Abstract

免疫系は、原発性癌細胞および転移性癌細胞の両方を含む腫瘍細胞を殺すように機能し得ることが十分に証明されている。確かに、免疫系が腫瘍性癌細胞の存在を認識する証拠は、腫瘍組織において浸潤するリンパ球の存在によって支持されている(Haskillら、Contemp.Top.Immunobiol.8:107−170(1978);VoseおよびMoore,Semin.Hematol.22:27−40(1985))。免疫細胞の存在にも関わらず、腫瘍は、しばしば打ち勝ち、そして生存するだけでなく、無制限の増殖を有して遠位部位に転移する。

ヒトDCの培養は、本明細書中の前に記載したように、および米国特許第5,788,963号(本明細書中に参考として援用される)中に記載されたように確立した。簡単に言うと、末梢血単核細胞(PBMC)を、Ficoll−Paque(Pharmacia,Uppsala,Sweden)上での標準的な遠心分離によってリンパ球に富む「軟膜」から得た。プラスチックに接着したPBMC(37℃で約1時間)を、2% 自己血清、50U/ml ペニシリン、50μg/ml ストレプトマイシン、2mM L−グルタミン、10mM HEPES、0.1mM 非必須アミノ酸、および1mM ピルビン酸を補充または補充していないAIM−V(「培養培地」と称する;これらは全てBoehringer Ingelheim,Biowhittaker,Verviers,Belgiumから)中、1000U/mlまたは500U/mlの各顆粒球マクロファージ−コロニー刺激因子(GM−CSF)(LeucomaxTM 1.11×107U/mg(Novartis,Basel,Switzerlandより))および500U/mlのインターロイキン−4(IL−4)(Schering−Plough Research Institute,Kenilworth,NJ)の存在下で、6〜7日間培養した。
表面抗原(Ag)発現を決定するために、細胞(50μl中105DC)を、完全培地中の一次モノクローナル抗体(mAb)を用いて標識し、その後、ヤギ抗マウスIgのFITC結合体化F(ab’)2フラグメント(Dako,Glostrup,Denmark)によって標識した。以下のモノクローナル抗体が使用され得るが、同じ特異性を有する多数の他の抗体が、周知である:G46−2.6(IgG1、抗HLA−ABC)、L243(IgG2a、抗HLA−DR)、HB−15a(IgG2b、抗CD83)、BU63(IgG1、抗CD86)。洗浄は、0.2% アルブミンを含むHBSS中であった。最後の洗浄後、細胞を、0.2% アルブミンおよび2% ホルムアルデヒドを含むHBSS中で保存した。サンプルを、FACScan(登録商標)(Becton−Dickinson,San Jose,CA)で分析した。データを、Becton Dickinson製のCellQueste(登録商標)ソフトウェアを用いて分析し、そして表示させた。
以下のように、BCGおよびLPSを使用して、DCのMHCクラスI負荷を刺激した:1〜100μgのLNCaP由来のPSMAまたは部分的に精製された組換えPSMA(rPSMA)を、滅菌マクロチップを用いたピペッティングによって6日目のDCの培養培地に添加した。同じときに、BCG(0.2〜1.6×106U/ml 終濃度;Tice−BCG,Organon Teknika,Durham,NC)およびLPS(40U/ml 終濃度)(複製のフラスコ中)を、同じ方法を用いて培養培地に添加した。次いで、CO2インキュベーターに戻す前に、培養培地を穏やかに混合した。
本実施例は、樹状細胞が、低温保存後にその機能を保持しているか否かを試験する。免疫療法アプローチは複数の処置を含むので、この特性は特に重要であり、そして各患者についての全てのDCを一度の調製の間に調製し、そして抗原を負荷し、次いで、その後の注入のために等分および低温保存することが好ましい。DCの凍結および解凍は、CD8+ T細胞アクチベーターとしてのそれらの効果を制限し得ることが可能であった。
GM−CSFおよび500U/ml IL−4の存在下で、7日間培養した。7日目に、単離されたDCを収集し、そして90%ウシ胎仔血清および10%ジメチルスルホキシドを用いて低温保存した。低温保存されたDCを、その後一定期間凍結させて保存し、37℃の水浴で解凍し、そして15mlポリプロピレンチューブに移し、そして1200rpmで5分間遠心分離した。次いで、解凍したDCを、10%熱不活性化ヒト血清を含む培地に再懸濁し、そして計数し、そして以下のように使用した。
この実施例では、癌患者から単離された樹状細胞を単離し、そして種々の濃度のBCGで処理した。数日の培養後、DCを、1)飲細胞運動により粒子を取り込む能力について、ならびに2)HLA−DR、CD86、CD40、CD83、CD80、およびHLA−クラスIを含む、特定の樹状細胞成熟マーカーの表面発現について、試験した。
