Abstract
Description
前記封入体中のTDP-43タンパクは異常リン酸化とユビキチン化を受けて凝集しており、その凝集過程では種々の細胞毒性を生じることが知られている。また、前記封入体を有する細胞では核内からTDP-43タンパクが消失しており、TDP-43の機能喪失も発症に寄与すると考えられている。
よって、TDP-43タンパクの核からの消失と細胞質での凝集化を伴う神経変性疾患は、TDP-43 プロテノパシーという概念で括られている(非特許文献1参照)。
すなわち、変異型だけでなく野生型TDP-43タンパクであっても、発現量が過剰になるとALSやFTLD様病態が引き起こされることが示されたのである。
これらの知見より、TDP-43タンパクの発現過多は、TDP-43 プロテノパシーの主原因と考えられている。そして、該疾患を治療するために、TDP-43タンパクの発現過多を是正するための方法が精力的に研究されている。
また、非特許文献6では、小胞体膜上に局在するシステインプロテアーゼであるカスパーゼ4がTDP-43タンパクの第174番目と175番目のアミノ酸間を切断すると、TDP-43のクリアランスが開始されることを報告している。
マイクロRNAは、miRNA-RISC複合体として特定のmRNAの3’UTRに結合し、該mRNAの分解又は翻訳阻害を引き起こしてその発現を抑制する。これまでに、種々の疾患に関わる遺伝子を標的とするマイクロRNAが多数同定され、該マイクロRNA又は該マイクロRNAを標的とする低分子核酸(anti-miR)を有効成分とする薬剤が開発されている(例として、特許文献2−4)。
TDP-43タンパクは、マイクロRNAと同様に、自身のmRNAの3’UTRに直接結合して該mRNAの分解を促進することが知られている(非特許文献8)。よって、TDP-43遺伝子に関しては、マイクロRNAに代わって、TDP-43タンパク自身が自らのmRNA量とタンパク量を制御している可能性も考えられている。さらに、TDP-43タンパクが既に発現過多となった細胞では、TDP-43を標的とするマイクロRNAがあったとしても、十分には機能しない可能性も懸念されている。
このような事情から、マイクロRNAを用いたTDP-43 プロテノパシーの治療方法の開発は容易ではないと考えられていた。
[1] 配列番号1で表されるヒトmiR-33、その変異体、ならびにそれらの前駆体からなる単離されたRNA、及び前記RNAをコードする単離された核酸、
からなる群より選ばれる1以上の核酸を含む、TDP-43 プロテノパシーの予防又は治療用組成物。
[2] 前記変異体が、前記ヒトmiR-33の5’側から第1位の塩基又は第9位以降の塩基における変異である、前記[1]に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
[3] 前記変異が、5塩基以下の置換、欠失、又は挿入である、前記[1]又は[2]に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
[4] 前記前駆体が、二本鎖miRNA及びpre-miRNAである、前記[1]−[3]のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
[5] 前記RNAをコードする単離された核酸が、ウイルスベクターに機能的にコードされた核酸である、前記[1]−[4]のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
[6] 前記ウイルスベクターが、レンチウイルスベクター又はアデノ随伴ウイルスベクターである、前記[5]に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
[7] 前記RNAが、修飾されたヌクレオチドを少なくとも1以上含む、前記[1]−[4]のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。

本明細書中の“NR_”、“NM_”、“NP_”とそれに続く数字は、NCBI(National Center for Biotechnology Information)データベースに標準配列(Reference Sequence)として登録されているnon-coding RNA遺伝子の塩基配列(NR_〜)、転写物の塩基配列(NM_〜)、アミノ酸配列(NP_〜)のIDを表す。なお、複数の標準配列が登録されているものについては、一のIDのみを例示する。
マイクロRNAはnon-coding RNAの一種で、部位及び時期特異的に発現して特定の遺伝子(=標的遺伝子)の発現を翻訳段階で抑制する、内在性の遺伝子発現制御因子である。
その生合成過程では、ゲノムからPrimary microRNA(pri-miRNAと略記)として転写された後、約60-100ヌクレオチド程度の領域が切り出されてPrecursor miRNA(pre-miRNAと略記)となる。pre-miRNAは、不完全に対合したステム部分とループ部分からなるヘアピン状の構造を形成したヘアピン型RNAである。pre-miRNAは細胞質に輸送され、RNaseIIIであるDicerによって当該ループ部分が切断されて二本鎖miRNAとなり、さらにプロセシングされて18−26bp程度の二本鎖成熟miRNAとなる。その後、二本鎖又は一本鎖の状態でRNA-induced silencing complex(RISCと略記)に取り込まれ、最終的に片方の成熟miRNAを内包するmiRNA-RISC複合体が形成される。RISCと複合体を形成した一本鎖成熟miRNAはguide鎖と呼ばれ、もう一方の(RISCと複合体を形成しなかった)一本鎖成熟miRNAはpassenger鎖と呼ばれる(Ha M., and Kim NV., Nat. Rev. Mol. Cell Biol., 15:509-524, 2014)。
よって、pre-miRNA、二本鎖miRNA、二本鎖成熟miRNAはすべてマイクロRNAの前駆体であり、これらの前駆体のいずれかをRNA分子として細胞に直接導入、又は発現ベクターを用いて細胞内で発現させることで、当該標的遺伝子の発現抑制効果を得ることができる。
また、本明細書における“pre-miRNA”には、当該guide鎖の配列を保持し、且つ、当該ヘアピン構造を保持し得るpre-miRNAバリアントがすべて包含される。それらのバリアントは、Dicerに切断されて最終的に当該guide鎖を生じ得る(すなわち、当該pre-miRNAとして機能する)からである。
本発明者らは、ヒトmiR-33が、TDP-43遺伝子の3’-UTR配列(NM_007375の第1379番目から第4216番目までのヌクレオチド配列)における第2108-2114番目の塩基配列(CAAUGCA)を標的配列としてその発現を抑制するマイクロRNAであることを明らかにした。さらに、ヒトmiR-33から当該シード配列(=5’側から第2-8位の塩基配列)以外の塩基配列を一部置換しても、前記TDP-43遺伝子を標的とする活性が損なわれないことを見出した。
よって、miR-33a、miR-33b、及び、当該シード配列以外の塩基配列が一部変異した変異型miR-33a及びmiR-33bは、いずれもTDP-43を標的とするマイクロRNAである。
miR-33a(microRNA 33a、Official Symbol:MIR33A)のguide鎖の塩基配列は配列番号2で表され、pre-miRNAの塩基配列は例えばNR_029507(配列番号10)で表される。当該Pre-miR-33aは、細胞内で下記化学式1で表される二次構造を形成することが明らかとなっている。
よって、前記pre-miR-33a(化学式1で表されるRNAとそのバリアント)と、それらから生じる二本鎖miR-33a及び二本鎖成熟miR-33aは、すべてmiR-33aの前駆体である。
miR-33b(microRNA 33b、Official Symbol:MIR33B)のguide鎖の塩基配列は配列番号3で表され、pre-miRNAの塩基配列は例えばNR_030361(配列番号11)で表される。当該Pre-miR-33bは、細胞内で下記化学式2で表される二次構造を形成することが明らかとなっている。
よって、前記pre-miR-33b(化学式2で表されるRNAとそのバリアント)と、それらから生じる二本鎖miR-33b及び二本鎖成熟miR-33bは、すべてmiR-33bの前駆体である。
本発明に用いることができるヒトmiR-33変異体は、TDP-43遺伝子の発現抑制活性を保持した変異体である。そのような変異体としては、当該シード配列以外の塩基配列に変異を有する変異体が挙げられる。当該変異は、塩基置換、欠失、挿入のいずれでもよく、5塩基以下の変異であることが好ましい。さらに好ましくは、4塩基以下、3塩基以下、2塩基以下、1塩基の塩基置換、欠失、又は挿入である。また、連続した5塩基、4塩基、3塩基、2塩基の塩基置換、欠失、又は挿入であってもよい。
前記変異部位は、最も好ましくは、配列番号1の塩基配列において5’側から第2-8位の塩基以外の塩基(=第1位の塩基、及び/又は第9位以降の塩基)であり、さらに第12-16位の塩基を除く部位(=第1位の塩基、第9-11位の塩基、及び/又は第17位以降の塩基)であるとが一層好ましい。当該第9-11位の塩基、及び/又は第17位以降の塩基を置換しても、TDP-43遺伝子の3’UTRに結合して発現を抑制する活性が損なわれないことを確認しているからである。
本発明にかかるマイクロRNAとその前駆体は、安定性の向上を目的として、修飾ヌクレオチド(ヌクレオチド類似体を含む)を好適に含むことができる。そのような修飾ヌクレオチドの例として、糖修飾リボヌクレオチド、骨格修飾リボヌクレオチド、及び塩基修飾リボヌクレオチド等が挙げられる。
上記のうち、2’-フルオロ、2’-O-メチル、又は2’-O-メトキシエチル修飾されたリボヌクレオチドを特に好適に用いることができる。
本発明における“単離された核酸”とは、“該核酸が由来する生物のゲノム内で直接隣接していた核酸配列から分離された核酸”を指す。よって、ゲノム中にコードされるmiR-33aは単離された核酸ではないが、ゲノムから転写されたpri-miR-33と以降のプロセシング産物(pre-miR-33a、二本鎖、二本鎖成熟、一本鎖成熟)はすべて単離された核酸である。また、人工的に合成された核酸は単離された核酸であり、PCR法を用いて増幅されたDNA断片も単離された核酸である。
これらの単離された核酸は、本発明にかかる組成物中で、核酸分子として独立して存在していてもよく、また、前記ゲノム内で隣接していた配列とは異なる配列に接続していてもよい。よって、前記人工的に合成された核酸又はPCR法で増幅されたDNA断片がプラスミドや発現ベクターに組み込まれた状態も、単離された核酸に相当する。
前記プロモーターとしては、SV(Simian virus)40プロモーター、CMV(Cytomegaro virus)プロモーター、β-Actinプロモーター、EF(elongation factor)1α プロモーター、CAGプロモーター等のRNA Polymerase II系プロモーター、及び、U6及びH1プロモーター等のRNA Polymerase III系プロモーターを好適に用いることができる。
なお、前記TDP-43を標的とするマイクロRNA等をコードする核酸は、DNA、RNAのいずれであってもよい。
本発明に用いることができるウイルスベクターの例としては、これらに限定されるものではないが、組換えアデノウイルス、レトロウイルス等が挙げられ、より具体的には、例えば、無毒化したレンチウイルス、アデノ随伴ウイルス、ヘルペスウイルス、ワクシニアウイルス、ポリオウイルス等が挙げられる。このうち、レンチウイルス及びアデノ随伴ウイルスが好ましく、最も好ましくはアデノ随伴ウイルスである。
前述したウイルスベクターは、当該プロモーターの改変も含めて、公知の方法で作製することができる。
本発明にかかるTDP-43 プロテノパシーの予防又は治療用組成物は、有効成分として、前記TDP-43を標的とするマイクロRNA、当該前駆体、及びそれらをコードする単離された核酸から選ばれる1以上の核酸を含むものである。本発明にかかる組成物は、前記有効成分の他に、医薬的に許容される担体、希釈剤、賦形剤、アジュバント等を含むことができ、通常の製薬に用いられる方法によって製造することができる。
これらの薬物送達用ナノ粒子は、当業者に周知の方法に従って製造することが可能である。
PLGAナノ粒子の製造と本発明にかかる有効成分の該粒子への封入は、例えば、特許4340744号に記載された方法に従って行っても良い。PLGAナノ粒子は、生体適合性、生体内分解性、及び徐放性に非常に優れることから、前述した本発明にかかる有効成分を内包するPLGAナノ粒子の分散液は、吸入薬剤、筋注薬剤、ステント等として用いることも可能である(Tsukada Y., et al, New Developments in Polyactic Acid Research, published by Nova Science Publishers、 Chapter 6、pp153-182、2015;塚田雄亮他、PLGAナノ粒子が拓く新たなDDS製品の開発と実用化、医薬ジャーナル、第50巻、第73-80頁、2014年、参照のこと)。
本発明にかかるTDP-43 プロテノパシーの予防又は治療用組成物は、当該分野において公知の方法を用いて患者に投与することができる。例えば、前記液体の組成物を、静脈注射や輸液によって全身投与してもよく、又は、定位固定注射、ニードルやカテーテル、浸透圧ポンプやインフュージョンポンプ、薬剤溶出型ステント等を用いて脳室又は髄腔内に局所的に投与してもよい。さらに、本発明にかかる組成物が前記有効成分をウイルスベクターにコードされる状態で含む場合には、筋肉(好ましくは骨格筋)への定位固定注射によって投与してもよい。
なお、本発明における“健常者”とは、TDP-43 プロテノパシーに罹患していない者の意である。
本発明にかかる組成物は、TDP-43 プロテノパシー患者及び該疾患が疑われる患者に対し、予防又は治療薬として好適に投与することができる。TDP-43 プロテノパシーは、TDP-43タンパクの構造的異常及び細胞内局在異常を伴う神経変性疾患で、代表的な疾患としては、大部分の孤発性ALS、多くのSOD1非関連家族性ALS、大部分の孤発性及び家族性FTLD-Uが挙げられる。
また、本発明にかかる組成物は、脳脊髄液中のTDP-43濃度が病的に高い患者に対し、TDP-43 プロテノパシーの予防薬として用いてもよい。
本発明においてニューロン(Neuron;N略記する場合がある)とは、β-III tubulin、NCAM、MAP2等のニューロンのマーカー遺伝子を1以上発現し、且つ、神経突起を有する細胞と定義される。また、本発明において運動ニューロン(Motor neuron;MNと略記する場合がある)とは、HB9、ChAT(choline acetyltransferase)等の運動ニューロンのマーカー遺伝子を1以上発現するニューロンと定義される。
ヒトmiR-33は、ヒトiPSC由来ニューロンが備えるTDP-43発現抑制因子であることから、ヒトに対する有効なTDP-43発現抑制剤と考えられる。
SREBP-1は脂肪酸代謝、SREBP-2はコレステロール代謝の中心的な転写因子である(Brawn M.S., and Goldstein J.L., Cell, 89:331-340, 1997)。ヒトmiR-33及びマウスmiR-33は、コレステロール逆輸送を担うATP-binding cassette transporter A1(ABCA1)を標的としており、miR-33の発現抑制は動脈硬化を改善することが示されている(Horie T., et al, Nat. Commun., 4:2883, 2013)。そのため、ヒトmiR-33のanti-miRは、コレステロール関連疾患の治療薬として非常に期待されている(例として、特許文献4)。
よって、antimiRでなく、ヒトmiR-33そのものを予防又は治療薬とする用途(方法)、さらには、該ヒトmiR-33を神経変性疾患の予防又は治療薬とする用途(方法)は、本発明において初めて見出されたものである。
・レポーター遺伝子の作製とアッセイ用細胞の作製
健常者由来iPSC株(201B7株)から抽出したtotal RNAを鋳型としてPCRを行い、ヒト野生型TDP-43遺伝子の3’UTR全長を含むDNA断片を得た。当該3’UTR領域をPNL2.2[NlucP/Hygro] vector(Promega社製)にクローニングして、SV40プロモーターに制御され、且つ、3’UTRとして前記ヒト野生型TDP-43 3’UTR全長を有するNlucPルシフェラーゼ遺伝子(=図1D中の“Normal”)を作製した。以降、当該NlucPルシフェラーゼ遺伝子を“野生型レポーター遺伝子”と呼ぶ場合がある。
さらに、In-Fusion HD cloning kit(Clontech社製)を用いて、前記野生型レポーター遺伝子の3’UTRから7mer-1A、7mer-m8、又は7mer-1Aと7mer-m8領域が欠失した3種類の変異型レポーター遺伝子を作製した(=図1D中の“Del1”、“Del2”、“Del1+2”)。
内部コントロール用レポーター遺伝子としては、チミジンキナーゼプロモーターに制御されるfireflyルシフェラーゼ遺伝子(pRL-TK vector、Promega社製)を用いた。
Tol2 transposon法を用いて、前記4種類のレポーター遺伝子のいずれかと、前記内部コントロール遺伝子がゲノムに挿入されたHEK293T細胞株を作製した。
201B7 iPSCから抽出したgenomic DNAを鋳型としてPCRを行い、配列番号10で表されるヒトpre-miR-33aを含むDNA断片を得た。当該pre-miR-33a領域を、pHL-H1-ccdB-mEF1a-RiHベクター(addgene社よりplasmid #60601として入手可能)にクローニングして、H1プロモーターによって転写開始が制御され、poly(T)7で転写終結するpre-miR-33aをコードするプラスミド(以降、“野生型miR-33a発現プラスミド”と呼ぶ)を作製した。当該野生型miR-33a発現プラスミドから前記pre-miR-33a転写単位(すなわち、H1プロモーターからpoly(T)7まで)を含むDNA断片を切り出して、Lenti-6PW(Life technologies社製)にサブクローニングし、H1プロモーターによって転写開始が制御され、poly(T)7で転写終結するpre-miR-33aをコードするレンチウイルス(以降、“H1::miR-33a Lenti”と呼ぶ)を作製した。
同様の手順により、H1プロモーターに制御され、poly(T)7で転写終結するpre-miR-33b(配列番号11)をコードするレンチウイルス(以降、“H1::miR-33b Lenti”と呼ぶ)を作製した。
コントロールとして、前記pre-miR-33aの代わりに、pcDNA6.2-GW/EmGFP-miR-neg Control vector(Invitrogen社製)の“pre-miRNA control insert”(配列番号12、PCRで増幅)をコードするLenti-6PW(以降、“H1::miR-con Lenti”と呼ぶ)を作製した。
前記野生型miR-33a発現プラスミドに変異を導入して、図2に示される4種類のmiR-33a変異体(Mutant1-4)を機能的にコードしたプラスミド(以降、“miR-33a変異体発現プラスミド”と呼ぶ)を作製した。
コントロールとして、前記pre-miR-33aの代わりに、前記”pre-miRNA control insert”をH1プロモーターの制御下にコードするプラスミド(以降、“miR-con発現プラスミド”と呼ぶ)を作製した。
実施例1では、前記アッセイ用HEK293T細胞株に前記H1::miR-33a Lenti又はH1::miR-con Lentiを感染させて、24時間後に、Nano-Glo及びOne-Glo luciferase assay system(Promeg社製)を用いて当該NlucPルシフェラーゼ及びfireflyルシフェラーゼの活性を測定した。NlucPルシフェラーゼ活性はfireflyルシフェラーゼ活性で標準化し、コントロール細胞(=H1::miR-con Lentiを感染させた細胞)に対する相対値として評価トした。
実施例2では、前記アッセイ用HEK293T細胞株に前記miR-33a変異体発現プラスミド又はmiR-con発現プラスミドをリポフェクションして、48時間後に、実施例1と同じ手法を用いてNlucPルシフェラーゼ及びfireflyルシフェラーゼの活性を測定・評価した。
米国マサチューセッツ工科大学から無償で提供されているCRISPR Design(http://crispr.mit.edu/)を用いて、miR-33a遺伝子のシード配列近傍に二本鎖切断を導入し得るguide RNAペア(配列番号13で表されるmiR-33a KO gRNA-1a、配列番号14で表されるmiR-33a KO gRNA-2a)をデザインし、発現ベクターにクローニングした。また、miR-33a遺伝子を含むゲノム領域をPCRで増幅し、loxP配列を両端に有するneomycin選択カセットの上流及び下流に該増幅された核酸を相同アームとして配したコンストラクト(以降、“ドナープラスミド2”と呼ぶ)を作製した。
前記5クローンにCreリコンビナーゼ発現プラスミドを導入した後、プライマーセット(P-a5とP-a6)を用いてgenomic PCRを行い、前記選択カセットが除去されたクローンを選択した。その後、塩基配列決定を行い、当該クローンのゲノムDNAから前記カセットが除去されていることと、miR-33a遺伝子内の当該シード配列の全長又は一部が欠失していることを確認した。
なお、前記プライマーP-a1からP-a6の塩基配列は、配列番号17から22として表1に示したものである。
SFEBq(quick embryoid body-like aggregate method)法を用いて、iPSCから運動ニューロン(Motor neuron、本明細書ではMNと略記することがある)へ分化誘導した(非特許文献7参照)。具体的には、ヒトiPSCを単一細胞に解離させ、Matrigen(BD Bioscience社製)でコートしたU型96-well plateを用いて、2μM dorsomorphin及びSB431542含有5% DFK medium(5% KSR medium(DFK 5%)、Dulbecco’s modified Eagle’s medium/Ham’s F12(Sigma-Aldrich社製)、5% KSR(Invitrogen社製)、Minimum essential medium-nonessential amino acids(Invitrogen社)、L-glutamine(Sigma-Aldrich社製)、2-mercaptoethanol(Wako))中で培養を開始し(=当該培養開始日を分化誘導開始日(すなわち、Day0)とする)、12日間培養して再凝集させた。その後、B27サプリメント、1μM レチノイン酸、1mM Smoothened agonistを含有するNeurobasal medium中で培養し、Day22に当該凝集塊(付着性の胚葉体)をMatrigel-coated dishに播種して接着させた。当該付着性の胚葉体は、10ng/ml BDNF、10ng/ml GDNF、10ng/ml NT-3を含有するNeurobasal medium中で培養した。Day35に、Accutase(Innovative Cell Technologies社製)を用いて前記胚葉体を単一細胞又は小塊に解離させ、P3 maturation stageのニューロンとして、500,000 cells/mlの細胞密度でMatrigel-coated 24-well dishに播種した。
当該方法では、Day35以降にMNに分化した細胞が得られ、Day50くらいにかけてMN数が増加する。なお、当該培養系は、MNとそれ以外のニューロンに分化した細胞を含む、ヘテロな細胞集団である(非特許文献7参照)。
前記手法3に従ってMNへの分化誘導を行ったiPSCに対し、MNに分化した細胞を生きたままGFP蛍光で識別できるように、HB9プロモーターに制御されるGFP遺伝子をコードしたレンチウィルス(HB9::GFP lentivirus、非特許文献7を参照)を感染させた。Day50にAccutaseを用いて単一細胞に解離させた後、FACS AriaII(BD Bioscience社製)に供し、FACS Diva software(BD Bioscience社製)で解析して、GFP陽性細胞(すなわち、MN)を分取した。また、7-aminoactionomycin-D(7-AAD、BD Bioscience社製)染色を行い、死細胞を除去した。得られたGFP陽性生細胞を、20,000-30,000 cells/wellの細胞密度でMatrige-coated 96-well platesに播種し、iPSC-MNの純培養系を作製した。
miRNeasy mini/macro kit(QIAGEN社製)を用いて細胞からtotal RNAを抽出し、TaqMan MicroRNA Assays(Applied Biosystemns社製)のhsa-miR-33a検出キット(Assay ID:000424、Assay Name:hsa-miR-33a)、hsa-miR-33b検出キット(Assay ID:001565、Assay Name:hsa-miR-33b)、U6 snRNA検出キット(Assay ID:001973、Assay Name:U6 snRNA)を用いて、miR-33a、miR-33b、U6 RNA量を各々測定した。得られたシグナルをStepOne software v2.1(Applied Biosystemns社製)を用いて解析し、miR-33a、miR-33bのシグナルを各々U6 snRNAのシグナルに対する相対値として表した。
TDP-43のmRNA量:細胞からmiRNeasy mini/macro kit(QIAGEN社製)を用いてtotal RNAを抽出し、human TDP-43-Fw(配列番号29)とhuman TDP-43-Rv(配列番号30)を用いてqRT-PCRを行い、得られたPCR産物のシグナルを定量化した。TDP-43 mRNA量は、GAPDHのシグナルに対する相対値として表した。
TDP-43のタンパク量:細胞又は組織を、1% SDS含有lysis buffer(Complete mini protease inhibitorとphosphatase inhibitor(Roche社製)を含む)中で超音波処理(sonication)して破砕し、細胞抽出液を得た。当該抽出液(タンパク20μg分)に対し、抗TDP-43抗体(ProteinTech社製、1:1000倍希釈)を用いてウェスタンブロット法を行い、TDP-43タンパク量を測定した。TDP-43タンパク量は、βアクチンのタンパク量に対する相対値として表した。
約500,000細胞分のiPSC-Nを遠心によって回収し、Cholestery Ester Assay Kit(BioVision社製)を用いて総コレステロール量を測定した。解析では、EnVision Multilabeled Reader(PerkinElmer社製)を用いて570nmの吸光度を測定した。
iPSC-MNの神経突起長の測定は、IN Cell Analyzer 2000又は6000(GE Healthcare社製)を用いて行った。視野中のGFP陽性神経突起の長さの総和を測定し、GFP陽性細胞数で除して、iPSC-MNあたりの神経突起長を算出した。各実験につき8視野ずつ解析し、平均値とその標準誤差を算出した。
ヒトTDP-43遺伝子の発現を抑制し得るマイクロRNAを同定するために、公共のマイクロRNA標的予測プログラムであるTargetScanversion v6.2(http://www.targetscan.org/)を用いて、ヒトTDP-43遺伝子(mRNA)の3’-UTR配列(NM_007375の第1379番目から第4216番目までのヌクレオチド配列、計2838nt)を解析した。TargetScanは、対象となるmRNA配列に対して既に登録されているマイクロRNAの標的配列を検索し、当該標的配列の異種ゲノム間における保存率と当該RNA分子間の熱力学的モデリング結果を考慮して、標的配列となり得る可能性を評価するプログラムである。解析結果を図1Aに示す。
そこで、ヒトmiR-33が当該配列を標的とし得るかどうか、3’UTR ルシフェラーゼ・レポーターアッセイ(手法1)を行って検討した。
TargetScanで予測されたmiR-33aとmiR-33bの標的配列は、ヒトTDP-43 mRNAの3’-UTR配列(全長2843nt)の1415-1421番目のヌクレオチドからなる”AAUGCAA”(以降、“7mer-1A”と呼ぶ)と、2108-2114番目のヌクレオチドからなる”CAAUGCA”(以降、“7mer-m8”と呼ぶ)の2つである(図1C)。
同様に、Del1を導入した細胞にH1::miR-33a Lentiを感染させた場合にも、NlucPルシフェラーゼ活性が有意に低下した(H1::miR-con Lentiを導入した細胞の約83%)。これに対し、Del2又はDel1+2を導入した細胞では、H1::miR-33a Lentiを感染させても、NlucPルシフェラーゼ活性は低下しなかった。これらの結果は、miR-33aによる前記レポーター遺伝子の発現抑制には、当該3’UTR中の7mer-8m領域が不可欠であることを示している。
よって、miR-33aはTDP-43遺伝子を標的とするマイクロRNAであり、該遺伝子の3’UTR中の7mer-8m領域がmiR-33aの標的配列であることが明らかとなった。
前述したように、マイクロRNAは標的遺伝子のmRNAと不完全に対合することで該mRNAの分解又は翻訳阻害を引き起こすが、その対合の態様はマイクロRNAと標的mRNAとの組み合わせによって様々である。典型的には、マイクロRNAの5’側から第2−8位にあるシード配列部分が標的mRNAと完全に対合し、該シード配列より3’側では部分的に対合して機能する。よって、シード配列以外に多少の変異(塩基置換、欠失、挿入等)を導入しても当該標的遺伝子の発現抑制因子としての機能が損なわれない場合が多く、なかにはシード配列の一部を置換しても機能するマイクロRNAも知られている(例として、特許文献3)。
そこで、ヒトmiR-33がTDP-43発現抑制因子として機能する場合に許容される配列変更について検討した。
このmiR-33aの塩基配列をベースとして、当該シード配列より3’側の塩基を置換した変異型マイクロRNA(3種類)をデザインした。具体的には、miR-33aの3’末端から3塩基を置換したMutant1(配列番号4)、5塩基を置換したMutant2(配列番号5)と、該シード配列の3’側に隣接する3塩基を置換したMutant3(配列番号6)である。さらに、シード配列内の5塩基を置換したMutant4(配列番号7)も作製した。
Mutant1-3は、Normalを導入した細胞で発現させると当該NlucPルシフェラーゼ活性を有意に低下させたが、Del1+2を導入した細胞で発現させても当該活性を変化させなかった。よって、miR-33aから該シード配列より3’側の塩基を5つまで置換しても、TDP-43 mRNAの3’UTRに作用して該発現を抑制する活性は保持されることが示された。
TDP-43は、ある種のマイクロRNAに対し、pri-miRNAからpre-miRNAの生成工程、及び/又はpre-miRNAから二本鎖成熟miRNAの生成工程を促進することが知られている(Kawahara Y. and Mieda-Sato A., Proc. Natl. Acad. Sci. USA, 109:3347-3352, 2012)。そこで、下記実験系を作製して、TDP-43の発現量がヒトmiR-33の発現量に及ぼす影響を解析した。
なお、本発明者らが手法5に従ってヒト健常者由来iPSC、ヒト健常者由来iPSCから分化誘導したニューロン、及びヒト脳脊髄サンプルにおけるmiR-33a及びmiR-33bの発現量を解析した結果、miR-33bの発現量は非常に微量(検出困難レベル)で、これらの細胞・組織ではmiR-33bは実質的に発現していないことが明らかとなった。よって、以降の解析では、ヒトmiR-33としてmiR-33aのみを解析した。
ヒトTDP-43タンパク(野生型又はM337V変異型)をテトラサイクリン誘導性プロモーターの制御下に発現するコンストラクト(図3A)を作製し、HEK293T細胞に導入して安定発現株を作製した。以降、当該野生型、M337V変異型TDP-43をコードするコンストラクトを導入した安定発現株を、WT TDP-43導入細胞、M337V TDP-43導入細胞と呼ぶ。
これら2種類の安定発現株の培地にdoxycycline(以降、DOXと略記、終濃度
1μg/ml)を添加して、TDP-43の発現を誘導した。DOX添加から48時間後にTDP-43のタンパク量(図3B)とmiR-33a量(図3C)を測定した結果を示す。WT TDP-43導入細胞、M337V TDP-43導入細胞ともに、DOX添加群ではTDP-43タンパク量が顕著に増加し(図3B)、且つ、miR-33a量が有意に減少していた(図3C)。詳細には、DOX非添加群のmiR-33a発現量と比べて、WT TDP-43導入細胞では約74%、M337V TDP-43導入細胞では約63%にmiR-33a発現量が減少した。なお、TDP-43タンパクの代わりにDAP-LYARタンパク(Ly-1 antibody reactive clone、DNA結合タンパク)をコードするコンストラクトを導入した安定発現株では、DOXを添加してLYARの発現を誘導しても、miR-33a発現量は有意には変化しなかった(データは非開示)。
この結果は、前記抗TDP-43抗体の免疫沈降分画にはpri-miR-33aが含まれていたことを示しており、前記細胞溶解液中でTDP-43とPri-miR-33aが特異的に結合していたことを示唆するものである。
次に、TDP-43発現量の減少がmiR-33aの発現に及ぼす影響を、ヒトニューロンで解析することを試みた。そのために、TALエフェクターヌクレアーゼ(Transcription Activator-Like Effector Nuclease;TALEN)システムを用いて、TDP-43遺伝子を機能的に欠失(knockout)したヒトiPSC株(TDP-43 KO iPSCと呼ぶ場合がある)を作製した。当該システムは、DNA結合配列を操作したTALENによってゲノム上の任意の箇所に二本鎖DNA切断を導入し、該DNA切断が外来DNA(本法ではドナープラスミド1)との相同組み換えによって修復される反応を利用して、ゲノム上の任意の部位に欠失、置換、挿入等の変異を導入する方法である(Miller J.C., et al, Nat. Biotechnol., 29:143-148, 2011)。
これらのTDP-43 KO-Nに対し、分化誘導から50日後(=Day50)に、TDP-43のmRNA量(図5A)、TDP-43のタンパク量(図5B、C)、miR-33aの発現量(図5D)、及び細胞内コレステロール量(図5E)を測定した。TDP-43 KO-NのTDP-43 mRNA量は、親株から分化したニューロン(201B7-N)と比べて約半分であり(詳細には、TDP-43 KO1-Nで約39%、TDP-43 KO2-Nで約56%、図5A)、TDP-43タンパク量も顕著に減少していた(TDP-43 KO1-Nで約65%、TDP-43 KO2-Nで約74%、図5C)。
よって、前記TDP-43 KO iPSC株は、ニューロンに分化した後もTDP-43の発現量は低いままであることが明らかとなった。
以上より、TDP-43は、ヒトニューロンにおいてもmiR-33aの発現を負に制御しており、ニューロン内においてTDP-43タンパク量とmiR-33a量は逆相関することが示された。
上記結果は、TDP-43が発現過多となった細胞では、miR-33aの発現が抑制されている可能性を示唆するものである。そこで、TDP-43 プロテノパシーモデルマウス及び該患者由来iPSCについて、miR-33aの発現量を解析した。
図6Cの結果は、TDP-43 プロテノパシー患者では、TDP-43タンパクとmiR-33aの発現量の均衡が崩れてTDP-43タンパクが優位となり(図6D、左側の状態)、miR-33aによるTDP-43発現抑制機構が損なわれている可能性を強く示唆している。そこで、miR-33a遺伝子を機能的に欠失したヒトiPSC株を作製して、ヒト運動ニューロンにおいてmiR-33aの発現が失われることの影響を解析することにした。
miR-33-knockout ヒトiPSC(以降、“miR-33a KO iPSC”と呼ぶ場合がある)の作製には、CRISPR-Cas9システムを用いた。当該システムは、Cas9エンドヌクレアーゼとRNA(=Cas9用guide RNA)の複合体が該guide RNAの配列依存的にゲノムDNAの特定部位に二本鎖切断を導入する反応と、該DNA切断が外来DNA(本法ではドナープラスミド2)との相同組み換えによって修復される反応を利用して、ゲノム上の任意の部位に欠失、置換、挿入等の変異を導入する方法である(Gaj T., et al, Trends Biotechnol., 31:397-405, 2013; Doudna J.A., et al, Science, 346, no.6213, 2014)。
さらに、核型解析を行い、前記5クローンの核型が正常であること(46+XX)を確認した(図7F)。また、前記5クローンからtotal RNAを抽出し、イントロン16がスプライスアウトされた正常なSREBP-2 mRNAが産生されていること(すなわち、当該ゲノムの改変はSREBP-2 mRNAのスプライシングに影響しないこと)を確認した。
前記バイアレリックなmiR-33a KO iPSC株から運動ニューロンの純培養系を作製して、miR-33aの発現消失がヒト運動ニューロンに及ぼす影響を解析した。
前記miR-33a KO 1、miR-33a KO 2、及び201B7をMNへ分化誘導し(手法3)、MNに分化した細胞をFACS法を用いて分取して(手法4)、MNの純培養系を作製した。以降、201B7、miR-33a KO 1、miR-33a KO 2から上記方法で得られたMNを、各々、“201B7-MN”、“miR-33a KO 1-MN”、“miR-33a KO 2-MN”と呼ぶ。
各MNのGFP蛍光顕微鏡写真を図8Aに示す。当該写真より、201B7-MNと比べてmiR-33a KO 1-MNは神経突起が弱々しく、また、miR-33a KO 2-MNは神経突起が少ない傾向があることがわかる。
その結果、miR-33a KO 1-MNでは、H1::miR-33a Lenti感染群の方がH1::miR-con Lenti感染群よりも神経突起が長く、miR-33aの供給によって神経突起の脆弱性が回復する傾向があることがわかった。さらに、miR-33a KO 2-MNでは、H1::miR-33a Lenti感染群の方がH1::miR-con Lenti感染群よりも神経突起長が有意に長く、miR-33aの供給によって神経突起の伸展不良が201B7-MNと同レベルにまで回復することが明らかとなった(図8D)。
分化誘導から36日後(=Day36)に前記H1::miR-33a Lenti又はH1::miR-con Lentiを感染させて、Day50にmiR-33a量(図8B)とTDP-43タンパク量(図8C)を測定した。まず、H1::miR-con Lenti感染群どうしの比較により、miR-33a KO-NではTDP-43のタンパク量が有意に増加することが明らかとなった。具体的には、201B7-Nと比べて、miR-33a KO 1-Nで約130%、miR-33a KO 2-Nで約140%に増加していた。
次に、H1::miR-33a Lenti感染群とH1::miR-con Lenti感染群との比較により、いずれのiPSC-Nでも、H1::miR-33a Lentiの感染によってTDP-43タンパク量が有意に減少することが明らかとなった。具体的には、201B7-Nで約70%、miR-33a KO 1-Nで約73%、miR-33a KO 2-Nで約50%に減少していた。
さらに、上記iPSC-Nについて細胞内コレステロール量を測定した結果、miR-33aの発現量と連動して変化することが確認された(図8E)。
すなわち、ヒトmiR-33は、TDP-43 プロテノパシーの予防又は治療薬になり得ることが示された。
Claims (9)
- 配列番号1で表されるヒトmiR-33、その変異体、ならびにそれらの前駆体からなる単離されたRNA、及び前記RNAをコードする単離された核酸、
からなる群より選ばれる1以上の核酸を含む、TDP-43 プロテノパシーの予防又は治療用組成物。 - 前記変異が、前記ヒトmiR-33の5’側から第1位の塩基又は第9位以降の塩基における変異である、請求項1に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記変異が、5塩基以下の置換、欠失、又は挿入である、請求項1又は2に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記前駆体が、二本鎖miRNA及びpre-miRNAである、請求項1−3のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記RNAをコードする単離された核酸が、ウイルスベクターに機能的にコードされた核酸である、請求項1−4のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記ウイルスベクターが、レンチウイルスベクター又はアデノ随伴ウイルスベクターである、請求項5に記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記RNAが、修飾されたヌクレオチドを少なくとも1以上含む、請求項1−4のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記1以上の核酸が、ナノ粒子に内包されている、請求項1−7のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
- 前記TDP-43 プロテノパシーが、SOD1非関連の筋萎縮性側索硬化症、及び、ユビキチン化封入体を有する前頭側頭葉変性症である、請求項1−8のいずれかに記載のTDP-43 プロテノパシーの予防又は治療用組成物。
