Abstract
Description
i)間葉系幹細胞について蛍光量子ドットと結合させたTat及びRGDペプチドで以前に示されたように(Shah等、2007年;Lei等、2008年;Jo等、2012年)、NSCを追跡するためにNSCを標識するため、若しくは特性(自己再生若しくは分化)を調節するため;又は
ii)神経膠芽腫細胞で以前に示されたように(Balzeau等、2013)、機能化したナノ粒子を使用する送達系としての
NFL-TBS40〜63ペプチドの使用を提案する。
神経幹細胞は、神経系の主要な表現型を生じる自己再生性の多能性細胞である。幹細胞は、環境からの外部刺激によって複数の細胞種に分化する能力が特徴である。これらは非対称細胞分裂を受けて2つの娘細胞になり、1つは特殊化されておらず、もう一つは特殊化される。NSCは、主にニューロン、星状膠細胞及び乏突起膠細胞に分化する(Hall及びWat、1989年;Reynolds及びWeiss、1992年;Azari等、2010年)。
神経幹細胞の標的化は、NSCが成人脳に存在し、そこで増殖し、自己再生して新たなニューロン、星状膠細胞及び乏突起膠細胞に分化することができるので、新たな再生戦略の開発に有望である。
その後の態様では、本発明は、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体を含有する医薬組成物を、それらを必要とする患者を治療するための方法で使用するために提供する。
- NFL-TBS40〜63(配列番号1)又はその生物学的に活性のある誘導体を含有する融合タンパク質、
- NFL-TBS40〜63ペプチド又はその生物学的に活性のある誘導体に直接結合した化合物又は生物学的物質、或いは、
- 適切な担体(例えば、ナノカプセル、リポソーム、ミセル)に含有される化合物又は生物学的物質であって、前記担体がNFL-TBS40〜63ペプチド又はその生物学的に活性のある誘導体に結合している化合物又は生物学的物質
であってもよい。
別の態様では、本発明は、インビボ又はインビトロにおいて、神経幹細胞を検出するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体の使用に関する。
a. 試料の細胞を適切な培地に懸濁すること、
b. NFL-TBS40〜63ペプチド又はその生物学的に活性のある誘導体を、試料の懸濁細胞と混合すること、
c. 前記NFL-TBS40〜63ペプチドを含有する細胞の割合を測定することを含み、
前記NFL-TBS40〜63ペプチドを含有する細胞の割合が、試料中の神経幹細胞の割合に対応する方法に関する。
1. フルオレセイン及びヘキサクロロ-フルオレセイン、テトラクロロ-フルオレセイン、カルボキシフルオレセイン(TAMRA)、CAL FLUOR(登録商標)( BIOSEARCH TECHNOLOGIES社製のCAL Fluor Green 520、CAL FLUOR Gold 540、CAL FLUOR ORANGE 560、CAL FLUOR RED 590、CAL FLUOR RED 635)、カルボキシフルオレセインのスクシニミジルエステル(6-カルボキシ-2',4,4',5',7,7'-ヘキサクロロフルオレセイン(HEX(商標)のスクシニミジルエステル)又は6-カルボキシ-4',5'-ジクロロ-2',7'ジメトキシフルオレセイン(JOE(商標)のスクシニミジルエステル)のような誘導体;
2. ローダミン及び5-若しくは6-カルボキシ-X-ローダミン(ROX)、N,N,N',N'-テトラメチル-6-カルボキシローダミンのような誘導体;
3. シアニン及びCy3、Cy3.5、Cy5、Cy5.5のような誘導体;
4. 4,4-ジフルオロ-5,7-ジフェニル-4-ボラ-3a,4a-ジアザ-s-インダセン-3-プロピオン酸、4,4-ジフルオロ-5,p-メトキシフェニル-4-ボラ-3a,4a-ジアザ-s-インダセン-3-プロピオン酸、4,4-ジフルオロ-5-スチリル-4-ボラ-3a,4-adiaz-a-S-インダセン-プロピオン酸、4,4-ジフルオロ-5,7-ジフェニル-4-ボラ-3a,4a-ジアザ-s-インダセン-3-プロピオン酸、4,4-ジフルオロ-5,p-エトキシフェニル-4-ボラ-3a,4a-ジアザ-s-インダセン 3-プロピオン酸及び4,4-ジフルオロ-5-スチリル-4-ボラ-3a,4a-ジアザ-S-インダセン-プロピオン酸のようなBODIPY(登録商標)発色団;
5. テキサスレッド(登録商標)及び誘導体;
6. APTS、HPTS(CASCADE BLUE(登録商標))のようなピレントリスルホン酸;並びに
7. エオジン及び誘導体が含まれる。
a) NFL-TBS40〜63ペプチド又はその生物学的に活性のある誘導体を含むアミノ酸配列を、直接又は適切な担体によって(ナノカプセル等)蛍光色素若しくは発光性色素で標識すること、
b) 前記アミノ酸配列を脳内に注射すること、
c) 神経幹細胞を明らかにするために特定の波長の光を(蛍光性色素若しくは発光性色素に応じて)、ペプチドを注射した脳領域に適用すること
を含む方法を提供する。
神経幹細胞の初代培養は、新生仔(1〜5日)又は成体(<4ヶ月)のウィスターラット脳の脳室下帯から得る。分離した細胞は、D-グルコース(Sigma社)25mmol/L、ピルビン酸Na 1mmol/L、HEPES 15mmol/L、5%ペニシリン/ストレプトマイシン(PAA社)及び1% B27(Gibco社)を補給し、EGF(Promega社)20ng/mLを含有するMEMアルファ(PAA)中で増殖させた。5〜7日後、幹細胞は浮遊するニューロスフェアを形成した。
FITC標識NFL-TBS40〜63ペプチドの内部移行を蛍光標示式細胞分取器(FACS、Becton Dickinson社)によって評価するために、ニューロスフェアを35mm皿に接種し、濃度を増加させたFITC標識NFL-TBS40〜63ペプチドを含有する培地中で、37℃で30分間培養した。FITC標識NFL-TBS40〜63ペプチドの細胞外シグナルを消失させるために、0.4%トリパンブルー(Sigma社)をFACS解析前に添加した。摂取機構を調べるために、ニューロスフェアは4℃で30分間予備インキュベートするか、又は細胞性ATPを欠乏させるためにアジ化ナトリウム10mmol/Lと一緒に2-デオキシ-D-グルコース6mmol/Lの存在下で予備インキュベートするか、又は様々な阻害剤と一緒に37℃で30分間予備インキュベートした。その後、FITC標識ペプチド20μmol/Lを細胞に37℃で30分間添加した。その後、遠心分離(700rpmで5分間)してから、細胞を機械的に分離し、PBSで2回洗浄し、次いでヨウ化プロピジウム(PI、Sigma社)50μg/mL中に再懸濁した。FITC標識ペプチドを取り込んだ蛍光陽性細胞をフローサイトメトリー(FACSCalibur、Becton Dickinson社)によって分析した。
NSC増殖に対するNFL-TBS40〜63ペプチドの効果を分析するために、1次ニューロスフェアを分離し、ウェル当たり5.103個の細胞を96ウェルマイクロプレート上のBD Cell-Tak(BD Biosciences社)に播種した。細胞は濃度を増加させたビオチン化NFL-TBS40〜63ペプチド又はコルヒチン1μg/mLと一緒に37℃で72時間処理した。PBSで洗浄後、細胞を-80℃で凍結させた。最後に、DNA濃度は、CyQUANT細胞増殖アッセイキット(Molecular Probes社)を使用して分析した。
神経幹細胞におけるNFL-TBS40〜63ペプチド摂取を視覚化するために、ニューロスフェアをBD Cell-Tak(BD Biosciences社)カバーガラス又はLabTekチャンバーに播種した。24時間後、ニューロスフェアをFITC標識NFL-TBS40〜63ペプチド20μmol/Lで30分、6時間又は72時間インキュベートした。PBSで洗浄した後、細胞を4%パラホルムアルデヒドで15分間固定し、PBSで3回洗浄した。次に、これらを0.5%トリトンX-100透過処理溶液で30分間インキュベートして、PBSで3回洗浄してから、ブロッキング溶液(0.1%トリトンX-100に溶かした1%牛血清アルブミン)で1時間インキュベートした。その後、ニューロスフェアをマウス抗α-チューブリン抗体(Sigma社)1/1000で、4℃で一晩インキュベートした。PBSで3回洗浄した後、Alexa568nm抗マウス抗体(Life Technologies社)1/200を2時間使用してチューブリンの位置を突き止めた。ニューロスフェアはPBSで3回洗浄してから、4'6-ジアミニド-2-フェニルインドール(DAPI、Sigma社)3μmol/Lを5分間添加した。PBSで洗浄した後、カバーガラスにProLong Gold antifade溶液(Life Technologies社)を加えた。染色したニューロスフェアは、倒立蛍光顕微鏡Leica又はLSM 700 Zeiss共焦点顕微鏡で観察し、画像はそれぞれMetamorph又はZen 2009ソフトウェアで分析した。
ニューロスフェア形成特性を調べるために、24ウェルプレートでEGF 20ng/mLを含有する培地にウェル当たり5.103個のSVZ細胞を接種した。細胞は、濃度を増加させたビオチン化ペプチドの非存在下若しくは存在下で、又はコルヒチン1μg/mLでインキュベートした。5〜7日後、1次ニューロスフェアの総数及び付着した1次ニューロスフェアの割合を各条件下で顕微鏡分析によって測定し、写真を倒立顕微鏡及びMetamorphソフトウェアで撮影した。次に、自己再生アッセイのため、1次ニューロスフェアを各条件下で収集し、単一細胞として分離し、EGF 20ng/mLを含有する培地に接種した。5日後、2次ニューロスフェアの総数を各条件下で顕微鏡分析によって計数した。
実験手順及び動物の世話は全てフランス政府の指針に則り、動物実験倫理の地方委員会(Comite d'Ethique en Experimentation Animale des Pays-de-la-Loire)の承認後実行した。
全実験は、少なくとも3回繰り返した。FACS解析のために、試料当たり20000事象を解析した。結果は、蛍光細胞の平均パーセントとして表し、データは平均の標準誤差(SEM)を有する棒グラフとして表した。統計学的解析は、Prism 3.00(GraphPad software社、San Diego、CA)を使用したスチューデントのt検定で実施した。アスタリスクは対照条件に対する有意なレベルを示す:*P<0.05;**P<0.01;***P<0.001。
新生仔及び成体ラットの神経幹細胞におけるNFL-TBS40〜63ペプチドの内部移行はまずインビトロ及びインビボにおいて調べ、次にその摂取機構を特徴付けた。その後、NSCの微小管ネットワーク、細胞周期及び生存能に対するペプチドの可能性のある分子及び細胞効果を分析した。最後に、ニューロスフェア形成、自己再生及び分化を含むNSCの基本的な特性に対するペプチド摂取の影響を評価した。
神経幹細胞は、濃度を増加させたカルボキシフルオレセインタグ付けペプチドで30分間インキュベートし、次にペプチド取り込みは高感度FACS技術を使用して評価した。図1Aは、新生仔ラットのNSCにおけるペプチドの用量依存摂取を示している。FITC-ペプチド5μmol/Lでは、新生仔NSCの38.72±9.08%がペプチドを内部移行し、40μmol/LではほとんどのNSCがペプチドを内部移行した(91.07±5.13%)。これらの実験で得られたシグナルが内部移行したFITC-ペプチドに対応するかどうかを評価するために、FACS読み取りの前に0.4%トリパンブルーを添加してFITC-ペプチドの表面結合蛍光を消失させた。結果は、消失させないものと類似のペプチドの用量依存摂取を示し、ペプチドの細胞内蛍光が裏付けられた(図1A)。更に、共焦点顕微鏡によって、ニューロスフェア中に存在するNSC並びに周辺の前駆細胞におけるペプチドの内部移行が裏付けられた(図1B)。
ペプチド摂取、特にペプチドの内部移行のよく知られている2つの主要な経路、エンドサイトーシス及び直接移動(Stewart等、2008年)に関与する分子機構を調べた。ペプチドを添加する前に細胞を4℃で、又はATP欠乏緩衝液中で30分間インキュベートし、更にそれらを30分間インキュベートすることによって、受動又は能動輸送をまず評価した。ペプチド摂取は、4℃又はATP欠乏によって影響を受けなかったので、NSCにおける内部移行は受動機構によって生じることが示された(図3A及び図3B)。能動輸送の使用を更に排除するために、よく知られた各エンドサイトーシス経路の一群の阻害剤並びにエンドサイトーシス機構に関与するシグナル伝達経路の阻害剤(PI3K、MAPK、成長因子受容体)を試験した。図3Cは、これらの阻害剤の存在下で、ペプチド摂取は影響を受けないことを示し、エンドサイトーシスによってNSCに内部移行しているのではないことが裏付けられた。
NSCの特徴は、インビトロにおいてニューロスフェアを形成し、自己再生する能力である。濃度を増加させたペプチドにNSCを5日間曝露したとき、100μmol/L以外では一次ニューロスフェアの数は未処理対照NSC培養物と比較して変化しなかった(図4A)。更に、結果は、1次ニューロスフェアのほとんどがペプチド40μmol/Lで付着性であることを示した。高濃度のペプチドでは、浮遊するニューロスフェアは認められなかった(図4C)。これらのニューロスフェアの画像は、ペプチド濃度の増加に相関した付着細胞の数の増加を示した(図4D)。
これらの観察を完全にするために、PSA-NCAM発現も分析した。このタンパク質の特徴は、細胞-細胞相互作用及び細胞外マトリックスとの細胞相互作用を制限することによって、及び細胞付着を止めることによって神経幹細胞を増殖させることである(Johnson等、2005年)。
ペプチドは10μmより高い濃度で神経膠芽腫細胞において微小管形成を中断させ、結果的に細胞周期を中断することが示された。しかし、このような効果は、ニューロン及び星状膠細胞のような正常細胞では認められなかった(Bocquet等、2009年;Berges等、2012年)。NSCにおけるペプチドの効果を調べるために、濃度を増加させたペプチドの存在下でNSCの細胞周期を、48時間FACS解析を使用することによって分析した。ペプチドが有っても無くても、NSCの細胞周期は変化せず、G1期の細胞が約60〜65%、S期が10%、G2/M期が7%であった。NSCの細胞周期は、微小管ネットワークを中断させる陽性対照として使用したコルヒチン1μg/mLの存在下でのみ影響を受けた(図5A)。これらの結果は、神経膠芽腫細胞とは対照的に、ペプチドがNSCの細胞分割に影響を及ぼさないことを示す(Berges等、2012年)。更に、共焦点顕微鏡によって、ペプチドで6時間インキュベートした後、NSCの微小管ネットワークは影響を受けなかったが、コルヒチンは微小管ネットワークに非常に影響を及ぼすことが示された(図5B)。ペプチドがNSCの生存能にあまり影響を及ぼさないことも示された。細胞は、濃度を増加させたペプチド又はコルヒチンで3日間処理し、アネキシンV-FITC及びPIで染色し、その後フローサイトメトリーによって分析した。生細胞の数は、コルヒチンで処理したとき強く低下したが、細胞生存能はペプチドの存在下では変わらなかった(図5C)。
Claims (15)
- インビトロにおいて化合物又は生物学的物質を神経幹細胞に標的化するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体を含む単離されたアミノ酸配列の使用。
- 前記化合物又は生物学的物質がNFL-TBS40〜63ペプチド又は前記誘導体に直接結合している、請求項1に記載の使用。
- 前記化合物又は生物学的物質がナノカプセル、リポソーム又はミセル中に含有されている、請求項1に記載の使用。
- 前記生物学的に活性のある誘導体が、NFL-TBS40〜63ペプチドの生物学的活性及び特異性を保持しているNFL-TBS40〜63ペプチドのバリアント又は断片である、請求項1から3のいずれか一項に記載の使用。
- インビボにおいて化合物又は生物学的物質を神経幹細胞に標的化するための方法において使用するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体を含む単離されたアミノ酸配列。
- 前記化合物又は生物学的物質がNFL-TBS40〜63ペプチド又は前記誘導体に直接結合している、請求項5に記載の単離されたアミノ酸配列。
- 前記化合物又は生物学的物質がナノカプセル、リポソーム又はミセル中に含有されている、請求項5に記載の単離されたアミノ酸配列。
- 前記生物学的に活性のある誘導体が、NFL-TBS40〜63ペプチドの生物学的活性及び特異性を保持しているNFL-TBS40〜63ペプチドのバリアント又は断片である、請求項5から7のいずれか一項に記載の単離されたアミノ酸配列。
- 前記化合物又は生物学的物質が神経変性障害又は脳癌を治療することを目的とする、請求項5から8のいずれか一項に記載の単離されたアミノ酸配列。
- インビボにおいてNSC分化が有利になるようにする又はNSC自己再生を低下させるために使用するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体を含む単離されたアミノ酸配列。
- 神経変性疾患を治療するために使用するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体を含む単離されたアミノ酸配列。
- インビトロにおいて神経幹細胞を検出するための、NFL-TBS40〜63ペプチド(配列番号1)又はその生物学的に活性のある誘導体の使用。
- 前記NFL-TBS40〜63ペプチド又は前記誘導体が標識されている、請求項12に記載の使用。
- インビトロにおいて生物学的試料を、神経幹細胞の有無について試験するための方法であって、
a.試料の細胞を適切な培地に懸濁すること、
b.NFL-TBS40〜63ペプチド又はその生物学的に活性のある誘導体を、試料の懸濁細胞と混合すること、
c.前記NFL-TBS40〜63ペプチドを含有する細胞の割合を測定することを含み、
前記NFL-TBS40〜63ペプチド又は前記誘導体を含有する細胞の割合が、前記試料中の神経幹細胞の割合に対応する方法。 - 前記NFL-TBS40〜63ペプチド又は前記誘導体が標識されている、請求項14に記載の方法。
