Abstract
Description
− Vx-001(NSCLCガンHLA-A2患者に向けられたTERT由来モノエピトープペプチド)は第IIb相試験中であり、
− Vx-006(これもまたHLA-A2患者に向けられた、普遍的腫瘍抗原TERT、MAGE-A及びHER-2/neuを標的する3エピトープポリペプチド)は第I相試験中である。
こうして、可能性のある各接合部を試験した。表2に示す可能な12のジペプチドを、HLA-B*0702トランスジェニックマウスにおいてインビボで、健常ドナーPBMC培養物においてインビトロで、次の能力:
− ジペプチドが効率的にプロセスされ、各最適化エピトープをロードした細胞を認識することができる特異的CTLを誘導する能力、
− 誘導CTLが腫瘍細胞表面に天然に存在する配列に対応する同族天然型ペプチドを認識する能力
について試験した。
PAProCによれば、両エピトープを生じ得る7T5HN3M(切断部位に二重ダッシュ)を除き、いずれのジペプチドも、プロテアソームにより正確にプロセスされないようである(予測される切断部位にダッシュ。表3)。
次いで、ジペプチドをHLA-B*0702トランスジェニックマウスにおいてインビボで試験した。実施例1に記載するように、組合せ7C1HN3M、7C1T5M、7T5HN3M、7M1HN3M及び7T5M1Mは、応答マウスの数に関して、対応する逆配列より良好な結果であった。7C1M1Mと7M1C1Mとは同じ結果であった。
これらインビボ実験データに基いて、以下の理論上の最適ポリペプチドを設計した:CEA188L9/TERT444A1/MAGE273L9/HER-2/neu246A1。
興味深いことに、この配列は、プロテアソームMAPPP切断予測モデルにより選択された配列とは異なる。なぜならば、7M1T5Mは、インビボで起きることとは反対に、正確にプロセスされないと予測されたからである。
最後に、ポリペプチドSPRLQLSNLAPRRLVQLLGPRALVETLAPKHSDCLA(配列番号23)を、1人のHLA-B*0702健常ドナーのPBMC培養物で試験した(実施例4)。結果は、該ポリペプチドの多重特異的CTL免疫応答を生じる能力を確証する。重要なことに、幾つかの天然型ペプチドが認識された。これら結果は、該ポリペプチドがプロテアソームにより効率的にプロセスされること及び誘導された免疫応答が多重特異的であることを確証する。
本発明によるポリペプチドにおいて、アミノ酸はL-アミノ酸又はD-アミノ酸のいずれであることもできる。
本発明によるポリペプチドは、該ポリペプチドをワクチン接種したHLA-B*0702トランスジェニックマウスの過半数において、CEA188、TERT444、MAGE273及びHER-2/neu246同族天然型ペプチドから選択される少なくとも1つ、好ましくは少なくとも2つのペプチドに対して特異的CD8+ T細胞応答を誘導する。
本発明によるポリペプチドはまた、健常HLA-B*0702ドナーのヒトPBMCを用いるインビトロアッセイにおいて、CEA188、TERT444、MAGE273及びHER-2/neu246天然型ペプチドから選択される少なくとも1つ、好ましくは少なくとも2つのペプチドに対して特異的CD8+ T細胞応答を誘導する。
− 各天然型ペプチドが1匹のHLA-B*0702トランスジェニックマウスで少なくとも1回認識されること;
− 配列番号23のポリペプチドが、健常HLA-B*0702ドナーのヒトPBMCを用いるインビトロアッセイにおいて、CEA188、TERT444、MAGE273及びHER-2/neu246同族天然型ペプチドから選択される少なくとも1つ、通常2以上のペプチドに対して特異的CD8+ T細胞応答を誘導すること;及び
− これら特異的応答が2人以上の健常HLA-B*0702ドナーのPBMCを用いて得られ、各最適化ペプチドがHLA-B*0702健常ドナーのヒトPBMCを用いるインビトロアッセイで少なくとも1回認識されること
も証明した。
1つの更に好適な実施形態において、本発明によるポリペプチドは、上記特性の全てを示す。このことは、下記の実験の部に記載のプロトコール及びアッセイを用いて当業者が容易に試験することができる。
本発明はまた、ペプチド送達ベクター及び上記ポリペプチドを含んでなる複合体に関する。本発明に従って用いることができるペプチド送達ベクターの例は、例えばWO2013/150338、EP1795539及びWO2014/053629として公表された特許出願に記載されたもののような細胞透過性ペプチド、百日咳菌(B. pertussis)のアデニレートシクラーゼ(Fayolleら,1999)、ジフテリア毒素(Fayolleら,1999)、炭疽毒素(Dolingら,1999)、シガトキシンのBサブユニット(Haicheurら,2000)のような細菌性毒素、及びハチ毒PLA2(Babonら,2005)、リポソーム、ビロゾーム(Bungenerら,2002)のような他のベクターなどである。
本発明は、実質的に任意のタイプのガン、例えば副腎皮質ガン、結腸直腸ガン、胆管ガン腫、膀胱ガン、骨ガン、脳及び中枢神経系ガン、乳ガン、子宮頸ガン、子宮内膜ガン、食道ガン、胃ガン、胃腸カルチノイド腫瘍、ホジキン病、非ホジキンリンパ腫、カポジ肉腫、腎臓ガン、頭部頸部ガン、肝臓ガン、肺ガン 中皮腫、卵巣ガン、膵臓ガン、陰茎ガン、下垂体ガン、前立腺ガン、網膜芽種、横紋筋肉腫、皮膚ガン(例えば、メラノーマ、非メラノーマ皮膚ガン)、睾丸ガン、胸腺ガン、甲状腺ガン、膣ガン、外陰ガン及び子宮ガンを治療(又はその再発を防止)するために用いることができる。
本発明によるポリペプチドとアジュバントとを乳化し、ガンのワクチン接種又は治療の必要がある患者に該ポリペプチドをインビボ投与する工程を含んでなるガンのワクチン接種又は治療法、及び上記の操作した樹状細胞又は複合体を個体に投与する工程を含んでなるワクチン接種又は治療法もまた本発明の一部である。
本発明による別の1つのキットは、少なくとも2用量の上記ポリペプチド又は複合体を含んでなる。ペプチドは、既にアジュバントを用いて製剤化されていてもよいし、されていなくてもよい。
ガンワクチン接種は、十分に効率的であるために数回の刺激を必要とするので、本発明によるキットは、3〜50、好ましくは6〜20用量の上記ポリペプチドを含んでなることができる。
上記キットの1つの好適な実施形態によれば、ポリペプチドの各用量は0.5〜10mgのポリペプチドを含んでなる。
実施例
実施例は以下の材料及び方法を用いて行った:
トランスジェニックマウス.HLA-B7 H-2クラス-Iノックアウトマウスは、以前(Rohrlichら,2003)に記載されており、F. Lemonnier(Institut Pasteur,Paris,France)の厚意により提供を受けた。
細胞.HLA-B*0702トランスフェクトヒトT2-B7細胞は、以前(Rohrlichら,2003)に記載されている。細胞は、ペニシリン ストレプトマイシン FCS 20%補充RPMI1640培養培地で生育させた。
ペプチド及びプラスミド.ペプチドはMillegen(Labege,France)又はEurogentec(Seraing,Belgique)が合成した。
HLA-B*0702トランスジェニックマウスにおけるインビボCTL誘導.HLA-B*0702トランスジェニックマウスの尾基部に、不完全フロイントアジュバント(IFA)又はMontanide ISA51VG(Seppic,Castres,France)中に乳化した200μgの最適化ジペプチド又は400μgのVbx-016及びHBVコアT13Lヘルパーペプチド(150μg)を、2週間間隔で2回、皮下ワクチン接種した。
最後のワクチン接種の7日後、脾臓を取り出し、T細胞をフィコール遠心分離により単離し、10μMの天然型又は最適化ペプチドの認識についてエクスビボで試験した。ポジティブコントロールはコンカナバリンAであり、ネガティブコントロールは培地のみであった。全ての条件を4連で試験した。IFNγ産生T細胞を、Diaclone Kit Murine IFNγ ELISpotを用いるELISpotにより定量した。
1)ネガティブコントロールと試験ペプチドとの間に10スポット/106細胞より大きい差が存在し、及び2)この2群間にt検定で統計学的有意差(p<0.05)が存在すれば、免疫応答をポジティブとみなした。
7日目から、培養物を、20IU/ml IL-2及び10ng/ml IL-7、1μg/ml 抗CD40及び500IU/ml IFNγの存在下、ペプチドロードDCで毎週再刺激した。
3回目の再刺激の後、CD8細胞を、20IU/ml IL-2を補充したAIMV中で7日間維持した。培養物をAIMVで一晩飢餓状態にした後、IFNγ ELISpotアッセイで試験した。
IFNγ ELISpotアッセイ.96ウェルの1培養物につき、24ウェルの4プールを集め、CD8細胞を計数し、ウェルあたり50,000のCD8を、10μMの各天然型モノペプチド又は10μMの(該ポリペプチドを構成する)改変モノペプチドをロードした25,000のT2B7と共にELISpotプレートに素早く移した。各条件について8回行った。ポジティブコントロールはフィトヘマグルチニンAであり、ネガティブコントロールは、HLA-B7分子に結合できない無関係のペプチドをロードしたT2B7であった。
各ジペプチドを用いて、HLA-B*0702トランスジェニックマウスに2週間間隔で2回ワクチン接種し、最適化ペプチド及びその同族天然型対応物の両方に対する免疫応答を誘導する能力について試験した。ジペプチドを表2に記載する。各実験において、最良の組合せを灰色で強調する。7C1HN3M、7C1T5M、7T5HN3M、7M1HN3M及び7T5M1Mは、応答マウスの数に関して、逆配列より良好な結果であった。7C1M1Mと7M1C1Mとは同じ結果であった。
ジペプチド実験で決定したVbx-016(配列番号23)が最適であるかどうかを評価するため、HLA-B*0702トランスジェニックマウスの尾基部に、不完全フロイントアジュバント(IFA)又はMontanide ISA51VG(Seppic,Castres,France)に乳化した400μgのVbx-016及び150μgのHBVコアT13Lヘルパーペプチドを、2週間間隔で2回、皮下ワクチン接種をした。
2回のワクチン接種後、全てのマウスが少なくとも1つの天然型ペプチドに応答し、13/14のワクチン接種マウスが2以上の同族天然型ペプチドに応答した。重要なことに、各天然型ペプチドは1匹のワクチン接種マウスで少なくとも1回は認識された。
トランスジェニックマウスで検証されたポリペプチドCEA188L9/TERT444A1/MAGE273L9/HER-2/neu246A1に存在する接合部がヒトで効率的にプロセスされるかどうかを評価するため、ヒトPBMCを、「方法」に記載したプロトコールに従って各ジペプチド(CEA188L9/TERT444A1、TERT444A1/MAGE273L9及びMAGE273L9/HER-2/neu246A1)で刺激した。同族天然型ペプチドの認識を、ジペプチドで刺激したCD8+(本試験用に4プールに分割)に由来する特異的IFNγ産生細胞の測定により評価した。興味対象のペプチドについて得られる平均スポット数が無関係のペプチドで得られる平均スポット数を上回った場合、t検定を行った;t検定値が0.05未満の場合、結果を有意にポジティブであるとみなして、下記表において灰色で強調した。
各ドナーにおいて、CTLは、最適化及び同族天然型ペプチド(及びMAGE-A273L9を共有するペプチドの4つの天然型)の両方を認識することができた。このことから、各接合部がプロテアソームにより良好にプロセスされることが確証される。
最後に、Vbx-016を用いて健常ドナーのヒトPBMCを刺激した。
興味対象のペプチドについて得られる平均スポット数が無関係のペプチドで得られる平均スポット数を上回った場合、t検定を行った;t検定値が0.05未満の場合、結果を有意にポジティブであるとみなして、下記表において灰色で強調した。多重特異的応答が含まれており、幾つかの天然型ペプチドが、誘導されたCTLにより認識された。このことから、該ポリペプチドがプロテアソームにより良好にプロセスされ、誘導された応答が多重特異的であることが確証される。

Claims (15)
- 配列SPRLQLSNLXXXAPRRLVQLLXXXGPRALVETLXXXAPKHSDCLA(配列番号24)を含んでなり、CEA188L9(配列番号7)、TERT444A1(配列番号9)、MAGE273L9(配列番号10)及びHER-2/neu246A1(配列番号8)エピトープがスペーサーXXX(式中、Xは任意のアミノ酸であるか又はアミノ酸なし)で分離されていることを特徴とするポリペプチド。
- 配列SPRLQLSNLAPRRLVQLLGPRALVETLAPKHSDCLA(配列番号23)を含んでなる請求項1に記載のポリペプチド。
- 配列SPRLQLSNLAPRRLVQLLGPRALVETLAPKHSDCLA(配列番号23)からなる請求項1又は2に記載のポリペプチド。
- 小胞体移行シグナル配列をN末端に更に含んでなる請求項1〜3のいずれか1項に記載のポリペプチド。
- ユビキチンをC末端部に更に含んでなる請求項1〜4のいずれか1項に記載のポリペプチド。
- 前記ポリペプチドをワクチン接種したHHDマウスの過半数においてCEA188、HER-2/neu246、TERT444、MAGE273 A6、MAGE273 V6及びMAGE273 I6からなる群より選択される少なくとも2つのエピトープに対するCD8+ T細胞応答を誘導することを特徴とする請求項1〜5のいずれか1項に記載のポリペプチド。
- 健常HLA-B*0702ドナーのヒトPBMCを用いるインビトロアッセイにおいてCEA188、HER-2/neu246、TERT444、MAGE273 A6、MAGE273 V6及びMAGE273 I6からなる群より選択される少なくとも2つのエピトープに対するCD8+ T細胞応答を誘導することを特徴とする請求項1〜6のいずれか1項に記載のポリペプチド。
- 請求項1〜7のいずれか1項に記載のポリペプチドをロードした単離樹状細胞。
- ペプチド送達ベクターと請求項1〜7のいずれか1項に記載のポリペプチドとを含んでなる複合体。
- 請求項1〜7のいずれか1項に記載のポリペプチド及び/又は請求項8に記載の樹状細胞及び/又請求項9に記載の複合体を含んでなる医薬組成物。
- HLA-B*0702表現型を有する患者におけるガン免疫療法に使用するための請求項1〜7のいずれか1項に記載のポリペプチド及び/又は請求項8に記載の樹状細胞及び/又請求項9に記載の複合体。
- 少なくとも1用量の請求項1〜7のいずれか1項に記載のポリペプチドと少なくとも1用量のアジュバントとを含んでなるキット。
- 少なくとも2用量の請求項1〜7のいずれか1項に記載のポリペプチドを含んでなるキット。
- 6〜20用量の請求項1〜7のいずれか1項に記載のポリペプチドを含んでなる請求項12又は13に記載のキット。
- ポリペプチドの各用量が0.5〜10mgのポリペプチドを含んでなる請求項12〜14のいずれか1項に記載のキット。
