Description
本願は、インターロイキン−6(以下、IL−6と称する)に対する親和性を有する改変ポリペプチド及び抗体又はその抗原結合フラグメントを含む複合体に関し、ここで、前記IL−6に対する親和性を有する改変ポリペプチドは、配列 EEX3X4AWX7EIHX11LPNLX16X17X18QX20X21AFIX25X26LX28X29を含む改変されたポリペプチドのクラスに属する。

炎症性リウマチ疾患では、IL−6誘発効果の競合的阻害が非常に有効であることが示されている。同様に、高親和性抗ヒトTNFモノクローナル抗体アダリムマブは、慢性関節リウマチを含むTNF誘発性炎症適応症の治療に臨床的に使用される。これらの2つの抗炎症効果が1つの分子に組み合わされ得るかどうかを調べるために、IL−6標的Z変異体及びTNF標的抗体アダリムマブと同じ結合特異性を有する抗体「Ada」に基づく異なる融合分子を設計し、以下の実施例に記載するように、インビトロ及びインビボで産生及び特性を決定した。
IL−6結合Z変異体の選択及びELISAスクリーニング
標的タンパク質、ヒト及びマウスIL−6のビオチン化:No−Weigh EZ−Link Sulfo−NHS−LC(Thermo Scientific、カタログ番号21327)を用いて、製造業者の推奨に従って12×モル過剰で添加して、hIL−6及びmIL−6(Peprotech、各カタログ番号200−06及び216−16)をビオチン化した。反応は室温(RT)で30分間行った。次に、Slide−a−lyzer透析カセット(Thermo Scientific、カタログ番号66333,3,500 MWCO)を用いて、製造元の指示に従って、リン酸緩衝生理食塩水(PBS、10mMリン酸、137mM NaCl、2.68mM KCl、pH7.4)に緩衝液を交換する。
IL−6結合Z変異体のファージディスプレイ選択:個々のクローンを、ビオチン化hIL−6及びmIL−6に対する4サイクルのファージディスプレイ選択の後に調製した。
IL−6結合Z変異体の産生及びインビトロ特性決定
Z変異体のサブクローニング:13のIL−6結合Z変異体のDNA、Z06777(配列番号1503)、Z06779(配列番号1504)、Z06789(配列番号1505)、Z06791(配列番号1506)、Z06792(配列番号1507)、Z06799(配列番号1508)、Z06802(配列番号1509)、Z06805(配列番号1510)、Z06809(配列番号1511)、Z06814(配列番号1512)、Z06829(配列番号1513)、Z06834(配列番号1514)、Z06844(配列番号1515)を、ライブラリーベクターpAY02592から増幅した。標準的な分子生物学技術(別の標的に結合するZ変異体について、国際公開第2009/077175号に本質的に記載されているように)を用いて、N末端His6タグを有する単量体Z変異体分子の構築のサブクローニング戦略を適用した。Z遺伝子フラグメントを発現ベクターpAY01448にサブクローニングして、コードされた配列MGSSHHHHHHLQ−[Z#####]−VDを得た。
培養及び精製:N末端His6タグを用いて構築された13のIL−6結合Z変異体(配列番号1503〜1515)を大腸菌で産生させた。280nmでの吸光度を測定することによって分光光度的に決定した、約2〜5gの細菌ペレット由来のIMAC精製タンパク質の量は、異なるIL−6結合Z変異体について約10mg〜20mgの範囲であった。ABD変異体PP013(配列番号1554)に融合された5つのZ変異体、約2.5gの細菌ペレットから2mg〜12mgが得られた。各最終タンパク質調製物のSDS−PAGE分析は、これらが主にIL−6結合Z変異体を含有することを示した。各IL−6結合Z変異体の正確な同一性及び分子量を、HPLC−MS分析によって確認した。
IL−6結合Z変異体の第一の成熟したライブラリーの設計及び構築
ライブラリー設計:ライブラリーは、実施例1及び2に記載のIL−6結合Z変異体の配列の選択に基づいた。新しいライブラリーでは、Z分子骨格中の12の可変位置は、特定のアミノ酸残基にバイアスを有し、1つの位置は、配列番号1503〜1551に定義されたZ変異体配列に基づく戦略に従って、一定に保たれた。スプリットプール合成を使用して、147bpのDNAリンカーを生成し、Z変異体アミノ酸配列の部分的に無作為化されたヘリックス1及び2をコードした。従って、制限部位XhoI及びSacIに隣接する5’−AA ATA AAT CTC GAG GTA GAT GCC AAA TAC GCC AAA GAA NNN NNN NNN GCG TGG NNN GAG ATC NNN NNN CTG CCT AAC CTC ACC NNN NNN CAA NNN NNN GCC TTC ATC NNN AAA TTA NNN GAT GAC CCA AGC CAG AGC TCA TTA TTT A−3’(配列番号1563;NNNと表示したランダム化コドン)は、DNA2.0(Menlo Park、CA、USA)に注文した。Z分子骨格における12の可変Z位置(9,10,11,14,17,18,24,25,27,28,32及び35)を含む新しいライブラリー中のアミノ酸残基の理論的分布を表6に示す。結果として得られる理論上のライブラリーサイズは、3.6×109変異体である。
ライブラリー構築:新しいライブラリーは、結合特性が確認されたIL−6結合Z変異体のセット(実施例1及び2)に基づいて設計された。設計されたライブラリーの理論上のサイズは、3.6×109個のZ変異体であった。大腸菌ER2738細胞への形質転換後の滴定によって決定されたライブラリーの実際のサイズは、3.5×109個の形質転換体であった。
第一の成熟したライブラリーからのZ変異体の選択、スクリーニング及び特徴付け
成熟IL−6結合Z変異体のファージディスプレイ選択:標的タンパク質hIL−6(R&D Systems、カタログ番号206−IL/CF)及びmIL−6(Abnova、カタログ番号P4346116)は、実施例1に記載のようにビオチン化した。実施例3に記載のZ変異体分子の新規ライブラリーを用いたファージディスプレイ選択は、以下の例外を除いて、本質的には実施例1に記載されるように、hIL−6及びmIL−6に対して4サイクルで実施した。選択時に、ウシ胎仔血清(FCS、Gibco、カタログ番号10108−165)及びヒト血清アルブミン(HSA、Albucult、Novozymes、カタログ番号230−005)を選択緩衝液に最終濃度10%及び1.5μMになるように添加した。選択に用いた全ての試験管及びビーズを、3%BSAを補充した0.1%のPBSTで予備ブロッキングした。サイクル1Aでは、SA−ビーズとファージストックのインキュベーションによって予備選択工程を実施した。全てのトラックに対し、サイクル1で選択容量は2mlであった。標的結合ファージ捕捉のために、4μgのビオチン化hIL−6又はmIL−6あたり1mgのビーズを使用した。
成熟したIL−6結合Z変異体のファージディスプレイ選択:各4サイクルを含む完全に並列の12トラックで選択した。異なる選択トラックは、標的濃度、標的型(hIL−6又はmIL−6)、選択時間、洗浄条件及び溶出緩衝液のpHを変えた。
IL−6結合Z変異体の第2成熟ライブラリーの設計及び構築
ライブラリー設計:ライブラリーは、実施例4に記載のヒトIL−6結合Z変異体の配列の選択に主に基づいた。新しいライブラリーでは、Z分子骨格中の13の可変位置は、主に第1の成熟からのZ変異体、すなわち、配列番号7、配列番号15〜89及び配列番号151〜871で定義された配列に基づく戦略に従って、特定のアミノ酸残基にバイアスを有した。無作為の二本鎖リンカーは、後に構築される二本鎖DNAの連結及び制限を用いて、トリヌクレオチド構築阻害の無作為セットの組み込みを可能にするColibra(商標)技術によって生成された。二本鎖DNAのライブラリーであり、制限部位XhoI及びSacIに隣接するZ変異体アミノ酸配列の部分的にランダム化されたヘリックス1及び2をコードする、5’−AA ATA AAT CTC GAG GTA GAT GCC AAA TAC GCC AAA GAA NNN NNN NNN GCT NNN NNN GAG ATC NNN NNN CTG CCG AAC CTG ACC NNN NNN CAG NNN NNN GCC TTC ATC NNN AAA TTA NNN GAT GAC CCA AGC CAG AGC TCA TTA TTT A−3’(配列番号1564;ランダム化コドンをNNNと表示する)をIsogenica(Essex、UK)から注文された。Z分子骨格中に8つの可変アミノ酸位置(10,11,14,18,24,25,27及び32)及び5つの定常アミノ酸位置(9,13,17,28及び35)を含む、新規ライブラリー中のアミノ酸残基の理論的分布は、表10に示す。得られた理論上のライブラリーサイズは、2.6×107変異体であった。
ライブラリー構築及びファージストック調製:新規ライブラリーは、結合特性が確認されたIL−6結合Z変異体のセット(実施例4)に基づいて設計された。設計されたライブラリーの理論上のサイズは、2.6×107個の変異体であった。大腸菌ER2738細胞への形質転換後の滴定によって決定されたライブラリーの実際のサイズは、1.8×109個の形質転換体であった。
第2の成熟ライブラリーからのZ変異体の選択、スクリーニング及び特徴付け
成熟したIL−6結合Z変異体の第2のファージディスプレイ選択:実施例4に記載のように、標的タンパク質hIL−6をビオチン化した。実施例5に記載のZ変異体分子の第2の成熟化ライブラリーを用いた、ファージディスプレイ選択は、以下の例外を除いて、本質的に実施例4に記載の通り、hIL−6に対して実施した。全てのトラックに対し、サイクル1で選択容量は4mlであった。サイクル2では、選択トラック1−2及び1−3を1本の共通のチューブで取り扱い、最後の1分間の洗浄後まで分離せず、その後それらを別々に処理した。サイクル4でも、選択トラック1−1−1−1から1−1−1−3、1−1−1−4から1−1−1−6、1−1−2−1から1−1−2−3及び1−1−2−4から1−1−2−6それぞれを共通のチューブに入れ、最後の1分間の洗浄後に3本の別々のチューブに分割した後、表11に要約した異なる洗浄戦略を用いて別々に処理した。実施例1に記載のように、グリシン−HCl、pH2.2を用いて結合したファージ粒子を溶出させた。選択サイクル間のファージ粒子の増幅は、基本的に実施例1に記載したように行った。
成熟したIL−6結合Z変異体の第2のファージディスプレイ選択:各トラックは4つのサイクルを含み、合計17の並行トラックで選択を実施した。選択トラックは、表11に概説されているように、標的濃度、選択時間及び洗浄条件が異なっていた。
IL−6結合Z変異体のサブセットのサブクローニング、生産及び特性決定
Z変異体の発現ベクターへのサブクローニング:14のIL−6結合Z変異体、Z11632(配列番号7)、Z14630(配列番号6)、Z14700(配列番号8)、Z14712(配列番号9)、Z14861(配列番号4)、Z14862(配列番号10)、Z14976(配列番号1)、Z14984(配列番号5)、Z15015(配列番号2)、Z15036(配列番号11)、Z15110(配列番号12)、Z15122(配列番号3)、Z15126(配列番号13)及びZ15142(配列番号14)のDNAは、ベクターpAY02592から増幅した。サブクローニングを実施例2に記載のように実施した。Z遺伝子フラグメントを発現ベクターpAY01448にサブクローニングし、コードされた配列MGSSHHHHHHLQ−[Z#####]−VDを得た。
ProteOn動態分析:6つのHis6タグ化IL−6結合Z変異体とヒトIL−6との相互作用を、異なる固定化Z変異体を含む表面上で様々な濃度のhIL−6を注入することによりProteOn装置で分析した。表面のリガンド固定化レベルは、それぞれ100〜220RUであった。1:1相互作用モデルを用いたhIL−6のZ変異体への結合に関する速度論的パラメータ(KD、ka(kon)及びkd(koff))の概要を表13に示す。
ABDとの融合におけるIL−6結合Z変異体のインビボ活性
IL−6標的Z変異体Z06814(配列番号1512)及び無関係の標的に結合する対照Z変異体Z04726(配列番号1535)をクローニングし、実施例2に記載のように、ABD変異体PP013(配列番号1554)と融合させた。Balb/cマウスの4つの群(n=8)に、5μg/kgのhIL−6(R&D Systems)の腹腔内(ip)投与9時間後に、Z06814−ABDを様々な用量(0,0.025,2.5又は25mg/kg体重)を皮下(sc)に注射した。第5群のマウス(n=8)は、25mg/kgのZ04726−ABDを受けた。マウスの2つのさらなる対照群は、それぞれPBS(n=4)及び25mg/kgのZ06814−ABD(n=8)を受けたが、その後IL−6注射はしなかった。20時間後、血液を心穿刺により採取し、血清を採取した。
Z変異体Z06814(配列番号1512)の抗関節炎効果を、IL−6誘発血清アミロイド−A(SAA)タンパク質放出をマウスモデルを用いて、インビボで評価した。4群のマウスに、5μg/kg体重のhIL−6を注射する9時間前に、0、0.025、2.5又は25mg/kg体重のIL−6結合Z06814−ABD融合タンパク質又は25mg/kgの対照Z04726−ABD融合タンパク質を与えた。さらに22時間後、SAAタンパク質のレベルを測定し、異なる群間で比較した。Z06814−ABD無し又は25mg/kgの対照Z04726−ABD融合物を投与していない動物では、血清中のSAAタンパク質レベルは約500〜600μg/mlのレベルまで増加した。PBSのみ(及びhIL−6なし)を与えた対照動物は、16〜64μg/mlの範囲のレベルを示した。Z06814−ABDを与えられた動物では、有意に低いSAAタンパク質レベルが用量依存的に測定された(図7)。最も高い用量のZ06814−ABD(25mg/kg体重)を与えられた群では、SAAタンパク質レベルは、hIL−6注射されていない動物ほど低かった。
複合体及び対照ポリペプチドの産生
抗体及び複合体の生成:IL−6及びTNFを標的とする4つの異なる複合体、並びにTNFに対する親和性を有する抗体を構築した。市販のモノクローナル抗体アダリムマブと同じCDR配列及び特異性を有する「Ada」と称する抗体を、重鎖(HC)及び軽鎖(LC)配列HCAda(配列番号1557)及びLCAda(配列番号1558)を用いて構築した。VDの代わりにアミノ酸残基AEで始まり、追加のC末端アミノ酸残基VDを有する、IL−6標的Z変異体Z06814(配列番号1512)部分は、柔軟な15残基(GGGGS)3を介し、HCAda又はLCAdaのN末端に遺伝的に融合し、それぞれZ06814−HCAda及びZ06814−LCAdaと表される複合体、又は同じ鎖のC末端に融合し、複合体HCAda−Z06814及びLCAda−Z06814が得られる。遺伝子合成、クローニング、CHO細胞における一過性遺伝子発現による産生及びプロテインAアフィニティークロマトグラフィーを用いる精製は、Evitria AG(スイス)によって実施した。複合体及びAdaの純度は、非還元条件下及び還元条件下の両方でSDS−PAGEにより分析した。15μgの各変異体を12ウェル4−12%NuPAGE(登録商標)ゲル(Life Technologies)にロードした。
複合体構築物の作製:4つの異なる産生複合体の各設計の模式図を図8に示す。作製された構築物を含むサンプルの非還元条件下、SDS−PAGEによる分析は、それらが高純度であることを示した。還元条件下でのSDS−PAGE分析は、Z変異体の抗体鎖への融合部位と一致する個々のサブユニットの予想されるサイズを確認した(図9)。

複合体Z06814−HCAda及びLCAda−Z06814、並びにAda及びHis6−Z06814の各10μg/ml溶液、すなわち各融合パートナーのみを10mM NaAc pH4.5緩衝液中で調製し、アミンカップリング化学反応を介して、Proteon GLCチップ(Bio−Rad)上に該タンパク質を固定するために使用した。得られた固定化レベルは、2つの複合体及びAdaについては〜1400〜2000RUであり、His6−Z06814については100〜200RUであった。一連の50nM、10nM、2nM、0.4nM及び0.08nM濃度のrhIL−6(R&D Systems)を注入し、応答を記録した。この実験を3回繰り返した。
2つの標的タンパク質IL−6及びTNFに対する親和性は、複合体Z06814−HCAda及びLCAda−Z06814並びに個々のサブユニットAda及びZ06814単独について測定した。IL−6及びTNFとの相互作用の動態パラメータ((KD、ka(kon)及びkd(koff))をそれぞれ表16及び17に要約する。
インビトロでの生物学的活性の分析
TF−1細胞を、10%FCS(Gibco)、Pen−Strep(Lonza)及び2ng/ml rhGM−CSF(R&D Systems)を添加したL−glut(Lonza)を含むRPMI1640中で培養した。使用前に、細胞をrhGM−CSFの非存在下でRPMI1640中にて2回洗浄した。次いで細胞を計数し、1ウェルあたり4×104細胞の密度で96ウェル平底プレートに分注した。別々のプレートにおいて、複合体Z06814−HCAda、Z06814−LCAda、HCAda−Z06814及びLCAda−Z06814の連続希釈物(400〜0.004nM又は10〜0.00001nMのいずれかの範囲);N末端His6タグを有する陰性対照ポリペプチドZ04726(配列番号1553);陽性対照ポリペプチドZ06814−ABD及びAda、及びAdaとZ06814−ABDとの混合物を調製した。サンプルを、0.099nM rhIL−6(R&D Systems、カタログ番号206−IL/CF)、0.023nM rhTNF(R&D Systems、カタログ番号210−TA)又は両方のサイトカインの組み合わせのいずれかの存在下でインキュベートした。プレ混合物を、TF−1細胞を含むウェルに移し、37℃、加湿5%CO2雰囲気下で72時間インキュベートした。インキュベーションの最後の4時間の間に、10μlのCCK−8(Fluka、Sigma Aldrich)をウェルごとに添加して、増殖細胞の数を決定した。吸光度は、マイクロプレートリーダー(Victor3、Perkin Elmer)を用いて450nm(Abs450)で測定した。細胞増殖に関するデータを、4パラメータの用量反応曲線に対する非線形回帰によって評価し、GraphPadPrismプログラムを用いて半最大阻害濃度(IC50)を決定した。阻害分子によるTF−1細胞のIL−6/TNF依存増殖の阻害は、Abs450から、細胞を含むがIL−6を含んでいない対照ウェルを除算したものであった。
第1セットの実験では、TF−1細胞のhIL−6又はTNF依存性増殖のいずれかを阻害する4つの異なる複合体Z06814−HCAda、Z06814−LCAda、HCAda−Z06814及びLCAda−Z06814の能力を試験した。対照として、Ada及びZ06814−ABDのみを含んだ。IL−6阻害試験では、4つの複合体並びにZ06814−ABD構築物の全てが、0.1〜1nMの範囲で推定されるIC50値を有する増殖阻害能力を示した。予想通り、このアッセイでは、TNF標的Adaは効果を示さなかった(図10A)。
インビボにおける生物学的活性の解析
7群のBalb/cマウス(n=6)に、70、7、0.7又は0(ビヒクル)mg/kgのZ06814−HCAda又はAdaを腹腔内(i.p.)に注射した。注射は、rhIL−6及びrhTNFの混合物(それぞれ2.5μg/kg)で腹腔内注射の9時間前に投与した。16時間後、眼窩穿刺により採血し、血清を採取した。採取した血清を製造者の指示に従ってELISA(Tridelta、カタログ番号KMA0021)によりマウスSAA含量を評価した。簡潔述べると、希釈した血清サンプルを抗SAA−HRPとともにSAAでプレコートしたプレートに添加した。プレートを1時間インキュベートし、次いで4回洗浄した。TMB基質を加え、20分後に停止溶液で反応を停止させた。吸光度は、マイクロプレートリーダー(Victor3、Perkin Elmer)を用いて450nmで測定した。
IL−6誘発血清アミロイド−A(SAA)タンパク質放出のマウスモデルを用いて、Z06814−HCAdaの抗関節炎効果をインビボで評価した。この結果は、高用量(70mg/kg)のTNF阻害抗体Ada単独では観察された血清SAAレベルが50%低下したが、同じ用量のZ06814−HCAdaは、使用した試験キットの定量限界(LOQ)よりも低い濃度まで血清SAAレベルを減少したことを示す(図11)。このことは、1分子中のIL−6シグナル伝達阻害ポリペプチドZ06814とTNF阻害Ada抗体との組み合わせが、二重作用が可能な複合体をもたらし、従ってIL−6及びTNF曝露の両方に対して治療軽減をもたらすことを示唆する。
IL−6及びTNFを標的とする成熟複合体の産生及び結合活性
IL−6及びTNF標的複合体の産生:IL−6及びTNFを標的とする3つの異なる複合体を構築した。市販のモノクローナル抗体アダリムマブと同じCDR配列及び特異性を有する「Ada2」と称される抗体を、重鎖(HC)及び軽鎖(LC)配列HC2Ada(配列番号1559)及びLCAda(配列番号1559)を用いて構築した。VDの代わりにアミノ酸残基AEで始まるIL−6標的Z変異体Z14976(配列番号1)部分は、柔軟な5残基(GGGGS)又は15残基(GGGGS)3リンカーをLCAdaのN末端へ介して遺伝的に融合して、それぞれZ14976−(GGGGS)−LCAda及びZ14976−(GGGGS)3−LCAdaと表示される複合体が得られる、又は、LCAdaのC末端に対して柔軟な5残基(GGGGS)リンカーを介して遺伝的に融合して、LCAda−(GGGGS)−Z14976と表示される複合体が得られる。重鎖及びZ変異体融合軽鎖構築物をそれぞれpcDNA3.1(Invitrogen)又はpOptiVEC(Invitrogen)にクローニングした。CHO−Sシステム(Invitrogen)を用いてIL−6及びTNF標的化複合体を生成し、プロテインAクロマトグラフィー(GE Healthcare、カタログ番号17−1279−01)を用いて精製した。複合体の純度を非還元(6%アクリルアミドゲル)及び還元条件(12%アクリルアミドゲル)の両方でSDS−PAGEにより分析した。
IL−6及びTNFを標的とする複合体の産生:非還元及び還元条件下で精製した複合体のSDSページ分析を図12に示す。還元条件下で行った分析(図12B)は、個々のサブユニットの予想されるサイズを確認した。

- TNF依存性シグナル伝達を阻害することができる、請求項7〜11のいずれか1項に記載の複合体。
- 前記TNF依存性シグナル伝達を阻害する半最大阻害濃度(IC50)が、最大で1×10-7M、例えば最大で1×10-8M、例えば最大で1×10-9M、例えば最大で1×10-10M、例えば最大で1×10-11Mである、請求項12に記載の複合体。

- 前記複合体又は組成物が、IL−6機能及び追加の抗原、例えばインビボでの免疫系の疾患又は障害に関連する抗原、又は癌に関連する抗原の機能を調節する、請求項20に記載の使用のための複合体又は組成物。
- IL−6関連障害、例えばIL−6及びTNFに関連する障害、例えば自己免疫疾患、炎症性疾患、癌及び腫瘍性疾患からなる群から選択される、例えば慢性炎症性疾患及び炎症誘発性癌から選択されるIL−6関連障害である、請求項20又は21に記載の使用のための複合体又は組成物。
- 前記障害が、慢性関節リウマチ;若年性関節リウマチ;若年性特発性関節炎;全身性若年性特発性関節炎;血管炎;乾癬性関節炎;乾癬;強直性脊椎炎;慢性炎症性腸疾患、例えばクローン病及び潰瘍性大腸炎;グレーブス病;ベーチェット病;ブドウ膜炎;巨細胞性動脈炎;多発性硬化症;全身性硬化症;全身性エリテマトーデス;多発性筋炎;リウマチ性多発性筋痛症;喘息;慢性閉塞性肺疾患;再発性多発軟骨症;膵炎;腹膜炎;腎炎;川崎病、シェーグレン症候群及び成人スティル病、からなる群から選択される、請求項22に記載の使用のための複合体又は組成物。
