完全ヒトモノクローナル抗体等のモノクローナル抗体の作成方法は当技術分野で公知である。そのような公知の任意の方法を本発明と関連して用いてヒトTie2に特異的に結合するヒト抗体を作ることができる。

比較目的のために、以下に説明する様々な実験に例示的なコントロール構築物(抗Tie2抗体)を含めた。コントロールIと称するす抗体は、米国特許第6,376,653号に記載の「12H8」のドメインに対応するアミノ酸配列を有するマウスの重鎖および軽鎖の可変ドメイン備えるキメラ抗Tie2抗体であった。この抗体の定常ドメインはヒトIgG4であった。
ヒトTie2に対するヒト抗体の作成
標準的方法(例えば米国特許第6,596,541号参照)に従って、ヒトTie2抗原を備えたVELOCIMMUNE(登録商標)マウスを免疫化することにより、様々なヒト抗Tie2抗体を作成した。この技術を用いて、様々な抗Tie2抗体を得た;この

ヒトモノクローナル抗Tie2抗体の結合親和性および動力学的定数を得た表面プラズモン共鳴
ヒトモノクローナル抗Tie2抗体の結合親和性および動力学的定数を表面プラズモン共鳴によって25℃および37℃で測定した(表1〜4)。Biacore2000またはT200機器上で測定を実施した。
期(T1/2)を、KD(M)=kd/Ka;およびT1/2(分)=(ln2/(60*kd))として動力学的速度定数から算出した。表1〜2に示すように、試験した両方の温度で様々な抗体がhTie2への高い親和性結合を示した。更に、H1M2055N、H1H2332B、H1H2337B、H1H2340BおよびH4H2055Nは、マウスおよびラットTie2への有意な結合を示した(表3〜4)。
Luminexビーズを用いるTIe2mAbのエピトープマッピング
抗Tie2抗体に結合するドメインを決定するために、様々なhTie2受容体細胞外ドメイン欠失構築物をluminex xMAPビーズに共有結合的に連結させた(107個のビーズ当たり各タンパク質10μg/ml)。構築物を図1に図示し、以下のよう
に命名した:hTie2(Ig1−Ig2−EGF)(配列番号7);hTie2(Ig2−EGF)(配列番号8);hTie2(EGF)(配列番号9);hTie2(Ig3−FN)(配列番号10);およびhTie2(FN)(配列番号11)。また、本実施例において、完全長hTie2−mFc(配列番号4)およびhTie1−hFc(配列番号12)の細胞外ドメイン構築物を試験した。
温(RT)で90分にわたってインキュベートし、または振とうしつつ4℃で終夜インキュベートした。ビーズを洗浄緩衝液(PBS+0.05%Tween20)で3回洗浄してPE(フィコエリトリン)標識抗ヒトカッパFab二次抗体またはPE標識抗マウスFab二次抗体を含有する結合緩衝液100μl中に再懸濁させ、振とうしつつ45分にわたってRTでインキュベートした。サンプルを2回以上洗浄し、Luminex L20
0またはFLEXMAP3D機器を用いて各ビーズに関して結合シグナル(MFI)を測定した。ヒトTie2に連結したビーズをポジティブ・コントロールとして用い、ヒトTie1に連結したビーズを用いてファミリー交差反応性を測定した。一般的に、500単位より大きいMFIシグナルは有意な結合を示す。結果を表5にまとめる。
H/D交換によるTie2へのH4H2055N結合のエピトープマッピング
実験を行なうことにより、H4H2055Nが相互作用するTie2のアミノ酸残基をより正確に限定した(H4H2055Nは、ハイブリドーマPTA−12295から生産した抗体H1M2055Nの完全ヒトIgG4バージョンである)。この目的のために、H/D交換エピトープマッピングを行なった。H/D交換法の概要は例えばEhring(1999)Analytical Biochemistry267(2):252〜259ページ;ならびにEngenおよびSmith(2001)Anal.Chem.73:256A〜265Aページに記載されている。
BS緩衝液中で5分また10分にわたって重水素化し、次いで2分にわたってインキュベートしてH4H2055Nビーズに結合させた。Tie2結合ビーズをPBS水性緩衝液(H2Oで調製)で洗浄し、PBS緩衝液中でオン−交換時間の半分にわたってインキュ
ベートした。オフ−交換後、氷冷した低pHのTFA溶液により、結合したTie2をビーズから溶出させた。次いで、溶出したTie2を、固定したペプシン(Thermo Scientific)により5分にわたって消化した。得られたペプチドをZipTip(登録商標)クロマトグラフィー・ピペット・チップを用いて脱塩し、UltrafleXtremeマトリックス支援レーザー脱離イオン化飛行時間型(MALDI−TOF)質量分析(MS)により速やかに分析した。
程(オフ−交換、ペプシン消化およびMS分析)を「オン−溶液/オフ−ビーズ」の手順に関して記載されているように行なった。検出したペプチド全ての重心値または平均質量対電荷比(m/z)を算出し、これら2組の実験の間で比較した。
7のアミノ酸139〜152および166〜175)は、同種のリガンドであるアンジオポエチン2に直接接触するTie2のアミノ酸領域内に位置することにも留意すべきである。従って、本実施例は、抗体H4H2055Nが、主に(Ig1ドメインのC末端部内での1個または2個の潜在的アミノ酸接触を有する)ヒトTie2のIg2ドメイン内の不連続エピトープに結合し、本明細書の実施例5および6に示すように、これらの領域への結合は、Tie2とAng2との間の相互作用を遮断するH4H2055Nの能力と関連することを示す。
Tie2とアンジオポエチンとの間の相互作用を遮断する抗Tie2抗体の能力の評価
Tie2がアンジオポエチン(Ang1、Ang2、Ang3およびAng4)と相互作用することが知られている。従って、Ang2に結合するTie2を遮断する抗Tie2抗体の能力を評価するために、最初の組の実験を行なった。この最初の組の実験では定量的遮断免疫アッセイを利用した。簡潔に言えば、6xHis(配列番号2)に融合した0.8nMのビオチン化ecto−Tie2溶液を段階希釈により約50nM〜0nMの範囲で抗Tie2抗体と前混合した。室温で1時間にわたりインキュベートした後、プレート被覆Bow−Ang2に結合したTie2ーHisの量をサンドイッチELISAにより測定した。Bow−Ang2(配列番号13)、即ち両末端でAng2の1つのFDドメインに挟まれたhIgG1のFcドメインを含むAng2構築物を96ウェルのマイクロタイター・プレート上において2μg/mlで被覆し、BSAで遮断した。HRPが接合したストレプトアビジンを用いて、プレートに結合したビオチン−Tie2−Hisを検出し、BD OptEIA(商標)(BD Biosciences Pharmingen、San Diego、カリフォルニア州)を用いて現像した。OD450nmの
シグナルを記録し、GraphPad Prismを用いてデータを分析することによりIC50の値を算出した。結果を表7にまとめる。IC50を、プレートに結合したBow−Ang2リガンドで検出可能なビオチン−Tie2−Hisを50%低減するのに必要な抗体の量として定義した。表7において、抗体の添加により、抗体がないコントロールと比較してBow−Ang2被覆表面に結合したTie2−Hisの量が増加する場合、抗体を「増強剤」と命名した。
Systems,Inc.,Minneapolis、ミネソタ州、Cat923−AN/CF;受託番号Q5HYA0)、hAng2(R&D Systems,Inc.,Minneapolis、ミネソタ州N、Cat623−AN/CF;受託番号O15123)、mAng3(R&D Systems,Inc.,Minneapolis、ミネソタ州、Cat738−AN/CF;受託番号Q9WVH6)またはhAng4(R&D Systems,Inc.,Minneapolis、ミネソタ州、Cat964−AN/CF;受託Q9Y264)を100nM含有するウェル中に5分にわたってセンサー先端部を配置した。実験の各工程での結合反応性をモニタリングし、モノクローナル抗体飽和受容体表面への様々なアンジオポエチンの結合をプロットした(図2)。図2に示
すように、H4H2055Nは、4種全てのアンジオポエチンのhTie2.mFc表面への結合を遮断した。コントロールIの抗体は、アンジオポエチンのTie2被覆表面へのいずれの結合も遮断しなかった。
アンジオポエチン介在Tie2シグナル伝達を遮断する抗Tie2抗体の能力の評価
本発明の抗hTie2mAbを更に特徴付けるために、Tie2誘導ルシフェラーゼ活性を遮断する抗体の能力を調べた。簡潔に言えば、HEK293細胞にhTie2を安定的に遺伝子導入し、Tie2を発現する細胞の上位10%をFACSにより単離した。次いで、これらの細胞に血清応答要素(SRE)依存型ルシフェラーゼ・レポーター・レンチウイルス(SA Biosciences)を形質導入し、アンジオポエチン−1(Bow−Ang1、配列番号14)およびアンジオポエチン−2(Bow−Ang2、配列番号13)の四量体に対する強い反応性を有するクローナル個体群(293−TIE2/SRE−Luc)を単離した。
インビボでの抗Tie2の抗腫瘍活性の評価
免疫不全マウスにおけるヒト腫瘍の異種移植片の増殖を阻害する例示的な抗Tie2抗体(H2M2055N)の能力に関して、Tie2抗体を試験した。簡潔に言えば、SCIDマウスにおいてヒト結腸直腸のHT29またはColo205腫瘍を増殖させた。腫瘍が触知できた場合(HT29に関しては100mm3およびColo205に関しては
400mm3)、Fcタンパク質(10mg/kg)または抗Tie2抗体(H1M20
55N;10mg/kg)により隔週で動物を処理した。処理の終了時(HT29に関しては31日およびColo205に関しては14日)に、腫瘍増殖阻害率(%TGI)を求め(表9)、腫瘍組織を摘出して血管密度の分析に利用した。CD31免疫組織化学により、30μm厚のOCT腫瘍部における血管密度を評価した。NIH画像ソフトウェアを利用して腫瘍組織部における血管密度の%面積を求めた。結果を図4〜7に図示する。
Claims (20)
- ヒトTie2に特異的に結合し、Tie2とTie2リガンドとの間の相互作用を遮断する単離抗体またはその抗原結合フラグメント。
- Tie2リガンドは、Ang1、Ang2、Ang3およびAng4からなる群から選択される、請求項1に記載の単離抗体または抗原結合フラグメント。
- 抗体またはその抗原結合フラグメントは、Tie2と、Ang1、Ang2、Ang3およびAng4の4つ全てとの間の相互作用を遮断する、請求項1に記載の単離抗体または抗原結合フラグメント。
- 抗体またはその抗原結合フラグメントは、ヒトTie2のIg1−Ig2−EGFドメイン(配列番号7)からなるポリペプチドまたはヒトTie2のIg2−EGFドメイン(配列番号8)からなるポリペプチドに特異的に結合する、請求項1〜3のいずれか1項に記載の単離抗体または抗原結合フラグメント。
- 抗体または抗原結合フラグメントは、配列番号7のアミノ酸96〜106、配列番号7のアミノ酸139〜152および配列番号7のアミノ酸166〜175からなる群から選択される1つまたはそれ以上のアミノ酸セグメント内に位置する1個またはそれ以上のアミノ酸と相互作用する、請求項1〜4のいずれか1項に記載の抗体または抗原結合フラグメント。
- 抗体または抗原結合フラグメントは、水素/重水素交換法で特定して、配列番号7のアミノ酸96〜106、配列番号7のアミノ酸139〜152および配列番号7のアミノ酸166〜175と相互作用する、請求項5に記載の抗体または抗原結合フラグメント。
- 抗体またはその抗原結合フラグメントは、げっ歯類のTie2およびヒトTie2と特異的に結合する、請求項1〜6のいずれか1項に記載の単離抗体または抗原結合フラグメント。
- げっ歯類のTie2は、マウスTie2またはラットTie2である、請求項7に記載の単離抗体または抗原結合フラグメント。
- 抗体またはその抗原結合フラグメントは、ヒト、マウスおよびラットTie2と特異的に結合する、請求項1〜6のいずれか1項に記載の単離抗体または抗原結合フラグメント。
- 抗体またはその抗原結合フラグメントは、受託番号PTA−12295またはPTA−12296でアメリカン・タイプ・カルチャー・コレクションに寄託された細胞株から生産される、請求項1〜9のいずれか1項に記載の単離抗体または抗原結合フラグメント。
- 受託番号PTA−12295またはPTA−12296でアメリカン・タイプ・カルチャー・コレクションに寄託された細胞株から生産される抗体とヒトTie2上の同じエピトープに結合する単離抗体またはその抗原結合フラグメント。
- ヒトTie2への結合に関して、受託番号PTA−12295またはPTA−12296でアメリカン・タイプ・カルチャー・コレクションに寄託された細胞株から生産される抗体と競合する単離抗体またはその抗原結合フラグメント。
- 請求項1〜12のいずれか1項に記載の抗体または抗原結合フラグメントと、薬学的に許容される担体または賦形剤とを含む医薬組成物。
- ヒトTie2に特異的に結合しTie2とTie2リガンドとの間の相互作用を遮断する抗体またはその抗原結合フラグメントを含む、患者の腫瘍の増殖阻害に用いられる医薬組成物。
- Tie2リガンドは、Ang1、Ang2、Ang3およびAng4からなる群から選択される、請求項14に記載の医薬組成物。
- 抗体またはその抗原結合フラグメントは、Tie2と、Ang1、Ang2、Ang3およびAng4の4つ全てとの間の相互作用を遮断する、請求項14に記載の医薬組成物。
