Abstract
Description
いくつかの実施形態において、加熱冷却モジュールは、熱電加熱冷却(TEHC)装置を含む。1つまたは複数のTEHC装置は、モジュールに一体化され得る。他の実施形態において、加熱冷却モジュールは、温度センサーをさらに備える。例示的な温度センサーは、測温抵抗体(RTD)、熱電対、サーモパイル、およびサーミスターである。いくつかの実施形態において、温度センサーは、RTDである。他の実施形態において、温度センサーは、より高い分解能と、より小さな温度範囲と、より大きな経時ドリフトとをもつサーミスターである。いくつかの実施形態において、加熱冷却装置のサーミスターは、電子アナログ・デジタル変換器(ADC:analog−to−digital convertor)に結合している。
本明細書で使用する「フローセル」という用語は、マイクロアレイ式検出装置を指す。いくつかの実施形態において、フローセルは、試料入口と試料出口とその中に位置するマイクロアレイとを含む反応チャンバを備える。いくつかの実施形態において、反応チャンバは、PCRなどの増幅反応と、同じチャンバ内でのハイブリダイゼーション反応との両方を実行できる増幅ハイブリダイゼーション複合反応チャンバである。いくつかの実施形態において、フローセルは、反応チャンバと流体連通した廃棄チャンバをさらに備える。いくつかの実施形態において、反応チャンバは、親水性物質で被覆されており、反応チャンバの完全な充填と反応チャンバから廃棄チャンバへの流体の流れとを促進する位置にある親水性表面をもつ。親水性表面は、試料入口から反応チャンバに液体が入るときに液体に接触し、マイクロアレイチャンバの完全な充填を可能にする。特定の実施形態において、反応チャンバは、様々な幅の延びたチャンネルの形状をもち、廃棄チャンバに直接接続されている。他の実施形態において、マイクロアレイチャンバは、廃棄チャンネルを通して廃棄チャンバに接続されている。
いくつかの実施形態において、反応チャンバ110が加熱冷却装置の伝熱面202上に位置するように、フローセル100が、加熱冷却モジュール200上に配置される。図4を参照されたい。いくつかの実施形態において加熱冷却モジュール200は、移動システムに搭載される。いくつかの実施形態において加熱冷却装置の伝熱面は光を吸収する。光吸収を達成する例示的な方法は、伝熱面202を黒色に塗装すること、黒色陽極酸化すること、または電気めっきにより黒色クロムで伝熱面202を被覆することを含む。光吸収は、マイクロアレイを像形成することと干渉し得る散乱を低減する。いくつかの実施形態において、熱サイクルは、像形成前に起こる。いくつかの実施形態において熱サイクルは、像形成と同時に起こる。
いくつかの実施形態において加熱冷却モジュールは、所望の温度への接近を加速する手段として、変化状態中に設定点温度が変化するように制御される。いくつかの実施形態において設定点は、所望の温度の上方−5℃〜5℃の範囲内に人為的に設定される。
薄膜RTD(Minco RTD Model S39)が、反応チャンバ(厚さ0.5mm)に組み込まれ、熱伝導ペーストで充填され、さらに、フラットブロックQuantaサーモサイクラー上に配置される。1つの反応チャンバが、厚さ1インチ(約2.54cm)のスチロフォーム断熱層を含み、他方は、断熱材を含まない。2つの反応チャンバが、順次、サーモサイクラー上に導入される。熱サイクルプロトコルは、88℃で60秒間の後、55℃で60秒間の30サイクルである。変性温度のみが描かれている。温度測定値は、20秒の移動平均を表す。図9からわかるように、反応チャンバが断熱されていない場合、88℃の設定点から1℃の温度オフセットがあり得る。
反応チャンバは、QuestarTM基材と、0.5mmの両面加圧粘着スペーサテープ(double-sided pressure sensitive adhesive spacer tape)と、被覆薄膜とを備える。反応チャンバの容量は、約50μLで充填される。反応チャンバは、入口孔と出口孔とを含む。
実施例2で説明される加熱冷却モジュール200は、反応チャンバ上に組立体を降下させるために使用される線形アクチュエータを備える機械装置に搭載されている(図5参照)。組立体は、反応チャンバ上に降下されるときに圧縮される4つのばねを含む。
N−アセチルシステイン、水酸化ナトリウムが消化された痰が、107cfu/mLのH37Ra細胞で修正された。均一化と溶解とは、米国特許第8,399,190号(その全体が参照により本明細書に組み込まれる)で説明される装置を使用して実現された。DNAの抽出は、米国特許第8,236,553号と第8,574,923号と(全体として参照により本明細書に組み込まれる)において説明される装置と方法とを使用して実現された。

- 薄膜熱電加熱冷却装置を含む加熱冷却モジュールと、
伝熱外面と内面におけるマイクロアレイとを有する増幅ハイブリダイゼーション複合反応チャンバを含む取り外し可能な検査モジュールと、
を備え、
前記熱電加熱冷却装置が、前記反応チャンバの前記伝熱外面と接触するように適応された伝熱面をもつ、
分子検査装置。 - 前記薄膜熱電加熱冷却装置が、ペルティエ素子である、
請求項1に記載の分子検査装置。 - 前記ペルティエ素子が、セラミックペルティエ素子である、

前記PCR装置が、伝熱面をもつ薄膜熱電加熱冷却装置を含む加熱冷却モジュールと、前記伝熱面の温度を調節するプログラム可能制御モジュールとを備える、
配置することと、
(b)前記薄膜熱電加熱冷却装置の前記伝熱面を前記反応チャンバの前記伝熱外面に接触させることと、
(c)前記制御モジュールに記憶されたPCRプログラムに基づいて、前記伝熱面を通して前記反応チャンバを加熱および冷却することによりPCRを遂行することと、
を含む、反応チャンバにおけるマイクロアレイでポリメラーゼ連鎖反応(PCR)を実施する方法。
