Description

最後に、IBD、特にクローン病の診断および治療上の検査で有用な、一連の差次的に発現されたmiRNAが提供される。
IBDの発症傾向を診断する方法
特定のマイクロRNA分子がIBDを有する患者では差次的に発現されるという発見は、そのような疾患の発症傾向を診断または検出する方法を提供する。図1に示した、他の差次的に発現されたmiRNAの発現レベルは、対象の生体サンプルで評価することができる。1つの実施形態では、少なくとも1、2、3、4、5、6、7、8、9、10、またはそれ以上のmiRNAの発現レベルが測定される。特定の実施形態では、発現量の差が最も大きく異なるmiRNAの発現が測定される。特定の実施形態では、前記患者から単離されたmiRNAの発現レベルを、事前にCDの存在を示すと判断されたレベルと比較している。特定の実施形態では、前記IBD関連miRNA濃度を患者自身のmiR−150および/またはmiR−342−3p(内部標準miRNA)濃度と比較し、それによって対照サンプルと比較する必要がなくなる。これに限定されるものではないが、体液(血清、胃腸液、および痰など)、すべての細胞タイプ(腸細胞、白血球、単核細胞など)、または体組織を含むあらゆるタイプの生体サンプルで、miRNA発現レベルを検出するアッセイを行うことができる。好ましい実施形態では、miRNA濃度が血清サンプルで評価される。
特定のマイクロRNA分子がIBDを有する患者では差次的に発現されるという発見は、そのような疾患の発症傾向を診断または検出する方法を提供する。図1に示した、他の差次的に発現されたmiRNAの発現レベルは、対象の生体サンプルで評価することができる。1つの実施形態では、少なくとも1、2、3、4、5、6、7、8、9、10、またはそれ以上のmiRNAの発現レベルが測定される。特定の実施形態では、発現量の差が最も大きく異なるmiRNAの発現が測定される。特定の実施形態では、前記患者から単離されたmiRNAの発現レベルを、事前にCDの存在を示すと判断されたレベルと比較している。特定の実施形態では、前記IBD関連miRNA濃度を患者自身のmiR−150および/またはmiR−342−3p(内部標準miRNA)濃度と比較し、それによって対照サンプルと比較する必要がなくなる。これに限定されるものではないが、体液(血清、胃腸液、および痰など)、すべての細胞タイプ(腸細胞、白血球、単核細胞など)、または体組織を含むあらゆるタイプの生体サンプルで、miRNA発現レベルを検出するアッセイを行うことができる。好ましい実施形態では、miRNA濃度が血清サンプルで評価される。
治療薬を開発するため、miRNAの調節を利用する方法
本発明者は、数種類のmiRNA分子がIBD患者で差次的に発現され、これは血清サンプルで検出可能であることを発見した。特に、これらのmiRNA分子は炎症の調節および制御に関与する様々なタンパク質と相同性を示すため、様々な異なる医学的疾患で観察される異常炎症経路を抑制または軽減する治療薬として利用することができる。そのような試薬は、IBDなどの炎症性疾患を含む様々な疾患の治療に有用である。
本発明者は、数種類のmiRNA分子がIBD患者で差次的に発現され、これは血清サンプルで検出可能であることを発見した。特に、これらのmiRNA分子は炎症の調節および制御に関与する様々なタンパク質と相同性を示すため、様々な異なる医学的疾患で観察される異常炎症経路を抑制または軽減する治療薬として利用することができる。そのような試薬は、IBDなどの炎症性疾患を含む様々な疾患の治療に有用である。

別の実施形態では、前記差次的に発現されたmiRNAが腸の炎症プロセスに関与していることを特定すれば、そのようなmiRNAの発現濃度が変化した実験マウス系を作成することができる。これらのmiRNA分子を発現したトランスジェニックマウスは、IBDの発症およびIBDへの進行における前記miRNAの役割を検討するためのモデル系を提供する。実験マウスにトランス遺伝子を導入する方法は当業者に周知である。一般的な方法は3つあり、1.初期胚に対象の外来遺伝子をコードするレトロウイルスベクターを組み込む、2.新たに受精した卵の前核にDNAを注入する、および3.初期胚に遺伝子操作した胚幹細胞を組み込むなどである。上述のトランスジェニックマウスの作成は、標的タンパク質が様々な細胞炎症プロセスに果たしている役割の分子的解明を促す。そのようなマウスは、動物モデル全体で想定される治療薬を研究するin vivoスクリーニングツールを提供し、本発明に含まれる。
差次的に発現されたmiRNAまたはその模倣体を有する医薬品を、例えば、経口または静脈内(i.v.)、筋肉内、皮下、眼窩内、鼻腔内、関節包内、腹腔内(i.p.)、大槽内、気管内(i.t.)、または関節内などの非経口、または受動または促進吸収を含む様々な経路で被験者に投与することができることは、当業者の理解するところである。同じ投与経路を、例えば、上述の小さな分子、核酸分子、ペプチド、抗体、およびポリペプチドなどの他の薬学的に有用な化合物に利用することができる。
望ましい場合は、そのような差次的に発現されたmiRNAまたは模倣体を有する医薬組成物もリポソーム、ミクロスフェア、微小気泡、または他のポリマー基質に組み込むことができる(Gregoriadis, Liposome Technology, Vols. I to III, 2nd ed., CRC Press, Boca Raton Fla. (1993))。例えば、リン脂質または他の脂質から成るリポソームは、作成および投与が比較的簡単な、薬学的に許容でき、代謝可能な非毒性担体である。
