Abstract
Description
a)そのオリゴマー複合体に対する中和抗体を惹起するのに好適な免疫スキームのもとで、本発明の蛋白質オリゴマー複合体を哺乳動物に投与するステップ、
b)その哺乳動物から、その中和抗体および/またはその中和抗体を産生する能力がある細胞を含む生物試料を得るステップ、および
c)その生物試料をその抗体の供給源として用いるステップ、
を含むということを特徴とする。
キメラ蛋白質BLS−Stx2の分子モデリング
キメラの設計は、PyMOL 1 .5プログラム(www.pymol.org)によってその理論上の構造をモデリングし、Stx2Bの構造のC末端(PDBコードIR4P)をBLSの結晶学的構造のN末端(PDBコードID10)と融合することによって行った。これは以前にLaplagneらによって記載されたものと同様の戦略である(Laplagne et al., 2004. Proteins 57:820-828)。BLSのN末端の最初の8残基のコード配列を、Stx2Bと両方の蛋白質を繋ぐフレキシブルG/S5ペプチド、10ペプチド、または15アミノ酸ペプチドリンカーとのコード配列によって置換した。Stx2BおよびBLSは両方とも同じC5対称性の五量体構造を形成し、各プロトマーは他の2つと相互作用する。分子モデリングによって得られたキメラの理論上の構造(図1)は、用いられる全てのリンカーが十分に長く、立体障害なしにBLSの五量体モジュール上へのStx2B五量体の集合体形成を可能にするということを示している。十量体BLS粒子において、2つのStx2B五量体は構造の頂部および底部において逆方向に提示されるであろう。このモデルにおいては、Stx2ホロ毒素のAサブユニットと相互作用するStx2BのC末端表面はBLSスキャフォールドに向いており、免疫システムとの相互作用から恐らく防御されるであろう一方で、グロボトリアオシルセラミド(Gb3)結合部位はまったく曝露されるであろうということは、注目に値する。
配列番号4を有する遺伝子を含むpET11a−BLS−Stx2B−L5(5アミノ酸リンカーを有する)プラスミドが、次のプロトコールによって構築された。
a)Brucella種のルマジンシンターゼ(BLS)のコード遺伝子をクローニングするために、BLS配列をB. abortusのゲノムDNAから特異的プライマーによるPCR増幅によって得、pET11aベクターにクローニングした(Novagen, USA)。カセットを製作するために、Brucella種のルマジンシンターゼ(BLS)のオープンリーディングフレームをコードする配列に対して部位特異的変異導入を実施した(Laplagne et. al., 2004)。2つの新たな制限部位をBLS遺伝子の5’領域に挿入した(5’末端の最初の2つのコドンにNsiI部位、BLSの天然アミノ酸配列の8および9残基を含む2つのコドンに1つのAflII部位)。
b)Stx2B蛋白質(配列番号1)をコードする配列を挿入するために、本発明者は、配列の5’のNsiI酵素の部位(下線)および配列の3’のAflII酵素の部位(下線)+5アミノ酸のリンカーをコードする配列(小文字)を有するStx2B蛋白質を増幅するオリゴヌクレオチドを設計した。Stx2B遺伝子(配列番号2)は、鋳型としてpGEM−Tベクター中の以前にクローニングされた配列からPCRによって得た(Capozzo et al., 2003. Infect Immun 71: 3971-3978)。
そのため, 次のオリゴヌクレオチドを作り、プライマーとしてPCRに用いた。
プライマーforward(配列番号10):
5' ATCAACATGCATGCGGATTGTGCTAAAGGT 3'
プライマーreverse 5(配列番号11):
5' TAAAATCTTAAGACCAGAACCAGAACCGTCATTATTAAACTGCAC 3'
PCR反応は0.2mMデオキシリボヌクレオチド三リン酸(dNTP)(Promega Inc.)、1.5mMのMgCl2(PBL(登録商標))、0.15ユニットのDNATaqポリメラーゼ(PBL(登録商標))、および0.5μΜプライマーによって行った。サイクルのプロファイルは、94℃2分間の1サイクル、92℃10秒間、62℃12秒間、および72℃30秒間の5サイクル、ならびに最後に72℃2分間の1サイクルであった。
PCR産物はアガロースゲルを用いてチェックし、次にQiagen抽出キット(Qiagen, UK)を用いて精製した。
c)pET11a−BLSプラスミドおよびStx2B−L5の精製PCR産物をNsiIおよびAflII制限酵素によって消化した。消化は3ユニットのNsiI(Fermentas Inc.)および3ユニットのAflII(Fermentas Inc)によってTango(商標)緩衝液2×(最終濃度)中で、4時間37℃で実施した。消化産物を5ユニットのDNAT4リガーゼ酵素(Fermentas Inc.)によって一晩16℃でライゲーションした。ライゲーションを大腸菌DH5αコンピテントセルに形質転換し、コロニーを特異的プライマー(forwardおよびreverse 5)によるPCRによってスクリーニングした。挿入はシーケンシングによって確認した。シーケンシング分析によって、Brucella種のルマジンシンターゼの最初の8アミノ酸がStx2B蛋白質の70アミノ酸(配列番号1)によって置換されているということが示された。よって配列番号3を有するBLS−Stx2B(L5)と呼ばれるキメラ融合蛋白質をコードする遺伝子が得られた。
pCI−neo−BLS−Stx2B−L5プラスミドを次のプロトコールによって構築した。
pET11aベクターにクローニングされた配列番号4のBLS−Stx2B(L5)の遺伝子の配列を、ベクターpCi−neo(Promega, USA)にサブクローニングした。そのため、次のオリゴヌクレオチドを作り、プライマーとしてPCRに用いた。ACCATG

BLS-DNA-chimera allプライマーはEcoRI酵素の制限部位(小文字)およびKozakコンセンサス配列(下線)を追加し、BLS-BackプライマーはNheI酵素の制限部位(小文字)を追加する。
かかる配列は、鋳型としてpET11a−BLS−Stx2B(L5)プラスミドを用いてPCRによって増幅した。PCR反応を実施例2Cに記載された通り行った。サイクルのプロファイルは、94℃5分間の1サイクル、94℃1分間、55℃1分間、および74℃1分間の40サイクル、ならびに最後に74℃8分間の1サイクルであった。PCR産物をEcoRIおよびNheI酵素によって消化し、pCI−neoベクターをEcoRIおよびXbaIによって消化した。EcoRI/Nhel消化は次の通り行った。5ユニットのNheI(Fermentas Inc.)、Tango(商標)緩衝液1×(最終濃度)中、4時間37℃。次に、5ユニットのEcoRI(Fermentas Inc.)およびTango(商標)緩衝液を式V=A/8に従って追加した。Vは追加されるべき緩衝液の体積であり、Aは反応の最初の体積である。消化は37℃でもう3時間インキュベーションした。EcoRI/XbaI消化は、4ユニットのEcoRIおよび4ユニットのXbaI(Fermentas Inc.)でTango(商標)緩衝液2×(最終濃度)中で、4時間37℃で行った。
次に、ライゲーション反応を(NheIおよびXbaI酵素は適合する末端を有する)実施例2Cに記載された通り実施した。ライゲーションを大腸菌DH5αコンピテントセルに形質転換し、コロニーを特異的プライマー(BLS-DNA-chimera all/BLS-Backおよびforward/reverse 5)によるPCRによってスクリーニングした。得られた構築物はシーケンシングによってチェックした。pCI−neo−BLS−Stx2B(L5)プラスミドを大腸菌DH5α細胞によって増幅し、さらに"mini prep"カラム(Qiagen, UK)によって精製した。DNAの純度および濃度は260/280nmの分光測色法によって評価した。
配列番号6を有する遺伝子を含むpET11a−BLS−Stx2B−L10(10アミノ酸リンカーを有する)プラスミドを次のプロトコールによって構築した。
a)Brucella種のルマジンシンターゼ(BLS)のコード遺伝子をクローニングするために、BLS配列をB. abortusのゲノムDNAから特異的プライマーによるPCR増幅によって得、pET11aベクター(Novagen, USA)にクローニングした。カセットを製作するために、Brucella種のルマジンシンターゼ(BLS)のオープンリーディングフレームをコードする配列に対して部位特異的変異導入を実施した(Laplagne et al., 2004. Proteins 57:820-828)。2つの新たな制限部位をBLS遺伝子の5’領域に挿入した(5’末端の最初の2つのコドンにNsiI部位、BLSの天然アミノ酸配列の8および9残基を含む2つのコドンに1つのAflII部位)。
b)Stx2B蛋白質(配列番号1)をコードする配列を挿入するために、本発明者は、配列の5’のNsiI酵素の部位(下線)および配列の3’のAflII酵素の部位(下線)+10アミノ酸のリンカーをコードする配列(小文字)を有するこのStx2B蛋白質をコードするヌクレオチド配列を含む配列を設計した。
Stx2B遺伝子(配列番号2)は、鋳型としてpGEM−Tベクター中の以前にクローニングされた配列からPCRによって得た(Capozzo et al., 2003. Infect Immunol 71: 3971-3978)。
そのため、次のオリゴヌクレオチドを作り、プライマーとしてPCRに用いた。

PCR反応を実施例2Cに記載された通り行った。サイクルのプロファイルは、1サイクルの94℃2分間、35サイクルの92℃10秒間、68℃12秒間、および72℃30秒間、ならびに最後の1サイクルの72℃2分間であった。
PCR産物はアガロースゲルを用いてチェックし、次にQiagen抽出キット(Qiagen, UK)を用いて精製した。
c)pET11a−BLSプラスミドおよび精製遺伝子をNsiIおよびAflII制限酵素によって消化し、DNAT4リガーゼ酵素によって16℃で実施例2(C)に記載された通りライゲーションした。ライゲーションを大腸菌DH5αコンピテントセルに形質転換し、コロニーを特異的プライマー(forwardおよびreverse 10)によるPCRによってスクリーニングした。挿入はシーケンシングによって確認した。シーケンシング分析によって、Brucella種のルマジンシンターゼの最初の8アミノ酸がStx2B蛋白質の70アミノ酸(配列番号1)によって置換されているということが示された。よって遺伝子が得られ、これは配列番号5を有するBLS−Stx2B(L10)と呼ばれるキメラ融合蛋白質をコードする。
pCI−neo−BLS−Stx2B−L10プラスミドを次のプロトコールによって構築した。
pET11aベクターにクローニングされた配列番号6のBLS−Stx2B(L10)の遺伝子の配列をpCi−neoベクター(Promega, USA)にサブクローニングした。そのため, 次のオリゴヌクレオチドを作り、プライマーとしてPCRに用いた。

BLS-DNA-chimera allプライマーはEcoRI酵素の制限部位(小文字)およびKozakコンセンサス配列(下線)を追加し、BLS-BackプライマーはNheI酵素の制限部位(小文字)を追加する。
かかる配列を、鋳型としてpET11a−BLS−Stx2B(L10)プラスミドを用いてPCRによって増幅した。PCRは実施例3に記載された通り実施した。PCR産物をEcoRIおよびNheI酵素によって消化し、pCI−neoベクターをEcoRIおよびXbaIによって実施例3に記載された通り消化した。次にライゲーション反応を(NheIおよびXbaI酵素は適合する末端を有する)実施例2(C)に記載された通り実施した。ライゲーションを大腸菌DH5αコンピテントセルに形質転換し、コロニーを特異的プライマー(BLS-DNA-chimera all/BLS-Backおよびforward/reverse 10)によるPCRによってスクリーニングした。得られた構築物をシーケンシングによってチェックした。pCI−neo−BLS−Stx2B(L10)プラスミドを大腸菌DH5α細胞によって増幅し、さらに"mini prep"カラム(Qiagen, UK)によって精製した。DNAの純度および濃度は260/280nmの分光測色法によって評価した。
配列番号8を有する遺伝子を含むpET11a−BLS−Stx2B(15アミノ酸リンカーを有する)プラスミドを次のプロトコールによって構築した。

BLS−Stx2B(5、10、および15アミノ酸リンカーを有する)キメラの精製
3つのpet11a−BLS−Stx2Bプラスミドを組み換え蛋白質の発現のための大腸菌BL21(DE3)コンピテントセルに形質転換した。細胞をアンピシリンを補ったLBブロス中で、37℃で撹拌しながら(250rpm)生育した。一晩培養物を新鮮なLB−アンピシリン培地によって1:100希釈し、OD600nm=1に達するまで生育した。この時点で、1mMイソプロピルβ−D−チオガラクトピラノシド(IPTG)(Sigma)を追加することによって培養物を誘導し、撹拌しながら(250rpm)28℃で3時間インキュベーションした。細菌を4℃で20分、15000gで遠心した。採取した細胞を50mMのTris−HCl(pH8.0)緩衝液中に再懸濁し、音波処理によって溶解した。
全てのStx2B−BLSキメラは封入体として発現された(図2A)。封入体を、一晩のインキュベーションによって、8M尿素、50mMのTris/HCl、5mMのEDTA(pH8)の緩衝液中で、室温で撹拌しながら可溶化した。蛋白質は1M尿素、50mMのTris/HCl、5mMのEDTA(pH8.5)緩衝液に対して12−14,000 MWCO膜(Spectrum Laboratories, Inc., Rancho Dominguez, CA, USA)によって透析した。可溶化した蛋白質は、UV/vis検出器(Gilson, model 152)に繋がったHPLC装置(Gilson model 320)を用い、Q-Sepharose(Pharmacia, GE Healthcare Life Sciences)カラムの陰イオン交換クロマトグラフィーによって精製した。溶出は1M尿素、50mMのTris/HCl、1mMのPMSF(pH8.5)の緩衝液中の0〜1MのNaClの直線勾配を用いて実施した。蛋白質を10,000 MWCO Centriprep(登録商標)濃縮システム(Millipore, Carrigtwohill, Co. Cork, Ireland)によって濃縮した。調製物の純度はドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動SDS−PAGE(15%、w/v)によって決定し(図2BおよびD)、標準的な方法によって定量した。蛋白質は各免疫以前にリン酸緩衝生理食塩水(PBS)に対して透析した。SDS−PAGEによって決定すると、蛋白質は予想される分子量を有しており(図2D)、抗BLSおよび抗Stx2抗体によってウェスタンブロットアッセイによって認識された(図2C)。
pCIneo−BLS−Stx2BL5、L10、およびL15による293T細胞のトランスフェクション
真核細胞内におけるBLS−Stx2Bの正しい発現を裏付けるために、PolyFect(登録商標)試薬(Qiagen Inc.)を用いてpCI−BLS−Stx2Bによって293T細胞をトランスフェクションした。簡略には、6×105細胞/ウェルを、5%CO2中の37℃の6ウェル培養プレートで、抗生物質−抗真菌薬(AA)および5%胎児ウシ血清(FBS, Natocor)を有するRPMI1640培地中で生育した。2μgのpCIneo−BLS−Stx2BをRPMI培地によって100μlまで希釈し、20μlのPolyFect(登録商標)試薬を追加した。複合体形成を可能にするために、混合物を室温で10分インキュベーションした。トランスフェクション混合物をRMPI培地によって1mlまで希釈して細胞に追加し、5%CO2中37℃で、AAおよび2%FBSを有するRPMI1640培地中で48時間インキュベーションした。48時間後に細胞をPBSによって採取し、1500rpmで5分間遠心した。細胞ペレットをSDS−PAGE試料緩衝液中に再懸濁し(細胞溶解のため)、ウェスタンブロット分析にかけた。細胞溶解蛋白質の結果を15%SDS−PAGEにかけ、ニトロセルロース膜に電気転写した。膜は、PBS−0.1%Tweenで希釈した5%ミルクによって室温で2時間ブロッキングし、3%ミルクのPBS−0.1%Tweenで希釈したBLS特異的な抗体(1:2000)と一緒に4℃で一晩インキュベーションした。膜を洗浄し、ペルオキシダーゼ標識ウサギ抗マウスIgG(1:3000)(Bio-Rad Laboratories, Hercules, CA, USA)と一緒に室温で2時間インキュベーションした。反応はECL溶液によって現像した(図3)。
図3は、全ての3つのpCIneo−BLS−Stx2B構築物が対応する蛋白質を真核細胞株293T中で発現できるということを示している。これはウェスタンブロットアッセイによって、予想された分子量(およそ25kDa)の特異的なバンドとして明らかにされた。
円偏光二色性による構造分析
精製キメラの二次構造を評価するために、遠UV(200〜250nmの波長範囲)の円偏光二色性(CD)シグナルが測定された。
BLS−Stx2Bの遠UV−CDスペクトルは、単離されたBLSおよびStx2BのCDシグナルの組み合わせからこの蛋白質について理論上計算されたものと実質的に同一であるということが示された。これらの結果は、3つのキメラの構造において、BLSおよびStx2B両方の二次構造の保持を示している(図4)。
静的光散乱による構造分析
BLS−Stx2Bの平均分子量(Mw)を、高速液体クロマトグラフィーシステム(Waters 486 UV検出器およびLKB 2142示差屈折率計を含む)にタンデムに繋がれたPrecision Detectors PD2010光散乱器によって決定した。一般的に、200〜400μlの各BLS−Stx2B−L5、L10、およびL15(0.5〜1mg/ml)をSuperdex 200 HR-10/30カラムにローディングし、1M尿素の50mMのTris/HCl、0.15MのNaCl(pH8)の緩衝液によって溶出した。溶出材料の90°の光散乱および屈折率シグナルをPCコンピュータ上に記録し、Precision Detectorsによって供給されたDiscovery32ソフトウェアによって分析した。90°光散乱検出器はウシ血清アルブミン(Mw:66.5kDa)を標準として用いて校正した。もたらされたMWと理論上のMWとの間の類似性によって、天然BLSの四次構造と一致する各キメラの構造の適切なフォールディングされたものが示された(図5)。
熱変性分析
PBS(pH7.0)緩衝液中のBLS−Stx2B−L5、L10、およびL15、BLS野生型、ならびにStx2B試料の熱によって誘導される変性を、温度の関数としてJASCO J-810分光偏光計によってそれらの蛋白質の222nmのCDシグナルを測定することによって追跡した。ペルチェシステム(Jasco)によって温度を増大させることによって試料を低速で加熱した。温度走査の範囲は4℃/分の速度で25〜100℃であった。222nmのモル楕円率を0.5℃毎に測定した。よって熱転移の中点温度を見かけ上の融解温度(Tm)として考慮した。キメラの適切なフォールディング構造は、単離されたSTX2BおよびBLSモジュールと比較した熱安定性の分析によってもまた支持された。これらの蛋白質の熱変性をCD分光によって分析した(図6)。BLSの構造は85℃まで安定である。この温度を超えると、蛋白質は89℃の見かけ上のTmで不可逆的プロセスによって協同的にアンフォールディングする。一方で、Stx2Bは50℃までは安定なままであり、次に60℃の見かけ上のTmで協同的で不可逆的にアンフォールディングする。
GB 3 結合試験
Stx2Bの五量体立体配座はGB3結合に必要であるので、Gb3結合ELISAアッセイを用いて、5、10、または15アミノ酸の長さのペプチドリンカーを有するキメラ蛋白質の形(それぞれBLS−Stx2B−L5、BLS−Stx2B−L10、およびBLS−Stx2B−L15)が集合体形成して天然ホロ毒素Bサブユニットの立体配置になったということを機能的に確認した。アッセイは以前に記載された通り実施した(Akenazi et al., 1989. J. ClinMicrobiol 27:1145-1150)。クロロホルム:メタノール(2:1)中に溶解されたGb3(Matreya, State College, PA)を用いて、96ウェルELISAプレート(Greiner Bio-One, Frickenhausen, Germany)をウェルあたり1μgでコーティングした。BLS−Stx2B免疫マウスの血清のプールを一次抗体として用い(1:1000希釈)、ペルオキシダーゼ標識ヤギ抗マウスIgG(Zymed, Invitrogen, Carisbad, CA, USA)を二次抗体として用いた(1:3000希釈)。反応をO−フェニレンジアミン(Sigma, St Louis, MO, USA)によって現像し、吸光度を492nmで読み取った。BLSおよび大腸菌BL21抽出物を非特異的結合の陰性対照として用いた。
投与量依存的な様式の陽性反応がキメラおよび組み換えStx2ホロ毒素では観察されたが、BLSでは観察されなかった(図7)。これらの発見から、BLS十量体の頂部のBサブユニットは正しく集合体形成されており、キメラ蛋白質の3つの形においてそれらのGb3結合能力を維持しているということが結論づけられる。
免疫中の抗体−力価の経時変化
単離された精製組み換えStx2Bサブユニット(Stx2B)と比較して液性免疫応答を改善するBLS−Stx2Bの能力を評価するために、マウスの群に、10アミノ酸の長さのG/Sリンカーペプチドを有するキメラ(BLS−Stx2B−L10)またはStx2Bをフロイントアジュバント(FA)の存在下でi.p.注射した。水酸化アルミニウムゲル(AH)懸濁液(1mg/ml)をStx2B−BLSと混合し、撹拌しながら室温で30分インキュベーションした。AH吸着抗原を10000gで10分遠心し、ペレットをPBS中に再懸濁した。
免疫システムを活性化し、異なる製剤またはワクチンレジメンによって抗体を生ずるBLS−Stx2Bの能力もまた評価するために、本発明者は、フロイントアジュバント(FA)もしくは水酸化アルミニウム(AH)と一緒に、アジュバントなしに、またはDNA−蛋白質プライム・ブーストスケジュールに従って、BLS−Stx2B−L10蛋白質によってマウスの群を免疫した。血清試料を予防接種後の異なる時間に採集し、特異的なIgG力価をELISAによって決定した。結果は、BLS−Stx2Bキメラが試験された全てのプロトコールにおいて外因性アジュバントなしでさえも免疫応答を活性化するということを示している(図8B)。一方で、DNA−蛋白質プライム・ブーストレジメンはStx2B特異的な抗体を遅く(lately)誘導したが(蛋白質ブーストの35日後)、この時点においてAHのBLS−Stx2B−L10と同じ力価に達した。アジュバント非存在下の蛋白質は、最大力価およびそれらに達する時間に関して最低の液性応答を誘導した。
プライム・ブースト免疫のためには、マウスに100μgのpCI−BLS−Stx2B−L10を0、14、および28日目に筋肉内(i.m.)経路によって後ろ足に注射し、次に最終回のi.p.ブースターを40日目に不完全FA中のBLS−Stx2B−L10によって実施した。
マウスを各免疫以前および15または30日毎に麻酔し、眼窩静脈叢の穿刺によって採血し、血清試料を得た。
予防接種マウスの血清試料中のStx2B特異的なIgGをELISAアッセイによって分析した。簡略には、96ウェルのMaxiSorpプレート(Greiner Bio-One, Frickenhausen, Germany)を0.5gの精製Stx2Bによってコーティングし、0.4%BSAのPBS−0.05%Tweenによって2時間、37℃でブロッキングした。次に、プレートを段階希釈したマウス血清と一緒に2時間、37℃でインキュベーションした。洗浄後に、プレートは、PBS−0.05%Tweenで希釈した(1:3000)ペルオキシダーゼ標識ヤギ抗マウスIgG(Zymed, Invitrogen, Carisbad, CA, USA)と一緒に37℃で1.5時間インキュベーションした。プレートを洗浄し、クエン酸/リン酸緩衝液中の2mg/mlのO−フェニレンジアミン(Sigma)および0.3%H2O2によって反応を現像した。反応を2MのH2SO4によって止め、492nmの吸光度をマイクロタイタープレートリーダーASYS UVM340(Biochrom Ltd., Cambridge, England)によって測定した。結果はエンドポイント力価として表し、免疫前血清試料の媒質±2×標準偏差(SD)よりも高い光学密度を有する最終希釈の逆数値として計算した。
抗体のサブタイピングおよび抗体の親和性
BLS−Stx2B−L10抗原は、分析された全てのプロトコールでIgG2aよりも高い(P<0.001)抗Stx2Bの特異的なIgG1力価を誘導した。ただしプライム・ブーストスケジュールでは、両方のIgGサブクラス間に重大な違いはなかった(図9)。IgGのサブタイピングのために、ELISAを上で記載された通りペルオキシダーゼ標識ヤギ抗マウスIgG1およびIgG2a(1:1000)(BD-Pharmigen, New Jersey, USA)を二次抗体として用いて行った。
FAまたはAHと一緒に製剤されたBLS−Stx2B−L10によって実施例14の通り免疫されたマウスの血清は、FA中の精製Stx2Bによって免疫されたマウスの血清よりもStx2Bに対する高い親和性を有する抗体を提示した。同様の親和性の結果が、表1Aのプライム・ブーストレジメンのアジュバントなしのBLS−Stx2B−L10によって免疫されたマウスから採集された血清で観察された。免疫マウスによって作られた抗体のStx2B親和性をチオシアン酸溶出ベースのELISAによって決定した。手順はStx2BのELISAについて記載したものと同様であったが、追加のステップを含んだ。正規化した濃度の血清と一緒のインキュベーションの後に、プレートを洗浄し、チオシアン酸アンモニウム(Anedra, San Fernando, Bs. As., Argentina)をPBSで希釈して、0〜4Mに亘る濃度でウェルに追加した。プレートを室温で15分静置した後に洗浄し、アッセイを続けた。結合した抗体の50%を解離させるのに必要なチオシアン酸アンモニウムの濃度を決定した。結合のパーセンテージを次の通り計算した。チオシアン酸アンモニウム存在下のOD492nm×100/チオシアン酸アンモニウム非存在下のOD492nm。
組み換えStx2(rStx2)に対する中和力価
Stx2中和抗体力価を決定するために、1CD50のrStx2(670pgのStx2、Vero細胞の50%を殺す細胞毒性量)と実施例14の通り免疫されたマウスの実験血清試料の段階希釈物とを、37℃で1時間、次に4℃で1時間プレインキュベーションした。104個のVero細胞を含む各ウェルに混合物を重層し、5%CO2中37℃で48時間インキュベーションした。細胞をPBSによって洗浄し、クリスタルバイオレット色素によって染色し、マイクロタイタープレートリーダー(Biochrom Ltd.)上で570nmフィルターによって読み取った。中和活性は、Vero細胞に対するStx2毒性の50%を遮断した最高希釈の逆数値として表した。
FAと一緒に製剤されたBLS−Stx2B−L10によって免疫されたマウスの全ての血清は、最高の中和力価を示した(P<0.001)。対照的に、単離されたStx2Bによって免疫されたマウスの血清は、FA中に製剤されたときであっても最低の中和活性を示した(表1)。
マウス血清の交差反応
この目的のために、Stx2またはStx2cもしくはStx2dバリアントを産生するEHECのヒト分離株の上清を、実施例14の通りBLS−Stx2B−L10+FAまたはStx2B+FAによって免疫されたマウスの血清と一緒にインキュベーションし、Vero細胞に対する毒性を評価した。本発明者は組み換えStx1(rStx1)に対する中和能力もまた評価した。BLS−Stx2B−L10によって免疫されたマウスの血清は、野生型Stx2およびそのバリアント、ならびにrStx1もまた強力に中和した。はっきりと対照的に、精製Stx2Bによって免疫した6匹のマウスのうち1匹から採取した血清試料のみが野生型Stx2を弱く中和でき、それらのいずれもStx2バリアントまたはrStx1を中和しなかった(表2)。
rStx2のチャレンジに対する免疫マウスの防御:エクスビボのrStx2中和活性
エクスビボアッセイのために、2.2ng/マウスのrStx2(100%致死量。1LD100)を、実施例14の通り免疫されたマウスの血清と一緒に37℃で1時間および4℃で1時間プレインキュベーションした。混合物を未処理の成体BALB/cマウスに静脈内(i.v.)接種し、生存率を観察した。この投与量のrStx2は、注射後96時間以内に100%の死亡率に至る。図10Aに示されている通り、異なるプロトコールのもとでBLS−Stx2B−L10によって免疫されたマウスから採取された血清と一緒のrStx2のインキュベーションによって、rStx2の毒性は完全に無効化された。一方で、Stx2B予防接種マウスの血清によってrStx2の毒性は予防されなかった。
rStx2による静脈内チャレンジと長期の応答
実施例14の通り免疫されたマウスを、最終回の免疫の50日後に1LD100のrStx2によるi.v.接種によってチャレンジした。図10Bは、BLS−Stx2B−L10予防接種マウスの100%およびStx2B予防接種マウスの33%がrStx2の致死量のチャレンジから生き残ったということを示している。用いたrStx2投与量によれば、BLSによって免疫された動物または非免疫マウスの0%がチャレンジから生き残った(図10B)。
生き残ったBLS−Stx2B免疫マウスの血清を長期採取して、特異的な抗体応答の持続期間を研究した(図11A)。全てのBLS−Stx2Bレジメンは長期持続的な免疫応答を誘導した(これはELISAの抗Stx2抗体として評価した)。マウスを、2LD100(4.4ng/マウス)のrStx2によって最終回の投与の8ヶ月後に、3LD100(6.6ng/マウス)によって最終回の投与の10ヶ月後に再チャレンジした(図11B)。全てのBLS−Stx2B−L10予防接種マウスは、最終回の免疫投与の8ヶ月後に2LD100によってチャレンジされたものおよび最終回の免疫投与の11ヶ月後の3LD100によるチャレンジから生き残った。
bls−stx2b−l10の1回投与による免疫によって作られた抗体の防御能力
BLS−Stx2B−L10蛋白質による免疫によって作られた抗体の防御能力をさらに分析するために、水酸化アルミニウム中に製剤されたBLS−Stx2B−L10の1回投与によって成体BALB/cマウスを免疫した(投与量は20μgのStx2Bと同等であった)。水酸化アルミニウムゲル(AH)懸濁液(1mg/ml)をBLS−Stx2B−L10と混合し、撹拌しながら室温で30分インキュベーションした。AH吸着抗原を10000gで10分遠心し、ペレットをPBS中に再懸濁した。血清試料を予防接種後の異なる時間に採集し、特異的なIgG力価をELISAによって決定した。予防接種マウスの血清試料中のStx2B特異的なIgGを実施例14に記載された通りELISAアッセイによって分析した。
図12は、BLS−Stx2B−L10の1回の投与が特異的なIgG液性応答を発生させるのに十分であるということを示している。
免疫応答の防御能力を試験するために、マウスを免疫の1ヶ月後に1LD100のrStx2によってチャレンジした。図13Aは、BLS−Stx2B−L10予防接種マウスの100%がrStx2の致死量のチャレンジから生き残った一方で、非免疫マウスの0%がチャレンジから生き残ったということを示している。
生き残ったマウスは、それぞれ免疫の2ヶ月後および免疫の4ヶ月後に3LD100および5LD100によって再チャレンジした。図13BおよびCは、全てのマウスが高い投与量のrStx2による両方のチャレンジから生き残ることができたということを示している。これは、BLS−Stx2B−L10による免疫によって作られた抗体の高い防御能力を示している。
Claims (42)
- 志賀毒素2サブユニットBのモノマーに対して少なくとも95%のアミノ酸配列同一性を有するペプチドを含み、前記のペプチドが直接的にまたはペプチドリンカーを介してBrucellaルマジンシンターゼのモノマーに融合されている、キメラ蛋白質。
- ペプチドが直接的にまたはペプチドリンカーを介してBrucellaルマジンシンターゼのモノマーのN末端に融合されている、請求項1に記載のキメラ蛋白質。
- Brucellaルマジンシンターゼのモノマーが、配列番号9に対して少なくとも95%の配列同一性を有する、請求項1または2に記載のキメラ蛋白質。
- Brucellaルマジンシンターゼのモノマーが、欠失と挿入との組み合わせが配列番号9に関して5アミノ酸よりも多くの伸長をもたらさないように、1つまたは2つ以上の置換、欠失、および/または挿入を最初の8つのN末端アミノ酸位置に有する、請求項1〜3のいずれか一項に記載のキメラ蛋白質。
- 志賀毒素2のサブユニットBのモノマーがペプチドリンカーによってBrucellaルマジンシンターゼのモノマーのN末端に連結される、請求項1〜4のいずれか一項に記載のキメラ蛋白質。
- ペプチドリンカーがG/Sフレキシブルペプチドである、請求項5に記載のキメラ蛋白質。
- ペプチドリンカーが2〜20アミノ酸の長さである、請求項5または6に記載のキメラ蛋白質。
- ペプチドリンカーが5〜15アミノ酸の長さである、請求項7に記載のキメラ蛋白質。
- 配列番号3、配列番号5、および配列番号7からなる群から選択されるアミノ酸配列を含む、請求項8に記載のキメラ蛋白質。
- 配列番号5のアミノ酸配列を含む、請求項9に記載のキメラ蛋白質。
- 請求項1〜10のいずれか一項に記載のキメラ蛋白質の五量体の二量体を含む、蛋白質オリゴマー複合体。
- 請求項1〜10のいずれか一項に記載のキメラ蛋白質をコードするポリヌクレオチド。
- 配列番号4、配列番号6、および配列番号8からなる群から選択されるヌクレオチド配列を含む、請求項12に記載のポリヌクレオチド。
- 配列番号6のヌクレオチド配列を含む、請求項13に記載のポリヌクレオチド。
- 請求項12に記載のポリヌクレオチドを含むベクター。
- 配列番号4、配列番号6、および配列番号8からなる群から選択されるヌクレオチド配列を含む、請求項15に記載のベクター。
