Abstract
Description
本発明は、ヒト多能性幹細胞を使用して、薬物によって誘導される催奇形のリスクを決定する方法に関する。
後期の薬物失敗を減少させるため、薬物によって誘導される毒性を予測するための改善されたインビトロ系が、製薬産業およびバイオテクノロジー産業によって必要とされている。具体的には、ヒト細胞を利用し、動物研究への依存を低下させることができる予測ハイスループットインビトロモデルが必要とされている。
(1)中内胚葉へ分化中の多能性幹細胞と化合物を接触させる工程;
(2)Sox17、EOMES、およびT-ブラキュリなどの1種以上の発生的に制御されるマーカーのタンパク質発現を測定することによって、胚発生の進行を検出する工程;
(3)該1種以上のマーカーのタンパク質発現の50%の阻害をもたらす該化合物のIC50濃度を決定する工程;ならびに
(4)評価している該化合物の催奇形リスクを確立するため、工程(3)において決定されたIC50濃度を、(a)公知の催奇形リスクを有する1種以上の化合物のIC50濃度および(b)公知の非催奇形リスクを有する1種以上の化合物のIC50濃度と比較する工程。
本明細書において使用されるように、移行句においても、または特許請求の範囲の本文においても、「含む(comprise)」および「含む(comprising)」という用語は、無制限の意味を有していると解釈されなければならない。即ち、その用語は、「少なくとも有する」または「少なくとも含む」という語句と共に同義的に解釈されなければならない。過程に関して使用された時、「含む」という用語は、その過程が、示された工程を少なくとも含むが、付加的な工程を含んでいてもよいことを意味する。化合物または組成物に関して使用された時、「含む」という用語は、その化合物または組成物が、示された特色または成分を少なくとも含むが、付加的な特色または成分も含んでいてよいことを意味する。

本発明の方法は、定方向分化中の処理された多能性幹細胞と対照多能性幹細胞との間で、中内胚葉形成のレベルを比較することによって、薬物によって誘導される催奇形のリスクを評価する。従って、ヒト多能性幹細胞の分化を中内胚葉へ差し向けるための最適条件を最初に確立しなければならない。概念実証として、本明細書の実施例に記載されたプロトコルを使用して、H9ヒト胚性幹細胞株を使用して、初期検証を実施した。
第一に、3日間の分化において40〜60%の中内胚葉形成を再現性よく達成する最適の初期細胞播種密度および増殖因子の濃度を同定した(図2を参照のこと)。中内胚葉を生成するための増殖因子処理の理想的な濃度および期間の軽微なプロトコル最適化によって、当業者は、大部分のヒト多能性細胞株で使用するためにこの方法を適応させることができる。方法は、薬物によって処理された試料と対照試料との間の中内胚葉形成の比率の比較に依存するため、40〜60%の分化がこれらの研究のための出発点として選ばれたが、効率がウェル間で一貫している限り、中内胚葉細胞の正確な割合は重要ではないはずである。
定方向分化のための条件を最適化した後、胎児への害を引き起こさないことが公知の化合物からの、公知の催奇形性化合物の層別化を可能にする用量応答関係を確立する。初期研究のため、本発明者らは、公知のインビボ効果を有する、30種の市販の化合物および40種の専有の化合物のパネルを使用した。中内胚葉へ分化中の細胞を含有している個々のウェルを、1日目および2日目に処理した(図3を参照のこと)。一枚のプレートにおいて、増加する濃度の各化合物を特定のウェルに適用した。さらに、各プレートは、陽性対照として、催奇形性であることが公知の化合物によって処理されたウェルを含有し、陰性対照として、溶媒によってのみ処理されたウェルも含有していた(図5のプレートマップの例を参照のこと)。
関心対象の多能性細胞株を使用して、催奇形リスクを測定するための所望の閾値が確立された後は、アッセイをよりハイスループットのリスク評価のために利用することができる。許容される予測性のレベルに相関する単一の濃度で、トリプリケートに、化合物を試験することができる。

組織培養プレートのマトリゲルコーティング:1:80希釈を使用してマトリゲルによって96ウェルをコーティングする
多能性幹細胞の調製:
細胞培養。H9ヒトES細胞(WiCell)を10uM Y-27632を含むmTeSR培地で解凍した。3百万個の細胞を、ES細胞最適化マトリゲル(BD Biosciences)によってあらかじめコーティングされた2枚の100mm組織培養等級ディッシュに播種した。Y-27632は播種の間にのみ必要とされ、播種後24時間以内に除去されるべきである。mTeSR培地を毎日交換した。細胞は3〜4日以内にほぼコンフルエントになるはずである。
細胞が約50%コンフルエントになった時、培地を除去し、PBSで濯ぎ、次いで、100mmプレート1枚当たり2mlのアキュターゼを添加した。細胞を3分以内に剥離するべきである。
直ちに10uM Y-27632を含む8mlのmTeSRを添加し、(2×100mmプレートからの)20mlを50mlチューブへ移した。6分間400×gでスピンした。上清を除去した。Y-27632を含む10mlのmTeSRを添加した。細胞ペレットを懸濁させるために上下にピペッティングした。生細胞数を計数し、百万細胞/mlで再懸濁させた。5000〜50,000細胞/96ウェルで細胞を播種した。具体的な細胞株および細胞のロットによって、最適の細胞密度を達成するため、この数は変動し得る。これらの実験については、15,000細胞/ウェルを播種した。2〜5日間、毎日、mTESR培地を交換した。
0日目。薬物を含む分化培地Iによる分化開始
細胞を96ウェルプレートに均等に分配しなければならない。細胞が96ウェルプレートのウェル内で約35%コンフルエンシーになった時(2〜5日後)、分化を開始した。培地を除去し、Advanced RPMIで4〜5回洗浄した。mTESRは、細胞を未分化状態に維持する様々な因子を極めて高濃度で含有しているため、mTESRを完全に洗浄除去することは、重要である。試験化合物を含むかまたは含まないDE分化培地Iを添加した。3分間500rpmでプレートをボルテックスした。24時間プレートを放置した。
プレートをAdvance RPMIで3回迅速に(およそ10分)洗浄した。化合物を含むかまたは含まないDE分化培地IIを添加した。3分間500rpmでプレートをボルテックスした。48時間プレートを放置した。細胞を、合計72時間、薬物によって処理したことに注意すること。
1. 96ウェルプレートから培地を除去した。上部の細胞片または死細胞を除去するため、DPBSによって温和に3回ウェルを洗浄した。37℃に予熱されたPBSで新鮮に調製された3%ホルムアルデヒドを添加した。これを細胞に15分間添加した。長い固定は、高いバックグラウンドまたは抗原改変を引き起こす可能性がある。PBSを除去するため、PBSで3回洗浄した。
2. 15分間0.1%トリトンX-100を含むPBSによって細胞を透過性化した。
3. 96ウェル当たり30ulのImageIT-FX溶液を適用した。700rpmで1分間ボルテックスした。室温で30分間インキュベートした。
4. ブロッキング緩衝液(10%ロバ血清、0.3%トリトンX-100、1%BSAを含むPBS)で1回洗浄し、次いで、ブロッキング緩衝液を添加し、20分間放置した。
5. ブロッキング緩衝液中のSox17抗体(1:10希釈)を添加した。室温で3時間インキュベートした。
6. 1%BSAを含むPBSで4回洗浄した。
7. DAPIまたはヘキスト33242(いずれも5ug/mlの最終濃度)を含む1%BSAを含むPBSを5分間添加した。後にDAPIを含むSlowFadeを使用する場合には、この手順は必要ない。
8. 各96ウェルに30ulのDAPIを含むSlowFadeを添加した。
Envisionプレートリーダによって557nmにおける蛍光強度を測定した。
上記実施例は、ヒト胚性幹細胞H9(Wisconsin Alumi Research Foundation、WARF)に適用される本発明を例示したものである。他の多能性幹細胞株へ方法を適用する時には、初期細胞播種密度、培地中の増殖因子の濃度、および分化の期間などの、株依存性の分化効率に影響する典型的な変数を決定しなければならない。上記のこの最適化過程において、本発明者らは、催奇形リスクの予測のための最適条件を同定するため、中内胚葉形成の効率、およびサリドマイドによって媒介される薬物効果をモニタリングした。
Claims (21)
- 以下の工程を含む、化合物の催奇形リスクを評価する方法:
(1)中内胚葉へ分化中の多能性幹細胞と化合物を接触させる工程;
(2)Sox17、EOMES、およびT-ブラキュリからなる群より選択される1種以上の中内胚葉マーカーのタンパク質発現を測定する工程;
(3)該1種以上のマーカーのタンパク質発現の50%の阻害をもたらす該化合物のIC50濃度を決定する工程;ならびに
(4)評価している該化合物の催奇形リスクを確立するため、工程(3)において決定されたIC50濃度を、(a)公知の催奇形リスクを有する1種以上の化合物のIC50濃度および(b)公知の非催奇形リスクを有する1種以上の化合物のIC50濃度と比較する工程。 - 多能性幹細胞が霊長類起源のものである、請求項1に記載の方法。
- 多能性幹細胞が胚性幹細胞である、請求項1〜2のいずれか一項に記載の方法。
- 多能性幹細胞がH9ヒト胚性幹細胞である、請求項1〜3のいずれか一項に記載の方法。
- 多能性幹細胞が誘導多能性幹細胞である、請求項1〜2のいずれか一項に記載の方法。
- 前記中内胚葉マーカーの少なくとも1種がSOX17である、請求項1〜5のいずれか一項に記載の方法。
- 前記中内胚葉マーカーの少なくとも1種がEOMESである、請求項1〜6のいずれか一項に記載の方法。
- 前記中内胚葉マーカーの少なくとも1種がT-ブラキュリである、請求項1〜7のいずれか一項に記載の方法。
- 測定される中内胚葉マーカーが、SOX17のタンパク質発現のみである、請求項1〜5のいずれか一項に記載の方法。
- 測定される中内胚葉マーカーが、SOX17およびEOMESのタンパク質発現のみである、請求項1〜5のいずれか一項に記載の方法。
- 測定される中内胚葉マーカーが、SOX17およびT-ブラキュリのタンパク質発現のみである、請求項1〜5のいずれか一項に記載の方法。
- タンパク質発現が免疫組織化学によって測定される、請求項1〜11のいずれか一項に記載の方法。
- タンパク質発現がフローサイトメトリーによって測定される、請求項1〜11のいずれか一項に記載の方法。
- タンパク質発現が蛍光顕微鏡検査によって測定される、請求項1〜11のいずれか一項に記載の方法。
- 多能性幹細胞の40〜60%が中内胚葉へ分化した後、工程(1)が実施される、請求項1〜14のいずれか一項に記載の方法。
- 工程(4)における公知の催奇形リスクを有する1種以上の化合物が、アクチノマイシンD、トレチノイン、シタラビン、ノコダゾール、ロテノン、トレチノイン、イソトレチノイン、ブロモデオキシウリジン、ドキソルビシン、ドルソモルフィン、サリドマイド、5-フルオロウラシル、ダサチニブ、ソラフェニブ、バルプロ酸、スニチニブ、ジプラシドン、ミアンセリン、バンデタニブ、ジエチルスチルベストロール、6-アミノニコチンアミド、リタンセリン、およびゲフィチニブからなる群より選択される、請求項1〜15のいずれか一項に記載の方法。
- 工程(4)における公知の催奇形リスクを有する1種以上の化合物が、サリドマイド、ミアンセリン、およびリタンセリンからなる群より選択される、請求項1〜15のいずれか一項に記載の方法。
- 工程(4)における公知の非催奇形リスクを有する1種以上の化合物が、エソメプラゾール、葉酸塩、カテキン、リスリド、ナイアシン、アスピリン、イブプロフェン、アシクロビル、ケタンセリン、ストレプトマイシン、メチルドパ、サッカリン、カフェイン、ペニシリン、およびテガセロッドからなる群より選択される、請求項1〜17のいずれか一項に記載の方法。
- 工程(4)における公知の非催奇形リスクを有する1種以上の化合物が、葉酸塩およびメチルドパからなる群より選択される、請求項1〜17のいずれか一項に記載の方法。
- 工程(4)における公知の催奇形リスクを有する1種以上の化合物が、サリドマイド、ミアンセリン、およびリタンセリンからなる群より選択され、かつ公知の非催奇形リスクを有する1種以上の化合物が、葉酸塩およびメチルドパからなる群より選択される、請求項1〜15のいずれか一項に記載の方法。
