Abstract
Description

A.本開示の1つまたは複数のリポカリンムテイン

研究の初めに、マウスの右側腹部に100μlの5×106 Caki-1細胞を、皮下に(s.c.)移植した。すべてのマウスを毎日観察して、健康および腫瘍サイズをモニタリングした。腫瘍が100〜150 mm3の平均容積に達した際に、96匹のマウスのうち80匹を、その腫瘍容積に基づいて選択し、投薬前に有効性群に無作為に割り当てた。各群は、10匹の腫瘍を有するマウスからなった(n=10/群)。処置群中の腫瘍を有するマウスを、無作為化後の日から開始して、媒体、TLC対照-PEG40(30mg/kg)、ならびにリポカリンムテイン(30mg/kg、15mg/kg、7.5mg/kg、2.5mg/kg、および0.75mg/kg)で(毎日腹腔内に(i.p.))処置した。マウスを、各投薬時に計量し、毎日記録した。図1は、Caki-1異種移植腫瘍モデルにおける、リポカリンムテイン(30 mg/kg、15 mg/kg、7.5 mg/kg、2.5 mg/kg、および0.75 mg/kg)群の腫瘍増殖阻害を示す。腫瘍容積の統計学的に有意な低減が、媒体群およびTLC対照と比較した際に、リポカリンムテイン処置群の各々において観察された。
Caki-1腫瘍を有するヌードマウスの体重を、リポカリンムテイン(SEQ ID NO: 1)を含む試験化合物の投与による変動についてモニタリングした。腫瘍を有するマウスを、媒体、TLC対照-PEG40(30mg/kg)、ならびにリポカリンムテイン(30mg/kg、15mg/kg、7.5mg/kg、2.5mg/kg、および0.75mg/kg)で(毎日腹腔内に(i.p.))処置した。体重を毎日記録し、すべてのマウスを毒性の徴候についてモニタリングした。すべての群が、(恐らく適応ストレスにより)処置の最初の週に類似したレベルの体重の減少を経験したが、その後、研究を通して体重が増加した。いかなる毒性の徴候も観察されなかった。図2に示されるように、媒体対照群とリポカリンムテイン群との間で体重パターンに差異は無く、化合物が試験されたすべての用量レベルで十分に耐容性を示すことを示唆した。
リン酸化Metおよび全MetのELISAベースの定量化を、媒体またはリポカリンムテイン(SEQ ID NO: 1)での処置の5日後に、Caki-1およびEBC-1細胞異種移植モデルから切除した腫瘍組織に対して行った。リポカリンムテインは、両方のリガンドに依存しないCaki-1およびEBC-1の細胞異種移植モデルにおいて、リン酸化Metおよび全内在性c-Metを有意に低減させることが、図3において実証された(P<0.01)。

リポカリンムテイン(SEQ ID NO: 1)の内部移行を評価するために、A549細胞に対して免疫蛍光顕微鏡検査を行った。カバーガラス上にプレーティングしたA549細胞を、SEQ ID NO: 1 Dylight633(500 nM)と共に1時間氷上でインキュベーションした。細胞を次に、1時間37℃でインキュベーションして、内部移行させた。核を、DAPIで青色に対比染色した(これは、各細胞内で大きな青色の円として視覚化され、単一細胞解析を手助けする)。免疫蛍光を、Leica TCS SP2 AOBS共焦点レーザー走査顕微鏡(Leica Microsystems)を用いて解析した。図5Aにおいて観察される強いピンク色の細胞内染色により示されるように、リポカリンムテイン(SEQ ID NO: 1)がc-Met受容体に結合し、かつ内部移行されることが、実証された。
H441腫瘍を有するマウスのmicroPET画像化を使用して、腫瘍特異的な89Zr-SEQ ID NO: 1の取り込みおよび時間に依存した器官分布を評価した。動物を、microPET Focus 220げっ歯類走査装置(CTI Siemens)を用いて画像化した。画像再構築後、インビボ定量化を、AMIDE Medical Image Data Examinerソフトウェア(バージョン0.9.1、Stanford University)で行った。試料およびプライミングされた標準を、ウェルタイプのガンマカウンターにおいて放射活性を計数し、物理的崩壊について補正した。エクスビボの組織活性を、1グラムの組織当たりの注射された用量のパーセンテージ(%ID/g)として表す。代表的な横断および冠状のmicroPET画像を、図6(A)においてトレーサー注射(50μg 89Zr- SEQ ID NO: 1(5 MBq))の6、24、48、および96時間後に示し、白色矢印が腫瘍取り込みを示す。腫瘍取り込みは、6〜24時間の間に増加した。microPETデータの定量化を、すべてのマウスにおいて血液プール、肝臓、脾臓、腎臓、および腫瘍取り込みについて行った。異種移植マウスにおける非標的組織のmicroPET走査解析により、50μg 89Zr-SEQ ID NO: 1の注射の6時間後に、高い血液プール、ならびに肝臓および腎臓のトレーサー取り込みが明らかになった。これらの器官における非特異的な取り込みは、注射の24、48、および96時間後に行われた走査に基づくと時間と共に減少し、トレーサー注射の96時間後までに増大した腫瘍:器官の比を結果としてもたらした(図6B)。特異的なc-Metに向けられた取り込みはまた、89Zr-Tlc対照と比較してH441およびU87-MG腫瘍においても見られた(図7)。A2780異種移植(c-Met陰性)により、89Zr-SEQ IDおよび89Zr-Tlc対照の両方のバックグラウンドの非特異的腫瘍取り込みが示された。データを、平均標準取り込み値(SUV平均)として表す。

対照試料中の検出可能なシグナル伝達標識の量に対する、対象から採取した試料中の検出可能なシグナル伝達標識の量の増大が、対象におけるc-Met発現の量の増大を示し、かつ対象におけるMetの量の増大が、対象におけるc-Metを発現する癌または腫瘍の進行を示唆し;かつ
対照試料中の検出可能なシグナル伝達標識の量に対する、対象から採取した試料中の検出可能なシグナル伝達標識の量の減少が、対象におけるc-Metの量の減少を示し、かつ対象におけるMetの量の減少が次に、対象におけるc-Metを発現する癌または腫瘍の退縮を示唆する、方法。

Claims (40)
- (i)c-Metに特異的なリポカリンムテインと、(ii)検出可能なシグナル伝達標識と、(iii)半減期延長部分とを含む、c-Metの存在を検出するための免疫画像化技術における使用に適した診断用組成物であって、
対象において、検出可能なシグナル伝達標識により視覚化されるような画像化結果を得るために必要とされる半減期を有する、診断用組成物。 - 対象において数時間から6日の範囲内の半減期を有する、請求項1に記載の診断用組成物。
- 対象において少なくとも約1、2、4、6、7、14、または21日の半減期を有する、請求項2に記載の診断用組成物。
- 検出可能なシグナル伝達標識が、活性を有するかまたは活性化可能である、請求項1〜3のいずれか一項に記載の診断用組成物。
- 診断上許容される担体および/または賦形剤をさらに含む、請求項1〜4のいずれか一項に記載の診断用組成物。
- c-Metを発現する細胞の表面から内部移行され得る、請求項1〜5のいずれか一項に記載の診断用組成物。
- 血清半減期延長部分が、対象において組成物に、画像化結果を得るために必要とされる時間に適合した半減期を与えることができる、請求項6に記載の診断用組成物。
- 画像化結果が腫瘍対非腫瘍の比である、請求項7に記載の診断用組成物。

- 検出可能なシグナル伝達剤が、ガンマカメラ画像形成剤(imageable agent)、PET画像形成剤、およびMRI画像形成剤からなる群より選択される、請求項1〜12のいずれか一項に記載の診断用組成物。
- 検出可能なシグナル伝達剤が、放射性核種、蛍光剤、発蛍光団、発色団、色原体、リン光剤、化学発光剤、および生物発光剤からなる群より選択される、請求項1〜12のいずれか一項に記載の診断用組成物。

- (a)対象の第1のコホートに、特定の半減期延長部分を含有する診断用組成物を投与する段階;
(b)対象の第2のコホートに、第1のコホートに関連する半減期延長部分とは異なる特定の半減期延長部分を含有する診断用組成物を投与する段階;
(c)それぞれの診断用組成物の半減期を測定する段階;および
(d)コホート中の対象と同じ種または異なる種由来の対象において画像化結果を得るために必要とされる時間に最も適した半減期延長部分を選択する段階
を含む、請求項1〜17のいずれか一項に記載の診断用組成物を選択するための方法。 - (a)細胞、組織、器官、または試料を、請求項1〜17のいずれか一項に記載の診断用組成物と接触させる段階;および
(b)器官または生物学的試料がc-Metを過剰発現しているか否かを判定する段階
を含む、細胞の表面上、細胞中、組織中、器官中、または生物学的試料中のc-Metの過剰発現の存在を検出するための方法。 - (a)対象から採取した生物学的試料を請求項1〜17のいずれか一項に記載の1つまたは複数の診断用組成物と接触させる段階;
(b)本開示の免疫画像化技術により、試料に結合した検出可能なシグナル伝達標識の量を測定する段階;および
(c)試料に結合した検出可能なシグナル伝達標識の量を対象におけるc-Metの量と相関させる段階
を含む、癌または腫瘍を有する対象においてc-Metを発現する癌または腫瘍の進行または退縮を判定する方法であって、
対照試料中の検出可能なシグナル伝達標識の量に対する、対象から採取した試料中の検出可能なシグナル伝達標識の量の増大が、対象におけるc-Met発現の量の増大を示し、かつ対象におけるMetの量の増大が、対象におけるc-Metを発現する癌または腫瘍の進行を示唆し;かつ
対照試料中の検出可能なシグナル伝達標識の量に対する、対象から採取した試料中の検出可能なシグナル伝達標識の量の減少が、対象におけるc-Metの量の減少を示し、かつ対象におけるMetの量の減少が次に、対象におけるc-Metを発現する癌または腫瘍の退縮を示唆する、方法。 - c-Metの過剰発現に関連する対象において疾患または状態を診断、予後判定、モニタリングするための、請求項1〜17のいずれか一項に記載の少なくとも1つの診断用組成物と、診断用組成物を使用するための1つまたは複数の指示書とを含む、診断用キット。
- リポカリンムテインの治療的有効量を、それを必要とする対象に投与する段階を含む、c-Metの活性化に関連する障害を処置、改善、または予防する方法における使用のためのリポカリンムテインであって、c-Metのシグナル伝達経路を遮断することができるかまたは遮断に寄与する、リポカリンムテイン。

- HGFのc-Metへの結合を含むc-Metシグナル伝達を遮断することができるかまたは遮断に寄与する、請求項22に記載の使用に従うリポカリンムテイン。
- HGFのアンタゴニストの治療的有効量を対象に投与する段階をさらに含む、請求項22に記載の使用に従うリポカリンムテイン。
- c-Met分子への結合時にc-Metを発現する細胞の表面から内部移行され得る、前記請求項のいずれか一項に記載の使用に従うリポカリンムテイン。
