The present invention relates to a catechol-degrading enzyme and a method for decomposing catechol in wastewater using the enzyme. Furthermore, the present invention relates to a catechol-degrading enzyme capable of decomposing catechol contained in the aqueduct discharged from a coke oven at a steel mill, and a method for decomposing the catechol in the aquatic water using the enzyme.
Aqueous water generated from a coke oven at an ironworks contains environmentally undesirable components such as phenols, thiocyanate and thiosulfuric acid, and a high concentration of ammonia. In the current water safety treatment system, organic substances such as phenols that are chemical oxygen demand (COD) components and sulfur compounds such as thiocyanate and thiosulfuric acid are removed by an activated sludge method (for example, Non-Patent Document 1). reference).
Among the microorganisms constituting the activated sludge that treats the low water, there are microorganisms that can decompose phenols, thiocyanate, thiosulfuric acid, and the like (see, for example, Patent Documents 1 and 2). In microorganisms that decompose these COD components, enzymes produced by the microorganisms are directly involved in the decomposition. However, it has not been clarified what enzymes are involved in the decomposition of phenols, thiocyanate, thiosulfate, etc. contained in the water.
The COD component contained in the highest concentration in the safety water discharged from the coke oven is phenol. This degradation of phenol is thought to be mainly due to oxidative degradation by aerobic microorganisms, and phenol is first converted to catechol, which is an intermediate reaction product, after initial oxygenation by hydroxylase. (See, for example, Non-Patent Document 2).
In addition, the reaction for cleaving the aromatic ring of catechol is generally considered to be a reaction that is difficult to proceed. In the oxidative degradation reaction of phenol, the cleavage reaction of the aromatic ring of catechol is considered to be rate-limiting. As an enzyme that degrades catechol, for example, 3-methylcatechol dioskinase (TodE) is known.
However, in the treatment of safe water, which has a special composition containing high concentrations of ammonia, sulfur compounds, etc., enzymes currently involved in the degradation of phenol produced by microorganisms in activated sludge responsible for the safe water treatment are currently available. The details of the catechol-degrading enzyme that catalyzes the reaction of cleaving the aromatic ring of catechol in the water are not known at all.
Therefore, in the present invention, a catechol-degrading enzyme capable of decomposing catechol even in low-pressure water having a special composition discharged from a coke oven can be isolated and used from the water-activated activated sludge, and further, the catechol-degrading enzyme An object is to provide a wastewater treatment method using a slag.
Therefore, the present inventors perform metagenomic analysis of activated sludge for treating aqueduct, and encode an enzyme involved in the degradation of catechol, which is a common intermediate reaction product in the degradation reaction of phenol in the aquatic treatment, and the enzyme. As a result of intensive studies on the DNA base sequence, the inventors have found a plurality of novel catechol-degrading enzymes that can be used for the treatment of aquatic water in the aquatic activated sludge, succeeded in using them, and completed the invention.

That is, the features of the present invention are as follows.
(1) A catechol-degrading enzyme encoded by DNA having a base sequence represented by any one of SEQ ID NOs: 1 to 43.

(4) The method for decomposing catechol in wastewater according to (2) or (3), wherein the wastewater containing the catechol is low-grade water discharged from a coke oven.
According to the present invention, it becomes possible to promote degradation of catechol contained in wastewater by an enzymatic reaction using a novel enzyme that has not been conventionally known.
In particular, the low water discharged from the coke oven contains a high concentration of phenol. By using the catechol-degrading enzyme of the present invention and the method for decomposing catechol in wastewater using the enzyme, a special composition is obtained. Even in the aquatic water, the decomposition of catechol can be promoted efficiently.
The present inventors have succeeded in elucidating the base sequence of DNA encoding a novel catechol-degrading enzyme from environmental DNA (metagenome) collected from activated water sludge treated with water. Furthermore, using the DNA of this base sequence, a novel catechol-degrading enzyme was produced by the host microorganism, and the catechol in the waste water was also successfully degraded.
First, a method for obtaining the catechol-degrading enzyme of the present invention will be described. The DNA of the base sequence shown in any one of SEQ ID NOs: 1 to 43 encoding the catechol-degrading enzyme of the present invention can be obtained by cloning the DNA from the water-reactive activated sludge of the steelworks.
For example, the present invention is obtained by transforming a host microorganism using a microorganism such as Escherichia coli as a host, a plasmid incorporating a DNA having a nucleotide sequence shown in any of SEQ ID NOs: 1 to 43, a phage incorporating a fosmid, and the like. The catechol-degrading enzyme of the present invention can be obtained by producing the catechol-degrading enzyme of a host microorganism.
The determination of whether or not it can be a host microorganism producing the catechol-degrading enzyme of the present invention and the confirmation of whether or not the transformed microorganism can produce the catechol-degrading enzyme can be performed as follows.
First, a catechol as a substrate is added to a cell extract obtained by disrupting cells of a host microorganism that has undergone transformation and culture. In the cell extract of a host microorganism that produces the catechol-degrading enzyme of the present invention, 2-hydroxymuconate semialdehyde is formed as a reaction product by cleavage of the aromatic ring of catechol, resulting in a yellow color. Therefore, by measuring the absorbance at 375 nm, it is possible to specify whether the host microorganism produces the catechol-degrading enzyme of the present invention. If the absorbance at 375 nm increases, catechol is decomposed to form 2-hydroxymuconate semialdehyde, and therefore, it can be determined that catechol decomposition has occurred by the catechol-degrading enzyme.
Next, a method for decomposing catechol contained in wastewater using the catechol-degrading enzyme specified by the base sequence shown in any of SEQ ID NOs: 1 to 43 obtained as described above will be described. The optimum temperature of the catechol-degrading enzyme of the present invention is 20 ° C. or more and 40 ° C. or less, and the optimum pH is 5 or more and 9 or less. When decomposing catechol in wastewater, it is desirable to use the catechol-degrading enzyme of the present invention under conditions that satisfy the conditions of temperature and pH.
In addition, there is no particular upper limit on the catechol concentration when attempting to degrade catechol in wastewater using the catechol-degrading enzyme of the present invention. Steelworks aqueducts contain high concentrations of phenol, but catechol may accumulate as an intermediate reactive organism when phenol is degraded. In the case of aqueous water, phenols that generate catechol may be contained up to about 1000 mg / L. However, even if all of these phenols are converted to a catechol concentration of 1000 mg / L, Treatment with catechol-degrading enzymes is applicable.
When decomposing catechol in wastewater, it is of course possible to use the host microorganism producing the catechol-degrading enzyme of the present invention as it is for degrading catechol. For example, when the catechol-degrading enzyme produced by the host microorganism is present in the cell membrane or periplasm of the cell surface or secreted outside the cell, the catechol-degrading enzyme can come into contact with water outside the cell. Can be used as is. In addition, when the catechol-degrading enzyme produced by the host microorganism is accumulated in the cells of the host microorganism, the catechol-degrading enzyme cannot contact the water outside the cell. A cell extract containing the catechol-degrading enzyme of the present invention can also be used.
When the catechol-degrading enzyme of the present invention is used to decompose catechol in wastewater, a catechol-degrading enzyme encoded by DNA having the base sequence shown in at least any one of SEQ ID NOs: 1 to 43 may be used. Although it is good, it does not matter even if it decomposes | disassembles using a some enzyme together. When only a single type of catechol-degrading enzyme is used, there is a possibility that the catechol-degrading activity is affected by the pH variation of the wastewater, the temperature change, the concentration of the catechol as a substrate, and the like. Therefore, it is considered that the use of a plurality of types of catechol-degrading enzymes having different optimal reaction conditions is considered to be less susceptible to such effects, and is more preferable. It is also possible to cause the microorganism to produce two or more catechol-degrading enzymes. In this case, it becomes possible by transforming the host microorganism using a plasmid, fosmid or the like to which two or more kinds of DNAs having the nucleotide sequences of SEQ ID NOs: 1 to 43 are bound. As the host microorganism, E. coli that can use a vector such as a plasmid is most easily used. Other genetically recombinable microorganisms such as yeast can also be used as host microorganisms. In addition, since the catechol-degrading enzyme of the present invention is derived from an aerobic microorganism that grows in an environment in which oxygen is present, it is desirable to aerate with air when degrading catechol using a host microorganism. .
The present invention will be described below with reference to examples.
Example 1 Degradation of catechol by a novel catechol-degrading enzyme (1)
DNA fragments encoding the nucleotide sequences of SEQ ID NOs: 1 to 43 (Sequence Listing and FIGS. 1 to 43) were purified and blunt-ended. It was then ligated to pCC1FOS fosmid with the chloramphenicol resistance gene. T4 DNA ligase (Takara Bio) was used for ligation to fosmid. The one ligated to this fosmid was subjected to in vitro packaging using MaxPlanx Lambda Packing Extracts. E. coli as a host microorganism. EPI-300T1R was used. Then, Escherichia coli transformed with lambda phage was selected by chloramphenicol resistance on an LB (LB / Cm) agar plate containing 12.5 μg / mL chloramphenicol.
E. coli colonies were picked and cultured at 37 ° C. for 18 hours in 1 ml of 2 × YT medium (2 × YT / Cm) containing 12.5 μg / mL chloramphenicol. Next, 200 microliters of the E. coli culture solution from which the colonies were collected was transferred to a new 800 microliter 2 × YT / Cm medium, mixed, and cultured at 37 ° C. for 30 minutes while shaking at 250 rpm. Microliter of Copy Control Induction Solution (Epicentre) was added. This increased the fosmid copy number in E. coli. Further, E. coli was cultured at 37 ° C. for 2 hours with vigorous shaking at 1200 rpm, and centrifuged at 3000 rpm for 15 minutes at 4 ° C. to precipitate and collect E. coli. The recovered E. coli was resuspended in 50 mM phosphate buffer (pH 7.5). Then 150 microliters of BugBuster Plus Benzoate Nuclease (Novagen) was added to lyse the E. coli cells. By centrifuging at 3000 rpm for 15 minutes at 4 ° C., the remaining lysed bacterial cell portion was precipitated, and the supernatant was obtained as a cell extract.
Next, it was confirmed as follows that this cell extract contains a catechol-degrading enzyme. Catechol (manufactured by SIGMA) serving as a substrate was added to and mixed with 100 microliters of cell extract at a rate of 5 microliters so that the final concentration was 0.5 mM. The reaction was conducted while gently shaking at 250 rpm at 25 ° C. The reaction time was 1 hour and 16 hours. Since the decomposition of catechol becomes yellow due to the formation of 2-hydroxymuconate semialdehyde, the absorbance at 375 nm was examined to examine the activity of the catechol-degrading enzyme.
The results are shown in Table 1. The catechol-degrading enzymes of each SEQ ID NO are also shown in Table 1 for subfamilies grouped according to their amino acid sequence homology. The subcategories of catechol-degrading enzymes were grouped based on the reports of
From the above results, it was confirmed that all the enzymes encoded by the DNA having the nucleotide sequences of SEQ ID NOS: 1 to 43 were catechol-degrading enzymes having catechol-degrading activity.
Example 2 Degradation of catechol by a novel catechol-degrading enzyme (2)
As catechol-degrading enzymes having different subfamily, SEQ ID NO: 19 (I. 1.C subfamily), SEQ ID NO: 12, 13, 14 (I.2.A subfamily), SEQ ID NO: 2, 43 (I.2.B) Subfamily), SEQ ID NO: 31, 38 (I.2.C subfamily), SEQ ID NO: 1, 3, 8, 32 (I.2.G subfamily), SEQ ID NO: 34 (I.3.M subfamily) The fosmid DNA containing the gene encoding each nucleotide sequence of SEQ ID NO: 6 (I. 3.N subfamily) was purified and blunt-ended. It was then ligated into pUC118 plasmid and E. coli E. coli. E. coli JM109 was transformed and E. coli was selected on LB (LB / Ap) agar plates containing 50 μg / mL ampicillin. Subsequently, colonies of E. coli were taken and cultured at 37 ° C. for 18 hours in 1 ml of LB medium (2 × LB / Ap / IPTG) containing 50 μg / mL ampicillin and 0.1 mM isopropyl β-D-thiogalactosidase (IPTG). did. Next, E. coli was precipitated and recovered by centrifugation at 3000 rpm for 15 minutes at 4 ° C. The recovered E. coli was resuspended in 50 mM phosphate buffer (pH 7.5). 150 microliters of BugBuster Plus Benzoate Nuclease (Novagen) was added to lyse the E.
The substrate catechol was added to and mixed with 100 microliters of the cell extract in 5 microliter increments so that the final concentration was 0.5 mM. The reaction was conducted while gently shaking at 250 rpm at 25 ° C. The reaction time was 1 hour.
Since the decomposition of catechol causes 2-hydroxymuconate semialdehyde to form a yellow color, the absorbance at 375 nm (absorption coefficient ε = 33000 M −1 cm −1 ) changes, so the absorbance was measured. The activity of catechol-degrading enzyme was calculated from the change in absorbance and the absorption coefficient. One unit of catechol-degrading activity means the activity of a catechol-degrading enzyme that produces 2-hydroxymuconate semialdehyde, which is a product of catechol-decomposing reaction of 1 micromole per minute. Catechol degradation activity was expressed in the above reaction units per gram of protein. The results are shown in Table 2.
As a control experiment, a known 3-methylcatechol dioxygenase (TodE) (I.3.B subfamily) gene, which is a catechol-degrading enzyme belonging to a different subfamily than the above-mentioned catechol-degrading enzymes, was similarly used. The catechol-degrading activity was also measured in the cell extract when produced in E. coli by the above method.
As described above, SEQ ID NO: 19 (I.1.1.C subfamily of the present invention belonging to a subfamily different from 3-methylcatechol dioxygenase (TodE) (I.3.B subfamily) which is a known catechol-degrading enzyme ), SEQ ID NO: 12, 13, 14 (I.2.A subfamily), SEQ ID NO: 2, 43 (I.2.B subfamily), SEQ ID NO: 31, 38 (I.2.C subfamily), sequence No. 1, 3, 8, 32 (I.2.G subfamily), SEQ ID NO: 34 (I.3.M subfamily), SEQ ID NO: 6 (I.3.N subfamily) Each encoded catechol-degrading enzyme was confirmed to have catechol-degrading enzyme activity.
Example 3 Degradation of Catechol in Aqueous Sequencing Encoding Base Sequences of SEQ ID NOs: 12 (I.2.A Subfamily), 18 (I.2.G Subfamily), 6 (I.3.N Subfamily) The fosmid DNA containing the gene was purified and blunt-ended.
Escherichia coli producing catechol-degrading enzyme as described above is placed in artificial water-free solution (composition is shown in Table 3) containing 100 mg / L of catechol so that the cell concentration is 5 × 10 5 cells / mL. Mix and treat for 24 hours at 25 ° C. with air aeration. As a control experiment, a test in which E. coli that does not transform and does not produce a catechol-degrading enzyme is treated under the same conditions at the same cell concentration (control 1), and a catechol-degrading enzyme having a subfamily different from that of the present invention. An E. coli producing 3-methylcatechol dioxygenase (TodE) (I.3.B subfamily) was also subjected to a test (control 2) that was treated under the same conditions at the same cell concentration.
As in Example 1, the degradation of catechol was measured by increasing the absorbance at 375 nm for 2-hydroxymuconate semialdehyde formed by cleavage of the aromatic ring of catechol. The results are shown in Table 4. In the table, those having an OD of 0.5 or more are indicated by ++, those having an OD greater than 0 and 0.5 or less are indicated by +, and those having an OD of 0 are indicated by-.
From Table 4, in the system containing Escherichia coli that produces each catechol-degrading enzyme of SEQ ID NOs: 12, 18, and 6, catechol was satisfactorily degraded in the aquatic water. In the system containing E. coli that does not produce the catechol-degrading enzyme of Control 1, catechol degradation did not proceed. Further, in the system containing E. coli that produces TodE, a known catechol-degrading enzyme of Control 2, compared to the system containing E. coli that produces the catechol-degrading enzyme of SEQ ID NOs: 12, 18, and 6 of the present invention, The catechol-degrading activity decreased in the aquatic water.
As described above, it was confirmed that the catechol-degrading enzyme of the present invention can promote the catechol-decomposing reaction even in the catechol in the wastewater having a special composition such as an aqueous solution.

(2) The method for decomposing catechol in wastewater, which is characterized.
(4) The method for decomposing catechol in wastewater according to (2) or (3), wherein the wastewater containing the catechol is low-grade water discharged from a coke oven.
First, a method for obtaining the catechol-degrading enzyme of the present invention will be described. The DNA of the nucleotide sequence shown in SEQ ID NO: 12 that encodes the catechol-degrading enzyme of the present invention can be obtained by cloning the DNA from the water-resistant activated sludge of the steelworks.
For example, by using a microorganism such as E. coli as a host and transforming the host microorganism using a plasmid incorporating a DNA having the nucleotide sequence shown in SEQ ID NO: 12 , a phage incorporating a fosmid, etc., the catechol-degrading enzyme of the present invention can be obtained. The catechol-degrading enzyme of the present invention can be obtained by producing it in a host microorganism.
Next, a method for decomposing catechol contained in wastewater using the catechol-degrading enzyme specified by the base sequence shown in SEQ ID NO: 12 obtained as described above will be described.
Further, when the catechol-degrading enzyme of the present invention is used to decompose catechol in wastewater, a catechol-degrading enzyme encoded by the DNA having the base sequence shown in SEQ ID NO: 12 may be used. Of course, it does not matter if it is used for the decomposition treatment. It is also possible to cause the microorganism to produce two or more catechol-degrading enzymes .
