試験2について説明する。試験1の検討において、ゆで時間が短い場合(10秒又は20秒)、葉に黒い部分が残ったものの、黒い部分が抜けた葉は、乾燥しても緑がきれいであることが判明した。そこで、試験2では、葉から黒い部分を抜きとる試験を行う。なお、当該黒い部分は、おかわかめ特有の褐変成分ではないかと見当をつけ、きざんで茹でることにより、葉の組織中に含まれる褐変成分を葉から抜き取りやすくすることにした。なお、おかわかめの葉の組織中にある褐変成分を取り除かなければ、葉の色はきれいな緑色にはならず、乾燥後には灰色または黒色となる。この褐変成分が活性酵素であるならば、クロロフィルの緑色の保持を妨げているとも考えられる。活性酵素を失活させるために適温まで加熱し、前述した黒い部分がどうなるのかを見極めることとした。

結果:上記試験区において、炭酸水素ナトリウム(重曹)水溶液で茹でると、濃い緑色を保持でき、粉砕工程を経た粉末にしても、緑色はそのまま保持できた。黒くならずきれいな緑色の葉を保持できたのは、おかわかめの葉を炭酸水素ナトリウムで茹でることにより、葉の表面が溶け、葉の内部組織中に存在する褐変成分が、湯の中に溶けだしたことが最も大きな理由であると考えられる。加えて、クロロフィルは炭酸水素ナトリウム(重曹)を加えてゆでるとフィトールが脱けでて緑色のクロロフィリンとなることにより、粉砕工程を経た後も緑色が保持できたものと思われる。なお、酢(酸性水溶液)でゆでると、葉の緑色は黄褐色になった。これは、酢(酸)を加えてゆでるとマグネシウムイオンが酸の水素イオンと置換し、マグネシウムイオンが脱けでて黄褐色のフィオフィチンとなるからであろう。
4.大葉を、1%炭酸水素ナトリウム(重曹)水溶液で茹でる(ゆで時間60秒)。

4と5については、大葉を使用した試験を行った。4については、1%炭酸水素ナトリウム(重曹)で大葉を60秒ゆでると、緑色になるもの一部くすみが残るもの、全く緑色が残らないものができる。しかし、葉の組織中にある褐変成分が抜け緑色だけが残るものが大部分であった。5については、ほぼ全数の大葉で褐変成分が抜け、鮮やかな緑色のみの葉になった。また、粉末でも、4と5において、色の差ははっきりと検出された。下記表1は、色差計を使用して、ブランチング処理後の葉の緑色の変化を示している。表中に示すとおり、L*は明るさ、a*は緑〜赤のレベル(a*値はマイナスの値(絶対値)が大きくなるほど緑色が強くなり、プラスの値が大きくなるほど赤色を呈するものと評価される)、b*は青〜黄のレベルを示している(b*値はプラスの値が大きくなるほど黄色が強くなり、マイナスの値(絶対値)が大きくなると青色を呈するものと評価される)。
この色差計による測定において、1%炭酸水素ナトリウム水溶液で120秒ゆでると、a*値が最も低い値となり、緑色が保持できることがわかった。また、本発明における緑色は緑色が濃く残り、黄色の変色が小さいことが理想とする緑色であることから−a*/b*の値で表現することが最適である。−a*/b*の値が大きいものほどより深い緑色である。葉の厚さが厚い物は褐変成分が抜けにくく、またより深い緑色になった。褐変成分を確実に抜き取るには、ゆで時間を長くする、きざんでからゆでるなどが効果的であった。

試験4について説明する。試験4は、炭酸水素ナトリウム水溶液の濃度とゆで時間を変えることにより、おかわかめの葉の緑色の保持を比較し、その相関を探るものである。この試験は、沸騰した炭酸水素ナトリウム水溶液1リットルに対し、おかわかめ葉を略50gゆでた結果である。ゆでた後のおかわかめ葉を乾燥し粉砕したおかわかめ粉末を色差計で測定した。その結果、表2の数値を得た。図5は、表2に基づくグラフであって、炭酸水素ナトリウム水溶液の濃度と処理時間とがおかわかめの葉の緑色(−a*/b*値)に与える影響を示すグラフである。すなわち、図5は、色差計を使用して、ブランチング処理後のおかわかめの葉の緑色の変化を示している。なお、試験4において、色差計として、日本電色工業株式会社のハンディ型分光色差計(NF777)を用いて、L*、a*、b*の各値の測定を行い、これらの測定値から−a*/b*値、−L*b*/a*値(黄化度)を計算した。おかわかめの葉の緑色は緑色が濃く残り、黄色の変色が小さいことが理想とする緑色であることから−a*/b*の値で表現することが最適である。
