<細胞培養および薬剤>
5つのヒト肺がん細胞株(NCI−H226、H292、H358、H520およびA549、および5つの結腸直腸がん細胞株(Caco2、SW48、LS180、LovoおよびHCT116)をATCC(Manassas,VA,USA)より購入して、1%ペニシリンおよびストレプトマイシンを含むRPMI1640またはDMEM(Mediatech Inc.,Manassas,VA,USA)中の10%ウシ胎児血清(FBS)中に保った。5つの頭頸部がん細胞株(UM−SCC1、UM−SCC4、UM−SCC6、UM−SCC11A、およびUM−SCC1483細胞)をミシガン大学から取得し、1%ヒドロコルチゾンで補充されたDMED中の10%FBS(Invitrogen,Carlsbad,CA,USA)中に保った。
抗HER−3抗体U1−59を用いた。
細胞を96穴プレートに播いて、複数回用量のU1−59に72時間曝露した。細胞増殖を、Cell Counting Kit−8(Dojindo Molecular Technologies,Gaithersbury,MD,USA)により調べた。
規定数の細胞を、6穴の組織培養プレートの各ウェル中に播いた。細胞が付着する時間(6時間)を与えた後、U1−59またはビヒクルコントロール(PBS)を規定濃度で加えた。4時間後に、プレートを、2、4、6、8Gyの照射量で照射した。播いてから10〜14日後、コロニーをクリスタルバイオレットで染色して、少なくとも50細胞を含むコロニーの数を判定し、生存率を計算した。U1−59単独により産生される細胞毒性に標準化後、生存曲線を作成した。示されるデータは、少なくとも3つの非依存性実験による平均±SDである。
細胞をU1−59(20μg/ml)とともに48時間インキュベートして、その後、6Gyの放射線を照射した。24時間後、細胞をトリプシン処理してPBSで洗浄し、90%エタノールに固定して4℃で一晩保管した。遠心分離によりエタノール除去後、細胞をPI染色バッファー(50μg/ml PI、100μg/ml Rnase A、0.1%Triton X−100)で染色した。細胞をFACSCaliburフローサイトメーターによりソーティングした(BD Biosciences,San Jose,CA,USA)。ヒストグラム解析をFlowJoソフトウェア(Tree Star Inc.,Ashland,OR,USA)で行なった。
アネキシンV/PIキットを用いてアポトーシスを検出した。処理に続いて、100μlの染色バッファー中に1×105細胞を含む細胞懸濁液を、5μlのアネキシンVおよびPIとともにインキュベートした。FACSCaliburフローサイトメーター(BD Biosciences,San Jose,CA,USA)により細胞をソーティングした。FlowJoソフトウェアで集団解析を行なった。
処理に続いて、バッファー(50mM HEPES(pH7.4)、150mM NaCl、0.1% Tween−20、10%グリセロール、2.5mM EGTA、1mM EDTA、1mM DTT、1mM PMSFおよび10μg/mlのロイペプチンおよびアプロチニン)で細胞を溶解した。標準的なブラッドフォード吸光度分析を用いてタンパク質を定量化した。等量のタンパク質をSDS−PAGEにより分画した。その後、タンパク質をPVDF膜に移して、適切な一次抗体とのインキュベーションにより分析した。HRP−コンジュゲート二次抗体とのインキュベーションおよびECL化学発光検出システムにより、タンパク質を検出した。NIH ImageJプログラムを用いてウェスタンのバンドの濃度測定をした。本試験で用いた抗体は以下であった:HER3、AKT、MAPK、ホースラディッシュペルオキシダーゼ−コンジュゲートヤギ抗ウサギIgGおよびヤギ抗マウスIgGは、Santa Cruz Biotechnology Inc.(Santa Cruz,CA,USA)より購入した。p−HER3(Tyr1289)、p−AKTおよびp−MAPKは、Cell Signaling Technology(Beverly,MA,USA)より取得し、α−チューブリンはCalbiochem(San Diego,CA,USA)より取得した。
およそ2×103細胞を、4穴のガラスチャンバースライド(Nalgene Nunc,Naperville,IL,USA)に播いた。48時間後、細胞をPBSで3回洗浄して、室温で15分間、2%ホルムアルデヒドで固定した。細胞を氷冷100%メタノール中で10分間、−20℃でインキュベートして、0.3% Triton X100溶液を含むPBS中、5%の正常血清で、室温で1時間ブロッキングして、p−HER3またはλ−H2AX抗体とともに、4℃で一晩インキュベートした。次に、細胞を、0.3% Triton X100および1% BSAを含むPBS中で、FITCがコンジュゲートされた適切な二次抗体とともに2時間インキュベートした。ProLong gold with DAPI(Invitrogen)を用いてスライドをマウントした。共焦点顕微鏡または蛍光顕微鏡により写真を取り込んだ。
無胸腺ヌードマウス(4〜6週齢;雄)を、ハーランラボラトリーズ(Indianapolis,IN,USA)から取得した。動物の全手順および維持は、ウィスコンシン大学の施設ガイドラインに従って行なった。0日目に、マウスの背横腹に左右両側に細胞を注入した(2×106細胞)。腫瘍が想定量に達した時点で、マウスを、1)IgG、2)U1−59、3)放射線照射、または、4)組み合わせで、単回量または分割量で処理した。デジタルキャリパーにより測定して、式(π)/6×(大直径)×(小直径)により計算した。
異種移植片腫瘍を中性ホルマリン中に固定して、パラフィン中に包埋した。
免疫組織化学染色を、PCNA、p−MAPK、p−AKT、p−S6およびγ−H2AXについて行なった。要するに、検体を脱パラフィンして通常どおりに再水和して、続いて、ウォーターバス中で、98℃で15分間、シトレートバッファーによりアンチジーン修復して、3%過酸化水素中で10分間インキュベーションして、3%BSAブロッキングを30分間行なった。染色は以下のとおりに行なった:推奨される抗体希釈剤中に希釈した一次抗体中で、4℃で一晩インキュベーションして、ビオチン化二次抗体中で室温で30分間インキュベーションして、Dakocytomation Liquid DAB+Substracte Chromogen Systemを用いてペルオキシダーゼを視覚化して、ヘマトキシリン中で対比染色して、通常どおりに乾燥および洗浄して、カバーガラスをマウントした。
異種移植片腫瘍増殖をグラフ化して、GraphPad Prism 5(GraphPad,San Diego,CA)を用いて解析した。
HER3は様々な固形腫瘍細胞株で発現される。
ヒト上皮成長因子受容体3(HER3)は、HERファミリーにとって重要な二量体化パートナーであり、発がんシグナリングパスウェイを活性化する。多くの固形腫瘍におけるその過剰発現は、予後不良につながっている。図1Aにおいて、我々は、HER3およびp−HER3の両方の発現について、非小細胞肺がん(H226、H292、H358、H520およびA549)、結腸直腸がん(Caco2、SW48、LS180、LovoおよびHCT116)および頭頸部扁平細胞がん(SCC1、SCC6、SCC11A、SCC38およびSCC1483)由来の15細胞株を特徴づけた。HNSCCおよびNSCLC細胞株の両方が活性化HER3を含み、HER3が、それらの腫瘍形成表現型を亢進し得ることを示す。
HER3は、内因性チロシンキナーゼ活性を欠く。しかしながら、ヘレグリン(ニューレグリン−1)などの多様なリガンドに結合する際に、HER3は、他のHERファミリーメンバー受容体とヘテロ二量体を形成して、細胞の増殖および生存に強く影響を与える様々なシグナリングパスウェイの活性化を開始することができる。U1−59は、結合してHER3発がんシグナリングパスウェイを不活性化する、完全にヒト化されたモノクローナル抗体である。図1Bでは、我々は、様々なHNSCCおよびNSCLC細胞株を上昇量のU1−59で処理すると、HER3およびAKTの両方の活性化をうまく阻害することができることを示す(図1B)。加えて、イムノブロッティングおよび免疫蛍光分析を用いて、我々は、HNSCC細胞株SCC6およびNSCLC細胞株H226の放射線治療は、HER3の活性化型を増加させることを見いだした(図2A、図2C、図2D、および図2F)。また、放射線治療後の一過的なHER3活性化は、SCC6およびH226細胞株の両方において、ERKおよびAKTの両方の活性化の増加も伴った(図2Aおよび図2D)。図2Bおよび図2Eでは、我々は、放射線照射により、SCC6およびH226細胞株の両方において、放射線治療の24時間後および48時間後の両方で、HER3、AKT、およびERKの継続的な活性化をもたらすことができることを示す。我々は続いて、放射線治療の前に、U1−59で24時間、SCC6またはH226細胞株を事前にインキュベーションすると、放射線により誘導されるHER3、ERK、およびAKTの活性化を防止することができることを示す(図2A、図2C、図2D、および図2F)。加えて、U1−59は、放射線治療後24時間および48時間の両方で、HER3、ERK、およびAKTの活性化の継続的な阻害を導くことができた(図2Bおよび図2E)。まとめると、これらのデータは、放射線治療の前にU1−59で細胞を事前にインキュベーションすると、放射線により誘導される細胞生存および増殖パスウェイの活性化を防止することができることを示唆する。
図2では、我々は、U1−59が、放射線により誘導されるHER3の活性化および後に続く下流のエフェクターERKおよびAKTを防止することができることを示し、したがって、我々は、U1−59がHNSCCおよびNSCLC細胞株の両方の放射線感受性を高めることができると仮説を立てた。第一に、我々は図3Aおよび図3Fにおいて、U1−59が、SCC6およびH226細胞株の両方の増殖を用量依存的な様式で阻害することができることを示す。放射線と組み合わせたU1−59のインビトロでの効果を評価するために、我々は、クローン原性分析を「材料および方法」に記載のとおりに行なった。我々は、U1−59で4時間、SCC6およびH226細胞の両方を事前インキュベートして、続いて、指示照射量で放射線を細胞に当てた。図3Bおよび図3Gに示すように、U1−59は、放射線曲線の傾斜を増大させ、SCC6およびH226細胞の両方に対する、放射線に応じたU1−59の増感効果を示した。コントロール曲線は、U1−59処理無しで放射線に曝された。
我々はさらに、U1−59はまた、HNSCCおよびNSCLC細胞株の、放射線により誘導される細胞周期の停止およびアポトーシスも亢進させ得ると仮説を立てた。これを調べるために、我々は、SCC6およびH226細胞の両方をU1−59で48時間事前インキュベートして、続いて、6Gyの放射線に曝した。放射線を照射して24時間後に、我々は、細胞周期の段階分布およびアポトーシスのレベルをPIおよびアネキシンV染色により分析した。図3Cおよび図3Hに示すように、コントロール細胞と比較して、U1−59で事前に処理した細胞はG1期停止を示し、一方で、放射線により処理した細胞はG2期停止を示した。我々は、いずれかの処理でS期の減少を観測する。G1期の細胞は放射線治療により感受性であり、一方でS期の細胞はより抵抗性であることが良く知られている。したがって我々は、U1−59とともに事前インキュベートして放射線処理した細胞において、アポトーシスが実際に亢進されることを、アネキシンV染色を介して示し(図3Dおよび図3I)、これは、U1−59により誘導されるG1期停止が、SCC6およびH226の両方の放射線感受性を促進した可能性があることを示唆する。加えて我々はさらに、放射線照射の前にU1−59で事前に処理されたSCC6およびH226細胞での、DNA二本鎖切断レベルの亢進を、γ−Η2ΑΧ染色を介して示す(図3Eおよび図3J)。概して、これらのデータは、放射線と組み合わせたU1−59の亢進した抗腫瘍効果を示す。
U1−59と組み合わせた放射線療法の有効性をさらに評価するために、我々は、SCC6、SCC1483およびH226細胞株を無胸腺マウスに接種した。腫瘍が100〜200mm3に達した時点で、腫瘍を単一線量および分割線量の両方の放射線照射に振り分けた。単一線量治療群では、異種移植片マウスは、IgG、U1−59(8mg/kg)、放射線(XRT16GyまたはXRT10Gy)または両方の組み合わせが与えられた(図4A)。分割線量治療群では、異種移植片マウスは、IgG、U1−59(100μg/マウス)、放射線(指示照射量で1週間に2回)、または組み合わせが与えられた(図4B)。合計3つの異種移植片モデルは、単一線量で治療されたマウスまたはIgGコントロールで治療されたマウスと比較して、単一線量または分割線量の放射線照射のいずれかとU1−59を組み合わせて治療した両方の場合に、腫瘍体積の著しい減少を示した。したがって、U1−59および単一線量または分割線量の放射線照射のいずれかで治療した異種移植片の両方とも、カプランマイヤー生存曲線分析を介して表されるように、明らかな延命効果を示す。
放射線治療と組み合わせたU1−59の増殖阻害効果をさらに分析するために、我々は、腫瘍治療されたSCC6におけるERK、AKT、S6キナーゼ、およびγ−H2AXの活性化を、免疫組織化学により分析した(図6)。我々は、U1−59および単一線量の放射線の両方で治療したSCC6細胞は、下流エフェクター分子ERK、AKT、およびS6キナーゼの活性化レベルを、より低くすることができることを示す。加えて、U1−59で治療された腫瘍は、PCNA陽性細胞のレベルの減少を示した。最終的に、U1−59および放射線の両方で治療したSCC6腫瘍において、γ−H2AXのレベルの増加が見られた。概して、これらのデータは、放射線照射と組み合わせたU1−59は、DNA損傷を亢進することに加えて、細胞生存および増殖パスウェイを効率的に阻害することができることを示す。これらのデータは、インビボにおける、放射線治療と組み合わせたU1−59の抗腫瘍効果の亢進に関する機構的証拠を提供する。
