Abstract
Description
The present invention relates to a method for producing 5-fluoro-1H-pyrazole-4-carboxylic acids.
5-Fluoro-1H-pyrazole-4-carboxylic acids are important compounds as medical pesticides and synthetic intermediates thereof. For example, 5-fluoro-N- (2-isopropylphenyl) -1,3-dimethyl-1H-pyrazole-4-carboxamide or N- (2-biphenylyl) -5-fluoro-1,3-dimethyl-1H-pyrazole -4-carboxamide is useful as a fungicide (Patent Documents 1 and 2).
In the conventional production of 5-fluoro-1H-pyrazole-4-carboxylic acids, 5-chloro-1H-pyrazoles are used as a raw material. For example, as a method for producing 5-fluoro-1H-pyrazole-4-carboxamides, a method is disclosed in which chlorine at the 5-position of 5-chloro-1H-pyrazole-4-carboxamides is fluorinated with potassium fluoride. (Patent Document 3). Further, the 5-position chlorine of 5-chloro-1H-pyrazole-4-carbaldehydes and 5-chloro-1H-pyrazole-4-carboxylic acid chlorides is fluorinated with potassium fluoride, and then the 5-formyl group or A method for converting a chloroformyl group into an N-arylcarbamoyl group is also disclosed (Patent Documents 4 and 5).
On the other hand, there has been no report on a method for producing 5-fluoro-1H-pyrazole-4-carboxylic acids by reacting α-trifluoromethyl-β-ketocarboxylic acids with hydrazines.
The production methods of 5-fluoro-1H-pyrazole-4-carboxylic acids described in Patent Documents 3 to 5 require heating at 150 ° C. or higher when fluorinating 5-position chlorine with potassium fluoride. . Furthermore, depending on the substrate, this fluorination step requires about 10 hours to complete the reaction, and the yield is only 20-30%.
An object of the present invention is to provide a method for producing 5-fluoro-1H-pyrazole-4-carboxylic acids in a high yield under milder conditions than conventional methods.
(1) As a result of intensive studies to solve the above problems, α-trifluoromethyl-β-ketocarboxylic acid and hydrazine may be reacted in the presence of a base in some cases to give 5-fluoro-1H-pyrazole- The inventors have found that 4-carboxylic acids can be produced, and have completed the present invention. That is, the present invention relates to the general formula (1)
By using the production method of the present invention, 5-fluoropyrazole-4-carboxylic acids that are useful as medical pesticides and synthetic intermediates thereof can be produced simply and efficiently.
