子はいずれも鋳型鎖の調製のために使用することができる。その後、当該鋳型鎖をコードするDNAに本発明の高親和力PDL-1の生成に必要な変更を導入する。
本発明の高親和力PDL-1分子は単独で使用してもよく、抱合体と共役またはほかの様態で結合してもよく、好ましくは共役の様態で結合してもよい。好ましくは、前記抱合体はT細胞受容体で、より好ましくは、前記T細胞受容体は高親和性のT細胞受容体である。
また、本発明は、薬学的に許容される担体と、本発明のPDL-1、または本発明のPDL-1複合体とを含有する薬物組成物を提供する。
また、本発明は、疾患を治療する方法であって、治療が必要な対象に本発明のPDL-1、または本発明のPDL-1複合体、または本発明の薬物組成物を施用することを含む方法を提供するが、特に、本発明のPDL-1分子はほかの分子と併用し、好ましくは、ほかの分子はImmTACまたはHATacである。
(1) 本発明は、PD-1に対して高親和力を有するPDL-1分子を得た。
(2) 本発明の高親和力PDL-1分子は有効にリンパ球の殺傷能力を向上させることができる。
以下の具体的な実施例によって、さらに本発明を説明する。これらの実施例は本発明を説明するために用いられるものだけで、本発明の範囲の制限にはならないと理解されるものである。下記実施例で具体的な条件が示されていない実験方法は、通常、たとえばSambrookおよびRussellら、「モレキュラー・クローニング:研究室マニュアル」(Molecular Cloning-A Laboratory Manual)(第三版)(2001) に記載の条件などの通常の条件に、あるいは、メーカーのお薦めの条件に従う。特に断らない限り、%と部は、重量で計算される。以下、実施例で使用された実験材料および試薬は、特に説明しない限り、いずれも市販品として得られる。
野生型PDL-1の細胞外アミノ酸配列およびヌクレオチド配列はそれぞれ配列番号1および2で、図1aおよび図1bに示すように、野生型PDL-1の細胞外配列を含む標的遺伝子をNcoIおよびNotIで二重酵素切断し、NcoIおよびNotIで二重酵素切断されたpET28aベクターに連結させた。連結物をE.coli DH5αに形質転換させ、カナマイシン含有LBプレートを塗布し、37℃で逆さまで一晩培養し、陽性クローンを取ってPCR選別を行い、陽性組換え体に対して配列決定を行い、配列が正確と確認されたら組換えプラスミドを抽出してE.coli BL21(DE3)に形質転換させ、発現に使用した。
BIAcore分析
BIAcore T200リアルタイム分析システムによって野生型PDL-1分子と PD-1の結合活性を検出した。抗ストレプトアビジンの抗体(GenScript)をカップリング緩衝液(10 mM酢酸ナトリウム緩衝液、pH 4.77)に入れた後、抗体に予めEDCおよびNHSで活性化させたCM5チップを通させ、抗体をチップの表面に固定化させ、最後にエタノールアミンの塩酸溶液で未反応の活性化表面をブロッキングし、カップリング過程を完成させ、カップリングレベルは約15,000 RUであった。
本実施例で使用されたPD-1のアミノ酸配列およびヌクレオチド配列はそれぞれ図3aおよび3bに示すように、その発現、再生および精製の過程は実施例1における野生型PDL-1の発現、再生および精製の過程と同様である。そのビオチン化の過程は以下の通りである。
Millipore限外ろ過チューブで精製されたPD-1分子を濃縮し、同時に緩衝液を10 mM Tris pH 8.0に置き換えた後、ビオチン化試薬として0.05 M Bicine pH 8.3、10 mM ATP、10 mM MgOAc、50 μM D-Biotin、100 μg/ml BirA酵素(GST-BirA)を入れ、室温で混合物を一晩インキュベートし、SDS-PAGEによって完全にビオチン化したか検出した。
Millipore限外ろ過チューブでビオチン化標識されたPD-1分子を1 mlに濃縮し、ゲルろ過クロマトグラフィーによってビオチン化されたPD-1を精製し、Akta精製装置(GE社)を使用し、ろ過されたPBSでHiPrepTM 16/60 S200 HRカラム(GE社)を予め平衡化し、1 mlの濃縮されたビオチン化PD-1分子をかけた後、PBSで1 ml/minの流速で溶離させた。ビオチン化されたPD-1分子は約10mlにおいて単峰として溶離して現れた。タンパク質を含有する成分を合併し、Millipore限外ろ過チューブで濃縮し、BCA法(Thermo)によってタンパク質濃度を測定し、ビオチン化されたPD-1分子を分けて-80 ℃で保存した。
本実施例の上記過程で検出された野生型PDL-1分子とPD-1分子の結合親和力のKD値は2.462E-05Mで、そのBIAcore結合グラフを図4に示す。
実施例1に記載の野生型PDL-1の細胞外配列を鋳型鎖とし、Liら((2005)Nature Biotech 23(3):349-354)によって開示されたファージディスプレイおよび選別の方法に従い、高親和力PDL-1の選別を行った。数回の選別後のファージライブラリーはいずれもPD-1と強い結合シグナルを持ち、中から単一のクローンを取り、配列分析を行った。
実施例1に記載の方法によって本発明の高親和性PDL-1分子を発現、再生および精製し、そして実施例2に記載の方法によってそのPD-1分子との親和力を測定した。本発明で得られた高親和力PDL-1分子とPD-1分子の親和力は野生型PDL-1分子とPD-1分子の親和力の少なくとも2倍で、そのアミノ酸配列は図5に示し、そのPD-1分子との親和力数値は下記表2に示す。
本実施例では非放射性細胞毒性試験によって殺傷作用を検証した。当該試験は51Cr放出細胞毒性試験の比色代替試験で、定量的に細胞分解後放出される乳酸脱水素酵素(LDH)を測定するものである。30-分間カップリングの酵素試験によって培養上清液における放出されたLDHを検出し、それによってテトラゾリド(INT)が赤色のホルマザン産物に転換される。形成する色の量は分解細胞の数に比例する。標準96ウェルプレートリーダーによって490 nmの吸光度のデーターを収集した。
ヒト免疫グロブリン結晶性フラグメント(human immunoglobulin fragment crystallizable、hIgG4Fc)で高親和力PDL1分子の二量体を構築した。具体的に、高親和力PDL-1の細胞外ヌクレオチド配列の3’末端とhIgG4Fcヌクレオチド配列の5’末端をオーバーラップPCRを連結し、融合タンパク質の核酸配列およびアミノ酸配列を得たが、 hIgG4Fcのアミノ酸配列および核酸配列はそれぞれ配列番号37および38で、図8aおよび図8bに示す。融合タンパク質配列を持つ標的遺伝子をEcoRIおよびNotIで二重酵素切断し、EcoRIおよびNotIで二重酵素切断されたpcDNA3.1(+)ベクターに連結した。連結物をTOP10に形質転換させ、アンピシリン含有LBプレートを塗布し、37℃で逆さまで一晩培養し、陽性クローンを取ってPCR選別を行い、陽性組換え体に対して配列決定を行い、配列が正確と確認されたら組換えプラスミドを抽出して293T細胞に一過性導入し、発現に使用した。野生型PDL1分子の二量体の構築方法は上記と同様で、野生型PDL1分子の二量体は本発明における発現手段はPDL1-hIgG4である。高親和力PDL1分子の二量体は本発明における発現手段は高親和力PDL1分子番号-hIgG4、たとえばL2D7-hIgG4である。

Claims (16)
- PDL-1分子であって、前記PDL-1分子のアミノ酸配列は、配列番号1で示されるアミノ配列に基づき、かつ、配列番号1で示されるアミノ配列に対して1個または複数個のアミノ酸残基の突然変異またはアミノ酸残基の挿入を行うことによって前記PDL-1分子を得るが、好ましくは、前記PDL-1分子のアミノ酸配列は、配列番号1で示されるアミノ配列と少なくとも90%の配列相同性を有し、より好ましくは、前記PDL-1分子とPD-1分子の親和力は、野生型PDL-1分子とPD-1分子との親和力の少なくとも2倍であることを特徴とする、前記PDL-1分子。
- 前記PDL-1分子における突然変異のアミノ酸残基の部位が、1〜3、35〜50、および/または95〜105番目のアミノ酸残基のうちの1個または複数個であり、ここで、アミノ酸残基の番号に配列番号1で示される番号が使用されることを特徴とする、請求項1に記載のPDL-1分子。
- 突然変異のアミノ酸残基の部位の数はnであり、ここで、1≦n≦15、好ましくは3≦n≦11、より好ましくは4≦n≦10であり、たとえばnは、5、6、7、8、9、10であってもよいことを特徴とする、請求項1に記載のPDL-1分子。
- 前記PDL-1分子における突然変異のアミノ酸残基の部位が、1F、36I、38Y、40E、47I、48Q、95R、97M、99S、および101Gのうちの1個または複数個を含むか、あるいは102Gの後ろに1個または複数個のアミノ酸残基の挿入があり、ここで、アミノ酸残基の番号に配列番号1で示される番号が使用され、
好ましくは、前記PDL-1分子は、1W;36Q、36E、36Tまたは36N;38F、38N、38Hまたは38A;40M、40F、40L、40Wまたは40Y;47L、47V、47F、47S、47Y、47Pまたは47A;48S;95T、95Vまたは95L;97L;99Gまたは99A;101D、101K、101Eまたは101H;102Gの後ろに挿入されたアミノ酸Gからなる群から選択される1個または複数個を含み、ここで、アミノ酸残基の番号に配列番号1で示される番号が使用され、
より好ましくは、前記PDL-1分子のアミノ酸配列は、配列番号5〜34から選ばれる一つであることを特徴とする、請求項1に記載のPDL-1分子。 - 請求項1〜4のいずれか一項に記載のPDL-1分子を含むことを特徴とする、融合タンパク質。
- 前記融合タンパク質が、さらにhIgG4Fcを含むことを特徴とする、請求項5に記載の融合タンパク質。
- 多価のPDL-1複合体であって、少なくとも2つのPDL-1分子を含み、かつそのうちの少なくとも1つのPDL-1分子が、請求項1〜4のいずれか一項に記載のPDL-1分子であるか、あるいは、少なくとも1つの請求項5に記載の融合タンパク質を含むことを特徴とする、前記複合体。
- 2価のPDL-1複合体、3価のPDL-1複合体または4価のPDL-1複合体であることを特徴とする、請求項7に記載の多価のPDL-1複合体。
- 請求項1に記載のPDL-1分子、請求項5に記載の融合タンパク質、あるいは、請求項7に記載の多価のPDL-1複合体をコードする核酸配列またはその相補配列を含むことを特徴とする、核酸分子。
- 請求項9に記載の核酸分子を含むことを特徴とする、ベクター。
- 請求項10に記載のベクターを含むか、あるいは染色体に外来の請求項9に記載の核酸分子が組み込まれていることを特徴とする、宿主細胞。
- 薬学的に許容される担体と、請求項1〜4のいずれか一項に記載のPDL-1分子、または請求項5に記載の融合タンパク質、または請求項7に記載のPDL-1複合体とを含むことを特徴とする、薬物組成物。
- 前記組成物が、さらにImmTACおよび/またはHATacを含むことを特徴とする、請求項12に記載の薬物組成物。
- 疾患を治療する方法であって、治療を必要とする対象に請求項1〜4のいずれか一項に記載のPDL-1分子、または請求項5に記載の融合タンパク質、または請求項7に記載のPDL-1複合体、または請求項12に記載の薬物組成物を施用することを含むことを特徴とする、前記方法。
- 請求項1〜4のいずれか一項に記載のPDL-1分子、または請求項5に記載の融合タンパク質、請求項7に記載のPDL-1複合体の使用であって、腫瘍を治療する薬物の製造に使用されることを特徴とする、前記使用。
- 請求項1〜4のいずれか一項に記載のPDL-1を製造する方法であって、
(i)請求項11に記載の宿主細胞を培養することによって、請求項1〜4のいずれか一項に記載のPDL-1分子を発現する工程と、
(ii) 前記のPDL-1分子を単離または精製する工程と、
を含むことを特徴とする、前記方法。
